1Clinical and biochemical indexes from 2019-nCoV infected patients linked to viral loads and lung injury显示文摘The outbreak of the 2019-nCoV infection began in December 2019 in Wuhan,Hubei province,and rapidly spread to many provinces in China as well as other countries.Here we report the epidemiological,clinical,laboratory,and radiological characteristics,as well as potential biomarkers for predicting disease severity in 2019-nCoV-infected patients in Shenzhen,China.All 12 cases of the 2019-nCoV-infected patients developed pneumonia and half of them developed acute respiratory distress syndrome (ARDS).The most common laboratory abnormalities were hypoalbuminemia,lymphopenia,decreased percentage of lymphocytes (LYM) and neutrophils (NEU),elevated C-reactive protein (CRP) and lactate dehydrogenase (LDH),and decreased CD8 count.The viral load of 2019-nCoV detected from patient respiratory tracts was positively linked to lung disease severity.ALB,LYM,LYM (%),LDH,NEU (%),and CRP were highly correlated to the acute lung injury.Age,viral load,lung injury score,and blood biochemistry indexes,albumin (ALB),CRP,LDH,LYM (%),LYM,and NEU (%),may be predictors of disease severity.Moreover,the Angiotensin Ⅱlevel in the plasma sample from 2019-nCoV infected patients was markedly elevated and linearly associated to viral load and lung injury.Our results suggest a number of potential diagnosis biomarkers and angiotensin receptor blocker (ARB) drugs for potential repurposing treatment of 2019-nCoV infection.Yingxia Liu Yang Yang Cong Zhang Fengming Huang Fuxiang Wang Jing Yuan Zhaoqin Wang Jinxiu Li Jianming Li Cheng Feng Zheng Zhang Lifei Wang Ling Peng Li Chen Yuhao Qin Dandan Zhao Shuguang Tan Lu Yin Jun Xu Congzhao Zhou Chengyu Jiang Lei Liu 2020Science China(Life Sciences)2020,63,3:157
2Protein trans-splicing based dual-vector delivery of the coagulation factor Ⅷ gene显示文摘A dual-vector system was explored for the delivery of the coagulation factor VIII gene,using intein-mediated protein trans-splicing as a means to produce intact functional factor VIII post-translationally.A pair of eukaryotic expression vectors,expressing Ssp DnaB intein-fused heavy and light chain genes of B-domain deleted factor VIII (BDD-FVIII),was constructed.With transient co-transfection of the two vectors into 293 and COS-7 cells,the culture supernatants contained (137±23) and (109±22) ng mL–1 spliced BDD-FVIII antigen with an activity of (1.05±0.16) and (0.79±0.23) IU mL–1 for 293 and COS-7 cells,respectively.The spliced BDD-FVIII was also detected in supernatants from a mixture of cells transfected with inteinfused heavy and light chain genes.The spliced BDD-FVIII protein bands from cell lysates were visualized by Western blotting.The data demonstrated that intein could be used to transfer the split factor VIII gene and provided valuable information on factor VIII gene delivery by dual-adeno-associated virus in hemophilia A gene therapy.ZHU FuXiang,LIU ZeLong,CHI XiaoYan & QU HuiGe Life Science College of Ludong University,Yantai 264025,China 2010Science China(Life Sciences)2010,53,6:27

5Total pollution effect of urban surface runoff显示文摘For pollution research with regard to urban surface runoff,most sampling strategies to date have focused on differences in land usage. With single land-use sampling,total surface runoffpollution effect cannot be evaluated unless every land usage spot is monitored. Through a new sampling strategy known as mixed stormwater sampling for a street community at discharge outlet adjacent to river,this study assessed the total urban surface runoffpollution effect caused by a variety of land uses and the pollutants washed off from the rain pipe system in the Futian River watershed in Shenzhen City of China.The water quality monitoring indices were COD(chemical oxygen demand),TSS(total suspend solid),TP(total phosphorus),TN(total nitrogen)and BOD(biochemical oxygen demand).The sums of total pollution loads discharged into the river for the four indices of COD,TSS,TN,and TP over all seven rainfall events were very different.The mathematical model for simulating total pollution loads was established from discharge outlet mixed stormwater sampling of total pollution loads on the basis of four parameters:rainfall intensity,total land area,impervious land area,and pervious land area.In order to treat surface runoffpollution,the values of MFF30(mass first flush ratio)and FF30(first 30%of runoffvolume) can be considered as split-flow control criteria to obtain more effective and economical design of structural BMPs(best management practices)facilities.LUO Hongbing LUO Lin HUANG Gu LIU Ping LI Jingxian HU Sheng WANG Fuxiang XU Rui HUANG Xiaoxue 2009Journal of Environmental Sciences2009,21,9:13
