1	视黄酸及Stra8基因联合作用对雄性鸡胚胎干细胞分化的影响显示文摘旨在研究视黄酸(All-trans retinoic acid,RA)及Stra8(Stimulated by retinoic acid gene 8)基因联合作用对雄性鸡胚胎干细胞(Chicken embryonic stem cells,cESCs)分化的影响。选用PCR方法鉴定雄性cESCs为研究对象,转染Stra 8基因,对Stra8基因阳性cESCs使用RA进行12d连续诱导,同时设立Stra8基因转染组、RA诱导组及空白组对照,各组均进行3次重复试验。细胞形态学、qRT-PCR检测RA及Sira8基因对cESCs分化的影响。结果表明。转染Stra8基因进行RA诱导,在诱导12d后,出现精原干细胞样的圆形小克隆,其他组cESCs出现类胚体或解体;qRT-PCR鉴定发现各组中干细胞标记基因表达量随着培养时间的延长逐渐下降,同时检测到生殖细胞相关基因表达,转染Stra8基因并使用RA诱导组中表达量均高于对照组(P<0.01),6d表达量达到最高,然后逐渐下降。Stra8基因和RA的联合作用能够促进cESCs向着生殖细胞方向的分化,不仅改变了原有细胞形态,同时出现了生殖相关基因的表达。通过该方法可以在体外建立一个。ESCs诱导分化模型,将为生殖细胞的发生、增殖、分化及调控机理的研究和人类不孕不育症的治疗奠定良好的基础。	张振韬王丹施青青 ELSAYED A K 张亚妮韦光辉朱睿左其生李东刘堃李碧春	2014	畜牧兽医学报2014，45，1：	3
