2GSDMB promotes non-canonical pyroptosis by enhancing caspase-4 activity显示文摘Gasdermin B (GSDMB) has been reported to be associated with immune diseases in humans, but the detailed molecular mechanisms remain unsolved. The N-terminus of GSDMB by itself, unlike other gasdermin family proteins, does not induce cell death. Here, we show that GSDMB is highly expressed in the leukocytes of septic shock patients, which is associated with increased release of the gasdermin D (GSDMD) N-terminus. GSDMB expression and the accumulation of the N-terminal fragment of GSDMD are induced by the activation of the non-canonical pyroptosis pathway in a human monocyte cell line. The downregulation of GSDMB alleviates the cleavage of GSDMD and cell death. Consistently, the overexpression of GSDMB promotes GSDMD cleavage, accompanied by increased LDH release. We further found that GSDMB promotes caspase-4 activity, which is required for the cleavage of GSDMD in non-canonical pyroptosis, by directly binding to the CARD domain of caspase-4. 3Mitochondrial ROS promote macrophage pyroptosis by inducing GSDMD oxidation显示文摘Disrupted mitochondrial membrane potential (MMP) and reactive oxygen species (ROS) generation are often associated with macrophage pyroptosis. It remains unclear how these forms of mitochondrial dysfunction relate to inflammasome activation and gasdermin-D (Gsdmd) cleavage, two central steps of the pyroptotic process. Here, we also found MMP collapse and ROS generation induced by Nlrp3 inflammasome activation as previous studies reported. The elimination of ROS alleviated the cleavage of Gsdmd, suggesting that Gsdmd cleavage occurs downstream of ROS release. Consistent with this result, hydrogen peroxide treatment augmented the cleavage of Gsdmd by caspase-1. Indeed, four amino acid residues of Gsdmd were oxidized under oxidative stress in macrophages. The efficiency of Gsdmd cleavage by inflammatory caspase-1 was dramatically reduced when oxidative modification was blocked by mutation of these amino acid residues. 4Roles of MAP kinase signaling pathway in oocyte meiosis显示文摘Mitogen-activated protein kinase (MAPK) is a family of Ser/Thr protein kinases expressed widely in eu-karyotic cells. MAPK is activated by a cascade of protein kinase phosphorylation and plays pivotal roles in regulating meiosis process in oocytes. As an important physical substrate of MAPK, p90rsk mediates numerous MAPK functions. MAPK was activated at G2/M transition during meiosis. Its activity reached the peak at M I stage and maintained at this level until the time before the pronuclear formation after fertilization. There is complex interplay between MAPK and MPF in the meiosis regulation. Furthermore, other intracel-lular signal transducers, such as cAMP, protein kinase C and protein phosphotase, ect., also regulated the activity of MAPK at different stages during meiosis in oocytes. 5Genomic footprints of Kam Sweet Rice domestication indicate possible migration routes of theDongpeople inChina andprovide resources for future rice breeding显示文摘The Dong people are one of China’s 55 recognized ethnic minorities,but there has been a long-standing debate about their origins.In this study,we performed whole-genome resequencing of Kam Sweet Rice(KSR),a valuable,rare,and ancient rice landrace unique to the Dong people.Through comparative genomic analyses of KSR and other rice landraces from south of the Yangtze River Basin in China,we provide evidence that the ancestors of the Dong people likely originated from the southeast coast of China at least 1000 years ago.Alien introgression and admixture in KSR demonstrated multiple migration events in the history of the Dong people.Genomic footprints of domestication demonstrated characteristics of KSR that arose from artificial selection and geographical adaptation by the Dong people.The key genes GS3,Hd1,and DPS1(related to agronomic traits)and LTG1 and MYBS3(related to cold tolerance)were identified as domestication targets,reflecting crop improvement and changes in the geographical environment of the Dong people during migration.A genome-wide association study revealed a candidate yield-associated gene,Os01g0923300,a specific haplotype in KSR that is important for regulating grain number per panicle.RNA-sequencing and quantitative reverse transcription-PCR results showed that this gene was more highly expressed in KSR than in ancestral populations,indicating that it may have great value in increasing yield potential in other rice accessions.In summary,our work develops a novel approach for studying human civilization and migration patterns and provides valuable genomic datasets and resources for future breeding of high-yield and climate-resilient rice varieties.Chunhui Liu Tianyi Wang Huicha Chen Xiaoding Ma Chengzhi Jiao Di Cui Bing Han Xiaobing Li Aixia Jiao Renchao Ruan Dayuan Xue Yanjie Wang Longzhi Han 2023Molecular Plant2023,16,2:3
