1Enhanced secondary pollution offset reduction of primary emissions during COVID-19 lockdown in China显示文摘To control the spread of the 2019 novel coronavirus(COVID-19),China imposed nationwide restrictions on the movement of its population(lockdown)after the Chinese New Year of 2020,leading to large reductions in economic activities and associated emissions.Despite such large decreases in primary pollution,there were nonetheless several periods of heavy haze pollution in eastern China,raising questions about the well-established relationship between human activities and air quality.Here,using comprehensive measurements and modeling,we show that the haze during the COVID lockdown was driven by enhancements of secondary pollution.In particular,large decreases in NOx emissions from transportation increased ozone and nighttime NO_(3)radical formation,and these increases in atmospheric oxidizing capacity in turn facilitated the formation of secondary particulate matter.Our results,afforded by the tragic natural experiment of the COVID-19 pandemic,indicate that haze mitigation depends upon a coordinated and balanced strategy for controlling multiple pollutants.Xin Huang Aijun Ding Jian Gao Bo Zheng Derong Zhou Ximeng Qi Rong Tang Jiaping Wang Chuanhua Ren Wei Nie Xuguang Chi Zheng Xu Liangduo Chen Yuanyuan Li Fei Che Nini Pang Haikun Wang Dan Tong Wei Qin Wei Cheng Weijing Liu Qingyan Fu Baoxian Liu Fahe Chai Steven JDavis Qiang Zhang Kebin He 2021National Science Review2021,8,2:47

5Correlation Between the Clinical Severity, Bacterial Load, and Inflammatory Reaction in Children with Mycoplasma Pneumoniae Pneumonia显示文摘Given the lack of defining features in the clinical manifestations and radiographic findings for children with mycoplasma pneumoniae pneumonia(MPP),quantitative polymerase chain reaction(qPCR)has become a useful diagnostic method.This study was performed to explore the relationship between the qPCR findings,clinical symptoms,and inflammatory markers in children with MPP.Four hundred children with MPP have been enrolled in this retrospective analysis.All clinical and analytical information,including mycoplasma pneumoniae(MP)PCR results,has been collected.Based on the PCR results,the patients were divided into groups with load values(copy number)<105(54 cases),2105 and<106(71 cases),2106 and<107(112 cases),>107 and<108(114 cases),and>108(49 cases).The clinical features(including symptoms and signs)and inflammatory indicators were compared among the groups.The incidence of high fever(above 39℃),thermal peak during the entire hospitalization period,fever duration,days of hospitalization,and plasma lactate dehydrogenase(LDH)levels were statistically correlated with the MP PCR load value in children with MPP.The analysis of relevance degree showed the correlative order as a thermal peak of hospitalization>duration of fever>period of hospitalization>LDH value>C-reactive protein value.The host immune response was significantly greater in the complication group than in the non-complication group.Chen ZHANG Qiao ZHANG Jie-lin DU Dan DENG Ye-lei GAO Cheng-lin WANG Hong-jie ZHAO Qian GUO Zhou FU Dai-yin TIAN 2020Current Medical Science2020,40,5:19
6Expansion and differentiation of human hepatocyte-derived liver progenitor-like cells and their use for the study of hepatotropic pathogens显示文摘The study of pathophysiological mecha nisms in huma n liver disease has bee n constrained by the in ability to expa nd primary hepatocytes in vitro while maintaining proliferative capacity and metabolic function. We and others have previously shown that mouse mature hepatocytes can be con verted to liver progenitor-like cells in vitro with defined chemical factors. Here we describe a protocol achieving efficient conversion of human primary hepatocytes into liver progenitor-like cells (HepLPCs) through delivery of developmentally relevant cues, including NAD +-dependent deacetylase SIRT1 signaling. These HepLPCs could be expanded sign ifica ntly duri ng in vitro passage. The expa nded cells can readily be con verted back into metabolically functional hepatocytes in vitro and upon tran splantati on in vivo. Under three-dime nsional culture conditions, differentiated cells gen erated from HepLPCs regai ned the ability to support infect io n or reactivatio n of hepatitis B virus (HBV). Our work dem on strates the utility of the con versi on betwee n hepatocyte and liver proge nitor-like cells for studying HBV biology and an tiviral therapies. 