| 1 | A high stable pH-temperature dual-sensitive liposome for tuning anticancer drug release显示文摘In order to improve the targeting and availability of liposomes to cancer cells,the temperature sensitivity of 1,2-Dipalmitoyl-sn-glycero-3-phosphocholine(DPPC)and the pH sensitivity of PASP in PASP-g-C8 are incorporated in a drug delivery system.A composite pH-temperature dual-sensitive liposomes(CPTLPs)was obtained as an efficient drug delivery system.The bionic bilayer is self-assembled by cholesterol/cationic temperature-sensitive lipids as base layer and pH-sensitive octylamine grafted poly aspartic acid(PASP-g-C8)as anchors coated outside.Cytarabine(CYT)was chosen as a model drug.SEM and DLS were used to observe the morphology characteristics of CPTLPs in different micro environment.The results demonstrated that the CPTLPs remained active in both normal(pH7.4 and 37°C)and tumor tissues(pH 5.0 and 42°C).As a stable colloidal system,the zeta potential of CPTSLs was−41.6 mV.In vitro drug-release experiments,the CTY encapsulated dual-sensitive liposomes,CPTSLs(+),not only have significant pH-temperature sensitivity but have more prolonged release in vitro than control groups.MTT tests results indicated that the cell apoptotic effects induced by CPTSLs(+)were nearly 30%higher than the naked drug CTY in HepG2 cells,and 20%lower apoptotic in vero cells.The CPTSLs(+)sustained a stable emulsion form,less toxic effects on normal cells,and exhibited a good pH-temperature sensitivity,thus expected to be a promising tumor targeting drug delivery. | Yilin Zhao Fuli Cai Xiangyi Shen Haijia Su | 2020 | Synthetic and Systems Biotechnology2020,5,2: | 2 |
| 2 | RNA-binding protein QKI regulates contact inhibition via Yes-associate protein in ccRCC显示文摘Contact inhibition adjusts organ size to the proper size and ensures the cultured cells growing to a monolayer.By regulating the downstream coordinator YAP,the evolutionarily conserved Hippo transduction pathway attunes cell growth and death in response to cell contact inhibition,polarity,self-renewal,and differentiation.Dysregulation of this pathway is involved in various diseases such as cancer.RNA-binding protein QKI regulates cell proliferation,metabolism,division,and immunity in various cancer models,but its role in cancer cell contact inhibition remains unclear.In this study,we aimed to clarify the relationship between QKI and YAP,and the role of their interaction in cell contact inhibition.We found a lower QKI expression level in sparse condition,whereas a higher expression level in confluent condition by western blot analysis and immunofluorescence assay.QKI knockdown elevated cell proliferation and invasion both in vitro and in vivo.Strikingly,the results of CCK-8 assay,colony formation assay,and transwell assay showed that the phenomenon was in accord with the expression level of pYAP and reverse with YAP.Higher levels of Wnt3a and β-catenin were also found in xenografts of QKI-knockdown clear cell renal cell carcinoma (ccRCC) CAKI-1 cells by western blot analysis and immumohistochemical staining.Finally,a positive correlation between QKI and pYAP was found in clinical specimens by immunohistochemistry.Thus,as a negative regulator of YAP,QKI attuned the cell contact inhibition,leading to inhibition of cancer cell proliferation and invasion through Wnt and GPCR pathway. | Zheng Zhu Di Wei Xi'an Li Fuli Wang Fei Yan Zibao Xing Zhao Yan Huanyu Lu Dongsheng Zhai Ziehen Ye Geng Zhang Ping Meng Yu Zheng Jiarui Yuan Zifan Lu Jianlin Yuan | 2019 | Acta Biochimica et Biophysica Sinica2019,51,1: | 2 |

| 8 | Dominant changes in the breast muscle lipid profiles of broiler chickens with wooden breast syndrome revealed by lipidomics analyses显示文摘Background: Chicken is the most consumed meat worldwide and the industry has been facing challenging myopathies. Wooden breast(WB), which is often accompanied by white striping(WS), is a serious myopathy adversely affecting meat quality of breast muscles. The underlying lipid metabolic mechanism of WB affected broilers is not fully understood.Results: A total of 150 chickens of a white-feathered, fast-growing pure line were raised and used for the selection of WB, WB + WS and control chickens. The lipids of the breast muscle, liver, and serum from different chickens were extracted and measured using ultra performance liquid chromatography(UPLC) plus Q-Exactive Orbitrap tandem mass spectrometry. In the breast, 560 lipid molecules were identified. Compared to controls, 225/225 of 560 lipid molecules(40.2%) were identified with differential abundance(DA), including 92/100 significantly increased neutral lipids and 107/98 decreased phospholipids in the WB/WB + WS groups, respectively. The content of monounsaturated fatty acids(MUFA) was significantly higher, and the polyunsaturated fatty acids(PUFA) and saturated fatty acids(SFA) were significantly lower in the affected breasts. In the liver, 434 lipid molecules were identified, and 39/61 DA lipid molecules(6.7%/14.1%) were detected in the WB and WB + WS groups, respectively. In the serum, a total of 529 lipid molecules were identified and 4/44 DA lipid molecules(0.8%/8.3%) were detected in WB and WB + WS group, respectively. Compared to controls, the content of MUFAs in the serum and breast of the WB + WS group were both significantly increased, and the content of SFAs in two tissues were both significantly decreased. Only five lipid molecules were consistently increased in both liver and serum in WB + WS group.Conclusions: We have found for the first time that the dominant lipid profile alterations occurred in the affected breast muscle. The relative abundance of 40.2% of lipid molecules were changed and is characteristic of increased neutral lipids and decreased phospholipids in the affected breasts. Minor changes of lipid profiles in the liver and serum of the affected groups were founded.
