Endovascular stent-induced alterations in host artery mechanical environments and their roles in stent restenosis and late thrombosis显示文摘Cardiovascular stent restenosis remains a major challenge in interventional treatment of cardiovascular occlusive disease.Although the changes in arterial mechanical environment due to stent implantation are the main causes of the initiation of restenosis and thrombosis, the mechanisms that cause this initiation are still not fully understood. In this article,we reviewed the studies on the issue of stent-induced alterations in arterial mechanical environment and discussed their roles in stent restenosis and late thrombosis from three aspects:(i)the interaction of the stent with host blood vessel, involve the response of vascular wall, the mechanism of mechanical signal transmission, the process of re-endothelialization and late thrombosis;(ii)the changes of hemodynamics in the lumen of the vascular segment and(iii)the changes of mechanical microenvironment within the vascular segment wall due to stent implantation. This review has summarized and analyzed current work in order to better solve the two main problems after stent implantation, namely in stent restenosis and late thrombosis, meanwhile propose the deficiencies of current work for future reference. | Jinxuan Wang Xuepu Jin Yuhua Huang Xiaolin Ran Desha Luo Dongchuan Yang Dongyu Jia Kang Zhang Jianhua Tong Xiaoyan Deng Guixue Wang | 2018 | Regenerative Biomaterials2018,5,3: | 13
A novel mechanism of inhibiting in-stent restenosis with arsenic trioxide drug-eluting stent: Enhancing contractile phenotype of vascular smooth muscle cells via YAP pathway显示文摘Objective:Arsenic trioxide(ATO or As2O3)has beneficial effects on suppressing neointimal hyperplasia and restenosis, but the mechanism is still unclear. The goal of this study is to further understand the mechanism of ATO's inhibitory effect on vascular smooth muscle cells(VSMCs).Methods and results:Through in vitro cell culture and in vivo stent implanting into the carotid arteries of rabbit,a synthetic-to-contractile phenotypic transition was induced and the proliferation of VSMCs was inhibited by ATO.F-actin filaments were clustered and the elasticity modulus was increased within the phenotypic modulation of VSMCs induced by ATO in vitro.Meanwhile,Yes-associated protein(YAP)nuclear translocation was inhibited by ATO both in vivo and in vitro. It was found that ROCK inhibitor or YAP inactivator could partially mask the phenotype modulation of ATO on VSMCs.Conclusions:The interaction of YAP with the ROCK pathway through ATO seems to mediate the contractile phenotype of VSMCs.This provides an indication of the clinical therapeutic mechanism for the beneficial bioactive effect of ATO-drug eluting stent(AES)on in-stent restenosis(ISR). | Yinping Zhao Guangchao Zang Tieying Yin Xiaoyi Ma Lifeng Zhou Lingjuan Wu Richard Daniel Yunbing Wang Juhui Qiu Guixue Wang | 2021 | Bioactive Materials2021,6,2: | 13
Effect of intraplaque angiogenesis to atherosclerotic rupture-prone plaque induced by high shear stress in rabbit model显示文摘Atherosclerotic prone-rupture plaque is mainly localized in the region of the entrance to the stenosiswith high shear stress and the reasons are largely unknown. Our hypothesis is that such a distributionof cells in atherosclerotic plaque may depend on the angiogenesis. Silastic collars inducedregions of high shear stress (20.6865.27 dynes/cm2) in the upstream flow and low shear stress(12.2561.28 dynes/cm2) in the downstream flow in carotid arteries. Compared with the low shearstress region, plaques in the high shear stress region showed more intraplaque haemorrhaging, less collagen and higher apoptotic rates of vascular smooth muscle cells;endothelial cells (ECs) inthe high shear stress region were characterized with integrity and high endothelial nitric oxidesynthase (eNOS) expression (1570.36345.5% vs 172.9649.9%). The number of intraplaque microvesselsis very high in the high shear stress region (1561.8 n/mm2 vs 3.560.4 n/mm2), and themicrovessels in the plaque show ECs were abnormal, with membrane blebs, intracytoplasmic vacuolesand leukocyte infiltration. Our current study reveals that the integrity of the endothelium andthe vulnerability of atherosclerotic plaques are simultaneously localized in high shear stress regions, and we provide evidence for the first time that microvessels in the intraplaque maybe responsiblefor rupture-prone plaque formation in the high shear stress region. | Juhui Qiu Daoxi Lei Jianjun Hu Tieying Yin Kang Zhang Donghong Yu Guixue Wang | 2017 | Regenerative Biomaterials2017,4,4: | 12