8Adsorption–desorption behavior of atrazine on agricultural soils in China显示文摘Adsorption and desorption are important processes that affect atrazine transport,transformation,and bioavailability in soils.In this study,the adsorption–desorption characteristics of atrazine in three soils(laterite,paddy soil and alluvial soil) were evaluated using the batch equilibrium method.The results showed that the kinetics of atrazine in soils was completed in two steps:a'fast' adsorption and a 'slow' adsorption and could be well described by pseudo-second-order model.In addition,the adsorption equilibrium isotherms were nonlinear and were well fitted by Freundlich and Langmuir models.It was found that the adsorption data on laterite,and paddy soil were better fitted by the Freundlich model;as for alluvial soil,the Langmuir model described it better.The maximum atrazine sorption capacities ranked as follows:paddy soil > alluvial soil > laterite.Results of thermodynamic calculations indicated that atrazine adsorption on three tested soils was spontaneous and endothermic.The desorption data showed that negative hysteresis occurred.Furthermore,lower solution pH value was conducive to the adsorption of atrazine in soils.The atrazine adsorption in these three tested soils was controlled by physical adsorption,including partition and surface adsorption.At lower equilibrium concentration,the atrazine adsorption process in soils was dominated by surface adsorption;while with the increase of equilibrium concentration,partition was predominant.Lin Yue ChengJun Ge Dan Feng Huamei Yu Hui Deng Bomin Fu 2017Journal of Environmental Sciences2017,29,7:16

10Expression of exogenous rat collagenase in vitro and in a rat model of liver fibrosis显示文摘AIM: The present study was conducted to test thehypothesis that the introduction of the collagenase geneinto tissue culture cells and into a rat model of liver fibrosiswould result in the expression of enzymatically active product.METHODS: FLAG-tagged full-length rat collagenase cDNAwas PCR amplified and cloned into a mammalian expressionvector. NIH3T3 cells were then transiently transfected withthis construct. Expression of exogenous collagenase mRNAwas assessed by RT-PCR, and the exogenous collagenasedetected by Western blotting using anti-FLAG monoclonalantibodyEnzymatic activity was detected by gelatinzymography. To determine the effects of exogenouscollagenase production in vivo, the construct was boundto glycosyl-poly-L-lysine and then transduced into rats thathad developed liver fibrosis as a result of CCI4 plus ethanoltreatment. The hepatic expression of the construct and itseffect on the formation of liver fibrosis were demonstratedusing RT-PCR and immunohistochemistry.RESULTS: It was found that exogenously expressed ratcollagenase mRNA could be detected in NTH3T3 cellsfollowing transfection. Enzymatic ally active collagenase couldalso be detected in the culture medium. The recombinantplasmid was also expressed in rat liver after in vivo genetransfer. Expression of exogenous rat collagenase correlatedwith decreased deposition of collagen types I and Ⅲ in thelivers of rats with experimentally induced liver fibrosis.CONCLUSION: The expression of active exogenous ratcollagenase could be achieved in vitro and in vivo. 11Effects of bowel rehabilitation and combined trophic therapy on intestinal adaptation in short bowel patients显示文摘AIM: To evaluate the effects of bowel rehabilitation and combined trophic therapy on intestinal adaptation in short bowel patients.METHODS: Thirty-eight patients with severe short-bowel syndrome (SBS) were employed in the present study, whose average length of jejunum-ileum was 35.8±21.2 cm. The TPN treatment was initiated early to attain positive nitrogen balance and prevent severe weight loss. The TPN composition was designated to be individualized and altered when necessary. Enteral feeding was given as soon as possible after resection and increased gradually. Meals were distributed throughout the day. Eight patients received treatment of growth hormone (0.14 mg/kg.day) and glutamine (0.3 g/kg.day) for 3 weeks. D-xylose test, 15N-Gly trace test and 13C-palmitic acid breath test were done to determine the patients' absorption capability.RESULTS: Thirty-three patients maintained well body weight and serum albumin concentration. The average time of follow-up for 33 survival patients was 5.9±4.3 years.Twenty-two patients weaned from TPN with an average TPN time of 9.5±6.6 months. Two patients, whose whole small bowel, ascending and transverse colon were resected received home TPN. An other 9 patients received parenteral or enteral nutritional support partly as well as oral diet. Three week rhGH+GLN therapy increased nutrients absorption but the effects were transient.CONCLUSION: By rehabilitation therapy, most short bowel patients could wean from parenteral nutrition. 13Emodin alleviates cardiac fibrosis by suppressing activation of cardiac fibroblasts via upregulating metastasis associated protein 3显示文摘Excess activation of cardiac fibroblasts inevitably induces cardiac fibrosis. Emodin has been used as a natural medicine against several chronic diseases. The objective of this study is to determine the effects of emodin on cardiac fibrosis and the underlying molecular mechanisms. Intragastric administration of emodin markedly decreased left ventricular wall thickness in a mouse model of pathological cardiac hypertrophy with excess fibrosis induced by transaortic constriction(TAC) and suppressed activation of cardiac fibroblasts induced by angiotensin II(AngII). Emodin upregulated expression of metastasis associated protein 3(MTA3) and restored the MTA3 expression in the setting of cardiac fibrosis. Moreover, overexpression of MTA3 promoted cardiac fibrosis;in contrast, silence of MTA3 abrogated the inhibitory effect of emodin on fibroblast activation. 