1Targeting the PI3K/Akt signaling pathway in gastric carcinoma: A reality for personalized medicine?显示文摘Frequent activation of phosphatidylinositol-3 kinases(PI3K)/Akt/m TOR signaling pathway in gastric cancer(GC) is gaining immense popularity with identification of mutations and/or amplifications of PIK3 CA gene or loss of function of PTEN,a tumor suppressor protein,to name a few; both playing a crucial role in regulating this pathway. These aberrations result in dysregulation of this pathway eventually leading to gastric oncogenesis,hence,there is a need for targeted therapy for more effective anticancer treatment. Several inhibitors are currently in either preclinical or clinical stages for treatment of solid tumors like GC. With so many inhibitors under development,further studies on predictive biomarkers are needed to measure the specificity of any therapeutic intervention. Herein,we review the common dysregulation of PI3K/Akt/m TOR pathway in GC and the various types of single or dual pathway inhibitors under development that might have a superior role in GC treatment. 3Pushover analysis of underground structures:Method and application显示文摘Pushover analysis is common because of its conceptual simplicity and computational attractiveness in computing seismic demand.Considering that traditional pushover analysis is restricted in underground structures due to the stark differences in the seismic response characteristics of surface structures,this paper proposes a pushover analysis method for underground structures and its application in seismic damage assessment.First,three types of force distribution are presented based on ground response analysis.Next,the target displacements and analysis models are established according to force-based and performance-based design.Then,the pushover analysis procedure for underground structures is described.Next,the applicability of pushover analysis to underground structures is verified by comparing the responses of a Chongwenmen subway station determined by the proposed procedure and by nonlinear response history analysis.In addition,two other points are made:that the inverted triangular distribution of effective earthquake acceleration is more practical than the other two distributions,and that performance-based design is more effective than force-based design.Finally,a cyclic reversal loading pattern based on one cycle of reversal loads as an earthquake event is presented and applied to the seismic damage assessment of underground structures.The results show that the proposed pushover analysis can be effectively applied to the seismic design and damage assessment of underground structures.LIU JingBo WANG WenHui DASGUPTA Gautam 2014Science China(Technological Sciences)2014,57,2:20
4Role of endoscopic therapy in early esophageal cancer显示文摘Esophageal carcinoma is a highly lethal cancer associated with high morbidity and mortality. Esophageal squamous cell carcinoma and esophageal adenocarcinoma are the two distinct histological types. There has been significant progress in endoscopic diagnosis and treatment of early stages of cancer using resection and ablation techniques, as shown in several trials in the recent past. Earlier detection of esophageal cancer and advances in treatment modalities have lead to improvement in the 5-year survival from 5% to about 20% in the past decade. Endoscopic eradication therapy is the preferred modality of treatment in cancer limited to mucosal layer of the esophagus as there is very low risk of lymph node metastasis, leading to high cure rates, low risk of recurrence and with few adverse effects. The most common adverse events seen are strictures, bleeding and rarely perforation which can be endoscopically managed. In patients with recurrent advanced disease or invasive tumor, esophagectomy with lymph node dissection remains the mainstay of treatment. There is debate on post-endoscopic surveillance with some studies suggesting closer follow up with upper endoscopy every 6 mo for the first 1-2 years and then annually for the 3 years while others recommending the appropriate action only if symptoms or other abnormalities develop. 