Initial corrosion behavior and mechanism of 7B04 aluminum alloy under acid immersion and salt spray environments显示文摘The initial corrosion behavior and mechanism of 7B04 aluminum alloy under acid immersion and salt spray environments(pH=3.5)are studied by Scanning Electron Microscope(SEM),optical microscope,Fourier Transform Infrared Spectroscopy(FT-IR),X-Ray Diffraction(XRD),potentiodynamic polarization,Electrochemical Impedance Spectroscopy(EIS),and Scanning Kelvin Probe(SKP).The results show that pitting corrosion occurs at the initial corrosion stage, and the potential difference between the second phase particles is the main cause of pitting.Pitting pits on different locations gradually expand and coalesce with the proceeding of corrosion. The main components of corrosion products are Al_(2)O_(3),Al(OH)_(3),and AlCl_(3),and the generation rate of the corrosion product layer under the salt spray environment is larger than that under acid immersion environment.Under both environments, the Volta potential distribution first disperses and then concentrates, while the charge transfer resistance first decreases and then increases with the corrosion time. The Volta potential gradually shifts in a positive direction, indicating that corrosion products have an inhibitory effect on corrosion. After the same corrosion time, the corrosion product layer resistance and the expectation of the Volta potential of the salt spray sample are higher than those of the immersion sample.Comparatively, the corrosion current density for the salt spray sample is significantly lower than the immersion sample, which indicates that the thicker the corrosion product layer, the stronger the inhibition of corrosion reaction. | Yangguang ZHANG Yueliang CHEN Yong ZHANG Guixue BIAN Chenguang WANG Andong WANG | 2022 | Chinese Journal of Aeronautics2022,35,1: | 8
Distinctive effects of CD34- and CD133-specific antibody-coated stents on re-endothelialization and in-stent restenosis at the early phase of vascular injury显示文摘It is not clear what effects of CD34-and CD133-specific antibody-coated stents have on reendothelialization and in-stent restenosis(ISR)at the early phase of vascular injury. This study aims at determining the capabilities of different coatings on stents(e.g.gelatin,anti-CD133 and anti-CD34 antibodies)to promote adhesion and proliferation of endothelial progenitor cells(EPCs).The in vitro study revealed that the adhesion force enabled the EPCs coated on glass slides to withstand flow-induced shear stress,so that allowing for the growth of the cells on the slides for 48 h.The in vivo experiment using a rabbit model in which the coated stents with different substrates were implanted showed that anti-CD34 and anti-CD133 antibody-coated stents markedly reduced the intima area and restenosis than bare mental stents(BMS)and gelatin-coated stents.Compared with the anti-CD34 antibody-coated stents, the time of cells adhesion was longer and earlier present in the anti-CD133 antibody-coated stents and anti-CD133 antibody-coated stents have superiority in re-endothelialization and inhibition of ISR.In conclusion, this study demonstrated that anti-CD133 antibody as a stent coating for capturing EPCs is better than anti-CD34 antibody in promoting endothelialization and reducing ISR. | Xue Wu Tieying Yin Jie Tian Chaojun Tang Junli Huang Yinping Zhao Xiaojuan Zhang Xiaoyan Deng Yubo Fan Donghong Yu Guixue Wang | 2015 | Regenerative Biomaterials2015,2,2: | 7
Inhibition of in-stent restenosis after graphene oxide double-layer drug coating with good biocompatibility显示文摘In this study,we designed a double layer-coated vascular stent of 316L stainless steel using an ultrasonic spray system to achieve both antiproliferation and antithrombosis. The coating included an inner layer of graphene oxide(GO)loaded with docetaxel(DTX)and an outer layer of carboxymethyl chitosan(CMC)loaded with heparin(Hep). The coated surface was uniform without aggregation and shedding phenomena before and after stent expanded. The coating treatment was able to inhibit the adhesion and activation of platelets and the proliferation and migration of smooth muscle cells, indicating the excellent biocompatibility and antiproliferation ability. The toxicity tests showed that the GO/DTX and CMC/Hep coating did not cause deformity and organ abnormalities in zebrafish under stereomicroscope. The stents with GO double-layer coating were safe and could effectively prevent thrombosis and in-stent restenosis after the implantation into rabbit carotid arteries for 4–12 weeks. | Shuang Ge Yadong Xi Ruolin Du Yuzhen Ren Zichen Xu Youhua Tan Yazhou Wang Tieying Yin Guixue Wang | 2019 | Regenerative Biomaterials2019,6,5: | 4
