1Serum cytokine profile in patients with hepatitis B e antigen-negative chronic active hepatitis B and inactive hepatitis B virus carriers显示文摘An insufficient cellular immune response seems to be critical for the immunopathogenesis of chronic hepatitis B virus infection.We have previously demonstrated no differences of T-lymphocyte subsets in blood between inactive hepatitis B s antigen(HBsAg) carriers and patients with HBeAg-negative chronic active hepatitis B.This study investigated the peripheral blood cytokine profile in patients with HBeAg-negative chronic active hepatitis B infection(Group A,n = 21) and inactive HBsAg carriers(Group B,n = 13).Serum cytokines [interferon(IFN)-γ,tumor necrosis factor-α,interleukin(IL)-1b,IL-4,IL-12,IL-10,IL-2,IL-5,IL-8] were analyzed by using flow cytometry.Patients with chronic active disease presented with significantly decreased levels of IFN-γ and IL-10 compared to inac-tive carriers(P = 0.048 and P = 0.008,respectively).In HBeAg-negative chronic active hepatitis B patients,a significant negative correlation of IFN-γ levels with serum hepatitis B viral load was noted(P = 0.021).In conclusion,patients with HBeAg-negative chronic active hepatitis B and HBsAg inactive carriers display a different cytokine profile.Decreased Th1 response observed in patients with chronic active hepatitis B could be implicated in the persistence of virus replication and ongoing progression of liver disease.Dimitra Dimitropoulou Marina Karakantza Georgios L Theodorou Lydia Leonidou Stelios F Assimakopoulos Athanasia Mouzaki Charalambos A Gogos 2013World Journal of Gastrointestinal Pathophysiology2013,4,1:13
2YODA-HSP90 Module Regulates Phosphorylation-Dependent Inactivation of SPEECHLESS to Control Stomatal Development under Acute Heat Stress in Arabidopsis显示文摘Stomatal ontogenesis,patterning,and function are hallmarks of environmental plant adaptation,especially to conditions limiting plant growth,such as elevated temperatures and reduced water availability.The specification and distribution of a stomatal cell lineage and its terminal differentiation into guard cells require a master regulatory protein phosphorylation cascade involving the YODA mitogen-activated protein kinase kinase kinase.YODA signaling results in the activation of MITOGEN-ACTIVATED PROTEIN KINASEs(MPK3 and MPK6),which regulate transcription factors,including SPEECHLESS(SPCH).Here,we report that acute heat stress affects the phosphorylation and deactivation of SPCH and modulates stomatal density.By using complem entary molecular,genetic,biochemical,and cell biology approaches,we provide solid evidence that HEAT SHOCK PROTEINS 90(HSP90s)play a crucial role in transducing heat-stress response through the YODA cascade.Genetic studies revealed that YODA and HSP90.1 are epistatic,and they likely function linearly in the same developmental pathway regulating stomata formation.HSP90s interact with YODA,affectits cellular polarization,and modulate the phosphorylation of downstream targets,such as MPK6 and SPCH,under both normal and heat-stress conditions.Thus,HSP90-mediated specification and differentiation of the stomatal cell lineage couples stomatal development to environmental cues,providing an adaptive heat stress response mechanism in plants.Despina Samakovli Tereza Ticha Tereza Vavrdova Miroslav Ovecka Ivan Luptovciak Veronika Zapletalova Anna Kucharova Pavel Krenek Yuliya Krasylenko Theoni Margaritopoulou Loukia Roka Dimitra Milioni George Komis Polydefkis Hatzopoulos Jozef Samaj 2020Molecular Plant2020,13,4:9
3Thermogenic protein UCP1 and UCP3 expression in non- small cell lung cancer: relation with glycolysis and anaerobic metabolism显示文摘Uncoupling protein 1(UCP1)is a proton transporter/channel residing on the inner mitochondrial membrane and is involved in cellular heat production.Using immunohistochemistry,we investigated the expression of UCP1 and UCP3 in a series of 98 patients with non-small cell lung cancer(NSCLC)treated with surgery.Expression patterns were correlated with histopathological variables,prognosis,and the expression of enzymes/proteins related to cell metabolism.Bronchial epithelium did not express UCP1 or UCP3,while alveolar cells strongly expressed UCP1.In tumors,strong expression of UCP1 and UCP3 was recorded in43/98(43.8%)and 27/98(27.6%)cases,respectively.UCP1 was significantly associated with squamous cell histology(P=0.05),whilst UCP3 was more frequently overexpressed in large cell carcinomas(P=0.08),and was inversely related to necrosis(P=0.009).In linear regression analysis,UCP1 was directly related to markers of glycolysis[hexokinase(HXKII)and phosphofructokinase(PFK1)]and anaerobic glucose metabolism[pyruvate dehydrogenase kinase(PDK1)and lactate dehydrogenase(LDH5)].UCP3 was directly linked with a glucose transporter(GLUT2),monocarboxylate transporter(MCT2),glycolysis markers(PFK1 and aldolase),and with the phosphorylation of pyruvate dehydrogenase(p PDH).Kaplan-Meier survival analysis showed that UCP3 was significantly related to poor prognosis in squamous cell carcinomas(P=0.04).UCP1 and UCP3 are overexpressed in a large subgroup of non-small cell lung tumors and their expression coincides with increased glucose