| 3 | Permian Alkaline Granites in Central Inner Mongolia and Their Geodynamic Significance显示文摘Alkaline granites (Rb-Sr ages 276-286 Ma)occurring in the Bayan Ul-East Ujimqin belt at the southern margin of the Siberian plate originated in a tensional tectonic environment about 60 Ma earlier than the Late Devonian to Early Carboniferous collision between the Siberian and Sino-Korean plates. They belong to post-orogenic A-type granites and may be used as an indicator of the end of the orogeny. At the northern margin of the Sino-Korean plate, however, only late-orogenic calc-alkaline granites occurred during the late Caboniferous-Permian, and alkaline syenites did not appear until the Late Triassic. | 8 | Matrine attenuates oxidative stress and cardiomyocyte apoptosis in doxorubicin-induced cardiotoxicity via maintaining AMPKα/UCP2 pathway显示文摘Oxidative stress and cardiomyocyte apoptosis are involved in the pathogenesis of doxorubicin(DOX)-induced cardiotoxicity. Matrine is well-known for its powerful anti-oxidant and anti-apoptotic capacities. Our present study aimed to investigate the effect of matrine on DOX-induced cardiotoxicity and try to unearth the underlying mechanisms. Mice were exposed with DOX to generate DOX-induced cardiotoxicity or normal saline as control. H9 C2 cells were used to verify the effect of matrine in vitro.DOX injection triggered increased generation of reactive oxygen species(ROS) and excessive cardiomyocyte apoptosis, which were significantly mitigated by matrine. Mechanistically, we found that matrine ameliorated DOX-induced uncoupling protein 2(UCP2) downregulation, and UCP2 inhibition bygenipin could blunt the protective effect of matrine on DOX-induced oxidative stress and cardiomyocyte apoptosis. Besides, 50-AMP-activated protein kinase α2(Ampkα2) deficiency inhibited matrine-mediated UCP2 preservation and abolished the beneficial effect of matrine in mice. Besides, we observed that matrine incubation alleviated DOX-induced H9 C2 cells apoptosis and oxidative stress level via activating AMPKα/UCP2, which were blunted by either AMPKα or UCP2 inhibition with genetic or pharmacological methods. | 13 | Efficiency and Inheritance of Targeted Mutagenesis in Maize Using CRISPR-Cas9显示文摘CRISPR/Cas(clustered regularly interspaced short palindromic repeats/CRISPR-associated proteins) is an adaptive immune system in bacteria and archaea to defend against invasion from foreign DNA fragments. Recently, it has been developed as a powerful targeted genome editing tool for a wide variety of species. However, its application in maize has only been tested with transiently expressed somatic cells or with a limited number of stable transgenic T0 plants. The exact efficiency and specificity of the CRISPR/Cas system in the highly complex maize genome has not been documented yet. Here we report an extensive study of the well-studied type II CRISPR-Cas9 system for targeted genome editing in maize, with the codon-optimized Cas9 protein and the short non-coding guide RNA generated through a functional maize U6 sn RNA promoter. Targeted gene mutagenesis was detected for 90 loci by maize protoplast assay, with an average cleavage efficiency of 10.67%. Stable knockout transformants for maize phytoene synthase gene(PSY1) were obtained. Mutations occurred in germ cells can be stably inherited to the next generation. Moreover, no off-target effect was detected at the computationally predicted putative off-target loci. No significant difference between the transcriptomes of the Cas9 expressed and non-expressed lines was detected. We present a new perturbation algorithm for enhancing security of multivariate public key cryptosystems (MPKCs). European Consortium SFLASH which derives from Matsumoto-Imai scheme is a fast signature scheme intended for low cost smart cards. It was broken with the differential cryptanalysis by Dubois et al. in 2007. | 15 | Radio Propagation and Wireless Coverage of LSAA-Based 5G Millimeter-Wave Mobile Communication Systems显示文摘Millimeter-wave(mm Wave) communications will be used in fifth-generation(5G) mobile communication systems, but they experience severe path loss and have high sensitivity to physical objects, leading to smaller cell radii and complicated network architectures. A coverage extension scheme using large-scale antenna arrays(LSAAs) has been suggested and theoretically proven to be cost-efficient in combination with ultradense small cell networks. To analyze and optimize the LSAA-based network deployments, a