| 1 | Clinical significance of K-ras and BRAF mutations in Chinese colorectal cancer patients显示文摘AIM:To identify and assess mutations in the K-ras and BRAF genes in a cohort of Chinese patients with colorectal cancer (CRC) for their association with various clinicopathological parameters and prognosis.METHODS:Genomic DNA was isolated from frozen tissues.Pyrosequencing analysis was conducted to detect mutations in the K-ras (codons 12,13,and 61) and BRAF genes (codon 600).Statistical analysis was carried out using SPSS-15.0 software.RESULTS:Among the 118 colorectal cancer patients,we detected 41 (34.7%) mutations in the K-ras gene.Mutation frequencies at codon 12 and codon 13 were 23.7% (28/118) and 10.2% (12/118),respectively.Only one patient harbored a point mutation at codon 61 (0.8%,1/118).Gender was the only factor that showed an obvious relationship with K-ras gene mutation (female 44.7% vs male 28.2%,P=0.037).Other clinicopathological features,such as age,location of the tumor,tumor differentiation,Tumor,Node and Metastases classification,and the Union for International Cancer Control staging,showed no positive relationship with K-ras gene mutations.No significant correlation was observed between the presence of K-ras mutations (codons 12,13,and 61) and the survival of the patients.BRAF mutations were rare,and only two patients (1.7%) harbored a detectable mutation at codon 600.CONCLUSION:K-ras gene mutation is a common event in our 118 Chinese CRC patients,with an obvious relationship with gender.However,it seems not to be an independent prognostic factor in CRC patients.The BRAF gene is rarely mutated in Chinese CRC patients. | Hong Shen Ying Yuan Han-Guang Hu Xiao-Xian Ye Mo- Dan Li Xian Zhong Wei-Jia Fang Shu Zheng | 2011 | World Journal of Gastroenterology2011,17,6: | 29 |
| 2 | Compound Danshen injection improves endotoxin-induced microcirculatory disturbance in rat mesentery显示文摘AIM: To investigate the effect of compound Danshen injection on lipopolysaccharide (LPS)-induced rat mesenteric microcirculatory dysfunctions and the underlying possible mechanism by an inverted intravital microscope and high-speed video camera system. METHODS: LPS was continuously infused through the jugular artery of male Wistar rats at the dose of 2 mg/kg per hour. Changes in mesenteric microcirculation,such as diameters of arterioles and venules,velocity of RBCs in venules,leukocyte rolling,adhesion and emigration,free radicals released from post-capillary venules,FITC- albumin leakage and mast cell degranulation,were observed through an inverted intravital microscope assisted with CCD camera and SIT camera. Meanwhile,the expression of adhesion molecules CD11b/CD18 and the production of free radical in neutrophils,and the expression of intercellular adhesion molecule 1 (ICAM-1) in human umbilical vein endothelial cells (HUVECs) were quantified by flow cytometry (FACS) in vitro. RESULTS: The continuous infusion with LPS resulted in a number of responses in microcirculation,including a significant increase in the positive region of venulestained with Monastral blue B,rolling and adhesion of leukocytes,production of oxygen radical in venular wall,albumin efflux and enhanced mast cell degranulation in vivo,all of which,except for the leukocyte rolling,were attenuated by the treatment with compound Danshen injection. Experiments performed in vitro further revealed that the expression of CD11b/CD18 and the production of oxygen free radical in neutrophils,and the expression of ICAM-1 in HUVECs were increased by exposure to LPS,and they were attenuated by compound Danshen injection. | 3 | Adverse Effects of Exposure to Fine Particulate Matters and Ozone on Gestational Hypertension显示文摘Gestational hypertension(GH)is a common complication during