1Identification of a novel HMW glutenin subunit and comparison of its amino acid sequence with those of homologous subunits显示文摘Aegilops tauschii is the donor of the D genome of common wheat (Triticum aestivum). Genetic variation of HMW glutenin subunits encoded by the Glu-1Dt locus of Ae. tauschii has been found to be higher than that specified by the Glu-1D locus in common wheat. In the present note, we report the identification of a novel HMW glutenin subunit, Dy13t, from Ae. tauschii. The newly identified subunit pos-sessed an electrophoretic mobility that was faster than that of the Dy12 subunit of common wheat. The complete ORF of encoding the Dy13t subunit contained 624 codons (excluding the stop codons). The amino acid sequence deduced from the Dy13t gene ORF was the shortest among those of the previ-ously reported subunits derived by the D genome. 3Transgenic expression of MYB15 confers enhanced sensitivity to abscisic acid and improved drought tolerance in Arabidopsis thaliana显示文摘Abiotic stresses cause serious crop losses. Knowledge on genes functioning in plant responses to adverse growth conditions is essential for developing stress tolerant crops. Here we report that transgenic expression of MYB15, encoding a R2R3 MYB transcription factor in Arabidopsis thaliana, conferred hypersensitivity to exogenous abscisic acid (ABA) and improved tolerance to drought and salt stresses. The promoter of MYB15 was active in not only vegetative and reproductive organs but also the guard cells of stomata. Its transcript level was substantially upregulated by ABA, drought or salt treatments. Compared with wild type (WT) control, MYB15 overexpression lines were hypersensitive to ABA in germination assays, more susceptible to ABA-elicited inhibition of root elongation, and more sensitive to ABA-induced stomatal closure. In line with the above findings, the transcript levels of ABA biosynthesis (ABA1, ABA2), signaling (ABI3), and responsive genes (AtADH1, RD22, RD29B, AtEM6) were generally higher in MYB15 overexpression seedlings than in WT controls after treatment with ABA. MYB15 overexpression lines displayed improved survival and reduced water loss rates than WT control under water deficiency conditions. These overexpression lines also displayed higher tolerance to NaCl stress. 4A life support-based comprehensive treatment regimen dramatically lowers the in-hospital mortality of patients with fulminant myocarditis: a multiple center study显示文摘Fulminant myocarditis(FM) has unacceptable high mortality. This study aimed to evaluate the therapeutic efficacy of a life support-based comprehensive treatment regimen(LSBCTR), a completely novel treatment regimen, for FM. A total of 169 FM patients recruited from January 2008 to December 2018 were divided into two groups: patients receiving LSBCTR(81 cases),which includes(i) mechanical life support(positive pressure respiration, intra-aortic balloon pump with or without extracorporeal membrane oxygenation),(ii) immunomodulation therapy using sufficient doses of glucocorticoids and immunoglobulins, and(iii) application of neuraminidase inhibitors, and those receiving conventional treatment(88 cases). The endpoints were in-hospital death and heart-transplantation. Of all the population, 44 patients(26.0%) died in hospitals. Inhospital mortality was 3.7%(3/81) for LSBCTR group and 46.6%(41/88) for traditional treatment(P<0.001). Early application of LSBCTR, mechanical life support, neuraminidase inhibitors, and immunomodulation therapy significantly contributed to reduction in in-hospital mortality. This study describes a novel treatment regimen for FM patients that dramatically reduces inhospital mortality. 5Chinese expert consensus statement on the diagnosis and treatment of fulminant myocarditis显示文摘Fulminant myocarditis is primarily caused by infection with any number of a variety of viruses. It arises quickly, progresses rapidly, and may lead to severe heart failure or circulatory failure presenting as rapid-onset hypotension and cardiogenic shock,with mortality rates as high as 50%–70%. Most importantly, there are no treatment options, guidelines or an expert consensus statement. Here, we provide the first expert consensus, the Chinese Society of Cardiology Expert Consensus Statement on the Diagnosis and Treatment of Fulminant Myocarditis, based on data from our recent clinical trial(NCT03268642). In this statement, we describe the clinical features and diagnostic criteria of fulminant myocarditis, and importantly, for the first time,we describe a new treatment regimen termed life support-based comprehensive treatment regimen. The core content of this treatment regimen includes(i) mechanical life support(applications of mechanical respirators and circulatory support systems,including intraaortic balloon pump and extracorporeal membrane oxygenation),(ii) immunological modulation by using sufficient doses of glucocorticoid, immunoglobulin and(iii) antiviral reagents using neuraminidase inhibitor. 