1Inhibition of KIT RNAi mediated with adenovirus in gastrointestinal stromal tumor xenograft显示文摘AIM: To investigate a therapeutic method for gastrointestinal stromal tumor (GIST) based on KIT RNA interference (RNAi) with AdMax adenovirus. METHODS: KIT short hairpin RNA (shRNA), whose lateral sides were decorated with restriction endonuclease sequences, was designed. T 4 DNA ligase catalyzed the joint of the KIT shRNA and the green fluorescent protein-containing PDC316-EGFP-U6 to form PDC316EGFP-U6-KIT. Homologous recombination of AdEGFPU6-KIT was performed with the AdMax system. Heterotopically transplanted GISTs were established in nude mice. AdEGFP-U6-KIT was intratumorally injected. The volume, inhibition ratio of tumor and CD117 expression of GIST graft tumor in nude mice were compared between test and control groups. RESULTS: The length of KIT shRNA was determined to be about 50bp by agarose electrophoresis. Gene se-quencing detected the designed KIT RNAi sequence in PDC316-EGFP-U6-KIT. After transfection with AdEGFPU6-KIT, 293 cells displayed green fluorescence. The physical and infective titers of AdEGFP-U6-KIT were 5 × 10 11 viral particles/mL and 5.67 × 10 7 plaque forming units/mL, respectively. The mean volume of the grafted tumor was significantly smaller in test mice than in control mice (75.3 ± 22.9 mm 3 vs 988.6 ± 30.5 mm 3 , t = -18.132, P < 0.05). The inhibition ratio of the tumors was 59.6% in the test group. CD117 positive expression was evident in two cases (20%) in the test group and 10 cases (100%) in the control group (χ 2 = 10.2083, P < 0.005). 2Assessing anxiety,depression,and stress among inpatients with cancer显示文摘Objective This study aimed to assess anxiety,depression,and stress among inpatients with cancer.Methods Two hundred thirty-five hospitalized patients with cancer were surveyed with the Depression Anxiety Stress Scales(simplified Chinese Version).The software program SPSS 25.0 was used for statistical analysis of the survey data.Results The average scores of depression,anxiety,and stress of inpatients with cancer were 12.17,11.84,and 13.98 respectively,which were higher than the normal range.The scores of anxiety and stress of inpatients with different caregivers were statistically different(P=0.024/0.036).The anxiety and stress scores of inpatients with spouses as caregivers were higher than those of inpatients with children as caregivers.There was a statistically significant difference in the incidence of stress between inpatients with cancer with religious beliefs and inpatients with cancer without religious beliefs(P=0.026),and those with religious beliefs had greater incidence of stress.The score of anxiety was significantly higher for inpatients with children than for inpatients without children(P=0.040).Conclusion The anxiety,depression,and stress levels of inpatients with cancer are relatively high.It is necessary to pay special attention to the psychological status of these patients during clinical diagnosis and treatment to improve their quality of life.Honghong Xu Ruotong Xue Yi Cheng Yujie Zhang Jie Rao Mei Liu Shiying Yu Lingxiang Liu Yiqian Liu Yongqian Shu Liuqing Yang Hanping Shi 2020Oncology and Translational Medicine2020,6,3:2

9Secure Application-Aware Service Differentiation in Public Area Wireless Networks显示文摘We are witnessing the increasing demand for pervasive Internet access from public area wireless networks (PAWNs). As their popularity grows, the inherent untrusted nature of public places and the diverse service requirements of end users are two key issues that need to be addressed. We have proposed two approaches to address these issues. First,the Home-based Authentication Protocol (HAP) that provides a framework by which to establish trust between a nomadic client and a service provider using a trusted third party (home). Second, we argue that the best-effort-based service model provided by many access points is not enough to satisfy the end user fairness and to maximize the wireless link utilization for a diverse user population. We have proposed an application-aware service differentiation (AASD) mechanism that takes both application semantics and user requirements into consideration. Our analysis of this framework shows several fruitful results. The total authentication latency increases with the number of clients but at a rate that is much less than linear increasing latency. 