| 1 | Establishment and evaluation of a peanut association panel and analysis of key nutritional traits显示文摘Breeding programs aim to improve the yield and quality of peanut(Arachis hypogaea L.); using association mapping to identify genetic markers linked to these quantitative traits could facilitate selection efficiency. A peanut association panel was established consisting of 268 lines with extensive phenotypic and genetic variation, meeting the requirements for association analysis. These lines were grown over 3 years and the key agronomic traits, including protein and oil content were examined. Population structure(Q) analysis showed two subpopulations and clustering analysis was consistent with Q-based membership assignment and closely related to botanical type. Relative Kinship(K) indicated that most of the panel members have no or weak familial relatedness, with 52.78% of lines showing K ? 0. Linkagedisequilibrium(LD) analysis showed a high level of LD occurs in the panel. Model comparisons indicated false positives can be effectively controlled by taking Q and K into consideration and more false positives were generated by K than Q. A preliminary association analysis using a Q t K model found markers significantly associated with oil, protein, oleic acid, and linoleic acid, and identified a set of alleles with positive and negative effects. | 2 | Carbon,nitrogen and phosphorus stoichiometry in Pinus tabulaeformis forest ecosystems in warm temperate Shanxi Province,north China显示文摘Although carbon(C), nitrogen(N), and phosphorous(P) stoichiometric ratios are considered good indicators of nutrient excess/limitation and thus of ecosystem health, few reports have discussed the trends and the reciprocal effects of C:N:P stoichiometry in plant–litter–soil systems. The present study analyzed C:N:P ratios in four age groups of Chinese pine, Pinus tabulaeformis Carr., forests in Shanxi Province, China: plantation young forests(AY,<20 year-old); plantation middle-aged forests(AM, 21–30 year-old); natural young forests(NY,<30 year-old); and natural middle-aged forests(NM,31–50 year-old). The average C:N:P ratios calculated for tree, shrub, and herbaceous leaves, litter, and soil(0–100 cm) were generally higher in NY followed by NM,AM, and AY. C:N and C:P ratios were higher in litter than in leaves and soils, and reached higher values in the litter and leaves of young forests than in middle-aged forests;however, C:N and C:P ratios were higher in soils of middle-aged forests than in young forests. N:P ratios were higher in leaves than in litter and soils regardless of stand age; the consistent N:P<14 values found in all forests indicated N limitations. With plant leaves, C:P ratios were highest in trees, followed by herbs and shrubs, indicating a higher efficiency in tree leaf formation. C:N ratios decreased with increasing soil depth, whereas there was no trend for C:P and N:P ratios. C:N:P stoichiometry of forest foliage did not exhibit a consistent variation according to stand age. | 4 | Molecular analysis of the chloroplast Cu/Zn-SOD gene(AhCSD2) in peanut显示文摘Superoxide dismutase(SOD, EC 1.15.1.1) plays a key role in response to drought stress, and differences in SOD activity changes among cultivars are important under drought conditions. We obtained the full-length DNA of the chloroplast Cu/Zn-SOD gene(Ah CSD2)from 11 allotetraploid cultivars and 5 diploid wild species in peanut. BLAST search against the peanut genome showed that the Ah CSD2 genes g CSD2-1 and g CSD2-2 are located at the tops of chromosome A03(A genome) and B03(B genome), respectively, and both contain 8exons and 7 introns. Nucleotide sequence analyses indicated that g CSD2-2 sequences were identical among all the tested cultivars, while g CSD2-1 sequences showed allelic variations.The amino acid sequences deduced from g CSD2-1 and g CSD2-2 both contain a chloroplast transit peptide and are distinguished by 6 amino acid(aa) residue differences. The other 2aa residue variations in the mature peptide regions give rise to three-dimensional structure changes of the protein deduced from the genes g CSD2-1 and g CSD2-2. Sequences analyses of cultivars and wild species showed that g CSD2-2 of Arachis hypogaea and g Aip CSD2(Arachis ipaensis) are identical, and despite the abundant polymorphic loci between g CSD2-1 of A.hypogaea and sequences from A genome wild species, the deduced amino acid sequence of Ah CSD2-1(A. hypogaea) is identical to that of Adu CSD2(Arachis duranensis), whereas