| 1 | Linking atmospheric pollution to cryospheric change in the Third Pole region: current progress and future prospects显示文摘The Tibetan Plateau and its surroundings are known as the Third Pole(TP). This region is noted for its high rates of glacier melt and the associated hydrological shifts that affect water supplies in Asia. Atmospheric pollutants contribute to climatic and cryospheric changes through their effects on solar radiation and the albedos of snow and ice surfaces; moreover, the behavior and fates within the cryosphere and environmental impacts of environmental pollutants are topics of increasing concern. In this review, we introduce a coordinated monitoring and research framework and network to link atmospheric pollution and cryospheric changes(APCC) within the TP region. We then provide an up-to-date summary of progress and achievements related to the APCC research framework, including aspects of atmospheric pollution’s composition and concentration, spatial and temporal variations, trans-boundary transport pathways and mechanisms, and effects on the warming of atmosphere and changing in Indian monsoon, as well as melting of glacier and snow cover. We highlight that exogenous air pollutants can enter into the TP’s environments and cause great impacts on regional climatic and environmental changes. At last, we propose future research priorities and map out an extended program at the global scale. | 2 | Update on the management of pancreatic cancer:Surgery is not enough显示文摘Pancreatic ductal adenocarcinoma(PDAC) represents the fourth cause of death in cancer and has a 5-year survival of < 5%.Only about 15% of the patients present with a resectable PDAC with potential to undergo 'curative' surgery.After surgery,local and systemic recurrence,is though very common.The median survival of resected patients with adjuvant chemotherapy after surgery is only 20-23 mo.This underscores the significant need to improve PDAC management strategies.Increased survival rate is dependent on new breakthroughs in our understanding of not at least tumor biology.The aim of this review is to update and comment on recent knowledge concerning PDAC biology and new diagnostics and treatment modalities.One fundamental approach to improve survival rates is by earlier and improved diagnosis of the disease.In recent years,novel blood-based biomarkers have emerged based on genetic,epigenetic and protein changes in PDAC with very promising results.For biomarkers to enter clinical practice they need to have been developed using adequate control groups and provide high sensitivity and specificity and by this identify patients at risk already in a pre-symptomatic stage.Another way to improve outcomes,is by employing neoadjuvant treatments thereby increasing the number of resectable cases.Novel systemic treatment regimes like FOLFIRINOX and nab-paclitaxel have demonstrated improvements in prolonging survival in advanced cases,but long-term survival is still scarce.The future improved understanding of PDAC biology will inevitably render new treatment options directed against both the cancer cells and the surrounding microenvironment. | Daniel Ansari Adam Gustafsson Roland Andersson | 2015 | World Journal of Gastroenterology2015,21,11: | 18 |

| 4 | High level of deoxycholic acid in human bile does not promote cholesterol gallstone formation显示文摘AIM: To study whether patients with excess deoxycholic acid (DCA) differ from those with normal percentage of DCA with respect to biliary lipid composition and cholesterol saturation of gallbladder bile.METHODS: Bile was collected during operation through puncturing into the gallbladder from 122 cholesterol gallstone patients and 46 gallstone-free subjects undergoing cholecystectomy. Clinical data, biliary lipids, bile acid composition,presence of crystals and nucleation time were analyzed.RESULTS: A subgroup of gallstone patients displayeda higher proportion of DCA in bile than gallstone free subjects.By choosing a cut-off level of the 90th percentile, a group of 13 gallstone patients with high DCA levels (mean 50percent of total bile acids) and a large group of 109 patients with normal DCA levels (mean 21 percent of total bile acids)were obtained. The mean age of the patients with high DCA levels was higher than that of the group with normal levels (mean age: 62 years vs45 years) and so was the mean BMI (28.3 vs. 24.7). Plasma levels of cholesterol and triglycerides were slightly higher in the DCA excess groups compared with those in the normal DCA group. There was no difference in biliary lipid composition, cholesterol saturation, nucleation time or occurrence of cholesterol crystals in bile between patients with high and normal levels of DCA.CONCLUSION: Gallstone patients with excess DCA were of older age and had higher BMI than patients with normal DCA. The two groups of patients did not differ with respect to biliary lipid composition, cholesterol saturation, nucleation time or occurrence of cholesterol crystals. | 11 | Bioengineering bacterial encapsulin nanocompartments as targeted drug delivery system显示文摘The development of Drug Delivery Systems(DDS)has led to increasingly efficient therapies for the treatment and detection of various diseases.DDS use a range of nanoscale delivery platforms produced from polymeric of inorganic materials,such as micelles,and metal and polymeric nanoparticles,but their variant chemical composition make alterations to their size,shape,or structures inherently complex.Genetically encoded protein nanocages are highly promising DDS candidates because of their modular composition,ease of recombinant production in a range of hosts,control over assembly and loading of cargo molecules and biodegradability.One example of naturally occurring nanocompartments are encapsulins,recently discovered bacterial organelles that have been shown to be reprogrammable as nanobioreactors and vaccine candidates.Here we report the design and application of a targeted DDS platform based on the Thermotoga maritima encapsulin reprogrammed to display an antibody mimic protein called Designed Ankyrin repeat protein(DARPin)on the outer surface and to encapsulate a cytotoxic payload.The DARPin9.29 chosen in this study specifically binds to human epidermal growth factor receptor 2(HER2)on breast cancer cells,as demonstrated in an in vitro cell culture model.The encapsulin-based DDS is assembled in one step in vivo by co-expressing the encapsulin-DARPin9.29 fusion protein with an engineered flavin-binding protein mini-singlet oxygen generator(MiniSOG),from a single plasmid in Escherichia coli.Purified encapsulin-DARPin_miniSOG nanocompartments bind specifically to HER2 positive breast cancer cells and trigger apoptosis,indicating that the system is functional and specific.The DDS is modular and has the potential to form the basis of a multi-receptor targeted system by utilising the DARPin screening libraries,allowing use of new DARPins of known specificities,and through the proven flexibility of the encapsulin cargo loading mechanism,allowing selection of cargo proteins of choice. | Alexander Van de Steen Rana Khalife Noelle Colant Hasan Mustafa Khan Matas Deveikis Saverio Charalambous Clare M.Robinson Rupali Dabas Sofia Esteban Serna Diana A.Catana Konstantin Pildish Vladimir Kalinovskiy Kenth Gustafsson Stefanie Frank | 2021 | Synthetic and Systems Biotechnology2021,6,3: | 2 |
