2A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 3Genome analysis of Taraxacum kok-saghyz Rodin provides new insights into rubber biosynthesis显示文摘The Russian dandelion Taraxacum kok-saghyz Rodin(TKS), a member of the Composite family and a potential alternative source of natural rubber(NR) and inulin, is an ideal model system for studying rubber biosynthesis. Here we present the draft genome of TKS, the first assembled NR-producing weed plant. The draft TKS genome assembly has a length of 1.29 Gb, containing 46 731 predicted protein-coding genes and68.56% repeats, in which the LTR-RT elements predominantly contribute to the genome enlargement. We analyzed the heterozygous regions/genes, suggesting its possible involvement in inbreeding depression.Through comparative studies between rubber-producing and non-rubber-producing plants, we found that enzymes of the mevalonate(MVA) pathway and rubber elongation might be critical for rubber biosynthesis, and several key isoforms have been isolated and shown to be predominantly expressed in the latex, indicating their crucial functions in rubber biosynthesis. Moreover, for two important families in rubber elongation, the CPT/CPTL and REF/SRPP families, diverse evolutionary tracks have been revealed. 4Natural history of covert hepatic encephalopathy: An observational study of 366 cirrhotic patients显示文摘AIM To explore the natural history of covert hepatic encephalopathy(CHE) in absence of medication intervention.METHODS Consecutive outpatient cirrhotic patients in a Chinese tertiary care hospital were enrolled and evaluated for CHE diagnosis. They were followed up for a mean of 11.2 ± 1.3 mo. Time to the first cirrhosis-related complications requiring hospitalization, including overt HE(OHE), resolution of CHE and death/transplantation, were compared between CHE and no-CHE patients. Predictors for complication(s) and death/transplantation were also analyzed.RESULTS A total of 366 patients(age: 47.2 ± 8.6 years, male: 73.0%) were enrolled. CHE was identified in 131 patients(35.8%). CHE patients had higher rates of death and incidence of complications requiring hospitalization, including OHE, compared to unimpaired patients. Moreover, 17.6% of CHE patients developed OHE, 42.0% suffered persistent CHE, and 19.8% of CHE spontaneously resolved. In CHE patients, serum albumin < 30 g/L(HR = 5.22, P = 0.03) was the sole predictor for developing OHE, and blood creatinine > 133 μmol/L(HR = 4.75, P = 0.036) predicted mortality. Child-Pugh B/C(HR = 0.084, P < 0.001) and OHE history(HR = 0.15, P = 0.014) were predictors of spontaneous resolution of CHE.CONCLUSION CHE exacerbates, persists or resolves without medication intervention in clinically stable cirrhosis. 6Simulated Microgravity Conditions and Carbon Ion Irradiation Induce Spermatogenic Cell Apoptosis and Sperm DNA Damage显示文摘Objective To investigate the effect of simulated microgravity and carbon ion irradiation(CIR) on spermatogenic cell apoptosis and sperm DNA damage to the testis of male Swiss Webster mice,and assess the risk associated with space environment.Methods Sperm DNA damage indicated by DNA fragmentation index(DFI) and high DNA stainability(HDS) was measured by sperm chromatin structure assay(SCSA).Apoptosis of spermatogenic cells was detected by annexin V-propidium iodide assay.Bax(the expression levels of p53) and proliferating cell nuclear antigen(PCNA) were measured by immunoblotting;p53 and PCNA were located by immunohistology.Results HDS,DFI,apoptosis index,and the expression levels of p53 and Bax were detected to be significantly higher in the experimental groups(P<0.05) compared with those in the control group;however,the PCNA expression varied to a certain degree.p53-and PCNA-positive expression were detected in each group,mainly in relation to the spermatogonic cells and spermatocytes.Conclusion The findings of the present study demonstrated that simulated microgravity and CIR can induce spermatogenic cell apoptosis and sperm DNA damage.Sperm DNA damage may be one of the underlying mechanisms behind male fertility decline under space environment.These findings may provide a scientific basis for protecting astronauts and space traveler's health and safety.LI Hong Yan ZHANG Hong MIAO Guo Ying XIE Yi SUN Chao DI Cui Xia LIU Yang LIU Yuan Yuan ZHANG Xin MA Xiao Fei XU Shuai GAN Lu ZHOU Xin 2013Biomedical and Environmental Sciences2013,26,9:10