absorption,intensified glycolysis,and anaerobic glucose usage.Whether UCPs are targets for therapeutic interventions in lung cancer is a hypothesis that demands further investigation.Alexandra Giatromanolaki Konstantina Balaska Dimitra Kalamida Christos Kakouratos Efthimios Sivridis Michael I. 4Life cycle assessment of open field and greenhouse cultivation of lettuce and barley显示文摘In the present paper,a life cycle assessment(LCA)study regarding barley and lettuce production in Spain(Barrax and Santomera regions)and Italy(Albenga region)in both open field(OF)and standard greenhouse(GH)cultivations was performed in order to evaluate energy consumption and environmental impacts.The study examines also the impact of the use of compost produced from agricultural wastes(AW).In this context,a detailed life cycle directory was created,based on site-specific experimental data,and used for a holistic cradle-to-gate LCA analysis using the GaBi 6 software package and specific related databases.In order to reveal the importance of system boundaries,factors that are often excluded from LCA studies,such as agricultural machinery manufacture,nursery production,waste management and raw materials transportation have been considered.According to the results of this study,the use of compost for fertilization of both crops is considered a good agronomic and ecological strategy in order to maintain productivity in terms of yield,especially in the case of greenhouse cultivation,and improve overall sustainability in the agricultural sector.Moreover,the phases of compost production,irrigation system,and greenhouse construction and operation were identified as the three main‘‘hotspots”with the highest environmental impact and energy contribution in all studied cases.Finally,improvements to reduce those impacts were proposed.Georgios Bartzas Dimitra Zaharaki Kostas Komnitsas 2015Information Processing in Agriculture2015,2,3:7
5Practical approaches to effective management of intestinal radiation injury:Benefit of resectional surgery显示文摘AIM:To study the outcome of patients undergoing surgical resection of the bowel for sustained radiation-induced damage intractable to conservative management.METHODS:During a 7-year period we operated on 17 cases (5 male,12 female) admitted to our surgical department with intestinal radiation injury (IRI).They were originally treated for a pelvic malignancy by surgical resection followed by postoperative radiotherapy.During follow-up,they developed radiation enteritis requiring surgical treatment due to failure of conservative management.RESULTS:IRI was located in the terminal ileum in 12 patients,in the rectum in 2 patients,in the descending colon in 2 patients,and in the cecum in one patient.All patients had resection of the affected region(s).There were no postoperative deaths,while 3 cases presented with postoperative complications (17.7%).All patients remained free of symptoms without evidence of recurrence of IRI for a median follow-up period of 42 mo (range,6-96 mo).CONCLUSION:We report a favorable outcome without IRI recurrence of 17 patients treated by resection of the diseased bowel segment.Nikolaos Perrakis Evangelos Athanassiou Dimitra Vamvakopoulou Maria Kyriazi Haris Kappos Nikolaos C Vamvakopoulos Iakovos Nomikos 2011World Journal of Gastroenterology2011,17,35:6

8Polycystic ovary syndrome and type 2 diabetes mellitus:A state-ofthe-art review显示文摘Polycystic ovary syndrome(PCOS)often coexists with a wide spectrum of dysglycemic conditions,ranging from impaired glucose tolerance to type 2 diabetes mellitus(T2D),which occur to a greater extent compared to healthy body mass index-matched women.This concurrence of disorders is mainly attributed to common pathogenetic pathways linking the two entities,such as insulin resistance.However,due to methodological flaws in the available studies and the multifaceted nature of the syndrome,there has been substantial controversy as to the exact association between T2D and PCOS which has not yet been elucidated.The aim of this review is to present the best available evidence regarding the epidemiology of dysglycemia in PCOS,the unique pathophysiological mechanisms underlying the progression of dysglycemia,the most appropriate methods for assessing glycemic status and the risk factors for T2D development in this population,as well as T2D risk after transition to menopause.Proposals for application of a holistic approach to enable optimal management of T2D risk in PCOS are also provided.Specifically,adoption of a healthy lifestyle with adherence to improved dietary patterns,such the Mediterranean diet,avoidance of consumption of endocrine-disrupting foods and beverages,regular exercise,and the effect of certain medications,such as metformin and glucagon-like peptide 1 receptor agonists,are discussed.Furthermore,the maintenance of a healthy weight is highlighted as a key factor in achievement of a significant reduction of T2D risk in women with PCOS.Sarantis Livadas Panagiotis Anagnostis Julia K Bosdou Dimitra Bantouna Rodis Paparodis 2022World Journal of Diabetes2022,13,1:4

