1SIZ1 negatively regulates aluminum resistance by mediating the STOP1–ALMT1 pathway in Arabidopsis显示文摘Sensitive to proton rhizotoxicity 1(STOP1) functions as a crucial regulator of root growth during aluminum(Al) stress. However, how this transcription factor is regulated by Al stress to affect downstream genes expression is not well understood. To explore the underlying mechanisms of the function and regulation of STOP1, we employed a yeast two hybrid screen to identify STOP1-interacting proteins. The SUMO E3 ligase SIZ1, was found to interact with STOP1 and mainly facilitate its SUMO modification at K40 and K212 residues. Simultaneous introduction of K40 R and K212 R substitutions in STOP1 enhances its transactivation activity to upregulate the expression of aluminum-activated malate transporter 1(ALMT1)via increasing the association with mediator 16(MED16) transcriptional co-activator. Loss of function of SIZ1 causes highly increased expression of ALMT1, thus enhancing Al-induced malate exudation and Al tolerance. Also, we found that the protein level of SIZ1 is reduced in response to Al stress. Genetic evidence demonstrates that STOP1/ALMT1 is epistatic to SIZ1 in regulating root growth response to Al stress. 4Initiation and maintenance of plant stem cells in root and shoot apical meristems显示文摘Plant stem cells are a small group of cells with a self-renewal capacity and serve as a steady supply of precursor cells to form new differentiated tissues and organs in plants.Root stem cells and shoot stem cells,which are located in the root apical meristem and in the shoot apical meristem,respectively,play a critical role in plant longitudinal growth.These stem cells in shoot and root apical meristems remain as pluripotent state throughout the lifespan of the plant and control the growth and development of plants.The molecular mechanisms of initiation and maintenance of plant stem cells have been extensively investigated.In this review,we mainly discuss how the plant phytohormones,such as auxin and cytokinin,coordinate with the key transcription factors to regulate plant stem cell initiation and maintenance in root and shoot apical meristems.In addition,we highlight the common regulatory mechanisms of both root and shoot apical meristems.Junxia Wang Yinghua Su Xiangpei Kong Zhaojun Ding Xian Sheng Zhang 2020aBIOTECH2020,1,3:1
5NAC1 regulates root ground tissue maturation by coordinating with the SCR/SHR-CYCD6;1 module in Arabidopsis显示文摘Precise spatiotemporal control of the timing and extent of asymmetric cell divisions(AcDs)is essential for plant development.In the Arabidopsis root,ground tissue maturation involves an additional AcD of the endo-dermis that maintains the inner cell layer as the endodermis and generates the middle cortex to the outside.Through regulation of the cell cycle regulator CYCLIND6;1(CYCD6;1),the transcription factors SCARECROW(SCR)and SHORT-ROOT(SHR)play critical roles in this process.In the present study,we found that loss of function of NAC1,a NAC transcription factor family gene,causes markedly increased periclinal cell divisions in the root endodermis.Importantly,NAC1 directly represses the transcription of CYCD6;1 by recruiting the co-repressor TOPLESS(TPL),creating a fine-tuned mechanism to maintain proper root ground tissue patterning by limiting production of middle cortex cells.Biochemical and genetic analyses further showed that NAC1 physically interacts with SCR and SHR to restrict excessive periclinal cell divisions in the endo-dermis during root middle cortex formation.Although NAC1-TPL is recruited to the CYCD6;1 promoter and represses its transcription in an SCR-dependent manner,NAC1 and SHR antagonize each other to regu-late the expression of CYCD6;1.Collectively,our study provides mechanistic insights into how the NAC1-TPL module integrates with the master transcriptional regulators SCR and SHR to control root ground tissue patterning by fine-tuning spatiotemporal expression of CYCD6;1 in Arabidopsis.Chuantian Xie Cuiling Li Fengxia Wang Feng Zhang Jiajia Liu Junxia Wang Xiansheng Zhang Xiangpei Kong Zhaojun Ding 2023Molecular Plant2023,16,4:1

