2Glucocorticoid receptor expression in neonatal rat cortex following recurrent seizures The role in developing brain injury显示文摘BACKGROUND: Studies have explored changes in neonatal rat glucocorticoid receptor (GR) expression changes following mature brain injury.OBJECTIVE: To investigate the temporal and special changes of GR during brain development in rats with recurrent seizures.DESIGN,TIME AND SETTING: A randomized,controlled animal experiment was performed at the Department of Pediatrics,Second Xiangya Hospital of Central South University,from February 2008 to March 2009.MATERIALS: Rabbit anti-rat GR monoclonal antibody was purchased from Santa Cruz Biotechnology,USA;goat anti-rabbit IgG was purchased from Zhongshan Goldenbridge Biotechnology,China.METHODS: A total of 48 Sprague-Dawley rats,7 days old,were randomly assigned to control and seizure groups,with 24 animals in each group.Seizures were induced by inhalant flurothyl.MAIN OUTCOME MEASURES: Changes in GR protein expression in the rat cerebral cortex were detected by Western blotting analysis and immunohistochemistry.RESULTS: GR expression in the cerebral cortex of control rats significantly increased with aging (P < 0.05),and varied in the frontal lobe,temporal lobe,and parietal lobe.GR was predominantly expressed in the cytoplasm early and rapidly increased in the nuclei.GR protein expression in the cerebral cortex after seizure was lower in the cytoplasm at 15 days and in nuclear protein at 19 days.CONCLUSION: GR expression displayed temporal and spatial changes in brain development.Recurrent seizures in neonatal rats cause abnormal GR expression and might play an important role in developing brain injury.Tao Bo Lu Yi Tuanmei Wang Jian Li Xingfang Li Dingan Mao 2010Neural Regeneration Research2010,5,2:2

12Automated retinal layer segmentation in optical coherence tomography images with intraretinal fluid显示文摘We propose a novel retinal layer segmentation method to accurately segment 10 retinal layers in optical coherence tomography(OCT)images with intraretinal fluid.The method used a fan filter to enhance the linear information pertaining to retinal boundaries in an OCT image by reducing the effect of vessel shadows and fluid regions.A random forest classifier was employed to predict the location of the boundaries.Two novel methods of boundary redirection(SR)and similarity correction(SC)were combined to carry out boundary tracking and thereby accurately locate retinal layer boundaries.Experiments were performed on healthy controls and subjects with diabetic macular edema(DME).The proposed method required an average of 415 s for healthy controls and of 482 s for subjects with DME and achieved high accuracy for both groups of subjects.The proposed method requires a shorter running time than previous methods and also provides high accuracy.Thus,the proposed method may be a better choice for small training datasets.Luquan Wang Xiaowen Li Yong Chen Dingan Han Mingyi Wang Yaguang Zeng Junping Zhong Xuehua Wang Yanhong Ji Honglian Xiong Xunbin Wei 2022Journal of Innovative Optical Health Sciences2022,15,3:0
13Gamma-aminobutyric acid A receptor gamma 2 subunit following Mg-free-induced seizures in cultured developing neurons显示文摘BACKGROUND:Studies have demonstrated that in vitro cultured cortical neurons from embryonic rats can produce spontaneous recurrent epileptiform discharges following transient Mg2+-free extracellular solution culture. OBJECTIVE: To explore gamma-aminobutyric acid A receptor (GABAAR) γ2 subunit expression follow-ing Mg2+-free-induced seizures in cultured developing neurons. DESIGN,TIME AND SETTING: Cellular and molecular biology. The in vitro experiment was performed at the Department of Pediatrics,Second Xiangya Hospital of Central Southern University between January 2007 and February 2008. MATERIALS: Cortical neurons of Wistar rats on gestational days 16-17 were used. Normal extracellular solution (pH 7.3) consisted of NaCl 145 mmol/L,KCl 2.5 mmol/L,HEPES 10 mmol/L,MgCl2 1 mmol/L,CaCl2 2 mmol/L,glucose 10 mmol/L,and glycine 0.01 mmol/L. In addition,there was no MgCl2 in the Mg2+-free extracellular solution. METHODS: Cortical neurons cultured for 6 days were exposed to normal extracellular solution (control group) and Mg2+-free media (Mg2+-free group) respectively for 3 hours,followed by continuous culture in DMEM solution. MAIN OUTCOME MEASURES: On days 1,7 and 12 after Mg2+-free treatment,real-time RT-PCR,im-munochemistry,and flow cytometry were used to detect GABAAR γ2 subunit expression. RESULTS: Compared with the control group,GABAAR γ2-positive cells decreased significantly on days 1 and 7 after Mg2+-free treatment (P<0.01),but significantly increased on day 12 (P<0.01). GABAAR γ2 subunit mRNA expression decreased significantly at 7 days Mg2+-free treatment when measured by real-time RT-PCR compared with the control group (P<0.05). CONCLUSION: GABAAR γ2 subunit expression is modified following Mg-free-induced seizures in cul-tured developing neurons. 14Predicting the severity of traffic accidents on mountain freeways with dynamic traffic and weather data显示文摘Traffic accident severity prediction is essential for dynamic traffic safety management.To explore the factors influencing the severity of traffic accidents on mountain freeways and to predict the severity of traffic accidents,four models based on machine learning algorithms are constructed using support vector machine(SVM),decision tree classifier(DTC),Ada_SVM and Ada_DTC.In addition,random forest(RF)is used to calculate the importance degree of variables and the accident severity influences with high importance levels form the RF dataset.The results show that rainfall intensity,collision type,number of vehicles involved in the accident and toad section type are important variables influencing accident severity.The RF feature selection method improves the classification performance of four machine leaming algorithms,resulting in a 9.3%,5.5%,7.2% and 3.6% improvement in prediction accuracy for SVM,DTC,Ada_SVM and Ada_DTC,respectively.The combination of the Ada_SVM integrated algorithm and RF feature selection method has the best prediction performance,and it achieves 78.9% and 88.4% prediction precision and accuracy,respectively.Juan Li Fengxiang Guo Yanning Zhou Wenchen Yang Dingan Ni 2023Transportation Safety and Environment2023,5,4:0

19Effects of progesterone on glutamate transporter 2 and gamma-aminobutyric acid transporter 1 expression in the developing rat brain after recurrent seizures显示文摘Seizures were induced by flurothyl inhalation. Rats were intramuscularly treated with progesterone after each seizure. Results demonstrated that glutamate transporter 2 and γ-aminobutyric acid transporter 1 expression levels were significantly increased in the cerebral cortex and hippocampus of the developing rat brain following recurrent seizures. After progesterone treatment, glutamate transporter 2 protein expression was upregulated, but γ-aminobutyric acid transporter 1 levels decreased. These results suggest that glutamate transporter 2 and γ-aminobutyric acid transporter 1 are involved in the pathological processes of epilepsy.
