2ALK,the Key Gene for Gelatinization Temperature,is a Modifier Gene for Gel Consistency in Rice显示文摘Gelatinization temperature(GT) is an important parameter in evaluating the cooking and eating quality of rice.Indeed,the phenotype,biochemistry and inheritance of GT have been widely studied in recent times.Previous map-based cloning revealed that GT was controlled by ALK gene,which encodes a putative soluble starch synthase II-3.Complementation vector and RNAi vector were constructed and transformed into Nipponbare mediated by Agrobacterium.Phenotypic and molecular analyses of transgenic lines provided direct evidence for ALK as a key gene for GT.Meanwhile,amylose content,gel consistency and pasting properties were also affected in transgenic lines.Two of four nonsynonymous single nucleotide polymorphisms in coding sequence of ALK were identified as essential for GT.Based on the single nucleotide polymorphisms(SNPs),two new sets of SNP markers combined with one cleaved amplified polymorphic sequence marker were developed for application in rice quality breeding.Zhenyu Gao Dali Zeng Fangmin Cheng Zhixi Tian Longbiao Guo Yan Su Meixian Yan Hua Jiang Guojun Dong Yuchen Huang Bin Han Jiayang Li Qian Qian 2011Journal of Integrative Plant Biology2011,53,9:13

4Deep Reinforcement Learning Based Joint Edge Resource Management in Maritime Network显示文摘Due to the rapid development of the maritime networks, there has been a growing demand for computation-intensive applications which have various energy consumption, transmission bandwidth and computing latency requirements. Mobile edge computing(MEC) can efficiently minimize computational latency by offloading computation tasks by the terrestrial access network. In this work, we introduce a space-air-ground-sea integrated network architecture with edge and cloud computing components to provide flexible hybrid computing service for maritime service. In the integrated network, satellites and unmanned aerial vehicles(UAVs) provide the users with edge computing services and network access. Based on the architecture, the joint communication and computation resource allocation problem is modelled as a complex decision process, and a deep reinforcement learning based solution is designed to solve the complex optimization problem. 5An efficient way to enhance the total nitrogen removal efficiency of the Anammox process by S^0-based short-cut autotrophic denitrification显示文摘In order to reduce the amount of NO_3^-–N generated by the Anammox process, and alleviate the competition between denitrification and Anammox for NO_2^-–N in a single reactor, the preference of S^0 for reacting with coexisting NO_2^-–N and NO_3^-–N in the sulfur autotrophic denitrifying(SADN) process and the coupling effect of short-cut SADN and the Anammox process were studied. The results showed that S^0 preferentially reacted with NO_3^-to produce NO_2^-–N, and then reacted with NO_2^-–N when NO_3^-–N was insufficient, which could effectively alleviate the competition between SADN bacteria(SADNB) and Anammox bacteria(An AOB) for NO_2^-–N. After 170 days of operation, coupling between short-cut S^0-SADN and the Anammox process was first successfully achieved. SADNB converted the NO_3^-–N generated by the Anammox process into NO_2^-–N, which was once again available to An AOB. The total nitrogen removal efficiency eventually stabilized at over 95%, and the effluent NO_3^-–N was controlled within 10 mg/L, when high NH_4^+–N wastewater was treated by the Anammox process. 7Partial cooperative spectrum sensing schedule in cognitive network显示文摘Cognitive radio has emerged as a promising technology for maximizing the utilization efficiency of the limited spectrum resources while accommodating the increasing amount of services and applications in wireless networks. One of the most important and critical components of the cognitive radio is spectrum sensing and accordingly, detection of primary users. Considering the hardware constraints existing in cognitive devices, based on the coarse estimation of channel occupancy, partial cooperative spectrum sensing with adaptive spectrum schedule scheme is proposed to increase the possibility to discover more spectrum opportunities promptly. Simulation results show the gain of sensing performance and the energy-saving feature of partial spectrum sensing. Special security scheme is designed to protect the reliability of sensing result from the false message attack. 