1Starch grain analysis reveals function of grinding stone tools at Shangzhai site, Beijing显示文摘Prehistoric groundstone tools including slabs (metate, mo-pan) and mullers (mano, mo-bang) are often considered to be tools that were used to process cereals in ancient China. Most archaeologists believe that groundstone tools are indicators of an agrarian economy. This study employed starch grain analysis to examine a slab and a muller excavated from the Shangzhai Site (7500-7000 cal a BP), Beijing. Identifiable starch grains recovered from the stone tools were classified into 9 genera and 12 species, most of which were from acorns and foxtail millets. Remains from the oak species Q. mongolicus, Q. acuttssima, and Q. dentate were identified as well as a few starch grains from broomcorn (Panicum miliaseum) and bean (Vigna sp.). Other starch grains were from unidentifiable roots and grasses. A variety of starch grains indicates that the grinding stone tools were used to process not only cereals, but also other seeds or roots that had to be ground or husked, thus bringing into question the aforementioned cereal hypothesis. Because the numbers of starch grains from nuts and millets were so large, both gathering and cultivation might have been important economic patterns before 7000 years ago in Beijing area. 5Early experiences on living donor liver transplantation in China: multicenter report显示文摘Background Because of the lack of brain death laws in China, the proportion of cadaveric organ donation is low. Many patients with end-stage liver disease die waiting for a suitable donor. Living donor liver transplantation (LDLT) would reduce the current discrepancy between the number of patients on the transplant waiting list and the number of available organ donors. We describe the early experience of LDLT in the mainland of China based on data from five liver transplant centers. Methods Between January 2001 and October 2003, 45 patients with end-stage liver disease received LDLT at five centers in China. The indication and timing, surgical techniques and complications, nonsurgical issues including rejection, infection, and advantages of LDLT in the series were reviewed. Actuarial patient and graft survival rates were calculated by using the Kaplan-Meier product-limit estimate. Statistical analysis was completed by using SPSS 10.0. Results All LDLT recipients were cirrhotic patients, except for one man with fulminant hepatic failure. Among the 45 cases of LDLT, 35 (77.8%) were performed in one center (the First Affiliated Hospital of Nanjing Medical University). The overall 1 and 3 year survival rate of the recipients was 93.1% and 92.0%, respectively. Of the 45 LDLT donors, there were 3 cases of biliary leakage, 2 subphrenic collections, 1 fat liquefaction around the incision and 1 biliary peritonitis after T tube removal. All donors recovered completely. Conclusions LDLT provides an excellent approach to addressing the problem of donor shortage in China even though the operation is complicated, uncompromising and difficult with respect to the safety of the donors and receptors. Despite early technical hurdles having been overcome, perfection of technique is still necessarily. 7Effects of thiamine on Trichothecium and Alternaria rots of muskmelon fruit and the possible mechanisms involved显示文摘The effects of thiamine against pink and black spot rots caused by Trichothecium roseum and Alternaria alternata and modulation on the metabolism of reactive oxygen species(ROS)and phenylpropanoid pathway were investigated in this paper.In vitro test indicated that thiamine significantly inhibited mycelia growth and spore germination of T.roseum and A.alternata.Thiamine at 100 mmol L^(–1) effectively inhibited lesion development of muskmelon fruit inoculated with T.roseum or A.alternata,enhanced production rate of O_2^(-.)and H_2O_2 content,activities of catalase(CAT),ascorbate peroxidase(APX)and glutathione reductase(GR)in muskmelon fruit.Thiamine also affect phenylpropanoid pathway in muskmelon fruit by increasing the activities of phenylalanine ammonia lyase(PAL)and peroxidase(POD),the content of total phenolic compounds,flavonoids and lignin.These results suggest that the effects of thiamine on pink and black spot rots in muskmelon fruits are associated with its direct fungitoxic against the pathogens and the modulation of O_2^(-.)and H_2O_2 production,eliminating enzymes and phenylpropanoid pathway.GE Yong-hong LI Can-ying Lü Jing-yi ZHU Dan-shi 2017Journal of Integrative Agriculture2017,16,11:4