Uptake of oxidative stress-mediated extracellular vesicles by vascular endothelial cells under low magnitude shear stress显示文摘Extracellular vesicles(EVs)are increasingly used as delivery vehicles for drugs and bioactive molecules, which usually require intravascular administration. The endothelial cells covering the inner surface of blood vessels are susceptible to the shear stress of blood flow.Few studies demonstrate the interplay of red blood cell-derived EVs(RBCEVs)and endothelial cells. Thus, the phagocytosis of EVs by vascular endothelial cells during blood flow needs to be elucidated. In this study, red blood cell-derived extracellular vesicles(RBCEVs)were constructed to investigate endothelial cell phagocytosis in vitro and animal models. Results showed that low magnitude shear stress including low shear stress(LSS)and oscillatory shear stress(OSS)could promote the uptake of RBCEVs by endothelial cells in vitro. In addition,in zebrafish and mouse models,RBCEVs tend to be internalized by endothelial cells under LSS or OSS.Moreover,RBCEVs are easily engulfed by endothelial cells in atherosclerotic plaques exposed to LSS or OSS.In terms of mechanism, oxidative stress induced by LSS is part of the reason for the increased uptake of endothelial cells.Overall, this study shows that vascular endothelial cells can easily engulf EVs in areas of low magnitude shear stress, which will provide a theoretical basis for the development and utilization of EVs-based nano-drug delivery systems in vivo. | Xian Qin Kun Zhang Juhui Qiu Nan Wang Kai Qu Yuliang Cui Junli Huang Li Luo Yuan Zhong Tian Tian Wei Wu Yi Wang Guixue Wang | 2022 | Bioactive Materials2022,7,3: | 3

Photocrosslinking silver nanoparticles–aloe vera–silk fibroin composite hydrogel for treatment of full-thickness cutaneous wounds显示文摘Damage to the skin causes physiological and functional issues. The most effective treatment approach is the use of wound dressings.Silk fibroin(SF)is a promising candidate biomaterial for regulating wound healing;however, its antibacterial properties and biological activity must be further improved. In this study,a photocrosslinking hydrogel was developed to treat full-thickness cutaneous wounds. The composite hydrogel(Ag-AV-SF hydrogel)was prepared by introducing the silver nanoparticles(AgNPs)and aloe vera(AV)as the modifiers. In vitro study exhibited great antibacterial ability, biocompatibility and cell-proliferation and-migration-promoting capacities. It also showed the pH-response releasing properties which release more AgNPs in a simulated chronic infection environment. The healing effect evaluation in vivo showed the healing-promoting ability of the Ag-AV-SF hydrogel was stronger than the single-modifiers groups, and the healing rate of it reached 97.02%on Day 21,higher than the commercial wound dressing, silver sulfadiazine(SS)cream on sale.Additionally, the histological and protein expression results showed that the Ag-AV-SF hydrogel has a greater effect on the pro-healing regenerative phenotype with M2 macrophages at the early stage, reconstructing the blood vessels networks and inhibiting the formation of scars. In summary, the Ag-AV-SF hydrogel developed in this study had good physical properties, overwhelming antibacterial properties, satisfactory biocompatibility and significantly promoting effect on cell proliferation, migration and wound healing.Overall, our results suggest that the Ag-AV-SF hydrogel we developed has great potential for improving the wound healing in clinical treatment. | Yangkun Liu JinChuan Fan MingQi Lv Kepeng She Jiale Sun Qingqing Lu Changhao Han SongTao Ding Shuang Zhao GuiXue Wang YuChan Zhang GuangChao Zang | 2021 | Regenerative Biomaterials2021,8,6: | 2