8Sugar Starvation Enhances Leaf Senescence and Genes Involved in Sugar Signaling Pathways Regulate Early Leaf Senescence in Mutant Rice显示文摘To clarify the complex regulatory relationship between changes in sugar content and leaf senescence during the grain-filling stage of rice,genotype-dependent differences in sugar content and the temporal transcriptional patterns of genes involved in sugar signaling pathways were determined in mutant rice exhibiting early leaf senescence and its wild type Zhefu 142.The effects of exogenous glucose or sucrose on the senescence of detached leaves under dark conditions were also investigated.Chlorophyll,soluble sugar,sucrose and fructose contents decreased,whereas electrolytic leakage and malondialdehyde levels increased in mutant leaves at the grain-filling stage.These results suggested that sugar starvation is positively correlated with the early leaf senescence of mutant plants.Detached leaf segments incubated in exogenous sugar solutions under dark conditions exhibited delayed senescence.The high expression of Hxk1 in leaves of mutant plants at the initial grain-filling stage suggested that Hxk1 is involved in the hexose-sensing process at the early stage of leaf senescence.The low expression levels of Hxk2 and Frk1 in the senescing leaves of mutant rice during the grain-filling stage are indicative of weakened hexose phosphorylation.In addition,the high expression levels of Su Sy1,Su Sy2 and Su Sy4 in leaves of mutant plants at the initial grain-filling stage are accompanied by the high transcript levels of SUT1,which favor sucrose translocation and remobilization from the early senescing leaves of mutant rice.The relatively reduced transcript levels of ch FBP,cy FBP,SPS1,SPS2 and SPS6 indicated that during the grain-filling stage,sucrose biosynthesis is weakened in the senescing leaves of mutant rice.LI Zhaowei ZHAO Qian CHENG Fangmin 2020Rice science2020,27,3:4
9Bispecific prodrug nanoparticles circumventing multiple immune resistance mechanisms for promoting cancer immunotherapy显示文摘Cancer immunotherapy is impaired by the intrinsic and adaptive immune resistance.Herein,a bispecific prodrug nanoparticle was engineered for circumventing immune evasion of the tumor cells by targeting multiple immune resistance mechanisms.A disulfide bond-linked bispecific prodrug of NLG919 and JQ1(namely NJ) was synthesized and self-assembled into a prodrug nanoparticle,which was subsequently coated with a photosensitizer-modified and tumor acidity-activatable diblock copolymer PHP for tumor-specific delivery of NJ.Upon tumor accumulation via passive tumor targeting,the polymeric shell was detached for facilitating intracellular uptake of the bispecific prodrug.NJ was then activated inside the tumor cells for releasing JQ1 and NLG919 via glutathione-mediated cleavage of the disulfide bond.JQ1 is a bromodomain-containing protein 4 inhibitor for abolishing interferon gamma-triggered expression of programmed death ligand 1.In contrast,NLG919 suppresses indoleamine-2,3-dioxygenase 1-mediated tryptophan consumption in the tumor microenvironment,which thus restores robust antitumor immune responses.Photodynamic therapy(PDT) was performed to elicit antitumor immunogenicity by triggering immunogenic cell death of the tumor cells.The combination of PDT and the bispecific prodrug nanoparticle might represent a novel strategy for blockading multiple immune evasion pathways and improving cancer immunotherapy.Jiayi Ye Bo Hou Fangmin Chen Shunan Zhang Muya Xiong Tianliang Li Yechun Xu Zhiai Xu Haijun Yu 2022Acta Pharmaceutica Sinica B2022,12,6:4
10Simultaneous determination of free methamphetamine,pethidine,ketamine and tramadol in urine by dispersive liquid–liquid microextraction combined with GC–MS显示文摘A simple and rapid dispersive liquid–liquid microextraction(DLLME)technique coupled with gas chromatography–ion trap mass spectrometry(GC–MS)was developed for the extraction and analysis of methamphetamine(MA),pethidine(PD),ketamine(KT)and tramadol(TD)from human urine.In this study,different parameters affecting the extraction process such as the type and volume of extraction solvent,type and volume of disperser solvent,extraction time and pH value and salt effect were studied and optimized.Under optimized conditions,the enrichment factor ranged from 185 to 226 and the average recovery ranged from 80.45%to 95.55%.The linear range was 10.0–1000.0 mg/L,the limit of detection and quantitation were in the range 0.43–1.96 mg/L and 1.44–6.53 mg/L,respectively.The relative standard deviations were in the range 1.98%–3.90%(n=7).The obtained results show that DLLME combined with GC–MS is a fast and simple method for the determination of MA,PD,KT and TD in human urine.Fangmin Xu Lingyun Liu 2019Forensic Sciences Research2019,4,2:3