8A novel real-time RT-PCR with TaqM an-MGB probes and its application in detecting BVDV infections in dairy farms显示文摘A real-time RT-PCR assay using Taq Man-MGB probes was developed to detect and type the bovine viral diarrhea virus(BVDV) in cattle.Universal primers and Taq Man-MGB probes were designed from the 5′-untranslated region of known pestiviral sequences.Prior to optimizing the assay, c RNAs were transcribed in vitro from the BVDV 1 and BVDV 2 RTPCR products to make standard curves.The detection limit of the assay was 1.72×102 copies for BVDV 1 and 2.14×102copies for BVDV 2.The specificity of the assay evaluated on several BVDV strains including bovine herpesvirus 1(BHV 1), foot and mouth disease virus(FMDV) and several classical swine fever virus(CSFV) strains showed specific detection of the positive virus over 40 cycles.The assay was highly reproducible with the coefficient of variance ranging from 1.04 to 1.33% for BVDV 1 and from 0.83 to 1.48% for BVDV 2, respectively.Using this method, we tested a total of 2 327 cattle from three dairy farms for the presence of BVDV persistently infected(PI) animals.In this assay, each RT-PCR template contained a mixture of ten samples from different animals.The occurrence rate of PI cattle in three farms ranging from 0.9 to 2.54% could represent partly the PI rates in cattle farm in China.In conclusion, using our real-time PCR assay, we could effectively detect and type BVDV and identify PI cattle in a rapid and cost-effective manner.ZHANG Yong-qiang LIU Hai-sheng WU Xiao-dong WANG Xiao-zhen LI Jin-ming ZHAO Yong-gang Lü Yan REN Wei-jie GE Sheng-qiang WANG Zhi-liang 2015Journal of Integrative Agriculture2015,14,8:3
9Non-junctional Cx32 mediates anti-apoptotic and pro-tumor effects via epidermal growth factor receptor in human cervical cancer cells显示文摘OBJECTIVE To investigate the role of connexin proteins(Cx),which form gap junctions(GJ),in progression and chemotherapeutic sensitivity of cervical cancer(CaC x).METHODS We analyze the expression of Cx26,Cx30,Cx32 and Cx43 in human specimens consisting of:Normal cervix(n=78),CaCx FIGO stageⅠ(n=148),CaCx FIGO stageⅡ(n=165).In CaCx cell lines,Hela-Cx32(induced expression by doxycycline),C-33A(endogenously express Cx32)and si Ha(transiently transfected plasmid with Cx32),we detected the role of Cx32 against tostreptonigrin/cisplatin-induced apopotosisin presence or absence of functional GJ through using GJ inhibitors or low density cultural.Furtherly,we observed the relativity of Cx32 and EGFR expression in human specimens.Also,we detected the role of EGFR signaling pathway in the process of Cx32 anti-apoptosis through suppressed EGFR expression by inhibitors or si RNA sequences in cell lines.RESULTS We firstly demonstrated the expression of Cx32 was highly upregulated and accumulated in cytoplasm in the CaCx specimens,and the degree of upregulation correlated with advanced FIGO stages.Thus,in three human cervical cell lines,Cx32 was shown to suppress apoptosis when GJ formation is inhibited.No matter in cases of CaCx or cell lines,Cx32 expression was highly correlated with expression of EGFR and the EGFR pathway is an essential component of the Cx32-induced anti-apoptotic effect.CONCLUSION Cx32,traditionally tumor suppressive protein,was shown to be tumor protective against chemotherapy through EGFR pathway in a GJ-independent way.Yi-fan ZHAO Yong-chang LAI Hui GE Yun-quan GUO Xue FENG Jia SONG Qin WANG Li-xia FAN Andrew L HARRIS Xi-yan WANG Liang TAO 2017中国药理学与毒理学杂志2017,31,10:3

11APOPTOSIS AND TAXOL PRODUCTION IN SUSPENSION CULTURES OF Taxus spp.CELLS显示文摘The hallmark of apoptosis, in suspension cultures of Taxus spp. cells induced by fungal extractive or by abiotic means, was studied by total DNA agarose gel electrophoresis and in situ end-labeling. The cleavage of nuclear DNA (nDNA) into oligonucleosomal fragments(DNA laddering) was a characteristic of apoptosis, which involved cell shrinkage, condensation of cytoplasm and tracheary elements differentiation. Terminal deoxynucleotidy transferase-mediated dUTP nick end in situ labeling (TUNEL) assay of Taxus spp. cells showed that fungal extractive or abiotic elicltors (Ce4+, Taxol, H2O2) induced TUNEL positive. Also, the increase of the apoptotic cell ratio was accompanied by the increase of secondary metabolites (especially Taxol). These results suggest that apoptosis may have some coincidence with biosynthesis of Taxol.