13Two-stage reproduction derived from cells of thallus could directly contribute to seeds for green tidal algal Enteromorpha(Ulva) prolifera/clathrata bloom,with disclosure of their ephemeral trait显示文摘Green tidal algal Enteromorpha species complete their life cycles by the isomorphic alternation of generations.The provenance of green tide caused by them in the western Yellow Sea has been disputed. The cell reproduction derived from adult thallus was observed on E. clathrata collected from Shantou City, Guangdong Province in this study. Subsequently, it further found that E. proliferia collected from Qingdao City, Shandong Province and Qinhuangdao City, Hebei Province, produced reproductive cells by somatic cells of its early infantile thallus or branch. The latter is functionally similar to that the seedlings of red alga Porphyra yezoensis produce the monospores, and could exquisitely explain the ephemeral or opportunistic trait and environmental adaptation ability of Enteromorpha species. Changes in growth conditions may induce the two types of cell reproduction.They contribute to the bloom, and can effectively reveal the seasonally occurring large-scale and on-year and offyear phenomenon. The latter may have played a decisive role in its formation. This paper analyses the legal status of the species name, the type of generation during bloom, ephemeral traits, the role of microscopic propagule, the area of origin, on-year and off-year phenomenon, early warning and prevention and control of the species, and so on. 14A feedback regulation between ARF7-mediated auxin signaling and auxin homeostasis involving MES17 affects plant gravitropism显示文摘Gravitropism is an essential adaptive response of land plants.Asymmetric auxin gradients across plant organs,interpreted by multiple auxin signaling components including AUXIN RESPONSE FACTOR7(ARF7),trigger differential growth and bending response.However,how this fundamental process is strictly maintained in nature remains unclear.Here,we report that gravity stimulates the transcription of METHYL ESTERASE17(MES17)along the lower side of the hypocotyl via ARF7-dependent auxin signaling.The asymmetric distribution of MES17,a methyltransferase that converts auxin from its inactive form methyl indole-3-acetic acid ester(MeI AA)to its biologically active form free-IAA,enhanced the gradient of active auxin across the hypocotyl,which in turn reversely amplified the asymmetric auxin responses and differential growth that shape gravitropic bending.Taken together,our findings reveal the novel role of MES17-mediated auxin homeostasis in gravitropic responses and identify an ARF7-triggered feedback mechanism that reinforces the asymmetric distribution of active auxin and strictly controls gravitropism in plants.Feng Zhang Cuiling Li Xingzhen Qu Jiajia Liu Zipeng Yu Junxia Wang Jiayong Zhu Yongqiang Yu Zhaojun Ding 2022Journal of Integrative Plant Biology2022,64,7:0
15Effects of simvastatin on ion channel currents in ventricular myocytes from rabbit with acute myocardial infarction显示文摘Objective To investigate the effects of simvastatin on membrane ionic currents in left ventricular myocytes after acute myocardial infarction (AMI,so as to explore the ionic mechanism of statin treatment for antiarrhythmia.Methods Fourty-five New Zeland rabbits were randomly divided into three groups:AMI group,simvastatin intervention group (statin group) and sham-operated control group (CON).Rabbits were infarcted by ligation of the left anterior descending coronary artery after administration of oral simvastatin 5 mg·kg-1·d-1 (Statin group) or placebo (AMI group)for 3 days.Twenty-four hours later,single ventricular myocytes were isolated enzymatically from the epicardial zone of the infracted region.Whole cell patch clamp technique was used to record membrane ionic currents,including sodium current (INa),L-type calcium current (ICa-L) and transient outward potassium current (Ito).Results①There was no significant difference in serum cholesterol concentration among three groups.