comprehensive survey of recent advances in statistical mmWave channel modeling is first presented in terms of channel parameter estimation, large-scale path loss models, and small-scale cluster models. Next, the measurement and modeling results at two 5G candidate mmWave bands(e.g., 28 GHz and 39 GHz) are reviewed and compared in several outdoor scenarios of interest, where the propagation characteristics make crucial contributions to wireless network designs. | 16 | Immunotherapeutical Potential of Mycobacterium Vaccae on M. Tuberculosis Infection in Mice显示文摘Tuberculosis remains the worldwide infectious disease. To identify the therapeutic potential of M. vaccae in treating tuberculosis,M. vaccae was injected into Mycobacterium tuberculosis(M. tuberculosis) infected mice. The optimal dose of M. vaccae(22.5 μg/mouse) treated mice showed lower pathological change index,spleen weight index,lung weight index and vital M. tuberculosis count than those of the untreated group. Treatment with M. vaccae enhanced the percentages of CD3+ and CD4+ T cells,IFN-γ+CD4+ T cells,innate immune cells including NK cells,NK1.1+ T cells and γδ T cells,and reduced the percentage of IL-4+CD4+ T cells. | 17 | The Involvement of Heat Shock Proteins in Murine Liver Regeneration显示文摘Partial hepatectomy (PHx) in mammals is a very common experimental model to investigate the process of liver regeneration. The surgery itself could give birth to a series of stresses, such as the temporary raise of body temperature and the ischaemia-reperfusion injury. Heat shock proteins (HSPs) were a family of stress-inducible proteins involved in maintaining cell homeostasis and regulating the immune system. In our study, we intended to investigate the expression and role of HSPs in liver regeneration. Using RT-PCR and Western blotting, we determined the expression in regenerating liver of HSP27, HSP60, HSP70 and HSP90 in mRNA level and protein level, respectively, with mice treated with sham operation as controls. We also used quercertin as an inhibitior of HSPs to explore their effects on liver regeneration. We found that hepatic expression of HSPs increased at the early phase of liver regeneration and declined to the constitutively low level later. Moreover, quercetin pretreatment delayed the progress of liver regeneration in mice via inhibition of HSPs. | 18 | Erucic acid from Isatis indigotica Fort. suppresses influenza A virus replication and inflammation in vitro and in vivo through modulation of NF-kB and p38 MAPK pathway显示文摘Isatis indigotica Fort.(Ban-Lan-Gen)is an herbal medicine prescribed for influenza treatment.However,its active components and mode of action remain mostly unknown.In the present study,erucic acid was isolated from Isatis indigotica Fort.,and subsequently its underlying mechanism against influenza A virus(IAV)infection was investigated in vitro and in vivo.Our results demonstrated that erucic acid exhibited broad-spectrum antiviral activity against IAV resulting from reduction of viral polymerase transcription activity.Erucic acid was found to exert inhibitory effects on IAV or viral(v)RNA-induced pro-inflam-matory mediators as well as interferons(IFNs).The molecular mechanism by which erucic acid with antiviral and anti-inflammatory properties was attributed to inactivation of NF-kB and p38 MAPK signaling.Furthermore,the NF-kB and p38 MAPK inhibitory effect of erucic acid led to diminishing the transcriptional activity of interferon-stimulated gene factor 3(ISGF-3),and thereby reducing IAV-triggered pro-inflammatory response amplification in IFN-β-sensitized cells.Additionally,IAV-or vRNA-triggered apoptosis of alveolar epithelial A549 cells was prevented by erucic acid.In vivo,erucic acid administration consistently displayed decreased lung viral load and viral antigens expression.Meanwhile,erucic acid markedly reduced CD8+cytotoxic T lymphocyte(CTL)recruitment,pro-apoptotic signaling,hyperactivity of multiple signaling pathways,and exacerbated immune inflammation in the lung,which resulted in decreased lung injury and mortality in mice with a mouse-adapted A/FM/1/47-MA(H1N1)strain infection.Our findings provided a mechanistic basis for the action of erucic acid against IAV-mediated inflammation and injury,suggesting that erucic acid may have a therapeutic potential in the treatment of influenza. | Xiaoli Liang Yuan Huang Xiping Pan Yanbing Hao Xiaowei Chen Haiming Jiang Jing Li Beixian Zhou Zifeng Yang | 2020 | Journal of Pharmaceutical Analysis2020,10,2: | 10 |