5Estimation of above-ground biomass and carbon stock of an invasive woody shrub in the subtropical deciduous forests of Doon Valley,western Himalaya,India显示文摘This study describes the different parameters used to derive the allometric equation for calculating the biomass of an invasive woody shrub Lantana camara L.from the subtropical conditions of western Himalaya.It identifies the most accurate and convenient method for biomass calculation by comparing destructive with nondestructive methodology.Different parameters were measured on a wide range of Lantana from different community levels for the non-destructive calculation of total aboveground biomass.Different explanatory variables were identified and measured such as basal diameter either as a single independent variable or in combination with plant height.The other suitable combinations of available independent variables include crown length,crown width,crown area,crown volume and coverage of the plant.Amongst the wide range of allometric equations used with different variables,the equation with D2 H as a variable was found to be the most suitable estimator of biomass calculation for Lantana.Sahastradhara,being the most disturbed area due to its high tourist activity round the year,showed maximum coverage(58.57 % ha-1),highest biomass(13,559.60 kg ha-1) and carbon density(6,373.01 kg ha-1)of Lantana.The degree of Lantana's invasiveness in subtropical conditions was also calculated on the basis of importance value index(IVI).The maximum IVI(22.77)and mean coverage(26.8 % ha-1) was obtained from the areas near Jolly Grant airport,indicating that physically disturbed areas are more suitable for the growth of Lantana,which may significantly increase shrub biomass.The importance of incorporating allometric equations in calculation of shrub biomass,and its role in atmospheric carbon assimilation has thus been highlighted through the findings of this study.Gautam Mandal S.P.Joshi 2015Journal of Forestry Research2015,26,2:8

10Seed Zone Properties and Crop Performance as Affected by Three No-Till Seeders for Permanent Raised Beds in Arid Northwest China显示文摘The no-till seeders of various soil opener configurations have been shown to produce various soil physical responses in relation to soil and climate conditions, thus affecting crop performance in permanent raised beds (PRB) systems. This is particularly important in arid Northwest China where large volumes of residue are retained on the soil surface after harvest. In Zhangye, Gansu Province, China, a field trial assessed the effects of three typical (powered-chopper, powered-cutter and powered-disc) PRB no-till seeders and one traditional seeder on soil disturbance, residue cover index, bulk density, fuel consumption, plant growth, and subsequent yield. In general, seedbed conditions and crop performance for PRB no- till seeders seeded plots were better than for traditional seeded plots. In PRB cropping system, the powered-chopper seeder decreased mean soil disturbance and increased residue cover index compared to powered-disc and -cutter seeders. However, the results indicated that soil bulk density was 2.3-4.8% higher, soil temperature was 0.2-0.6℃ lower, and spring wheat emergence was 3.2-4.7% less. This was attributed to greater levels of residue cover and firmer seedbeds. Spring maize and wheat performance in the powered-cutter and -disc treatments was better (non-significant) than powered-chopper treatment. So powered disc no-till seeder, which generally provided the best planting condition and the highest yield, appeared to be the suitable seeder in heavy residue cover conditions. Considering the precision requirements for soil disturbance and residue cover, the powered strip-chopping no-till seeder could be a suitable option for PRB cropping system in Northwest China. 11Effects of 42-year long-term fertilizer management on soil phosphorus availability, fractionation,adsorption–desorption isotherm and plant uptake in flooded tropical rice显示文摘Soil phosphorus(P) fractionation, adsorption, and desorption isotherm, and rice yield and P uptake were investigated in flooded tropical rice(Oryza sativa L.) following 42-year fertilizer and manure application. The treatments included low-input [unfertilized control without N, P, or K(C0N0)], farmyard manure(FYM)(C1N0), NP(C0NP), NPK(C0NPK), FYM + NP(C1NP), and high-input treatment, FYM + NPK(C1NPK). Grain yield was increased significantly by 74%over the control under the combined application of FYM + NPK. However, under low- and high-input treatments, yield as well as P uptake was maintained at constant levels for 35 years.During the same period, high yield levels and P uptake were maintained under the C0 NP, C0 NPK,and C1 NPK treatments. These are unique characteristics of a tropical flooded ecosystem, which is a self-sustaining system for rice production. The Fe–P fraction was highest compared to the Ca–P and Al–P fractions after 42 years of fertilizer application and was significantly higher under FYM + NPK treatment. The P adsorption capacity of soil was highest under the low-input treatment and lowest under long-term balanced fertilization(FYM + NPK). In contrast, P desorption capacity was highest under NPK and lowest in the control treatment. 