| 2 | Quantifying Responses of Winter Wheat Physiological Processes to Soil Water Stress for Use in Growth Simulation Modeling显示文摘A deep understanding of crop-water eco-physiological relations is the basis for quantifying plant physiological responses to soil water stress. Pot experiments were conducted to investigate the winter wheat crop-water relations under both drought and waterlogging conditions in two sequential growing seasons from 2000 to 2002, and then the data were used to develop and validate models simulating the responses of winter wheat growth to drought and waterlogging stress. The experiment consisted of four treatments, waterlogging (keep 1 to 2 cm water layer depth above soil surface), control (70%-80% field capacity), light drought (40%-50% field capacity) and severe drought (30%-40% field capacity) with six replicates at five stages in the 2000-2001 growth season. Three soil water content treatments (waterlogging, control and drought) with two replicates were designed in the 2001-2002 growth season. Waterlogging and control treatments are the same as in the 2000-2001 growth season. For the drought treatment, no water was supplied and the soil moisture decreased from field capacity to wilting point. Leaf net photosynthetic rate, transpiration rate, predawn leaf water potential, soil water potential, soil water content and dry matter weight of individual organs were measured. Based on crop-water eco-physiological relations, drought and waterlogging stress factors for winter wheat growth simulation model were put forward. Drought stress factors integrated soil water availability, the sensitivity of different development stages and the difference between physiological processes (such as photosynthesis, transpiration and partitioning). The quantification of waterlogging stress factor considered different crop species, soil water status, waterlogging days and sensitivity at different growth stages. | 3 | Alcoholic liver disease:Utility of animal models显示文摘Alcoholic liver disease(ALD) is a major cause of acute and chronic liver injury. Extensive evidence has been accumulated on the pathological process of ALD during the past decades. However, effective treatment options for ALD are very limited due to the lack of suitable in vivo models that recapitulate the full spectrum of ALD. Experimental animal models of ALD, particularly rodents, have been used extensively to mimic human ALD. An ideal animal model should recapitulate all aspects of the ALD process, including significant steatosis, hepatic neutrophil infiltration, and liver injury. A better strategy against ALD depends on clear diagnostic biomarkers, accurate predictor(s) of its progression and new therapeutic approaches to modulate stop or even reverse the disease. Numerous models employing rodent animals have been established in the last decades to investigate the effects of acute and chronic alcohol exposure on the initiation and progression of ALD. Although significant progress has been made in gaining better knowledge on the mechanisms and pathology of ALD, many features of ALD are unknown, and require further investigation, ideally with improved animal models that more effectively mimic human ALD. | 5 | Molecular Epidemiological Analysis of Echovirus 19 Isolated From an Outbreak Associated With Hand, Foot, and Mouth Disease (HFMD) in Shandong Province of China显示文摘Objective To elucidate the genetic characterization and molecular epidemiological features of Echovirus 19(E19)isolates collected from an outbreak associated with hand,foot and mouth disease(HFMD)in Tai'an city of Shandong Province of China from July to September,2003.Methods Thirty seven Echovirus 19 isolates were isolated from stool specimens and throat swabs collected during the outbreak,then major capsid(VP1)genomic sequence was determined,and phylogenetic tree was done based on the VP1 sequences among these 37 and other E19 viruses deposited in the Genbank.Also a representative strain named CHN-SD03-TN12 was selected for sequencing of 5'-untranslated regions(5'-UTR).Results The identity rate was about 98.9%-100% among all these 37 E19 viruses.The genetic relationships between these 37 E19 isolates and other strains reported were also depicted.The identity rate was about 78.4%-78.9% compared with E19 reference strain Burke.The substitutions in the sequence of 5'-UTR resulted in changes in the conjectural properties of 5'-UTR of E19 viruses.Conclusion The genetic features of E19 viruses isolated during the outbreak in Shandong Province in 2003 may be associated with a genetic and antigenic drift that changes the virulence of the Shandong isolates,but the molecular changes in Shandong E19 viruses contributing to their phenotype remain to be further illuminated.However,the sequences described in this paper substantiate the changes taken place in capsid VP1 and 5'UTR regions.These substitutions may contribute to their tropism and virulence,and play a significant role in pathogenesis and clinical manifestations of the disease. | ZHEN ZHU WEN-BO XU AI-QIANG XU HAI-YAN WANG YONG ZHANG LI-ZHI SONG Hui-LiYANG YANLI FENGJI | 2007 | Biomedical and Environmental Sciences2007,20,4: | 22 |
