1The effect of carbon nanotubes on osteogenic functions of adipose-derived mesenchymal stem cells in vitro and bone formation in vivo compared with that of nano-hydroxyapatite and the possible mechanism显示文摘It has been well recognized that the development and use of artificial materials with high osteogenic ability is one of the most promising means to replace bone grafting that has exhibited various negative effects.The biomimetic features and unique physiochemical properties of nanomaterials play important roles in stimulating cellular functions and guiding tissue regeneration.But efficacy degree of some nanomaterials to promote specific tissue formation is still not clear.We hereby comparatively studied the osteogenic ability of our treated multiwalled carbon nanotubes(MCNTs)and the main inorganic mineral component of natural bone,nano-hydroxyapatite(nHA)in the same system,and tried to tell the related mechanism.In vitro culture of human adiposederived mesenchymal stem cells(HASCs)on the MCNTs and nHA demonstrated that although there was no significant difference in the cell adhesion amount between on the MCNTs and nHA,the cell attachment strength and proliferation on the MCNTs were better.Most importantly,the MCNTs could induce osteogenic differentiation of the HASCs better than the nHA,the possible mechanism of which was found to be that the MCNTs could activate Notch involved signaling pathways by concentrating more proteins,including specific bone-inducing ones.Moreover,the MCNTs could induce ectopic bone formation in vivo while the nHA could not,which might be because MCNTs could stimulate inducible cells in tissues to form inductive bone better than nHA by concentrating more proteins including specific bone-inducing ones secreted from M2 macrophages.Therefore,MCNTs might be more effective materials for accelerating bone formation even than nHA.Zhipo Du Xinxing Feng Guangxiu Cao Zhending She Rongwei Tan Katerina E.Aifantis Ruihong Zhang Xiaoming Li 2021Bioactive Materials2021,6,2:3

6LAMC2 regulates proliferation, migration, and invasion mediated by the Pl3K/AKT/mTOR pathway in oral显示文摘Background:Oral squamous cell carcinoma(OSCC)is a common malignant tumor.Recently,Laminin Gamma 2(LAMC2)has been shown to be abnormally expressed in OSCC;however,how LAMC2 signaling contributes to the occurrence and development of OSCC and the role of autophagy in OSCC has not been fully explored.This study aimed to analyze the role and mechanism of LAMC2 signaling in OSCC and the involvement of autophagy in OSCC.Methods:To explore the mechanism by which LAMC2 is highly expressed in OSCC,we used small interfering RNA(siRNA)to knock down LAMC2 to further observe the changes in the signaling pathway.Furthermore,we used cell proliferation assays,Transwell invasion assays,and wound-healing assays to observe the changes in OSCC proliferation,invasion,and metastasis.RFP-LC3 was used to detect the level of autophagy intensity.A cell line-derived xenograft(CDX)model was used to detect the effect of LAMC2 on tumor growth in vivo.Results:This study found that the level of autophagy was correlated with the biological behavior of OSCC.The downregulation of LAMC2 activated autophagy and inhibited OSCC proliferation,invasion,and metastasis via inhibiting the PI3K/AKT/mTOR pathway.Moreover,autophagy has a dual effect on OSCC,and the synergistic downregulation of LAMC2 and autophagy can inhibit OSCC metastasis,invasion,and proliferation via the PI3K/AKT/mTOR pathway.Conclusions:LAMC2 interacts with autophagy to regulate OSCC metastasis,invasion,and proliferation via the PI3K/AKT/mTOR pathway.LAMC2 down-regulation can synergistically modulate autophagy to inhibit OSCC migration,invasion,and proliferation.FAYU SHAN LANLAN LIANG CHONG FENG HONGBAO XU ZIROU WANG WEILI LIU LINGLING PU ZHAOLI CHEN GANG CHEN XINXING WANG 2023Oncology Research2023,31,4:1

14High-lying excited states in ^10Be from the ^9Be(^9Be,^10Be)^8Be reaction显示文摘A transfer-reaction experiment of ~9Be(~9Be,^(10)Be)~8Be was performed at a beam energy of 45 Me V.Excited states in ^(10)Be up to 18.80 Me V are produced using missing mass and invariant mass methods.Most of the observed high-lying resonant states,reconstructed from theα+~6He and t+~7Li decay channels,agree with the previously reported results.In addition,two new resonances at 15.6 and 18.8 Me V are identified from the present measurement.The 18.55 Me V state is found to decay into both the t + ~7Lig:s: and t + ~7Li?(0.478 MeV) channels, with a relative branching ratio of 0:93 ± 0:33. 17MFAP2 promotes the progression of oral squamous cell carcinoma by activating the Wnt/β-catenin signaling pathway through autophagy显示文摘Microfibrillar-associated protein 2(MFAP2)is a small glycoprotein that is involved in vascular development and metabolic disease.The present study aims to explore the regulatory role of MFAP2 in the development and progression of oral squamous cell carcinoma(OSCC),including the underlying mechanisms.MFAP2 expression and its association with the progression of OSCC are explored using bioinformatics.MFAP2 expression in OSCC tissues is detected by immunohistochemical staining.SCC15 cell migration,invasion,apoptosis,proliferation,and viability are detected by wound healing,Transwell,flow cytometry,colony formation,and cell counting kit-8 assays.An in vivo experiment is used to detect tumor formation.Western blot analysis is used to determine MFAP2’s regulatory role in autophagy and the Wnt/β-catenin signaling pathway.MFAP2 is highly expressed in SCC15 cells and OSCC tissues,which correlates positively with the poor prognosis of patients with OSCCs.Functionally,MFAP2 promotes oncogenic autophagy to increase cell invasion,migration,and proliferation but inhibits apoptosis in SCC15 cells and promotes tumor growth in vivo.Mechanistically,MFAP2 upregulates autophagy and Wnt/β-catenin signaling to stimulate OSCC development.Intriguingly,regulation of Wnt/β-catenin signaling dependent on autophagy contributes to the malignant behaviors of SCC15 cells.MFAP2 could serve as a novel biomarker for OSCC and could affect OSCC tumorigenesis and development via autophagic regulation of Wnt/β-catenin signaling.Hao Zhang Si Shen Chong Feng Gang Chen Xinxing Wang 2023Acta Biochimica et Biophysica Sinica2023,55,9:0