ROS-responsive biomimetic nanoparticles for potential application in targeted anti-atherosclerosis显示文摘The development of nanomedicines provides new opportunities for the treatment of atherosclerosis(AS)due to their great advantages such as the improved drug solubility, enhanced bioavailability and reduced side effects.Despite these advantages, nanomedicines are still facing some challenges. The problems remain in the short circulation life, lack of specific targeting and poor drug release controllability. In order to overcome the shortages of conventional nanomedicines, the combination of biomimetic strategy with smart nanoagents has been proposed. In light with the high reactive oxygen species(ROS)level in AS microenvironment and the fact that macrophages play a critical role in the pathogenesis of AS,we fabricated ROS-responsive biomimetic nanoparticles(NPs),which camouflaged macrophage membrane(MM)on ROS-responsive NPs loaded with rapamycin(RNPs)for potential application in AS therapy. The resulting ROSresponsive biomimetic NPs(MM/RNPs)exhibited favorable hydrodynamic size with negative surface charge, retained the functional proteins from MM,and showed ROS-responsive drug release.Because of the biomimetic camouflaging on surface,MM/RNPs could effectively escape from macrophages uptake and target to inflammatory endothelial cells.Meanwhile,MM/RNPs could inhibit the proliferation of macrophages and smooth muscle cells in vitro.Furthermore, the MM-coated NPs were found to be nontoxic in both cytotoxicity assay and in vivo toxicity evaluation.Consequently, these results demonstrated that MM/RNPs could be a potential candidate of drug delivery system for safe and effective anti-AS applications. | Dan Tang Yi Wang Andy Wijaya Boyan Liu Ali Maruf Jinxuan Wang Jianxiong Xu Xiaoling Liao Wei Wu Guixue Wang | 2021 | Regenerative Biomaterials2021,8,4: | 2
Two-stage degradation and novel functional endothelium characteristics of a 3-D printed bioresorbable scaffold显示文摘Bioresorbable scaffolds have emerged as a new generation of vascular implants for the treatment of atherosclerosis, and designed to provide a temporary scaffold that is subsequently absorbed by blood vessels over time.Presently, there is insufficient data on the biological and mechanical responses of blood vessels accompanied by bioresorbable scaffolds(BRS)degradation. Therefore,it is necessary to investigate the inflexion point of degradation, the response of blood vessels, and the pathophysiological process of vascular,as results of such studies will be of great value for the design of next generation of BRS.In this study, abdominal aortas of SD rats were received 3-D printed poly-l-actide vascular scaffolds(PLS)for various durations up to 12 months. The response of PLS implanted aorta went through two distinct processes:(1)the neointima with desirable barrier function was obtained in 1 month, accompanied with slow degradation, inflammation, and intimal hyperplasia;(2)significant degradation occurred from 6 months, accompanied with decreasing inflammation and intimal hyperplasia, while the extracellular matrix recovered to normal vessels which indicate the positive remodeling. These in vivo results indicate that 6 months is a key turning point.This“two-stage degradation and vascular characteristics”is proposed to elucidate the long-term effects of PLS on vascular repair and demonstrated the potential of PLS in promoting endothelium function and positive remodeling, which highlights the benefits of PLS and shed some light in the future researches, such as drug combination coatings design. | Tieying Yin Ruolin Du Yang Wang Junyang Huang Shuang Ge Yuhua Huang Youhua Tan Qing Liu Zhong Chen Hanqing Feng Jie Du Yazhou Wang Guixue Wang | 2022 | Bioactive Materials2022,7,4: | 2
Multi-platform omics analysis reveals molecular signature for COVID-19 pathogenesis, prognosis and drug target discovery显示文摘Disease progression prediction and therapeutic drug target discovery for Coronavirus disease 2019(COVID-19)are particularly important,as there is still no effective strategy for severe COVID-19 patient treatment.Herein,we performed multi-platform omics analysis of serial plasma and urine samples collected from patients during the course of COVID-19.Integrative analyses of these omics data revealed several potential therapeutic targets, such as ANXA1 and CLEC3B.Molecular changes in plasma indicated dysregulation of macrophage and suppression of T cell functions in severe patients compared to those in non-severe patients.Further,we chose 25 important molecular signatures as potential biomarkers for the prediction of disease severity. The prediction power was validated using corresponding urine samples and plasma samples from new COVID-19 patient cohort, with AUC reached to 0.904 and 0.988,respectively.In conclusion, our omics data proposed not only potential therapeutic targets, but also biomarkers for understanding the pathogenesis of severe COVID-19. | Yuming Li Guixue Hou Haibo Zhou Yanqun Wang Hein Min Tun Airu Zhu Jingxian Zhao Fei Xiao Shanwen Lin Dongdong Liu Dunrong Zhou Lang Mai Lu Zhang Zhaoyong Zhang Lijun Kuang Jiao Guan Qiushi Chen Liyan Wen Yanjun Zhang Jianfen Zhuo Fang Li Zhen Zhuang Zhao Chen Ling Luo Donglan Liu Chunke Chen Mian Gan Nanshan Zhong Jincun Zhao Yan Ren Yonghao Xu | 2021 | Signal Transduction and Targeted Therapy2021,6,5: | 1