| 6 | In vitro differentiation of adipose-derived stem cells and bone marrow-derived stromal stem cells into neuronal-like cells显示文摘Adipose-derived stem cells and bone marrow-derived stromal stem cells were co-cultured with untreated or Aβ1-40-treated PC12 cells, or grown in supernatant derived from untreated or Aβ1-40-treated PC12 cells. Analysis by western blot and quantitative real-time PCR showed that protein levels of Nanog, Oct4, and Sox2, and mRNA levels of miR/125a/3p were decreased, while expression of insulin-like growth factor-2 and neuron specific enolase was increased. In comparison, the generation of neuron specific enolase-positive cells was most successful when adipose-derived stem cells were co-cultured with Aβ1-40-treated PC12 cells. Our results demonstrate that adipose-derived stem cells and bone marrow-derived stromal stem cells exhibit trends of neuronal-like cell differentiation after co-culture with Aβ1-40-treated PC12 cells. | 7 | Relationship between matrix metalloproteinase-2 mRNA expression and clinicopathological and urokinase-type plasminogen activator system parameters and prognosis in human gastric cancer显示文摘AIM: To investigate the relationship between matrix metalloproteinase-2 (MMP-2) mRNA expression and clinicopathologic and urokinase-type plasminogen activator (uPA) system parameter and prognosis in human gastric cancer.METHODS: Expression of MMP-2 mRNA, uPA, and uPA-R mRNA in tumor tissues and ≥5 cm adjacent normal tissues from 67 cases of gastric cancer was studied using RTPCR and Northern blot respectively. Survival analyses were done using the Kaplan-Meier method. RESULTS: The expression rates of MMP-2 mRNA, uPA and uPA-R mRNA in tumor tissues (31%, 41%, and 51%, respectively) were significantly higher than those in ≥5 cm adjacent tissues (19%, 11%, and 9%; χ2 = 4.59, 43.58, and 53.24 respectively, P<0.05, 0.0001, and 0.0001, respectively). Expression of MMP-2 mRNA was significantly correlated with lymph node metastasis (metastasis:61.9%, no metastasis: 39.1%, χ2= 7.61, P<0.05), Lauren's classification of diffuse/mixed types: 54.2%, intestinal type: 26.3%, χ2 = 4.25, P<0.05, expression of uPA and uPA-R mRNA (uPA+: 55.1%, uPA-: 22.2% and uPA-R+: 54.9%, uPA-R-: 18.8%, x2= 5.72 and 6.40 respectively, P<0.05). Kaplan-Meier survival analysis of MMP-2 mRNA expression did not show significant difference in all 67 cases, but revealed an association of the expression of MMP-2 mRNA, uPA, and uPA-R mRNA with worse prognosis (P = 0.0083, 0.0160, and 0.0094, respectively). | 12 | Clinical characterization and diagnosis of cystic fibrosis throughexome sequencing in Chinese infants with Bartter-syndrome-like hypokalemia alkalosis显示文摘Cystic fibrosis (CF) is a fatal autosomal-recessive disease caused by mutations in the CFtransmembrane conductance regulator (CFTR) gene. CF is characterized by recurrent pulmonary infectionwith obstructive pulmonary disease. CF is common in the Caucasian population but is rare in the Chinesepopulation. The symptoms of early-stage CF are often untypical and may sometimes manifest as Bartter syndrome(BS)-like hypokalemic alkalosis. Therefore, the ability of doctors to differentiate CF from BS-like hypokalemicalkalosis in Chinese infants is a great challenge in the timely and accurate diagnosis of CF. In China, sporadic CFhas not been diagnosed in children younger than three years of age to date. Three infants, who were initiallyadmitted to our hospital over the period of June 2013 to September 2014 with BS-like hypokalemic alkalosis, werediagnosed with CF through exome sequencing and sweat chloride measurement. The compound heterozygousmutations of the CFTR gene were detected in two infants, and a homozygous missense mutation was found in oneinfant. Among the six identified mutations, two are novel point mutations (c.1526G 〉 C and c.3062C 〉 T) that arepossibly pathogenic. The three infants are the youngest Chinese patients to have been diagnosed with sporadic CFat a very early stage. Follow-up examination showed that all of the cases remained symptom-free after earlyintervention, indicating the potential benefit of very early diagnosis and timely intervention in children with CF.Our results demonstrate the necessity of distinguishing CF from BS in Chinese infants with hypokalemic alkalosisand the significant diagnostic value of powerful exome sequencing for rare genetic diseases. | 13 | Epidermal growth factor improves intestinal morphology by stimulating proliferation and differentiation of enterocytes and mTOR signaling pathway in weaning piglets显示文摘Epidermal growth factor(EGF) has been shown to improve piglet intestinal morphology and epithelial recovery. In an attempt to further understand the mechanisms behind these improvements, this study tested the hypothesis that dietary EGF may affect intestinal morphology by stimulating the proliferation and differentiation of enterocytes in weaning piglets. In piglets receiving200 μg kg^-1 EGF, crypt depth and villus height increased(P<0.05). Adding 400 μg kg^-1 EGF increased villus height-to-crypt depth ratio(P<0.05), but reduced crypt depth(P<0.05). Dietary supplementation with 200 μg kg^-1 EGF significantly increased the number of Ki67-positive cells(P<0.01) and tended to increase the mRNA level of proliferating cell nuclear antigen(P<0.10).However, this supplementation decreased