②The peak INa current density (at-30 mV) was significantly decreased in AMI group (-23.26±5.18) compared with CON (-42.78±5.48,P<0.05),while it was significantly increased in Statin group (-39.23±5.45) compared with AMI group (P<0.01);The peak ICa-L current density (at 0 mV) was significantly decreased in AMI group (-3.23±0.91) compared with CON (-4.56±1.01,P<0.05),while it was significantly increased in Statin group (- 4.18±0.95) compared with AMI group (P<0.05);The Ito current density(at +60 mV) was significantly decreased in AMI group(10.41±1.93)compared with CON (17.41±3.13,P<0.01),while it was significantly increased in Statin group(16.21±2.42)compared with AMI group (P<0.01).Conclusions AMI induces significant down-regulation of INa,ICa-L and Ito.Pretreatment with simvastatin could attenuate this change without lowering the serum cholesterol level,suggesting that simvastatin reverse this electrical remodeling,thus contributing to the ionic mechanism of statin treatment for antiarrhythmia.Chao Ding~1 Xianghua Fu~2 Li Yang~1 Huixiao Chen~1 Junxia Li~1 Yuying Zhao~1 Jie Li~1 Jie Wang~1 1 Department of Cardiology, Bethune International Peace Hospital of Chinese PLA, Shijiazhuang Hebei 050082 China 2 Department of Cardiology, The Second Hospital, Hebei Medical University, Shijiazhuang Hebei 050000, China 2008Journal of Geriatric Cardiology2008,5,3:0
16A Systemic Review and Meta-analysis of the Effect of SARS-CoV-2 Infection on Sperm Parameters显示文摘Objective.Several studies examined the putativefets of SARS-Cov-2infectionon spem parametersHoweve,the resuls remain controversial.In this study,we conducted the most up-to-date systematic review and meta-analysis to investigate the effect of SARS-CoV-2 infection on sperm quality in COVID-19-positive and COVID-19-negative male participants.Method.Seven databases were searched forliterature released through June 10,2022,containing estimates for the outcomes of interest Using a random-fects model(REM)or a fixed-ffects model(FEM),we analyzed the pooled results.The quality of all included studies was asssed by the Newcastle-Ottawa scale In addition,we performed aquanttative and subgroup analysis of semen data across all included studies.Results.Fourteen studies were extracted from 10 publications,involving a total of 1174 participates for meta-analysis.Sperm parameters of 521 COVID-19 male patients and 653 controls were analyzed.In 8 case-control studies,the pooled mean dfference(MD)of total sperm motility was-5.37%(95%confidence interval(CI):-8.47 to-2.28:<0.05),suggesting that total motility was significantly impaired inmale COVID-19 cases.Subgroup analysis showed a significant decreasein semen volume,sperm concentration,and total motility in 238 patients with arecovery timeof less than 90 days.Moreover,in the other 6 included pre-to post-COVID-19 studies,the pooled MDs of sperm concentration,total sperm count,total motility,progressivemotility,and normal morphology were6.54×10^(6)/ml(95%CI:-10.27 to-2.81;p<0.05),38.89×10^(6)(95%CI:-59.20 to-18.58;p<0.05),-7.21%(95%CI:-14.36 to-0.07;p<0.05),-5.12%(95%CI:-8.71 to-1.53;p<0.05),and-1.52%(95%CI:-2.88 to-0.16;p<0.05),respectively,which indicate SARS-CoV-2 infection significantly affected these five sperm parameters.Conclusion.Our results revealed that SARS-CoV-2 infection was significantly correlated with decreased sperm quality.Of six sperm parameters,total motility and sperm concentration were the most significantly decreased parameters.These results suggest a possible negative influence of SARS-CoV-2 infection on testicular function and male fertility.Given the potential detrimental effect of SARS-CoV-2 on semen quality,male reproductive health should be monitored closely in patients with COVID-19.This trial is registered with CRD42021275823.Xi Chen Jinli Ding Miao Liu Kai Xing Peng Ye Junxia Min Yan Zhang Tailang Yin 2022Research2022,,2:0