6Microsatellite DNA analysis proves nucleus of interspecies reconstructed blastocyst coming from that of donor giant panda显示文摘A method for DNA isolation from early development of blastocyst and further analysis of nuclear and mitochondrial DNA was developed in present study. Total DNA was prepared from interspecies reconstructed blastocyst and a giant panda specific microsatellite locus g010 was successfully amplified. DNA sequencing of the PCR product showed that two sequences of reconstructed blastocysts are the same as that of positive control giant panda. 7Effects of ubiquitin-protea-some pathway on mouse sperm capacitation, acrosome reaction and in vitro fertilization显示文摘Chlortetracycline (CTC) fluorescence patterns were used to study changes in the patterns B and AR of mouse sperm after incubation with reagents that would block the UPP. They were the monoclonal antibody againstubiquitinated proteins——UCPi; the polyclonal antibodyagainst ubiquitin-anti-Ub, and a special inhibitor againstproteasome——ALLN. Furthermore, we treated the capaci-tated sperm or the eggs with these reagents separately and tested whether the normal in vitro fertilization was blocked or not. 8Serial nuclear transfer improves the development of interspecies reconstructed giant panda (Aluropoda melanoleucd) embryos显示文摘Interspecies somatic nuclear transfer (NT) may provide a new approach for preservation of the endangered rare species. Previous interspecies cloning studies have shown that a nucleus from a quiescent somatic cell supports early development of reconstructed embryos in the ooplasm from another species. In this study, we transferred nonqui-escent somatic cells from a giant panda into the perivitelline space of the enucleated rabbit oocytes. After electrofusion (at the rate of 71.6%) and electrical activation, 4.2% of the panda-rabbit reconstructed embryos developed to blastocyst in vitro. For improving the development rate of reconstructed embryos, we used serial NT in this study, i.e. blas-tomeres from reconstructed morulae were transferred into the perivitelline space of the enucleated rabbit oocytes. The fusion rates in the groups of serial I, serial Ⅱ and serial Ⅲ were 79.5%, 84.1% and 78.0%, respectively, having no difference with that of somatic group. 16MAPK regulates cell cycle progression in pig oocytes and fertilized eggs显示文摘Oocytes collected from prepubertal gilt ovaries were matured in vitro (IVM), and fertilized in vitro (IVF) or electrically activated. Phosphorylation of mitogen-activated protein kinase (MAPK) was detected with SDS-PAGE and Western blotting, and translocation of ERK2 was observed with immunofluorescent cytochemistry. We found that the quantity of MAPK kept unchanged during oocyte maturation. There was no phosphorylated MAPK in porcine oocytes at the germinal vesicle (GV) stage; a little MAPK was phosphorylated at 20 h of IVM; a high level phosphory-lation was detected at 30 h, while MAPK phosphorylation decreased at 36 h; and then MAPK phosphorylation increased again to the peak level from 40 to 60 h. ERK2 translocated from the peripheral cytoplasm to inner cytoplasm and nuclear area during oocyte maturation. There was nearly no phosphorylated MAPK at 18 and 20 h of electrically activated oocytes, but phosphorylation increased at 22 h. 19Interfamily pregnancy and expression of CD57, CD68 in deciduas between golden hamster and mouse显示文摘Pregnancy between different species is one of the key steps to interspecific somatic cell cloning. Although interspecific clone embryos have been constructed, they could not develop to birth after being transferred to recipi-ents. In order to clarify the mechanism of this phenomenon, interfamily pregnancy between golden hamste (Mesocricetus auratus) and mouse (Mus musculus) was studied. Co-culture results indicated that the adhesion ratios of golden hamster blastocysts on mouse uterine epithelia monolayer 12, 24, 48 and 72 h after co-culture were all significantly lower than those of mouse blastocysts. The outgrowth ratios of golden hamster blastocysts on mouse uterine epithelia monolayer 48, 72 h after co-culture were both significantly lower than those of mouse blastocysts (P < 0.01). Golden hamster抯 blastula could be implanted and develop to D 11 of pregnancy after being transferred to mouse uterus (the 7th day after embryo transfer). Compared to the transfer of mouse embryo to mouse uterus, the successful ratio of interfamily embryo transfer was lower and the bulk of fetus was smaller than that of intraspecific fetus. Compared to intraspecific preg-nancy of mouse, the remote decidual tissue of interfamily pregnancy on D8 is looser. At the same time, expressions of CD57 and CD 68 in remote deciduas were both higher than those in the secondary deciduas in both intraspecific and interfamily pregnancy. However, expressions of the two molecules in interfamily pregnancy were lower than those in intraspecific pregnancy. These results showed that interfam-ily pregnancy could be established between golden hamster and mouse. But the development of fetus in interfamily pregnancy was slower than that in intraspecific pregnancy.