10Nutrition in Alzheimer's disease:a review of an underappreciated pathophysiological mechanism显示文摘Alzheimer's disease(AD)is the leading cause of dementia in older individuals and is an escalating challenge to global public health.Pharmacy therapy of AD is one of the well-funded areas;however,little progress has been made due to the complex pathogenesis.Recent evidence has demonstrated that modifying risk factors and lifestyle may prevent or delay the incidence of AD by 40%,which suggests that the management should pivot from single pharmacotherapy toward a multipronged approach because AD is a complex and multifaceted disease.Recently,the gut-microbiota-brain axis has gained tremendous traction in the pathogenesis of AD through bidirectional communication with multiple neural,immune,and metabolic pathways,providing new insights into novel therapeutic strategies.Dietary nutrition is an important and profound environmental factor that influences the composition and function of the microbiota.The Nutrition for Dementia Prevention Working Group recently found that dietary nutrition can affect cognition in AD-related dementia directly or indirectly through complex interactions of behavioral,genetic,systemic,and brain factors.Thus,considering the multiple etiologies of AD,nutrition represents a multidimensional factor that has a profound effect on AD onset and development.However,mechanistically,the effect of nutrition on AD is uncertain;therefore,optimal strategies or the timing of nutritional intervention to prevent or treat AD has not been established.Thus,this review summarizes the current state of knowledge concerning nutritional disorders,AD patient and caregiver burden,and the roles of nutrition in the pathophysiology of AD.We aim to emphasize knowledge gaps to provide direction for future research and to establish optimal nutrition-based intervention strategies for AD.Jiwei Jiang Hanping Shi Shirui Jiang Anxin Wang Xinying Zou Yanli Wang Wenyi Li Yuan Zhang Mengfan Sun Qiwei Ren Jun Xu 2023Science China(Life Sciences)2023,66,10:0
11AIM-CICs: an automatic identification method for cell-in-cell structures based on convolutional neural network显示文摘Whereas biochemical markers are available for most types of cell death, current studies on non-autonomous cell death by entosis rely strictly on the identification of cell-in-cell structures (CICs), a unique morphological readout that can only be quantified manually at present. Moreover, the manual CIC quantification is generally over-simplified as CIC counts, which represents a major hurdle against profound mechanistic investigations. In this study, we take advantage of artificial intelligence technology to develop an automatic identification method for CICs (AIM-CICs), which performs comprehensive CIC analysis in an automated and efficient way. The AIM-CICs, developed on the algorithm of convolutional neural network, can not only differentiate between CICs and non-CICs (the area under the receiver operating characteristic curve (AUC) > 0.99), but also accurately categorize CICs into five subclasses based on CIC stages and cell number involved (AUC > 0.97 for all subclasses). 14Highly sensitive detection of daminozide using terahertz metamaterial sensors显示文摘In order to solve the problems of low sensitivity and complex sample pretreatment in traditional detection,a method was proposed in this study to detect daminozide using terahertz combined with a metamaterial sensor,which enables real-time and label-free molecular detection with high sensitivity.The correlation between the transmission frequency shift and absorbance of daminozide solution at different concentrations was analyzed.The simulation and experimental results showed that this metamaterial sensor could achieve highly sensitive sensing of daminozide solutions at 0.05 mg/L.The maximum quality factor(Q_(max))of the two peaks could reach 5.78 and 13.05,and the maximum figure of merit(FOM)of the two peaks can reach 0.82 and 1.72.The maximum sensitivity of two resonance peaks reached 38.148 GHz/(mg·L)and 133.516 GHz/(mg·L)when the concentration of daminozide in the solution was 2000 mg/L.There was an obvious positive correlation between the transmission and transmission,frequency shift and frequency shift,and absorbance and absorbance of the resonance peak of daminozide solutions.Therefore,this platform not only opens up new possibilities for the microanalysis of the chemical composition of substances in solutions but also provides a valuable reference for the design of other metamaterial-based sensors in the field of food safety.Hanping Mao Xiaoxue Du Yuting Yan Xiaodong Zhang Guoxin Ma Yafei Wang Yong Liu Bin Wang Xiaoyue Yang Qiang Shi 2022International Journal of Agricultural and Biological Engineering2022,15,6:0