13Dual-targeting prodrug nanotheranostics for NIR-Ⅱfluorescence imaging-guided photo-immunotherapy of glioblastoma显示文摘Glioblastoma(GBM) therapy is severely impaired by the blood-brain barrier(BBB) and invasive tumor growth in the central nervous system.To improve GBM therapy,we herein presented a dual-targeting nanotheranostic for second near-infrared(NIR-Ⅱ) fluorescence imaging-guided photoimmunotherapy.Firstly,a NIR-Ⅱ fluorophore MRP bearing donor-acceptor-donor(D-A-D) backbone was synthesized.Then,the prodrug nanotheranostics were prepared by self-assembling MRP with a prodrug of JQ1(JPC) and T7 ligand-modified PEG5k-DSPE.T7 can cross the BBB for tumor-targeted delivery of JPC and MRP.JQ1 could be restored from JPC at the tumor site for suppressing interferon gamma-inducible programmed death ligand 1 expression in the tumor cells.MRP could generate NIR-Ⅱ fluorescence to navigate 808 nm laser,induce a photothermal effect to trigger in-situ antigen release at the tumor site,and ultimately elicit antitumor immunogenicity.Photo-immunotherapy with JPC and MRP dual-loaded nanoparticles remarkably inhibited GBM tumor growth in vivo.The dual-targeting nanotheranostic might represent a novel nanoplatform for precise photo-immunotherapy of GBM.Fenglin Li Yi Lai Jiayi Ye Madiha Saeed Yijing Dang Zhifeng Zou Fangmin Chen Wen Zhang Zhiai Xu 2022Acta Pharmaceutica Sinica B2022,12,9:2
14Software defined industrial network architecture for edge computing offloading显示文摘The integration of the Internet and the traditional manufacturing industry makes the industrial Internet of things(IIoT) as a popular research topic. However, traditional industrial networks continue to face challenges of resource management and limited raw data storage and computation capacity. A novel software defined industrial network(SDIN) architecture was proposed to address the existing drawbacks in IIoT such as resource utilization, data processing and storage, and system compatibility. The architecture is developed based on the software defined network(SDN) architecture, combining hierarchical cloud and fog computing and content-aware caching technologies. Based on the SDIN architecture, two types of edge computing strategies in industrial applications are discussed. Different scenarios and service requirements are considered. 15Suppression of starch synthase I(SSI) by RNA interference alters starch biosynthesis and amylopectin chain distribution in rice plants subjected to high temperature显示文摘Based on known cDNAs of rice starch synthase isoforms,we constructed dsRNA interference vectors for starch synthase I(SSI)to produce transgenic plants containing starch with a moderately high amylose content.We investigated the effect of SSI suppression on grain quality traits,starch biosynthesis,and amylopectin chain distribution in rice plants exposed to two different temperature regimes.The activities and transcripts of BEs,DBEs,and other SS isoforms were further investigated to clarify the effect of SSI suppression on these key enzymes and their specific isoforms under different temperature treatments.Suppression of SSI by RNAi altered grain starch component and amylopectin chain distribution,but it exerted only a slight effect on total starch content(%)and accumulation amount(mg kernel?1)and on starch granule morphology and particle size distribution.Under normal temperature(NT),insignificant differences in kernel weight,chalky kernel proportion,chalky degree,and starch granule morphology between SSI-RNAi line and its wild type(WT)were observed.However,amylose content(AC)level and granule-bound starch synthase(GBSS)activity in rice endosperms were markedly increased by SSI-RNAi suppression.The chalky kernel proportion and chalky degree of SSIRNAi lines were significantly higher than those of WT under high temperature(HT)exposure at filling stage.Inhibition of SSI by RNAi affected amylopectin chain distribution and raised starch gelatinization temperature(GT)in two ways:directly from the SSI deficiency itself and indirectly by reducing BEIIb amounts in an SSI-deficient background.The deficiency of SSI expression led to an alteration in the susceptibility of grain chalkiness occurrence and starch gelatinization temperature to HT exposure,owing to a pleiotropic effect of SSI deficiency on the expression of other genes associated with starch biosynthesis.Qian Zhao Xiaoxia Du Zhanyu Han Yu Ye Gang Pan Muhammad-Asad-Ullah Asad Qifa Zhou Fangmin Cheng 2019The Crop Journal2019,7,5:2
