2Tomographic PIV investigation of coherent structures in a turbulent boundary layer flow显示文摘Tomographic particle image velocimetry was used to quantitatively visualize the three-dimensional coherent structures in the logarithmic region of the turbulent boundary layer in a water tunnel.The Reynolds number based on momentum thickness is Reθ = 2 460.The instantaneous velocity fields give evidence of hairpin vortices aligned in the streamwise direction forming very long zones of low speed fluid,which is flanked on either side by highspeed ones.Statistical support for the existence of hairpins is given by conditional averaged eddy within an increasing spanwise width as the distance from the wall increases,and the main vortex characteristic in different wall-normal regions can be reflected by comparing the proportion of ejection and its contribution to Reynolds stress with that of sweep event.The pre-multiplied power spectra and two-point correlations indicate the presence of large-scale motions in the boundary layer,which are consistent with what have been termed very large scale motions(VLSMs).The three dimen-sional spatial correlations of three components of velocity further indicate that the elongated low-speed and highspeed regions will be accompanied by a counter-rotating roll modes,as the statistical imprint of hairpin packet structures,all of which together make up the characteristic of coherent structures in the logarithmic region of the turbulent boundary layer(TBL).Zhan-Qi Tang Nan Jiang Andreas Schroder Reinhard Geisler 2012Acta Mechanica Sinica2012,28,3:14
3MicroRNA-regulated viral vectors for gene therapy显示文摘Safe and effective gene therapy approaches require targeted tissue-specific transfer of a therapeutic transgene.Besides traditional approaches, such as transcriptional and transductional targeting, micro RNA-dependent posttranscriptional suppression of transgene expression has been emerging as powerful new technology to increase the specificity of vector-mediated transgene expression. Micro RNAs are small non-coding RNAs and often expressed in a tissue-, lineage-, activation- or differentiation-specific pattern. They typically regulate gene expression by binding to imperfectly complementary sequences in the 3' untranslated region(UTR) of the m RNA. To control exogenous transgene expression, tandem repeats of artificial micro RNA target sites are usually incorporated into the 3' UTR of the transgene expression cassette, leading to subsequent degradation of transgene m RNA in cel s expressing the corresponding micro RNA. This targeting strategy, first shown for lentiviral vectors in antigen presenting cells, has now been used for tissue-specific expression of vector-encoded therapeutic transgenes, to reduce immune response against the transgene, to control virus tropism for oncolytic virotherapy, to increase safety of live attenuated virus vaccines and to identify and select cell subsets for pluripotent stem cell therapies, respectively. 4A novel miR167a-OsARF6-OsAUX3 module regulates grain length and weight in rice显示文摘Grain size is one of the most important factors that control rice yield,as it is associated with grain weight(GW).To date,dozens of rice genes that regulate grain size have been isolated;however,the regulatory mechanism underlying GW control is not fully understood.Here,the quantitative trait locus qGL5 for grain length(GL)and GW was identified in recombinant inbred lines of 9311 and Nipponbare(NPB)and fine mapped to a candidate gene,OsAUX3.Sequence variations between 9311 and NPB in the OsAUX3 promoter and loss of function of OsAUX3 led to higher GL and GW.RNA sequencing,gene expression quantification,dual-luciferase reporter assays,chromatin immunoprecipitation-quantitative PCR,and yeast one-hybrid assays demonstrated that OsARF6 is an upstream transcription factor regulating the expression of OsAUX3.OsARF6 binds directly to the auxin response elements of the OsAUX3 promoter,covering a single-nucleotide polymorphism site between 9311 and NPB/Dongjin/Hwayoung,and thereby controls GL by altering longitudinal expansion and auxin distribution/content in glume cells.Furthermore,we showed that miR167a positively regulate GL and GW by directing OsARF6 mRNA silencing.Taken together,our study reveals that a novel miR167a-OsARF6-OsAUX3 module regulates GL and GW in rice,providing a potential target for the improvement of rice yield.Jiyue Qiao Hongzhen Jiang Yuqing Lin Lianguang Shang Mei Wang Dongming Li Xiangdong Fu Markus Geisler Yanhua Qi Zhenyu Gao Qian Qian 2021Molecular Plant2021,14,10:8
5Microfluidic actuators based on temperature-responsive hydrogels显示文摘The concept of using stimuli-responsive hydrogels to actuate fluids in microfluidic devices is particularly attractive,but limitations,in terms of spatial resolution,speed,reliability and integration,have hindered its development during the past two decades.By patterning and grafting poly(N-isopropylacrylamide)PNIPAM hydrogel films on plane substrates with a 2μm horizontal resolution and closing the system afterward,we have succeeded in unblocking bottlenecks that thermo-sensitive hydrogel technology has been challenged with until now.In this paper,we demonstrate,for the first time with this technology,devices with up to 7800 actuated micro-cages that sequester and release solutes,along with valves actuated individually with closing and opening switching times of 0.6±0.1 and 0.25±0.15 s,respectively.Two applications of this technology are illustrated in the domain of single cell handling and the nuclear acid amplification test(NAAT)for the Human Synaptojanin 1 gene,which is suspected to be involved in several neurodegenerative diseases such as Parkinson’s disease.The performance of the temperature-responsive hydrogels we demonstrate here suggests that in association with their moderate costs,hydrogels may represent an alternative to the actuation or handling techniques currently used in microfluidics,that are,pressure actuated polydimethylsiloxane(PDMS)valves and droplets.Loïc D'Eramo Benjamin Chollet Marie Leman Ekkachai Martwong Mengxing Li Hubert Geisler Jules Dupire Margaux Kerdraon Clémence Vergne Fabrice Monti Yvette Tran Patrick Tabeling 2018Microsystems & Nanoengineering2018,4,1:3
