It is determined that the products of discontinuous precipitation transformation below 277℃ are not theequilibrium phase constituents, but the metastable phases made up of α phase, Zn phase and CuZn4 phase. | 5 | Quercetin promotes human epidermal stem cell proliferation through the estrogen receptor/β-catenin/c-Myc/cyclin A2 signaling pathway显示文摘Skin epidermal stem cells(EpSCs)play an important role in wound healing.Quercetin is a phytoestrogen reported to accelerate skin wound healing,but its effect on EpSCs is unknown.In this study,we investigated the effect of quercetin on human EpSC proliferation and explored the underlying mechanisms.We found that quercetin at 0.1~1μM significantly promoted EpSC proliferation and increased the number of cells in S phase.The pro-proliferative effect of quercetin on EpSCs was confirmed in cultured human skin tissue.Mechanistic studies showed that quercetin significantly upregulated the expressions ofβ-catenin,c-Myc,and cyclins A2 and E1.Inhibitor forβ-catenin or c-Myc significantly inhibited quercetin-induced EpSC proliferation.Theβ-catenin inhibitor XAV-939 suppressed quercetin-induced expressions ofβ-catenin,c-Myc,and cyclins A2 and E1.The c-Myc inhibitor 10058-F4 inhibited the upregulation of c-Myc and cyclin A2 by quercetin.Pretreatment of EpSCs with estrogen receptor(ER)antagonist ICI182780,but not the G protein-coupled ER1 antagonist G15,reversed quercetin-induced cell proliferation and upregulation ofβ-catenin,c-Myc,and cyclin A2.Collectively,these results indicate that quercetin promotes EpSC proliferation through ER-mediated activation ofβ-catenin/c-Myc/cyclinA2 signaling pathway and ER-independent upregulation of cyclin E1 and that quercetin may accelerate skin wound healing through promoting EpSC proliferation.As EpSCs are used not only in clinic to treat skin wounds but also as seed cells in skin tissue engineering,quercetin is a useful reagent to expand EpSCs for basic research,skin wound treatment,and skin tissue engineering. | Zhaodong Wang Guangliang Zhang Yingying Le Jihui Ju Ping Zhang Dapeng Wan Qiang Zhao Guangzhe Jin Hao Su Jinwei Liu Jiaxuan Feng Yi Fu Ruixing Hou | 2020 | Acta Biochimica et Biophysica Sinica2020,52,10: | 3 |
| 6 | Highly sensitive piezoresistive pressure sensors based on laser-induced graphene with molybdenum disulfide nanoparticles显示文摘Wearable pressure sensors have drawn significant attention because of their extensive applications in motion detection, tactile sensing, and health monitoring. However, the complex manufacturing process and high cost of active materials make low-cost,large-scale production elusive. In this work, we report a flexible piezoresistive pressure sensor assembled with two 3D laserinduced graphene(LIG) foam electrodes on a polyimide thin film from a simple laser scribing process in the ambient environment. The design of the air gap between the two foam electrodes allows the sensor to showcase a low limit of detection of 0.274 Pa, which provides favorable sensing performance in motion detection and wrist pulse monitoring. The addition of spherical MoS2 nanoparticles between the two foam electrodes further enhances the sensitivity to 88 k Pa-1 and increases the sensing range to significantly outperform the previous literature reports. | 7 | HIV-1 Vif suppresses antiviral immunity by targeting STING显示文摘HIV-1 infection-induced cGAS–STING–TBK1–IRF3 signaling activates innate immunity to produce type I interferon(IFN).The HIV-1 nonstructural protein viral infectivity factor(Vif)is essential in HIV-1 replication,as it degrades the host restriction factor APOBEC3G.However,whether and how it regulates the host immune response remains to be determined.In this study,we found that Vif inhibited the production of type I IFN to promote immune evasion.HIV-1 infection induced the activation of the host tyrosine kinase FRK,which subsequently phosphorylated the immunoreceptor tyrosine-based inhibitory motif(ITIM)of Vif and enhanced the interaction between Vif and the cellular tyrosine phosphatase SHP-1 to inhibit type I IFN.Mechanistically,the association of Vif with SHP-1 facilitated SHP-1 recruitment to STING and inhibited the K63-linked ubiquitination of STING at Lys337 by dephosphorylating STING at Tyr162.However,the FRK inhibitor D-65495 counteracted the phosphorylation of Vif to block the immune evasion of HIV-1 and antagonize infection.These findings reveal a previously unknown mechanism through which HIV-1 evades antiviral immunity via the ITIM-containing protein to inhibit the posttranslational modification of STING.These results provide a molecular basis for the development of new therapeutic strategies to treat HIV-1 infection. | Yu Wang Gui Qian Lingyan Zhu Zhuo Zhao Yinan Liu Wendong Han Xiaokai Zhang Yihua Zhang Tingrong Xiong Hao Zeng Xianghui Yu Xiaofang Yu Xiaoyan Zhang Jianqing Xu Quanming Zou Dapeng Yan | 2022 | Cellular & Molecular Immunology2022,19,1: | 2 |
