2Introduction of rice chitinase gene into wheat via low energy Ar^+ beam implantation显示文摘A chitinase gene (RCH8) in plasmid vector pCAMBIA1308 was delivered into 3 wheat cultivars (Yangmai 158, Wan 9210, Wanmai 32) by low energy Ar+ beam-mediated method. Preliminary calli from treated mature embryos were first selected on hygromycin (Hm, 20 or 30 mg/L) containing medium. After the resistant calli formed, they were transferred to the regeneration medium with 10 or 20 mg/L Hm. All the three wheat varieties obtained trans-genie plants. PCR and PCR-Southern assays showed that most plants regenerated from the resistant calli were positive transgenic plants. Southern blot of the positive green plants confirmed stable integration of alien DNA into wheat genome. The plant transformation frequencies varied with the variety and ion dose implanted. Wanmai 32 possessed the highest transformation frequency, reaching 3.8% at a suitable implantation dose. The transformation frequency of Yangmai 158 and Wan 9210 varied from 0.5% to 2.5% and from 0.5% to 1.4%, respectively. 3Effects of operational factors on soluble microbial products in a carrier anaerobic baffled reactor treating dilute wastewater显示文摘The effects of feed strength, hydraulic residence time (HRT), and operational temperatures on soluble microbial product (SMP) production were investigated, to gain insights into the production mechanism. A carrier anaerobic baffled reactor (CABR) treating dilute wastewater was operated under a wide range of operational conditions, namely, feed strengths of 300–600 mg/L, HRTs of 9– 18 h, and temperatures of 10–28°C. Generally, SMP production increased with increasing feed strength and decreasing temperature. At high temperature (28°C), SMP production increased with decreasing HRT. As the temperature was decreased to 18 and 10°C, the SMP production was at its peak for 12 h HRT. Therefore, temperature could be an important determinant of SMP production along with HRT. A higher SMP to soluble chemical oxygen demand (SCOD) ratio was found at high temperature and long HRT because of complete volatile fatty acid degradation. SMP accounted for 50%–75% of the SCOD in the last chamber of the CABR. 7Tetrahymena Gene Expression Database (TGED):A resource of microarray data and co-expression analyses for Tetrahymena显示文摘Tetrahymena thermophila is a model eukaryotic organism. Functional genomic analyses in Tetrahymena present rich opportunities to address fundamental questions of cell and molecular biology. The Tetrahymena Gene Expression Database (TGED; available at http://gffzz5d146715fb2e495ehopwp0uv5uw9q6u0b.ffgz.tsg.suse.edu.cn) is the first expression database of a ciliated protozoan. It covers three major physiological and developmental states: growth, starvation, and conjugation, and can be accessed through a user-friendly web interface. The gene expression profiles and candidate co-expressed genes for each gene can be retrieved using Gene ID or Gene description searches. Descriptions of standardized methods of sample preparation and the opportunity to add new Tetrahymena microarray data will be of great interest to the Tetrahymena research community. 8Toxicogenomic investigation of Tetrahymena thermophila exposed to dichlorodiphenyltrichloroethane (DDT),tributyltin (TBT),and 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)显示文摘Dichlorodiphenyltrichloroethane (DDT),tributyltin (TBT),and 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) are persistent in the environment and cause continuous toxic effects in humans and aquatic life.Tetrahymena thermophila has the potential for use as a model for research regarding toxicants.In this study,this organism was used to analyze a genome-wide microarray generated from cells exposed to DDT,TBT and TCDD.To accomplish this,genes differentially expressed when treated with each toxicant were identified,after which their functions were categorized using GO enrichment analysis.The results suggested that the responses of T.thermophila were similar to those of multicellular organisms.Additionally,the context likelihood of relatedness method (CLR) was applied to construct a TCDD-relevant network.The T-shaped network obtained could be functionally divided into two subnetworks.The general functions of both subnetworks were related to the epigenetic mechanism of TCDD.Based on analysis of the networks,a model of the TCDD effect on T.thermophila was inferred.Thus,Tetrahymena has the potential to be a good unicellular eukaryotic model for toxic mechanism research at the genome level.CHANG Yue FENG LiFang MIAO Wei 2011Science China(Life Sciences)2011,54,7:3