15MicroTom Metabolic Network: Rewiring Tomato Metabolic Regulatory Network throughout the Growth Cycle显示文摘Tomato(Solanum lycopersicum)is a major horticultural crop worldwide and has emerged as a preeminent model for metabolic research.Although many research efforts have focused on the analysis of metabolite differences between varieties and species,the dynamics of metabolic changes during the tomato growth cycle and the regulatory networks that underlie these changes are poorly understood.In this study,we integrated high-resolution spatio-temporal metabolome and transcriptome data to systematically explore the metabolic landscape across 20 major tomato tissues and growth stages.In the resulting MicroTom Metabolic Network,the 540 detected metabolites and their co-expressed genes could be divided into 10 distinct clusters based on their biological functions.Using this dataset,we constructed a global map of the major metabolic changes that occur throughout the tomato growth cycle and dissected the underlying regulatory network.In addition to verifying previously well-established regulatory networks for important metabolites,we identified novel transcription factors that regulate the biosynthesis of important secondary metabolites such as steroidal glycoalkaloids and flavonoids.Our findings provide insights into spatiotemporal changes in tomato metabolism and generate a valuable resource for the study of metabolic regulatory processes in model plants.Yan Li Yang Chen Lu Zhou Shengjie You Heng Deng Ya Chen Saleh Alseekh Yong Yuan Rao Fu Zixin Zhang Dan Su Alisdair R.Fernie Mondher Bouzayen Tao Ma Mingchun Liu Yang Zhang 2020Molecular Plant2020,13,8:10
16Generation of GGTA1 biallelic knockout pigs via zinc-finger nucleases and somatic cell nuclear transfer显示文摘Genetically modified pigs are valuable models of human disease and donors of xenotransplanted organs.Conventional gene targeting in pig somatic cells is extremely inefficient.Zinc-finger nuclease(ZFN)technology has been shown to be a powerful tool for efficiently inducing mutations in the genome.However,ZFN-mediated targeting in pigs has rarely been achieved.Here,we used ZFNs to knock out the porcineα-1,3-galactosyl-transferase(GGTA1)gene,which generates Gal epitopes that trigger hyperacute immune rejection in pig-to-human transplantation.Primary pig fibroblasts were transfected with ZFNs targeting the coding region of GGTA1.Eighteen mono-allelic and four biallelic knockout cell clones were obtained after drug selection with efficiencies of 23.4%and 5.2%,respectively.The biallelic cells were used to produce cloned pigs via somatic cell nuclear transfer(SCNT).Three GGTA1 null piglets were born,and one knockout primary fibroblast cell line was established from a cloned fetus.Gal epitopes on GGTA1 null pig cells were completely eliminated from the cell membrane.Functionally,GGTA1 knockout cells were protected from complement-mediated immune attacks when incubated with human serum.This study demonstrated that ZFN is an efficient tool in creating gene-modified pigs.GGTA1 null pigs and GGTA1 null fetal fibroblasts would benefit research and pig-to-human transplantation.BAO Lei CHEN HaiDe JONG UiMyong RIM CholHo LI WenLing LIN XiJuan ZHANG Dan LUO Qiong CUI Chun HUANG HeFeng ZHANG Yan XIAO Lei FU ZhiXin 2014Science China(Life Sciences)2014,57,2:9

18Trends in the Different Grades of Precipitation over South China during 1960–2010 and the Possible Link with Anthropogenic Aerosols显示文摘Using observed daily precipitation data to classify five levels of rainy days by strength in South China(SC), with an emphasis on the Pearl River Delta(PRD) region, the spatiotemporal variation of different grades of precipitation during the period 1960–2010 was analyzed and the possible link with anthropogenic aerosols examined. Statistical analysis showed that drizzle and small precipitation has significantly decreased, whereas medium to heavy precipitation has increased slightly over the past 50 years(although not statistically significant). Further data analysis suggested that the decline in drizzle and small precipitation probably has a strong link to increased concentrations of anthropogenic aerosols produced by large-scale human activities related to the rapid socioeconomic development of the PRD region. 20SIMULATIONS OF THE THREE-DIMENSIONAL TOTAL DISSOLVED GAS SATURATION DOWNSTREAM OF SPILLWAYS UNDER UNSTEADY CONDITIONS显示文摘Dams are often operated in a way to discharge over the spillways,which would cause a high dissolved concentration of air and be harmful to fish.The bubble transfer and the water surface transfer play an important role in affecting the concentration of the Total Dissolved Gas(TDG).Based on recent numerical simulations of the total dissolved gas saturation,in this article,a two-phase TDG transport equation is adopted to develop an unsteady three-dimensional(3-D) two-phase flow Computational Fluid Dynamics(CFD) model,including a number of parameters such as water depth,pressure and air volume entrained.This model is used to predict the hydrodynamics and the TDG distribution under unsteady discharge conditions.Good agreement between measured and numerical results is obtained for a case study.FU Xiao-li LI Dan ZHANG Xiao-feng 2010Journal of Hydrodynamics2010,22,4:8