the expression level of intestinal fatty acid-binding protein(P<0.05). Piglets fed with400 μg kg^-1 EGF had an increased mRNA level of intestinal alkaline phosphatase(P<0.05). The phosphorylation of m TOR(mammalian target of rapamycin) was observed in the 200 μg kg^-1 EGF group. | 16 | Miniature ultrasonic probes for diagnosis and treatment of digestive tract diseases显示文摘AIM: To investigate the clinical value of miniature ultrasonic probes (MUPs) for the diagnosis and treatment of digestive tract diseases.METHODS: Endoscopic ultrasonography (EUS) was performed for patients with its indications with 7.5-20 MHz MUPs and double-cavity electronic endoscope. According to the diagnosis of MUPs, patients who had indications of treatment received endoscopic resection or surgical excision.Postoperative histological results were compared with the preoperative diagnosis of MUPs. A few patients without endoscopic resection or surgical excision were periodically followed up with MUPs.RESULTS: A total of 537 patients were examined by MUPs, of them, 256 were diagnosed with gastrointestinal submucosal lesions, 146 with pseudo-submucosal lesions,50 with digestive tract cancers, 17 with peptic ulcer, 11 with cholecystolithiasis, 8 with chronic pancreatitis, and 2 with achalasia and 47 were diagnosed as normal. After MUPs examinations, 220 patients received endoscopic resection or surgical excision, and the postoperative histological results of 211 patients were completely consistent with the preoperative diagnosis of MUPs. The diagnostic accuracy of MUPs was 95.9%. The result of follow-up with MUPs indicated that gastrointestinal leiomyoma, lipoma,phlebangioma and cyst were unchanged within 1-2 years.The patients who received endoscopic resection or centesis did not have any complications.CONCLUSION: MUPs are of value in diagnosing gastrointestinal submucosal lesions, staging of digestive tract cancers and biliary-pancreatic diseases. | 18 | External use of Ruyanneixiao cream efficiently blocks precancerous mammary lesions by interfering with glycolysis induced by inhibition of hypoxia inducible factor-1α, hexokinase 2, phosphofructokinase, and pyruvate kinase M2 expression显示文摘OBJECTIVE:To investigate the effect of Ruyanneixiao cream(RYNX) on the expression of hypoxia inducible factor-1α(HIF-1α), hexokinase 2(HK2),phosphofructokinase(PFK), and pyruvate kinase M2(PKM2) mRNA and protein in MCF-10 AT cells and in an animal model of precancerous mammary lesions.METHODS:Following treatment of MCF-10 AT cells with RYNX, tamoxifen(TAM) and YC-1 for 48 h,HIF-1α, HK2, PFK, PKM2 mRNA and protein expression was analyzed.Fifty female SD rats were randomly divided into control, model, TAM, and highand low-dose RYNX groups, with 10 rats in each group.A precancerous mammary lesion model was established for all groups except the control group.High-and low-dose RYNX cream containing TAM was coated on the breasts of animals in the corresponding groups.The rat mammary tissue was removed in the 10 th week and HIF-1α, HK2, PFK,PKM2 mRNA and protein was analyzed.RESULTS:In vitro analyses demonstrated that, compared with the matrix group, HIF-1α, HK2, PFK,PKM2 mRNA and protein expression was significantly decreased in the RYNX group(P < 0.05).Compared with the YC-1 + RYNX group, HK2, PFK,and PKM2 protein expression was significantly reduced in the RYNX group.HIF-1α, HK2, PFK, and PKM2 protein expression was increased significantly in the model group(P < 0.05) compared with the control group, while HIF-1α, HK2, PFK, and PKM2 mRNA and protein expression was significantly decreased in both the high-and low-dose RYNX groups(P < 0.05), with the effect being greater in the high-dose group.CONCLUSION:RYNX can block precancerous breast lesions by decreasing the expression of HK2,PFK, and PKM2 mRNA and protein via inhibition of HIF-1α mRNA and protein overexpression in a dose-dependent manner. | Li Xiaobo Ma Min Zhang Guijuan Ma Yi Liao Rui Chen Ruixue Yan Xianxin Bie Fengjie Huang Maojie Liang Shijie | 2017 | Journal of Traditional Chinese Medicine2017,37,2: | 5 |

| 20 | Biogas production from microbial-alkali pretreated corn stover by solid-state anaerobic digestion显示文摘Corn stover pretreated by Ceriporiopsis subvermispora was used as substrate to enhance biogas production via solid state anaerobic digestion(SS-AD).Effects of temperature,inoculum volume and alkaline concentration on the biogas production profermance were investigated.The results showed that pretreatment of corn stover by C.subvermispora benefited the biogas production.The optimal conditions were temperature 35°C,inoculum 10:1(w/w)and adding 5.0%of alkali,under which the maximum productions were obtained for daily and cumulative biogas of 14.07 L/kg VS and 211.09 L/kg VS.During the SS-AD process,the total solids(TS)and volatile solid(VS)were significantly degraded.The volatile fatty acids(VFA)concentration increased to 106 mmol/L and alkalinity decreased by about 23%.The results collectively suggested that microbial-alkali pretreatment of corn stove combining mesophilic condition could be a promising way to produce biogas in SS-AD. | Zhao Pengxiang Cui Fengjie Bu Lingxi Jiang Jianxin | 2015 | International Journal of Agricultural and Biological Engineering2015,8,5: | 4 |