pregnancy.GH is regarded as a potential public health challenge for pregnant women and infants.Limited evidence has linked ambient air pollution to an increased GH risk.However,most of the studies were conducted in developed countries,with inconsistent results obtained.The present study was performed to explore whether exposure to particulate matters with an aerodynamic diameter≤2.5(PM2.5)and ozone(O3)was related to elevated odds of GH in a Chinese population.This population-based cohort study involved 38115 pregnant women in Wuhan,China.All information was collected from the Wuhan Maternal and Child Health Management Information System,using standardized quality control.The daily air pollutant data for PM2.5 and O3 were obtained from the 20 monitoring stations of the Wuhan Environmental Monitoring Center during 2014.The nearest monitor approach was applied to individual exposure assessment of PM2.5 and O3 for each participant.After adjusting for major confounders and other air pollutants,a 10μg/m^3 increase in PM25 and O3 concentrations was found to correlate to a 1.14-fold[95%confidence interval(95%CI):1.09,1.20]and a 1.05-fold(95%CI:1.02,1.07)increase in GH risk,respectively.Additionally,stronger relationships between GH risk and PM25 and O3 exposure were observed in women who conceived in winter and summer,respectively.These findings suggest that air pollutants may contribute to the development of GH. | Rong YANG Dan LUO Yi-ming ZHANG Ke HU Zheng-min QIAN Li-qin HU Long-jiao SHEN Hong XIAN Juliet Iwelunmor Su-rong MEI | 2019 | Current Medical Science2019,39,6: | 7 |
| 4 | The Association of Socioeconomic Status with the Burden of Cataract-related Blindness and the Effect of Ultraviolet Radiation Exposure: An Ecological Study显示文摘Objective To assess the association of socioeconomic status with the burden of cataract blindness in terms of year lived with disability(YLD) rates and to determine whether ultraviolet radiation(UVR) levels modify the effect of socioeconomic status on this health burden.Methods National and subnational age-standardized YLD rates associated with cataract-related blindness were derived from the Global Burden of Disease(GBD) study 2017. The human development index(HDI) from the Human Development Report was used as a measure of socioeconomic status.Estimated ground-level UVR exposure was obtained from the Ozone Monitoring Instrument(OMI)dataset of the National Aeronautics and Space Administration(NASA).Results Across 185 countries, socioeconomic status was inversely associated with the burden of cataract blindness. Countries with a very high HDI had an 84% lower age-standardized YLD rate [95%confidence interval(CI): 60%–93%, P < 0.001] than countries with a low HDI;for high-HDI countries, the proportion was 76%(95% CI: 53%–88%, P < 0.001), and for medium-HDI countries, the proportion was48%(95% CI: 15%–68%, P = 0.010;P for trend < 0.001). The interaction analysis showed that UVR exposure played an interactive role in the association between socioeconomic status and cataract blindness burden(P value for interaction = 0.047).Conclusion Long-term high-UVR exposure amplifies the association of poor socioeconomic status with the burden of cataract-related blindness. | 15 | 采用常压室温等离子体诱变技术获得一株高产雄甾-4-烯-3,17-二酮的突变菌株Mycobacterium neoaurum ZADF-4(英文)显示文摘目的:获得一株高产雄甾-4-烯-3,17-二酮(AD)的Mycobacterium neoaurum突变株。创新点:获得了一株3-甾酮-Δ1-脱氢酶(KSDD)酶活缺陷型的高产AD的诱变菌株Mycobacterium neoaurum ZADF-4,并采用菌落显色法筛选KSDD酶活缺陷型M.neoaurum突变株。方法:(1)诱变方法:采用常压室温等离子体(ARTP)诱变技术来处理出发菌株M.neoaurum ZAD。ARTP诱变条件如下:功率40 W,气流量12.5 L/min,辐射距离1 cm,样品体积10μl,辐射时间为60、90、120、150和180 s;致死率统计优化后,最适辐射时间为150 s,致死率为90%~96%。(2)筛选方法:将ARTP诱变处理后的菌株点种在硝酸纤维滤膜上,30°C培养2 d,然后将长有菌落的滤膜小心取出并漂浮在4 mg/ml二氯靛酚(DCPIP)溶液(0.1 mmol/L磷酸缓冲液p H 7.0),30°C培养1 d直到全部菌落染成蓝色。