Aco CSD2(Arachis correntina) and Aca CSD2(Arachis cardenasii) both have 2 aa differences in the transit peptide region compared with Ah CSD2-1(A. hypogaea). Based on the Peanut Genome Project, promoter prediction revealed many stress-related cis-acting elements within the potential promoter regions(pp-A and pp-B). pp-A contains more binding sites for drought-associated transcriptional factors than pp-B. | 5 | Erosive Wear and Wear Mechanism of in situ TiC_P/Fe Composites显示文摘The base structure of in situ TiCp/Fe composites fabricated under industrial condition was changed by differentheat treatments. Erosive wear tests were carried out and the results were compared with that of wear-resistant whitecast iron. The results suggest that the wear resistance of the in situ TiCP/Fe composite is higher than that ofwear-resistant white cast iron under the sand erosive wear condition. The wear mechanism of the wear-resistantwhite cast iron was a cycle process that base surface was worn and carbides were exposed, then carbides was brokenand wear pits appeared. While the wear mechanism of in situ TiCP/Fe composite was a cycle process that basesurface was worn and TiC grains were exposed and dropped. The wear resistance of in situ TiCP/Fe composite waslower than that of wear-resistant white cast iron under the slurry erosive wear condition. | 6 | Effects of Exogenous Nitric Oxide and 24-Epibrassinolide on the Physiological Characteristics of Peanut Seedlings Under Cadmium Stress显示文摘Cadmium(Cd) is highly toxic to plants, animals, and humans. Limited information is available on the role of nitric oxide(NO)and/or 24-epibrassinolide(EBR) in response of plants to Cd stress. In this study, a hydroponic experiment was performed to investigate the effects of NO and/or EBR on peanut plants subjected to Cd stress(200 μmol L^(-1)) with sodium nitroprusside(SNP, an exogenous NO donor)(250 μmol L^(-1)) and/or EBR(0.1 μmol L^(-1)) addition. The results showed that Cd exposure inhibited plant growth, and this stress was alleviated by exogenous NO or EBR, and especially the combination of the two. Treatment with Cd inhibited the growth of peanut seedlings, decreased chlorophyll content, and significantly increased the Cd concentration in plants. Furthermore, the concentration of reactive oxygen species(ROS) markedly increased in peanut seedlings under Cd stress, resulting in the accumulation of malondialdehyde(MDA) and proline in leaves and roots. Under Cd stress, applications of SNP, EBR, and especially the two in combination significantly reduced the translocation of Cd from roots to leaves, increased the chlorophyll content, decreased the concentrations of ROS, MDA, and proline, and significantly enhanced the activities of superoxide dismutase(SOD), peroxidase(POD), and catalase(CAT) in peanut seedlings. Exogenous NO and/or EBR also stimulated the activities of nitrate reductase(NR)and nitric oxide synthase(NOS) and increased the contents of antioxidants, such as ascorbic acid(AsA) and reduced glutathione(GSH). Furthermore, exogenous NO and/or EBR enhanced Cd accumulation in the cell wall and thus decreased Cd distribution in the organelles in the roots. The concentrations of calcium(Ca), iron(Fe), magnesium(Mg), and zinc(Zn) were also regulated by exogenous NO or EBR, and especially by the two in combination. | 17 | Preparation, optimization, and characterization of chitosancoated solid lipid nanoparticles for ocular drug delivery显示文摘The present study aimed to develop and optimize chitosan coated solid lipid nanoparticles(chitosan-SLNs)encapsulated with methazolamide. Chitosan-SLNs were successfully prepared by a modified oil-in-water emulsification-solvent evaporation method with glyceryl monostearate as the solid lipid and phospholipid as the surfactant. Systematic screening of formulation factors was carried out. The optimized formula for preparation was screened by orthogonal design as well as Box-Behnken design with entrapment efficiency, particle size and zeta potential as the indexes. The entrapment efficiency of the optimized formulation(methazolamide-chitosan-SLNs)prepared was(58.5± 4.5)%,Particle size(247.7 ± 17.3) nm and zeta potential(33.5 ±3.9) mV. Transmission electron microscopy showed homogeneous spherical particles in the nanometer range. A prolonged methazolamide in vitro release profile was obtained in the optimized chitosan-SLNs suspension compared with methazolamide solution. No ocular damages were observed in the susceptibility test on albino rabbits.