10Antidiabetic treatment, stroke severity and outcome显示文摘Ischemic stroke is a leading cause of mortality and long-term disability worldwide. Given the detrimental effects of acute stroke, several neuroprotective agents have been evaluated in these patients. However, the benefits of the evaluated agents appear to be limited and none is currently recommended for clinical use. On the other hand, prior treatment with agents that are used for the primary and secondary prevention of stroke, including statins and antiplatelets, has been associated with better outcome in patients who experience an acute stroke. In contrast, there are limited data as to whether prior treatment with antidiabetic agents is beneficial in diabetic patients who suffer a stroke. In this context, the findings of a recent study that showed reduced stroke size following pretreatment with linagliptin, a dipeptidyl peptidase-4(DDP-4) inhibitor, compared with glimepiride, in both diabetic and non-diabetic mice, appear promising. Despite these preclinical findings suggesting neuroprotective effects of DPP-4 inhibitors in acute stroke, it is still unclear whether these actions will also be observed in humans. Of note, two recent large randomized, placebo-controlled studies did not show any effect of DPP-4 inhibitors on cardiovascular events, including stroke. Several other ongoing trials are evaluating the effects of DPP-4 inhibitors on cardiovascular morbidity and mortality. 12Inhibition of IKK-NFκB pathway sensitizes lung cancer cell lines to radiation显示文摘Objective:Cancer cell radioresistance is a stumbling block in radiation therapy.The activity in the nuclear factor kappa B(NFκB)pathway correlates with anti-apoptotic mechanisms and increased radioresistance.The IKK complex plays a major role in NFκB activation upon numerous signals.In this study,we examined the interaction between ionizing radiation(IR)and different members of the IKK-NFκB pathway,as well as upstream activators,RAF1,ERK,and AKT1.Methods:The effect of 4 Gy of IR on the expression of the RAF1-ERK-IKK-NFκB pathway was examined in A549 and H1299 lung cancer cell lines using Western blot analysis and confocal microscopy.We examined changes in radiation sensitivity using gene silencing or pharmacological inhibitors of ERK and IKKβ.Results:IKKα,IKKγ,and IκBαincreased upon exposure to IR,thereby affecting nuclear levels of NFκB(phospho-p65).ERK inhibition or si RNA-mediated down-regulation of RAF1 suppressed the post-irradiation survival of the examined lung cancer cell lines.A similar effect was detected on survival upon silencing IKKα/IKKγor inhibiting IKKβ.Conclusions:Exposure of lung cancer cells to IR results in NFκB activation via IKK.The genetic or pharmacological blockage of the RAF1-ERK-IKK-NFκB pathway sensitizes cells to therapeutic doses of radiation.Therefore,the IKK pathway is a promising target for therapeutic intervention in combination with radiotherapy.Avgi Tsolou Maria Liousia Dimitra Kalamida Stamatia Pouliliou Alexandra Giatromanolaki MichaelKoukourakis 2017Cancer Biology & Medicine2017,14,3:2

19Assessment of groundwater contamination risk in an agricultural area in north Italy显示文摘In the present study a specific approach is followed,considering the Pesticide DRASTIC and Susceptibility index(SI)methods and a GIS framework,to assess groundwater vulnerability in the agricultural area of Albenga,in north Italy.The results indicate'high'to'very high'vulnerability to groundwater contamination along the coastline and the middle part of the Albenga plain,for almost 49%and 56%of the total study area for Pesticide DRASTIC and SI methods,respectively.These sensitive regions depict characteristics such as shallow depth to groundwater,extensive deposits of alluvial silty clays,flat topography and intensive agricultural activities.The distribution of nitrates concentration in groundwater in the study area is slightly better correlated with the SI(0.728)compared to Pesticide DRASTIC(0.693),thus indicating that both methods are characterized by quite good accuracy.Sensitivity analysis was also performed to acknowledge statistical uncertainty in the estimation of each parameter used,assess its impact and thus identify the most critical parameters that require further investigation in the future.Depth to water is the parameter that exhibited the largest impact on the Pesticide DRASTIC vulnerability index followed by the impact of the vadose zone and topography.On the other hand,the SI method is more sensitive to the removal of the topography parameter followed by the aquifer media and the depth to water parameters.Georgios Bartzas Federico Tinivella Luca Medini Dimitra Zaharaki Kostas Komnitsas 2015Information Processing in Agriculture2015,2,2:2
20Targeted therapies for pancreatic adenocarcinoma: Where do we stand, how far can we go?显示文摘Pancreatic adenocarcinoma(usually referred to aspancreatic cancer) is a highly lethal and aggressive malignancy with a disease-related mortality almost equaling its incidence, and one of the most challenging cancers to treat. The notorious resistance of pancreatic cancer not only to conventional cytotoxic therapies but also to almost all targeted agents developed to date, continues to puzzle the oncological community and represents one of the biggest hurdles to reducing the death toll from this ominous disease.