72018 update of expert consensus statement on antiplatelet therapy in East Asian patients with ACS or undergoing PCI显示文摘East Asians are the most populous race in the world and their health status is an important global issue.Compared with Caucasian populations, East Asian patients have a different benefit/risk ratio when using antithrombotic treatment. Despite this observation, treatment strategies in East Asian patients are mostly based on the American and European guidelines. Despite a lower platelet inhibitory response to clopidogrel, East Asian patients show a similar or even a lower rate of ischemic event occurrence and higher bleeding risk compared with Caucasian patients. For potent P2Y_(12) inhibitors(ticagrelor and prasugrel),East Asian patients have shown less favorable net clinical benefits compared with Caucasian patients,which may be related to differences in pharmacokinetic/pharmacodynamic profiles and therapeutic zone of antiplatelet effect. This updated consensus mainly focuses on state-of-the-art and current controversies in the East Asian population. 8Three-step semiquantum secure direct communication protocol显示文摘Quantum secure direct communication is the direct communication of secret messages without need for establishing a shared secret key first.In the existing schemes,quantum secure direct communication is possible only when both parties are quantum.In this paper,we construct a three-step semiquantum secure direct communication(SQSDC)protocol based on single photon sources in which the sender Alice is classical.In a semiquantum protocol,a person is termed classical if he(she)can measure,prepare and send quantum states only with the fixed orthogonal quantum basis{|0,|1}.The security of the proposed SQSDC protocol is guaranteed by the complete robustness of semiquantum key distribution protocols and the unconditional security of classical one-time pad encryption.Therefore,the proposed SQSDC protocol is also completely robust.Complete robustness indicates that nonzero information acquired by an eavesdropper Eve on the secret message implies the nonzero probability that the legitimate participants can find errors on the bits tested by this protocol.In the proposed protocol,we suggest a method to check Eves disturbing in the doves returning phase such that Alice does not need to announce publicly any position or their coded bits value after the photons transmission is completed.Moreover,the proposed SQSDC protocol can be implemented with the existing techniques.Compared with many quantum secure direct communication protocols,the proposed SQSDC protocol has two merits:firstly the sender only needs classical capabilities;secondly to check Eves disturbing after the transmission of quantum states,no additional classical information is needed.ZOU XiangFu QIU DaoWen 2014Science China(Physics,Mechanics & Astronomy)2014,57,9:12

14An experiment-library least-squares method on on-line coal element contents analysis显示文摘A new experiment-library least-squares(experiment-LLs) method used in the neutron inelastic-scattering and thermal-capture analysis(NITA) technique for on-line coal analysis was developed,which has significantly decreased the non-linear radiation effects.In this method,sixty samples with preset elemental contents were made,from whose experimental spectra twenty single-element spectrum libraries corresponding to twenty kinds of coal were built using the least-squares method.The spectrum of the unknown sample was analyzed based on these twenty libraries to estimate its element contents.With the initial estimated result,the procedure of developing library and analysis was iterated to build a new library with the closest element contents to the unknown sample.Hence the experiment-LLs method can reduce non-linear radiation effects.The experiment-LLs method was performed on an improved coal analysis system which was equipped with a long-life 14 MeV pulsed-neutron generator,a bulk BGO detector with a temperature controller,a moisture meter and coal-smoothing device.The precisions of this system for ash content,water content,volatile content and calorific value have reached 1.0wt%,0.5wt%,1.0wt%,350 kJ/kg,respectively.WANG HuiDong LU JingBin LU YuPing YANG Dong MA KeYan YANG Kang LIU YuMin CHENG DaoWen 2012Science China(Physics,Mechanics & Astronomy)2012,55,11:6