6Analysis for retrieval and validation results of FY-3 Total Ozone Unit (TOU)显示文摘Retrieval experiment was made for global total column ozone using the first year measurements of Total Ozone Unit (TOU) on board the second generation polar orbiting meteorological satellite of China, FY-3/A. The retrieval results were analyzed and validated by comparison with AURA/OMI, Meteop/GOME-2 global ozone products and ground-based ozone measurement data. The qualititative comparisons over the globe especially over Antarctica and the Tibetan Plateau show that the spatial and temporal distribution characteristics are consistent with OMI and GOME-2 products. The quantitative comparisons with ground-based measurements and AURA/OMI ozone product were made over 74 stations, the TOU total ozone retrieval has a 3% rms relative error compared with AURA/OMI ozone product and 4.2% rms relative error with ground-based measurements. The maximum difference between satellite retrieval and ground-based measurements was found in the Antarctica ozone hole. 7FY-3 satellite Ultraviolet Total Ozone Unit显示文摘FY-3 satellites are Chinese second-generation polar orbit meteorological satellite series. Ultraviolet Total Ozone Unit (TOU) is one of the main payloads on FY-3 satellite and the first instrument for daily global coverage of total ozone monitoring in China. The main purpose of TOU is to measure the Earth backscatter ultraviolet radiation for retrieving daily global map of atmospheric ozone. TOU will provide the important parameters for environmental monitoring, climate forecasting and global climate changing research. At present, the in-orbit testing of TOU has accomplished and won a consummation, and it will be delivered to National Satellite Meteorological Center for the operational phase of the project. In this paper we introduce the instrument of TOU and its measuring principle, in general. We also analyze the recent working status of the instrument, including the sensitivity, measuring precision of solar irradiance, diffuser degradation and wavelength drift. The inversion results show that TOU can provide good global ozone maps, and a comparison with the OMI total ozone product shows that their RMS deviation is about 5%. 8Vertical ozone profiles deduced from measurements of SBUS on FY-3 satellite显示文摘The FY-3 satellite is a second-generation Chinese polar-orbit meteorological satellite.The Solar Backscatter Ultraviolet Sounder (SBUS),one of the main payloads on the FY-3 satellite,is the first Chinese ozone-monitoring instrument on a meteorological satellite.As part of the in-orbit validation of FY-3,we carried out a retrieval trial using measurements from SBUS during the period of 17-30 July,2008,and compared those data with measurements and retrieved profiles from the National Oceanic and Atmospheric Administration (NOAA) satellite SBUV/2.The results show that the precision of the measurements and retrieved profiles are quite good.The averaged relative difference percentages of the ozone profiles retrieved from SBUS and those from SBUV/2 are within ±7%.HUANG FuXiang LIU NianQing ZHAO MingXian WANG ShuRong HUANG Yu 2010Chinese Science Bulletin2010,55,10:9
9The effect of a secretion-enhanced heavy chain on improving intein-based dual-vector co-delivery of a full-length factor Ⅷ gene显示文摘Treatment of hemophilia A by gene therapy is adversely affected by inefficient FVIII secretion and the large FVIII gene,which is difficult to package in the promising adeno-associated virus (AAV) vectors.Inhibited secretion of FVIII is caused mainly by inefficient secretion of its heavy chain.Previously,we have employed a protein splicing-based dual-vector to co-transfer a B-domain-deleted FVIII (BDD-FVIII) gene,suggesting that the light chain,covalently ligated to a co-expressed heavy chain can improve the secretion of spliced BDD-FVIII.However,its level of secretion was affected by inefficient secretion the heavy chain.Here,we studied the effect of a mutant heavy chain with L303E/F309S substitutions,which enhance FVIII secretion on the heavy chain itself and spliced FVIII when using a