9THF_1 mutations lead to increased basal and wound-induced levels of oxylipins that stimulate anthocyanin biosynthesis via COI_1 signaling in Arabidopsis显示文摘Mutants defective in chloroplast development or photosynthesis are liable to accumulate higher levels of anthocyanin in photo-oxidative stress.However,regulatory mechanisms of anthocyanin biosynthesis in the mutants remain unclear.Here,we investigated the mechanism by which the deletion of thylakoid formation1(THF1) leads to an increased level of anthocyanin in Arabidopsis thaliana L.Physiological and genetic evidence showed that the increased level of anthocyanin in thf1 is dependent on coronatine-insensitive1(COM)signaling.Our data showed that thf1 had higher levels of basalα-linolenic acid(α-LeA),and methyl jasmonate(JA)-induced α-LeA and 12-oxophytodienoic acid(OPDA) than the wild type(WT).Consistently,expression levels of phospholipase genes including pPLAIIα and PLA-Iγ1 were elevated in thf1.Furthermore,inhibition of lipase activity by bromoenol lactone,a specific inhibitor of plant pPLA,led to producing identical levels of anthocyanins in WT and thf1 plants.Interestingly,OPDA biosynthesis was triggered by light illumination in isolated chloroplasts,indicating that new protein import into chloroplasts is not required for OPDA biosynthesis.Thus,we conclude that the elevated anthocyanin accumulation in thf1 is attributed to an increase in JA levels.This JA-mediated signaling to coordinate plant metabolism and growth in stress may be conserved in other photosensitive mutants.Yi Gan Hong Li Ye Xie Wenjuan Wu Maoyin Li Xuemin Wang Jirong Huang 2014Journal of Integrative Plant Biology2014,56,9:8
10Measurement of boiling heat transfer coefficient in liquid nitrogen bath by inverse heat conduction method显示文摘Inverse heat conduction method (IHCM) is one of the most effective approaches to obtaining the boiling heat transfer coefficient from measured results. This paper focuses on its application in cryogenic boiling heat transfer. Experiments were conducted on the heat transfer of a stainless steel block in a liquid nitrogen bath, with the assumption of a 1D conduction condition to realize fast acquisition of the temperature of the test points inside the block. With the inverse-heat conduction theory and the explicit finite difference model, a solving program was developed to calculate the heat flux and the boiling heat transfer coefficient of a stainless steel block in liquid nitrogen bath based on the temperature acquisition data. Considering the oscillating data and some unsmooth transition points in the inverse-heat-conduction calculation result of the heat-transfer coefficient, a two-step data-fitting procedure was proposed to obtain the expression for the boiling heat transfer coefficients. The coefficient was then verified for accuracy by a comparison between the simulation results using this expression and the verifying experimental results of a stainless steel block. 12Metabolomics mechanism of traditional soy sauce associated with fermentation time显示文摘Given that fermentation time has significant impact on quality and flavor of soy sauce,a global understanding of the metabolic mechanism as fermentation time prolonged is essential for producing satisfactory and consistent quality of traditional soy sauce.Herein,the metabolic compounds changes of soy sauce associated with fermentation time up to 10 years were comprehensively investigated by using Chinese traditional fermented soy sauce(CTFSS)as a demonstration.Results showed that formaldehyde nitrogen,total soluble nitrogen(TSN),non-salt soluble solids,amino acids,free 5’-nucleotides and volatile compounds in CTFSS changed obviously during fermentation.Specifically,glutamic acid and aspartic acid were prominent in CTFSS.Continuous decrease in content of hypoxanthine(Hx)was found from 1 M(1-month soy sauce)to7 M(7-months soy sauce).Furthermore,a significant opposite tendency for changes between some volatile compounds and amino acids was indicated that there was a transformation between these two components.Therefore,a better understanding to the influence of fermentation time on soy sauce had been proposed.As the formation and conversion of amino acids and sugars might be mainly responsible for flavor formation in CTFSS,the ratio of these two reactions rate led the metabolism to be divided into three steps,degradation,conversion and balance.Li Zhu Siyu He Ying Lu Jianhong Gan Ningping Tao Xichang Wang Zaoli Jiang Yuanxiang Hong Changhua Xu 2022Food Science and Human Wellness2022,11,2:7
13Tetrandrine citrate eliminates imatinib-resistant chronic myeloid leukemia cells in vitro and in vivo by inhibiting Bcr-Abl/β-catenin axis显示文摘Objective:To evaluate the effects of tetrandrine citrate, a novel tetrandrine salt with high water solubility, on the growth of imatinib (IM)-resistant chronic myeloid leukemia (CML) in vitro and in vivo, and reveal action molecular mechanisms. Methods:Cell viability in vitro was measured using methyl thiazolyl tetrazolium (MTT) assay. CML cell growth in vivo was assessed using a xenograft model in nude mice. Bcr-Abl and β-catenin protein levels were determined using Western blotting. Bcr-Abl messenger RNA (mRNA) was measured by reverse transcription polymerase chain reaction (RT-PCR). Flow cytometry (FCM) was used to determine cell cycle status. Results:Tetrandrine citrate inhibited the growth of IM-resistant K562 cells, primary leukemia cells, and primitive CD34 + leukemia cells, and their inhibition concentration that inhibited 50% of target cells (IC 50 ) ranged from 1.20 to 2.97 μg/ml. In contrast, tetrandrine citrate did not affect normal blood