15HMW glutenin subunits in multiploid Aegilops species: composition analysis and molecular cloning of coding sequences显示文摘The Aegilops genus contains species closely related to wheat. In common with wheat, Aegilops species accumulate high molecular weight (HMW) glutenin subunits in their endospermic tissue. In this study, we investigated the composition of HMW glutenin subunits in four multiploid Aegilops species using SDS-PAGE analysis. Furthermore, by working with Ae. ventricosa, we established an efficient ge-nomic PCR condition for simultaneous amplification of DNA sequences coding for either x- or y-type HMW glutenin sub-units from polyploid Aegilops species. Using the genomic PCR condition, we amplified and subsequently cloned two DNA fragments that may code for HMW glutenin subunits in Ae. ventricosa. 17Assessment of the individual and combined effects of Rht8 and Ppd-D1a on plant height, time to heading and yield traits in common wheat显示文摘Grain yield in cereal crops is a complex trait controlled by multiple genes and influenced by developmental processes and environment. Here we report the effects of alleles Rht8 and Ppd-D1 a on plant height, time to heading, and grain yield and its component traits. Association analysis and quantitative trait locus mapping using phenotypic data from 15 environments led to the following conclusions. First, both Rht8 and Ppd-D1 a reduce plant height. However, Ppd-D1 a but not Rht8 causes earlier heading.Second, both Rht8 and Ppd-D1 a promote grain yield and affect component traits. Their combined effects are substantially larger than those conferred by either allele alone.Third, promotion of grain yield by Rht8 and Ppd-D1 a is through increasing fertile spikelet number. We speculate that Rht8 and Ppd-D1 a act independently and additively in control of plant height, grain yield and yield component. 18Pandemonium Breaks Out:Disruption of Salicylic Acid-Mediated Defense by Plant Pathogens显示文摘Salicylic acid (SA) or 2-hydroxybenoic acid is a phenolic plant hormone that plays an essential role in plant defense against biotrophic and semi-biotrophic pathogens. In Arabidopsis, SA is synthesized from chorismate in the chloroplast through the ICS1 (isochorismate synthase I) pathway during pathogen infectioru The transcription co-activator NPR1 (Non-Expresser of Pathogenesis-Related Gene 1), as the master regulator of SA signaling, interacts with transcription factors to induce the expression of anti-microbial PR (Pathogenesis-Related) genes. To establish successful in fections, plant bacterial, oomycete, fun gal, and viral pathogens have evolved at least three major strategies to disrupt SA-mediated defense? The first strategy is to reduce SA accumulation directly by converting SA into its inactive derivatives. The second strategy is to interrupt SA biosynthesis by targeting the ICS1 pathway. In the third major strategy, plant pathogens deploy different mechanisms to interfere with SA downstream signaling. The wide array of strategies deployed by plant pathogens highlights the crucial role of disruption of SA-mediated plant defense in plant pathogenesis. 19Nuclear miR-665 aggravates heart failure via suppressing phosphatase and tensin homolog transcription显示文摘Although numerous miRNAs have been discovered,their functions in the different subcellular organelles have remained obscure.In this study,we found that miR-665 was enriched in the nucleus of cardiomyocytes,and then investigated the underlying role of nuclear miR-665 in heart failure.RNA fluorescence in situ hybridization assays in human heart tissue sections and primary cardiomyocytes showed that miR-665 was localized in the nucleus of cardiomyocytes.Increased expression of nuclear miR-665 was observed not only in the cardiomyocytes isolated from the heart of mice treated in vivo by transverse aortic constriction(TAC),but also in phenylephrine(PE)-treated cultured cardiomyocytes in vitro.To further explore the role of miR-665 in heart failure,a type 9 recombinant adeno-associated virus(rAAV)system was employed to manipulate the expression of miR-665 in mice.Overexpression of miR-665 aggravated TAC-induced cardiac dysfunction,while down-expression of miR-665 showed opposite effects.Bioinformatic prediction and biological validation confirmed that the PTEN(phosphatase and tensin homolog)gene was one of the targets of miR-665 in the nucleus.Furthermore,restoring PTEN expression significantly eliminated the destructive effects of miR-665 over-expression in TAC-induced cardiac dysfunction.Our data showed that nuclear miR-665 aggravates heart failure via inhibiting PTEN expression,which provided a therapeutic approach for heart failure.Jiahui Fan Xudong Zhang Xiang Nie Huaping Li Shuai Yuan Beibei Dai Jiabing Zhan Zheng Wen Jiangang Jiang Chen Chen Daowen Wang 2020Science China(Life Sciences)2020,63,5:4

Wheat grain quality(WGQ) is central to food processing and nutritional value, and is a decisive factor for consumer acceptance and commercial value of wheat cultivars. Hence, improvement in WGQ traits is top priority for both conventional and molecular wheat breeding. In this review we will focus on two important WGQ traits, grain milling and end-use, and will summarize recent progress in China. Chinese scientists have invested substantial effort in molecular genetic and genomic analysis of these traits and their effects on end-use properties. The insights and resources generated have contributed to the understanding and improvement of these traits. As high-quality genomics information and powerful genome engineering tools are becoming available for wheat, more fundamental breakthroughs in dissecting the molecular and genomic basis of WGQ are expected.