protein splicing-based split-delivery of a full-length FVIII gene.Eukaryotic vectors expressing Ssp DnaB intein-fused mutant heavy and light chains were transiently co-transfected into cultured COS-7 cells.A spliced FVIII protein was seen in co-transfected cells by Western blot analysis.The heavy chain was secreted by cells transfected with the mutant heavy chain gene alone at (39±11) ng/mL and this secretion increased to (123±13) ng/mL when cells were co-transfected with the light chain gene,which was greater than the secretion of wild-type heavy chain.The amount of spliced FVIII in the culture supernatant of co-transfected cells was (86±14) ng/mL,with an activity of (0.61±0.08) IU/mL,which was greater than that of wild-type FVIII co-transfected cells.Spliced FVIII and bioactivity were also detected in the combined culture supernatant of cells individually transfected with mutant heavy and light chain gene at a higher level than that of combined wild-type heavy and light chain transfections.This suggested that the heavy chain with improved secretion markedly increased the efficacy of protein splicing-based split delivery of the full-length FVIII gene using a dual-vector.These results encourage the transfer of this technology to an animal model using a dual-AAV vector.ZHU FuXiang YANG ShuDe LIU ZeLong MIAO Jing QU HuiGe CHI XiaoYan 2011Chinese Science Bulletin2011,56,2:8
10Monitoring of the 2011 spring low ozone events in the Arctic region显示文摘Data from FY-3B SBUS and NOAA SBUV/2 were used to monitor ozone levels in the Arctic region from March 1 to April 5, 2011. Results revealed a significant ozone depletion in the area, with total ozone levels between 200-250 DU. The ozone levels recorded were 100-200 DU below normal, and in some parts the levels were as low as 200 DU, indicating a mini ozone-hole. During the sampling period, the ozone depletion area underwent identifiable expansion and detraction, a rotation around the North Pole from the west to the east, and a longitudinal movement from the Pole, spreading to the mid latitudes. The effects of these rare low ozone events were not only felt in the Arctic, but also extended to densely populated areas between Europe and the middle of Russia. In this region, rapidly increasing levels of ultraviolet radiation were detected at the Earth's surface. 11Monitoring on subsidence due to repeated excavation with DInSAR technology显示文摘DInSAR technology was used to monitor subsidence caused by underground coal mining activities in mountainous area, with multi source SAR data, including 8 EnviSAT C-band and 4 ALOS L-band, and 4 programmed TerraSAR-X dataset. The results revealed that 2-pass DInSAR technique sometimes failed to retrieve the mining-caused subsidence due to spatial and/or temporal de-correlation. We also noticed that there existed residual topographic phase after the compensation with SRTM DEM, which could almost overwhelm the subsidence information when the perpendicular baseline was relatively large. Based on the mining materials, analysis was made on the shape of subsidence area. For the well geocoded results from TerraSAR-X, confirmed by GPS surveying results of corner reflectors, we tried to extract the advance distance of influence besides the subsidence area. Due to the big deformation gradient over stopingfaces, the X-band SAR data could not capture the maximum value subsidence revealed by GPS survey in our preliminary results, the same as C-band EnviSAT data. 13Elevated plasma levels of selective cytokines in COVID-19 patients reflect viral load and lung injury显示文摘A recent outbreak of pneumonia in Wuhan,China was found to be caused by a 2019 novel coronavirus(2019-nCoV or SARS-CoV-2 or HCoV-19).We previously reported the clinical features of 12 patients with2019-n Co V infections in Shenzhen,China.To further understand the pathogenesis of COVID-19 and find better ways to monitor and treat the disease caused by 2019-n Co V,we measured the levels of 48 cytokines in the blood plasma of those 12 COVID-19 patients.Thirty-eight out of the 48 measured cytokines in the plasma of 2019-n Co V-infected patients were significantly elevated compared to healthy individuals.Seventeen cytokines were linked to 2019-nCoV loads.Fifteen cytokines,namely M-CSF,IL-10,IFN-α2,IL-17,IL-4,IP-10,IL-7,IL-1 ra,G-CSF,IL-12,IFN-γ,IL-1α,IL-2,HGF and PDGF-BB,were strongly associated with the lung-injury Murray score and could