cells under the same conditions, and IC 50 values were about 10.12-13.11 μg/ml. Oral administration of tetrandrine citrate caused complete regression of IM-resistant K562 xeno-grafts in nude mice without overt toxicity. Western blot results revealed that treatment of IM-resistant K562 cells with tetrandrine citrate resulted in a significant decrease of both p210 Bcr-Abl and β-catenin proteins, but IM did not affect the Bcr-Abl protein levels. Proteasome inhibitor, MG132, did not prevent tetrandrine-mediated decrease of the p210 Bcr-Abl protein. RT-PCR results showed that tetrandrine treatment caused a decrease of Bcr-Abl mRNA. FCM analysis indicated that tetrandrine induced gap 1 (G 1 ) arrest in CML cells. Conclusions:Tetrandrine citrate is a novel orally active tetrandrine salt with potent anti-tumor activity against IM-resistant K562 cells and CML cells. 14The Lyman-alpha Solar Telescope(LST) for the ASO-S mission——Ⅲ. data and potential diagnostics显示文摘The Lyman-alpha Solar Telescope(LST)is one of the three payloads onboard the Advanced Space-based Solar Observatory(ASO-S)mission.It aims at imaging the Sun from the disk center up to 2.5 R⊙targeting solar eruptions,particularly coronal mass ejections(CMEs),solar flares,prominences/filaments and related phenomena,as well as the fast and slow solar wind.The most prominent speciality of LST is the simultaneous observation of the solar atmosphere in both Lyαand white light(WL)with high temporospatial resolution both on the solar disk and the inner corona.New observations in the Lyαline together with traditional WL observations will provide us with many new insights into solar eruptions and solar wind.LST consists of a Solar Corona Imager(SCI)with a field of view(FOV)of 1.1–2.5 R⊙,a Solar Disk Imager(SDI)and a full-disk White-light Solar Telescope(WST)with an identical FOV up to 1.2 R⊙.SCI has a dual waveband in Lyα(121.6±10 nm)and in WL(700±40 nm),while SDI works in the Lyαwaveband of 121.6±7.5 nm and WST works in the violet narrow-band continuum of 360±2.0 nm.To produce high quality science data,careful ground and in-flight calibrations are required.We present our methods for different calibrations including dark field correction,flat field correction,radiometry,instrumental polarization and optical geometry.Based on the data calibration,definitions of the data levels and processing procedures for the defined levels from raw data are described.Plasma physical diagnostics offer key ingredients to understand ejecta and plasma flows in the inner corona,as well as different features on the solar disk including flares,filaments,etc.Therefore,we are making efforts to develop various tools to detect the different features observed by LST,and then to derive their physical parameters,for example,the electron density and temperature of CMEs,the outflow velocity of the solar wind,and the hydrogen density and mass flows of prominences.Coordinated observations and data analyses with the coronagraphs onboard Solar Orbiter,PROBA-3,and Aditya are also briefly discussed.Li Feng Hui Li Bo Chen Ying Li Roberto Susino Yu Huang Lei Lu Bei-Li Ying Jing-Wei Li Jian-Chao Xue Yu-Tong Yang Jie Hong Jian-Ping Li Jie Zhao Wei-Qun Gan Yan Zhang 2019Research in Astronomy and Astrophysics2019,19,11:7
15High-strength wrought magnesium alloy with dense nano-scale spherical precipitate显示文摘This paper reported the influences of Yb addition on the precipitate and mechanical properties of wrought magnesium alloy ZK60. The ingots of ZK60-1.78Yb (wt%, 0.26 at%) alloys were cast using permanent mould and extruded at 370℃ . By means of TEM and HRTEM, it was observed that Yb affected the precipitate and precipitation of ZK60-1.78Yb alloys significantly. Dynamic precipitation occurred in the as-extruded alloy and spherical nano-scale precipitate with high density and homoge-neity exhibited in the aged alloys. The precipitate particles were about 5-20 nm in diameter, 10-30 nm in average space length. 16Necessity and feasibility for an ET-based modern water resources management strategy: A case study of soil water resources in the Yellow River Basin显示文摘The necessity and feasibility of an ET-based modern water resources management was analyzed to improve assessment of critical water resources scarcity in the region/basin. This analysis was based on the whole water cycle process and its analysis object is evapotranspiration (ET), a main consumption component in the water resources dynamic transformation process. A case study was undertaken by selecting soil water resources in the Yellow River Basin and employing the WEP-L distributed hydrological model with physics mechanisms. This paper discusses the amount and consumption efficiency of soil-water resources according to completely simulated results of water cycle elements throughout the basin. Results indicate that it is important for the ET-based modern water resources management strategy to alleviate water resources scarcity because it may not only avoid unused water wasting but also improve water use efficiency.