trans-2-Enoyl-CoA Reductase Tecr-Driven Lipid Metabolism in Endothelial Cells Protects against Transcytosis to Maintain Blood-Brain Barrier Homeostasis显示文摘The transport and metabolism of lipids in cerebrovascular endothelial cells(ECs)have been hypothesized to regulate blood-brain barrier(BBB)maturation and homeostasis.Long-chain polyunsaturated fatty acids(LCPUFAs)as the important lipids components of cell membranes are essential for the development and function of BBB,but the direct links of lipid metabolism and ECs barrier function remain to be established. Here,we comprehensively characterize the transcriptomic phenotype of developmental cerebrovascular ECs in single-cell resolution and firstly find that trans-2-enoyl-CoA reductase(Tecr),a verylong-chain fatty acid synthesis,is highly expressed during barriergenesis and decreased after BBB maturation.EC-specific knockout of Tecr compromises angiogenesis due to delayed vascular sprouting.Importantly,EC-specific deletion of Tecr loss restrictive quality of vascular permeability from neonatal stages to adulthood, with high levels of transcytosis, but maintains the vascular tight junctions. Moreover, lipidomic analysis shows that the expression of Tecr in ECs is associated with the containing of omega-3 fatty acids, which directly suppresses caveolae vesicles formation. These results reveal a protective role for Tecr in BBB integrity and suggest that Tecr as a novel therapeutic target in the central nervous system(CNS)diseases associated with BBB dysfunction. | Jinxuan Wang Jianxiong Xu Guangchao Zang Tao Zhang Qi Wu Hongping Zhang Yidan Chen Yi Wang Weixi Qin Shuang Zhao Erdai Qin Juhui Qiu Xiaojuan Zhang Lin Wen Yeqi Wang Guixue Wang | 2022 | Research2022,,3: | 1
Remodeling tumor immunosuppressive microenvironment via a novel bioactive nanovaccines potentiates the efficacy of cancer immunotherapy显示文摘The clinical outcomes of cancer nanovaccine have been largely impeded owing to the low antigen-specific T cell response rate and acquired resistance caused by the immunosuppressive tumor microenvironment(TME).Here,we reported a tumor acidity-responsive nanovaccine to remodel the immunosuppressive TME and expand the recruitment of tumor infiltrating lymphocytes(TILs)using hybrid micelles(HM),which encapsulated colony stimulating factor 1 receptor(CSF1-R)inhibitor BLZ-945 and indoleamine 2,3-dioxygenase(IDO)inhibitor NLG-919 in its core and displayed a model antigen ovalbumin(OVA)on its surface(denoted as BN@HM-OVA).The bioactive nanovaccine is coated with a polyethylene glycol(PEG)shell for extending nanoparticle circulation. The shell can be shed in response to the weakly acidic tumor microenvironment. The decrease in size and the increase in positive charge may cause the deep tumor penetration of drugs. We demonstrated that the bioactive nanovaccine dramatically enhance antigen presentation by dendritic cells(DCs)and drugs transportation into M1-like tumor-associated macrophages(TAMs)and tumor cells via size reduction and increasing positive charge caused by the weakly acidic TME.Such bioactive nanovaccine could remodel the immunosuppressive TME into an effector T cells favorable environment, leading to tumor growth inhibition in prophylactic and therapeutic E.G7-OVA tumor models.Furthermore, combining the bioactive nanovaccine with simultaneous anti-PD-1 antibody treatment leads to a long-term tumor inhibition, based on the optimal timing and sequence of PD-1 blockade against T cell receptor. This research provides a new strategy for the development of efficient cancer immunotherapy. | Xiaoxue Xie Yi Feng Hanxi Zhang Qingqing Su Ting Song Geng Yang Ningxi Li Xiaodan Wei Tingting Li Xiang Qin Shun Li Chunhui Wu Xiaojuan Zhang Guixue Wang Yiyao Liu Hong Yang | 2022 | Bioactive Materials2022,7,10: | 1