be used to predict the disease severity of2019-n Co V infections by calculating the area under the curve of the receiver-operating characteristics.Our results suggest that 2019-nCoV infections trigger extensive changes in a wide array of cytokines,some of which could be potential biomarkers of disease severity of 2019-n Co V infections.These findings will likely improve our understanding of the immunopathologic mechanisms of this emerging disease.Our results also suggest that modulators of cytokine responses may play a therapeutic role in combating the disease once the functions of these elevated cytokines have been characterized.Yingxia Liu Cong Zhang Fengming Huang Yang Yang Fuxiang Wang Jing Yuan Zheng Zhang Yuhao Qin Xiaoyun Li Dandan Zhao Shunwang Li Shuguang Tan Zhaoqin Wang Jinxiu Li Chenguang Shen Jianming Li Ling Peng Weibo Wu Mengli Cao Li Xing Zhixiang Xu Li Chen Congzhao Zhou William J Liu Lei Liu Chengyu Jiang 2020National Science Review2020,7,6:6

15Study on the Metallogenetism of Sub-mantle Plume and Mantle Branches in the Gold Mineralization Concentration Area of Northwest Jiaodong Peninsula显示文摘The northwest of Jiaodong peninsula is characterized by its greatest gold concentration area, enormous gold reserves and limited time interval(115±5Ma) of gold mineralization in China. Studies show that the east area of north China began its mantle plume evolution since the Yanshanian movement, and the Laiyang sub-mantle plume and its surrounding mantle branches such as those in Guojiadian, Aishan and Panshidian were formed in the mineralization concentration area of northwestern Jiaodong peninsula. With the development of mantle plume → sub-mantle plume → mantle branch → favorable expending structures, the deep-derived hydrothermal fluid moved up to the favorable places, such as brittle-ductile or ductile-brittle shearing zones, the internal and external contact zones of intrusions, dense fracturing zones and the contact zones between dikes and countryrocks, and concentrated into deposits. Then the gold mineralization concentration zones that are centralized on mantle branches were formed. 16Combustion synthesis and magnetic investigation of nanosized CoFe_2O_4显示文摘Nanosized Co spinel ferrite was prepared by a combustion method. The grain size of resultant ferrite can becontrolled by the reaction conditions. Magnetic investigation revealed that the products with an average grain size of 4 nm behave as superparamagnetic materials and have zero coercive force; whereas the samples with an average grain size of 85 nm (near the size of critical single domain) have the maximal coercive force. 17The Effect of Netting Solidity Ratio and Inclined Angle on the Hydrodynamic Characteristics of Knotless Polyethylene Netting显示文摘Knotless polyethylene(PE) netting has been widely used in aquaculture cages and fishing gears, especially in Japan. In this study, the hydrodynamic coefficient of six knotless PE netting panels with different solidity ratios were assessed in a flume tank under various attack angles of netting from 0?(parallel to flow) to 90?(perpendicular to flow) and current speeds from 40 cm s^(-1) to 130 cm s^(-1). It was found that the drag coefficient was related to Reynolds number, solidity ratio and attack angle of netting. The solidity ratio was positively related with drag coefficient for netting panel perpendicular to flow, whereas when setting the netting panel parallel to the flow the opposite result was obtained. For netting panels placed at an angle to the flow, the lift coefficient reached the maximum at an attack angle of 50? and then decreased as the attack angle further increased. The solidity ratio had a dual influence on drag coefficient of inclined netting panels. 19Kinetics for hydrogen production by methanol steam reforming in fluidized bed reactor显示文摘Hydrogen is one of the best energy carriers.Fluidized bed reactor provides a promising approach for hydrogen production. To describe the hydrogen generating rate with methanol steam reforming in fluidized bed reactor quantitatively, dual-rate kinetic models of the reactions with exponent form were developed, including that of steam reforming reaction(SR) and decomposition reaction(DE).The reaction rate per unit mass of catalyst was related to partial pressures of components. The exponentials in kinetic equations were obtained by linear least-squares method based on the experimental data.