9ATP-binding cassette transporter enhances tolerance to DDT in Tetrahymena显示文摘The reuse of dichlorodiphenyltrichloroethane(DDT) as an indoor residual spray was permitted by the World Health Organization in 2007, and approximately 14 countries still use DDT to control disease vectors. The extensive exposure of insects to DDT has resulted in the emergence of DDT resistance, especially in mosquitoes, and the mechanism for this resistance in mosquitoes has been widely reported. Spraying can also introduce DDT directly into surface water, and DDT can subsequently accumulate in microorganisms, but the mechanism for the resistance to DDT degradation in microorganisms is unclear. Using whole-genome microarray analysis, we detected an abcb15 gene that was up-regulated in a specific manner by DDT treatment in T. thermophile. The deduced ABCB15 peptide sequence had two transmembrane domains(TMDs) and two nucleotide-binding domains(NBDs) to form the structure TMD-NBD-TMD-NBD, and each NBD contained three conserved motifs: Walker-A, C-loop, and Walker-B, which indicated the T. thermophila abcb15 was a typical ABC transporter gene. The expression of ABCB15 fused with a C-terminal green fluorescent protein was found to be on the periphery of the cell, suggesting that ABCB15 was a membrane pump protein. In addition, cells with abcb15 partially knocked down(abcb15-KD) grew slower than wild-type cells in the presence of 256 mg L-1 DDT, indicating the tolerance of abcb15-KD strain to DDT exposure was decreased. 11A model of suitability evaluation of tourism development for the suburban mining wasteland and its empirical research显示文摘The model of suitability evaluation of tourism development for suburban mining wasteland was established in Fengquan District of Xinxiang City, with measurable methods to provide the basis for its tourism development and landscape planning. The construction and evaluation of the model was conducted with AHP method from the follow-ing four aspects: ecological importance, landscape importance, economic importance and social importance, vali-dated with the case. In the established evaluation system, weight values of six indexes are higher than 0.05, among which the weight of industrial characteristics is the highest, and the weight of area, pollution conditions, air quality conditions, landscape diversity and topography were also higher. There are still seven indicators with weight values of higher than 0.02, including land price, transportation accessibility, geological conditions, water conditions, finan-cial security, distance to cities and similar competition. Weight values of five indexes are lower than 0.02, including government function, vegetation conditions, soil conditions, urban economic level and major tourism environment. We have tried to set a weighting set by AHP to avoid the shortcomings in ordinary weighting process, to perform the tourism evaluation with a maturely-developed method and solve the multi-indicator evaluation problems which in-volve the fuzziness and variability of scaling effectively. 12Rational Quasi-Interpolation Approximation of Scattered Data in R^(3)显示文摘This paper is concerned with a piecewise smooth rational quasi-interpolation with algebraic accuracy of degree(n+1)to approximate the scattered data in R 3.We firstly use the modified Taylor expansion to expand the mean value coordinates interpolation with algebraic accuracy of degree one to one with algebraic accuracy of degree(n+1).Then,based on the triangulation of the scattered nodes in R^(2),on each triangle a rational quasi-interpolation function is constructed.The constructed rational quasi-interpolation is a linear combination of three different expanded mean value coordinates interpolations and it has algebraic accuracy of degree(n+1).By comparing accuracy,stability,and efficiency with the C^(1)-Tri-interpolation method of Goodman[16]and the MQ Shepard method,it is observed that our method has some computational advantages.Renzhong Feng Lifang Song 2018Numerical Mathematics(Theory,Methods and Applications)2018,11,1:2
13Nonfunctional alleles of long-day suppressor genes independently regulate flowering time显示文摘Due to the remarkable adaptability to various environments, rice varieties with diverse flowering times have abeen domesticated or improved from Oryza rufipogon.Detailed knowledge of the genetic factors controlling flowering time will facilitate understanding the adaptation mechanism in cultivated rice and enable breeders to design appropriate genotypes for distinct preferences. In this study,four genes(Hd1, DTH8, Ghd7 and OsPRR37) in a rice long-day suppression pathway were collected and sequenced in 154, 74,69 and 62 varieties of cultivated rice(Oryza sativa)respectively. Under long-day conditions, varieties with nonfunctional alleles flowered significantly earlier than those with functional alleles. However, the four genes have different genetic effects in the regulation of flowering time: Hd1 and Os PRR37 are major genes that generally regulate rice flowering time for all varieties, while DTH8 and Ghd7 only regulate regional rice varieties. Geographic analysis and network studies suggested that the nonfunctional alleles of these suppression loci with regional adaptability were derived recently and independently. Alleles with regional adaptability should be taken into consideration for genetic improvement.
