| 1 | A SURVEY OF STACKELBERG DIFFERENTIAL GAME MODELS IN SUPPLY AND MARKETING CHANNELS显示文摘Stackelberg differential game models have been used to study sequential decision making in noncooperative games in diverse fields.In this paper,we survey recent applications of Stackelberg differential game models to the supply chain management and marketing channels literatures.A common feature of these applications is the specification of the game structure:a decentralized channel composed of a manufacturer and independent retailers,and a sequential decision procedure with demand and supply dynamics and coordination issues.In supply chain management,Stackelberg differential games have been used to investigate inventory issues,wholesale and retail pricing strategies,and outsourcing in dynamic environments.The underlying demand typically has growth dynamics or seasonal variation.In marketing,Stackelberg differential games have been used to model cooperative advertising programs,store brand and national brand advertising strategies,shelf space allocation,and pricing and advertising decisions.The demand dynamics are usually extensions of the classical advertising capital models or sales-advertising response models.We begin by explaining the Stackelberg differential game solution methodology and then provide a description of the models and results reported in the literature. | Xiuli HE Ashutosh PRASAD Suresh P. | 2 | Transcriptional cofactors Ski and SnoN are major regulatorsof the TGF-β/Smad signaling pathway in health and disease显示文摘The transforming growth factor-β(TGF-β)family plays major pleiotropic roles by regulating many physiological processes in development and tissue homeostasis.The TGF-βsignaling pathway outcome relies on the control of the spatial and temporal expression of>500 genes,which depend on the functions of the Smad protein along with those of diverse modulators of this signaling pathway,such as transcriptional factors and cofactors.Ski(Sloan-Kettering Institute)and SnoN(Ski novel)are Smadinteracting proteins that negatively regulate the TGF-βsignaling pathway by disrupting the formation of R-Smad/Smad4 complexes,as well as by inhibiting Smad association with the p300/CBP coactivators.The Ski and SnoN transcriptional cofactors recruit diverse corepressors and histone deacetylases to repress gene transcription.The TGF-β/Smad pathway and coregulators Ski and SnoN clearly regulate each other through several positive and negative feedback mechanisms.Thus,these cross-regulatory processes finely modify the TGF-βsignaling outcome as they control the magnitude and duration of the TGF-βsignals.As a result,any alteration in these regulatory mechanisms may lead to disease development.Therefore,the design of targeted therapies to exert tight control of the levels of negative modulators of the TGF-βpathway,such as Ski and SnoN,is critical to restore cell homeostasis under the specific pathological conditions in which these cofactors are deregulated,such as fibrosis and cancer. | Angeles C.Tecalco-Cruz Diana G.Rios-Lopez Genaro Vazquez-Victorio Reyna E.Rosales-Alvarez Marina Macias-Silva | 2018 | Signal Transduction and Targeted Therapy2018,3,1: | 9 |
| 3 | Cognitive disorder and dementia in type 2 diabetes mellitus显示文摘Insulin,a key pleiotropic hormone,regulates metabolism through several signaling pathways in target tissues including skeletal muscle,liver,and brain.In the brain,insulin modulates learning and memory,and impaired insulin signaling is associated with metabolic dysregulation and neurodegenerative diseases.At the receptor level,in aging and Alzheimer’s disease(AD)models,the amount of insulin receptors and their functions are decreased.Clinical and animal model studies suggest that memory improvements are due to changes in insulin levels.Furthermore,diabetes mellitus(DM)and insulin resistance are associated with age-related cognitive decline,increased levels ofβ-amyloid peptide,phosphorylation of tau protein;oxidative stress,pro-inflammatory cytokine production and dyslipidemia. Recent evidence shows that deleting brain insulin receptors leads to mildobesity and insulin resistance without influencing brain size and apoptosis development.Conversely, deleting insulin-like growth factor 1 receptor (IGF-1R) affects brain size anddevelopment, and contributes to behavior changes. Insulin is synthesized locally in the brain andis released from the neurons. Here, we reviewed proposed pathophysiological hypotheses toexplain increased risk of dementia in the presence of DM. Regardless of the exact sequence ofevents leading to neurodegeneration, there is strong evidence that mitochondrial dysfunctionplays a key role in AD and DM. A triple transgenic mouse model of AD showed mitochondrialdysfunction, oxidative stress, and loss of synaptic integrity. These alterations are comparable tothose induced in wild-type mice treated with sucrose, which is consistent with the proposal thatmitochondrial alterations are associated with DM and contribute to AD development. Alterationsin insulin/IGF-1 signaling in DM could lead to mitochondrial dysfunction and low antioxidantcapacity of the cell.