12Design and Experiment of ф-type-knots Knotters on Chinese Small Square Balers显示文摘Since the knotters on the Chinese rectangular balers are imported from outside of the country,Chinese knotters with independent intellectual property rights is far away from being closed.In order to harvest a large quantity of straw in a short period on the small-scale lands of China,basic requirements on the knotters are summarized.Mathematical model of the knotter is also determined uniquely.Furthermore,theф-type-knots knotter equipped on the Chinese square baler to form theфtype knots is designed.Knotting rate experiments of theф-type-knots knotter on the test bench and in the wheat/maize straws covered fields are carried out to check the knotting performances of the knotter.The parameters of the formed knots are also tested.The experiments results show that the knotting rate of theф-type-knots knotter reaches 100.0%on the test bench without straws,while reaches 99.6%in the wheat straws covered field and 100.0%in the maize straws covered field.The average maximum force in the knotting process is 194.7 N in the lab experiment.The length out of the knots formed in lab is 15.9%–20.6%lower than the knots formed in the field experiment.The breaking force of the knots formed in the field is 115.9%–167.2%higher than the knots formed in lab due to the higher preload and interactions with the compacted bales.Highly relevant relationships exist between the breaking force of the formed knots and the maximum force in the forming process of the knots in the lab experiment.The designed knotter breaks out the embarrassing situation of the domestic knotters which don’t have independent intellectual property rights,and promotes the development of Chinese knotter technology,and the mathematical model is helpful for designing new type of knotters.LI Hui HE Jin WANG Qingjie LI Hongwen RASAILY Rabi Gautam CAO Qingchun ZHANG Xiangcai 2014Chinese Journal of Mechanical Engineering2014,27,1:5
13Association of NFKB1 gene polymorphism (rs28362491) with levels of inflammatory biomarkers and susceptibility to diabetic nephropathy in Asian Indians显示文摘AIM To investigate the association of NFKB1 gene-94 ATTG insertion/deletion(rs28362491) polymorphism with inflammatory markers and risk of diabetic nephropathy in Asian Indians.METHODS A total of 300 subjects were recruited(100 each), normoglycemic,(NG); type 2 diabetes mellitus(T2DM) without any complications(DM) and T2 DM with diabetic nephropathy [DM-chronic renal disease(CRD)]. Analysis was carried out by polymerase chain reaction-restriction fragment length polymorphism and ELISA. Pearson's correlation, analysis of variance and logistic regression wereused for statistical analysis.RESULTS The allelic frequencies of-94 ATTG insertion/deletion were 0.655/0.345(NG), 0.62/0.38(DM) and 0.775/0.225(DM-CRD). 14Garcinol sensitizes human head and neck carcinoma to cisplatin in a xenograft mouse model despite downregulation of proliferative biomarkers显示文摘OBJECTIVE Platinum compounds such as cisplatin and carboplatin are frequently used as the first-line chemotherapy for the treatment of the head and neck squamous cell carcinoma(HNSCC).In the present study,we investigated whether garcinol,apolyisoprenylated benzophenone can chemosensitize HNSCC to cisplatin.METHODS The effect of garcinol and cisplatin on HNSCC was assessed by MTT,Western blotting,real time PCR,FACS,immunohistochemistry,DNA binding assay and xenograft mouse model.RESULTS We found that garcinol inhibited the viability of a panel of diverse HNSCC cell lines,enhanced the apoptotic effect of cisplatin,suppressed constitutive as well as cisplatin-induced NF-κB activation,and downregulated the expression of various oncogenic gene products(cyclin D1,Bcl-2,survivin and VEGF).In vivo study showed that administration of garcinol alone(0.5 mg·kg-1,ip five times/week)significantly suppressed the growth of the tumor,and this effect was further increased by cisplatin.Both the markers of proliferation index(Ki-67)and microvessel density(CD31)were downregulated in tumor tissues by the combination of cisplatin and garcinol.The pharmacokinetic results of garcinol indicated that good systemic exposure was achievable after ip administration of garcinol at 0.5and 2mg·kg-1 with mean peak concentration(cmax)of 1825.4 and 6635.7nmol·L-1 in the