然后将该滤膜取出,漂浮在250 mmol/L AD溶液(2%甲醇和50 mmol/L Tris p H 7.0缓冲液),室温放置15 min左右,观察菌落颜色变化。KSDD在底物AD存在时会脱氢产生雄甾-1,4-二烯-3,17-二酮(ADD)和H+,H+可以使被DCPIP染成蓝色的菌株褪色。因此,酶活缺陷型的菌株会仍保持蓝色,而酶活高的菌株会褪色为黄色(图3)。(3)对获得的潜在的高产AD菌株进行进一步的酶活检测以及产量验证,以期获得最优的突变株。结论:获得了4株具有潜在的高产AD能力的菌株,其中,最优的突变株ZADF-4的KSDD酶活相较于出发菌株ZAD下降了81.2%(图4),活性胶也证明其KSDD酶活相较于出发菌株下降明显(图5)。薄层色谱法(TLC)和高效液相色谱法(HPLC)实验证明突变株ZADF-4中,AD的产量有了明显的提高(图6和图7),提高到了(6.28±0.11)g/L,AD/ADD提高到8:1,AD的摩尔产率达到60.3%(表1)。对出发菌株ZAD和突变株ZADF-4的ksdd基因进行克隆和序列比对,发现ZADF-4的ksdd序列在5'端缺失9个核苷酸(atgttctac),导致3个氨基酸(MFY)的缺失;还发生了两个点突变,其中一个是无义突变(g.15a>6t),另一个是有义突变(g.413c>404t),并引起了相应位置上的氨基酸变化(p.138S>135L)。上述的基因突变及其引起的氨基酸序列的变化可能是引起M.neoaurum ZADF-4中KSDD酶活降低及AD产量提高的主要原因。 | Chao LIU Xian ZHANG Zhi-ming RAO Ming-long SHAO Le-le ZHANG Dan WU Zheng-hong XU Hui LI | 2015 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2015,16,4: | 1 |
| 16 | Independent variations in genome-wide expression, alternative splicing, and DNA methylation in brain tissues among castes of the buff-tailed bumblebee, Bombus terrestris显示文摘Caste differentiation in social hymenopterans is an intriguing example of phenotypic plasticity. However, the co-ordination among gene regulatory factors to mediate caste differentiation remains inconclusive. In this study, we determined the role of gene regulation and related epigenetic processes in pre-imaginal caste differentiation in the primitively eusocial bumblebee Bombus terrestris. By combining RNA-Seq data from Illumina and Pac Bio and accurately quantifying methylation at whole-genomic base pair resolution, we found that queens, workers, and drones mainly differentiate in gene expression but not in alternative splicing and DNA methylation. Gynes are the most distinct with the lowest global level of whole-genomic methylation and with the largest number of caste-specific transcripts and alternative splicing events. By contrast, workers exhibit few uniquely expressed or alternatively spliced genes. Moreover, several genes involved in hormone and neurotransmitter metabolism are related to caste differentiation, whereas several neuropeptides are linked with sex differentiation. Despite little genome-wide association among differential gene expression, splicing, and differential DNA methylation, the overlapped gene ontology(GO) terms point to nutrition-related activity. | 19 | First-principles study on the alkali chalcogenide secondary compounds in Cu(In,Ga)Se_2 and Cu_2ZnSn(S,Se)_4 thin film solar cells显示文摘The beneficial effect of the alkali metals such as Na and K on the Cu(In,Ga)Se_2(CIGS) and Cu_2ZnSn(S,Se)_4(CZTSSe) solar cells has been extensively investigated in the past two decades, however, in most of the studies the alkali metals were treated as dopants. Several recent studies have showed that the alkali metals may not only act as dopants but also form secondary phases in the absorber layer or on the surfaces of the films. Using the first-principles calculations, we screened out the most probable secondary phases of Na and K in CIGS and CZTSSe, and studied their electronic structures and optical properties. We found that all these alkali chalcogenide compounds have larger band gaps and lower VBM levels than CIGS and CZTSSe, because the existence of strong p-d coupling in CIS and CZTS pushes the valence band maximum(VBM) level up and reduces the band-gaps, while there is no such p-d coupling in these alkali chalcogenides. This band alignment repels the photo-generated holes from the secondary phases and prevents the electron-hole recombination. Moreover, the study on the optical properties of the secondary phases showed that the absorption coefficients of these alkali chalcogenides are much lower than those of CIGS and CZTSSe in the energy range of 0-3.4 e V, which means that the alkali chalcogenides may not influence the absorption of solar light.