| 8 | Effect of Cu on Discontinuous Precipitation of a Symmetrical AlZn Alloy显示文摘The effect of Cu additions on discontinuous precipitation of AlZn alloy were studied with optical microscopy, scanning electron microscopy and X-ray diffraction. It is found that the effect of addition of 2 at. pct Cu on cellular nucleation site is not remarkable, while the effect of aging temperature on cellular nucleation site of AlZn and AlZn-2Cu alloys is fairly obvious. The cell growth rate of discontinuous precipitation of AlZn alloy is remarkably accelerated with the addition of 2 at. pct Cu. The discontinuous precipitation microstructure of AlZn alloy is not apparently affected with the addition of 2 at. pct Cu and the cellular front is still fine microstructure of spinodal decomposition. The phase constituents consist of a fcc Al-rich phase, a hcp Zn-rich phase and a CuZn4 phase. | 15 | Single-cell transcriptome analysis reveals a cancer-associated fibroblast marker gene signature in hepatocellular carcinoma that predicts prognosis显示文摘Background and aims:Hepatocellular carcinoma(HCC)is one of the leading causes of cancer death.Multi-pathway combination therapy is used to treat HCC,and immunotherapy is also a routine part of treatment.As a major component of the tumor microenvironment(TME),cancer-associated fibroblasts(CAFs)actively participate in cancer progression through complex functions.However,because CAFs dynamically change during cancer development,most of the current treatment strategies targeting CAFs fail.We created a prognostic CAF marker gene signature(CAFMGS)to investigate the utility of CAFs as a prognostic factor and therapeutic target.Methods:Gene Expression Omnibus(GEO)single-cell RNA sequencing(Sc-RNA-seq)data were analyzed to identify CAF marker genes in HCC.The Cancer Genome Atlas(TCGA)database was used as a training cohort to construct the CAFMGS model and the International Cancer Genome Consortium(ICGC)dataset was used to validate the CAFMGS.Results:Marker genes in the CAFMGS model were(0.0001-SPP1),(0.0084-VCX3A),(0.0015-HMGA1),(0.0082-PLOD2),and(0.0075-CACYBP).The CAFMGS_score was separated into high-risk and low-risk groups based on the median of the patients’OS.Univariate and multivariate analyses confirmed that CAFMGS_score was an independent prognostic factor in the training group.CAFMGS_score was a more accurate prognostic indicator compared with clinicopathological score and tumor mutational burden score.Conclusion:CAFMGS offers a fresh perspective on stromal cell marker genes in HCC prognosis and expands our knowledge of CAF heterogeneity and functional diversity,perhaps paving the way for CAF-targeted immunotherapy in HCC patients. | Hao Chi Dapeng Chen Yuliang Zhang Zilin Cui Yi Bai Yamin Zhang | 2023 | iLIVER2023,2,1: | 0 |

| 17 | Effect of riboflavin deficiency on intestinal morphology,jejunum mucosa proteomics,and cecal microbiota of Pekin ducks显示文摘This study was to determine the effects of riboflavin deficiency(RD)on intestinal development,jejunum mucosa proteome,cecal short-chain fatty acids(SCFA)profiling,and cecal microbial diversity and community of starter Pekin ducks.Male white Pekin ducks(1 d old,n=240)were allocated into 2 groups,with 12 replicates and 10 birds per replicate in each group.For 21 d,all ducks had ad libitum access to either an RD or a riboflavin adequate(control,CON)diet,formulated by supplementing a basal diet with 0 or 10 mg riboflavin per kg of diet,respectively.Compared to the CON group,growth retardation,high mortality,and poor riboflavin status were observed in the RD group.Furthermore,RD reduced the villus height and the ratio of villus height to crypt depth of jejunum and ileum(P<0.05),indicating morphological alterations of the small intestine.In addition,dietary RD enhanced relative cecum weight and decreased cecal SCFA concentrations(P<0.05),including propionate,isobutyrate,butyrate,and isovalerate.The jejunum mucosa proteomics showed that 208 proteins were upregulated and 229 proteins were downregulated in the RD group compared to those in the CON group.Among these,RD mainly suppressed intestinal absorption and energy generation processes such as glycolysis and gluconeogenesis,fatty acid beta oxidation,tricarboxylic acid cycle,and oxidative phosphorylation,leading to impaired ATP generation.In addition,RD decreased the community richness and diversity of the bacterial community in the cecum of ducks.Specifically,RD reduced the abundance of butyrate-producing bacteria in the cecum(P<0.05),such as Eubacterium coprostanoligenes,Prevotella and Faecalibacterium.Dietary RD resulted in growth depression and intestinal hypofunction of Pekin ducks,which could be associated with impaired intestinal absorption and energy generation processes in intestinal mucosa,as well as gut microbiota dysbiosis.These findings contribute to our understanding of the mechanisms of intestinal hypofunction due to RD. | Yaxi Xu Bo Zhang Rui Zhao Kexin Gao Suyun Liang Yongbao Wu Yongsheng Hao Dapeng Liu Zhanbao Guo Jian Hu Zhengkui Zhou Ming Xie Jing Tang | 2023 | Animal Nutrition2023,,1: | 0 |