mouse serum,respectively.CONCLUSION Overall,our results suggest that garcinol can indeed potentiate the effects of cisplatin by negative regulation of various inflammatory and proliferative biomarkers.Feng Li Muthu K. 15Impact of gout on in-hospital outcomes of acute coronary syndrome-related hospitalizations and revascularizations: Insights from the national inpatient sample显示文摘BACKGROUND Previous studies have established a role of gout in predicting risk and prognosis of cardiovascular diseases. However, large-scale data on the impact of gout on inpatient outcomes of acute coronary syndrome (ACS)-related hospitalizations and post-revascularization is inadequate. AIM To evaluate the impact of gout on in-hospital outcomes of ACS hospitalizations, subsequent healthcare burden and predictors of post-revascularization inpatient mortality. METHODS We used the national inpatient sample (2010-2014) to identify the ACS and goutrelated hospitalizations, relevant comorbidities, revascularization and postrevascularization outcomes using the ICD-9 CM codes. A multivariable analysis was performed to evaluate the predictors of post-revascularization in-hospital mortality. RESULTS We identified 3144744 ACS-related hospitalizations, of which 105198 (3.35%) also had gout. The ACS-gout cohort were more often older white males with a higher prevalence of comorbidities. Coronary artery bypass grafting was required more often in the ACS-gout cohort. Post-revascularization complications including cardiac (3.2% vs 2.9%), respiratory (3.5% vs 2.9%), and hemorrhage (3.1% vs 2.7%) were higher whereas all-cause mortality was lower (2.2% vs 3.0%) in the ACSgout cohort (P < 0.001). An older age (OR 15.63, CI: 5.51-44.39), non-elective admissions (OR 2.00, CI: 1.44-2.79), lower household income (OR 1.44, CI: 1.17- 1.78), and comorbid conditions predicted higher mortality in ACS-gout cohort undergoing revascularization (P < 0.001). Odds of post-revascularization inhospital mortality were lower in Hispanics (OR 0.45, CI: 0.31-0.67) and Asians (OR 0.65, CI: 0.45-0.94) as compared to white (P < 0.001). However, postoperative complications significantly raised mortality odds. Mean length of stay, transfer to other facilities, and hospital charges were higher in the ACS-gout cohort. 17Prophylactic Effect of Gossypin Against Percutaneously Administered Sulfur Mustard显示文摘Objective To evaluate the protective efficacy of gossypin (3,3',4',5,7,8-hexahydroxyflavone 8-glucoside) by administering it intraperitoneally, for dose, time, and vehicle dependent effects against sulphur mustard (SM), administered through percutaneous route in mice. Methods SM (diluted in PEG-300) was administered percutaneously. The protective efficacy of gossypin was evaluated by administering it intraperitoneally (50, 100, 200, and 400 mg/kg), in various vehicles (water, PEG-300 and DMSO), and time intervals (30 min prior, simultaneous and 2 h post). The time dependent protection of gossypin (200 mg/kg in PEG-300; i.p.) was also evaluated using selected biochemical variables (GSH, GSSG, MDA, total antioxidant status, Hb, WBC count, RBC count, glutathione peroxidase, glutathione reductase, and superoxide dismutase) and liver histology. The protection of gossypin by oral route was also evaluated against percutaneously administered SM. Results The protection against systemic toxicity of SM (LD50 8.1 mg/kg) was better when gossypin was given with PEG-300 (8.0 folds) than DMSO (5.7 folds). No protection was observed when gossypin was administered with water. Good protection (8.0 folds) was observed when gossypin was administered (200 mg/kg in PEG-300; i.p.) at 30 min prior or simultaneous to SM exposure, but no protection was observed when gossypin was administered 2 h post to SM exposure. A significant weight loss was observed 7 days after SM administration (2 LD50), with a significant increase in RBC and Hb. A significant decrease in total antioxidant status of plasma, liver GSH and GSSG levels, and in the activities of glutathione peroxidase, glutathione reductase and superoxide dismutase was also observed 7 days after SM administration. SM treated mouse liver also showed necrosis. A significant protection was observed when gossypin (200 mg/kg in PEG-300; i.p.) was administered either as a pretreatment (30 min before) or simultaneous treatment, and not as a post treatment (2 h). The protective efficacy of gossypin was better through oral route when administered with DMSO (4.8 folds) than with PEG-300 (2.4 folds). No protection was observed when gossypin was administered orally with water.
