Anthropogenic emission inventories in China:a review显示文摘The development of reliable anthropogenic emission inventories is essential for both understanding the sources of air pollution and designing effective air-pollution-control measures in China. However, it is challenging to quantify emissions in China accurately, given the variety of contributing sources, the complexity of the technology mix and the lack of reliable measurements. Over the last two decades, tremendous efforts have been made to improve the accuracy of emission inventories, and significant improvements have been realized. More reliable statistics and survey-based data have been used to reduce the uncertainties in activity rates and technology distributions. Local emission factors and source profiles covering various sources have been measured and reported. Based on these local databases, improved emission inventory models have been developed for power plants, large industrial plants and the residential, transportation and agricultural sectors. In this paper, we review the progress that has been made in developing inventories of anthropogenic emissions in China. We first highlight the major updates that have been made to emission inventory models and the underlying data by source category. We then summarize the sector-based estimates of emissions of different species contained in current inventories. The progress that has been made in the development of model-ready emissions is also presented. Finally, we suggest future directions for further improving the accuracy of emission inventories in China. | Meng Li Huan Liu Guannan Geng Chaopeng Hong Fei Liu Yu Song Dan Tong Bo Zheng Hongyang Cui Hanyang Man Qiang Zhang Kebin He | 2017 | National Science Review2017,4,6: | 100

Single-cell RNA-seq highlights intra-tumoral heterogeneity and malignant progression in pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer featured with high intra-tumoral heterogeneity and poor prognosis. To comprehensively delineate the PDAC intra-tumoral heterogeneity and the underlying mechanism for PDAC progression, we employed single-cell RNA-seq (scRNA-seq) to acquire the transcriptomic atlas of 57,530 individual pancreatic cells from primary PDAC tumors and control pancreases, and identified diverse malignant and stromal cell types, including two ductal subtypes with abnormal and malignant gene expression profiles respectively, in PDAC. We found that the heterogenous malignant subtype was composed of several subpopulations with differential proliferative and migratory potentials. Cell trajectory analysis revealed that components of multiple tumor-related pathways and transcription factors (TFs) were differentially expressed along PDAC progression. Furthermore, we found a subset of ductal cells with unique proliferative features were associated with an inactivation state in tumor-infiltrating T cells, providing novel markers for the prediction of antitumor immune response. Together, our findings provide a valuable resource for deciphering the intra-tumoral heterogeneity in PDAC and uncover a connection between tumor intrinsic transcriptional state and T cell activation, suggesting potential biomarkers for anticancer treatment such as targeted therapy and immunotherapy. | Junya Peng Bao-Fa Sun Chuan-Yuan Chen Jia-Yi Zhou Yu-Sheng Chen Hao Chen Lulu Liu Dan Huang Jialin Jiang Guan-Shen Cui Ying Yang Wenze Wang Dan Guo Menghua Dai Junchao Guo Taiping Zhang Quan Liao Yi Liu Yong-Liang Zhao Da-Li Han Yupei Zhao Yun-Gui Yang Wenming Wu | 2019 | Cell Research2019,29,9: | 24
Modified Da-chai-hu Decoction regulates the expression of occludin and NF-κB to alleviate organ injury in severe acute pancreatitis rats显示文摘Modified Da-chai-hu Decoction(MDD), a traditional Chinese medicinal formulation, which was empirically generated from Da-chai-hu decoction, has been utilized to treat severe acute pancreatitis(SAP) for decades. The aim of the present study was to explore its potential organprotective mechanism in SAP. In the present study, rat SAP model was induced by retrograde injection of 3.5% sodium taurocholate into the biliopancreatic duct, MDD(23.35 g/kg body weight, twelve times the clinical dose) were orally given at 2 h before and 10 h after injection. At 12 h after model induction, blood was taken from vena cava for analysis of amylase, diamine oxidase(DAO), pulmonary surfactant protein-A(SP-A), and C-reactive protein(CRP). Histopathological change of pancreas, ileum and lung was assayed by H&E staining, myeloperoxidase(MPO) activity were determinated using colorimetric assay, and the expressions of occludin and nuclear factor-κB(NF-κB) were detected by real-time RT-PCR and western blot, respectively. In addition, the tissue concentrations of tumor necrosis factor-α(TNF-α), interleukin-1β(IL-1β), and monocyte chemoattractant protein-1(MCP-1) were measured by enzyme-linked immunosorbent assay(ELISA). The results showed that in SAP rats, MDD significantly alleviated histopathological damage, depressed the MPO activity and the concentrations of TNF-α, IL-1β, and MCP-1 of pancreas, ileum and lung, and reduced the serum levels of amylase [(3283.4±585.5) U·L^(-1) vs(5626.4±795.1) U·L^(-1)], DAO[(1100.1±334.3) U·L^(-1) vs(1666.4±525.3) U·L^(-1)] and CRP [(7.6±1.2) μg·mL^(-1) vs(17.8±3.8) μg·mL^(-1)]. However, the serum SP-A concentration [(106.1±16.6) pg·mL^(-1) vs(90.1±14.9) pg·mL^(-1)] was elevated when treated SAP rats with MDD. Furthermore, MDD increased the occludin expression and reduced the NF-κB expression in pancreas, ileum and lung of SAP rats. Our findings suggested that MDD administration was an effective therapeutic approach for SAP treatment. It could up-regulate occludin expression to protect intercellular tight junction and down-regulate NF-κB expression to inhibit inflammatory reaction of pancreas, ileum and lung. | ZHAO Guang ZHUO Yu-Zhen CUI Li-Hua LI Cai-Xia CHEN Sha-Yan LI Dan LIU Jun-Hong LI Di-Hua CUI Nai-Qiang ZHANG Shu-Kun | 2019 | Chinese Journal of Natural Medicines2019,17,5: | 23
Pathways of China's PM;air quality 2015–2060 in the context of carbon neutrality显示文摘Clean air policies in China have substantially reduced particulate matter(PM;) air pollution in recent years, primarily by curbing end-of-pipe emissions. However, reaching the level of the World Health Organization(WHO) guidelines may instead depend upon the air quality co-benefits of ambitious climate action. Here,we assess pathways of Chinese PM;air quality from 2015 to 2060 under a combination of scenarios that link global and Chinese climate mitigation pathways(i.e.global 2℃-and 1.5℃-pathways,National Determined Contributions(NDC) pledges and carbon neutrality goals) to local clean air policies. We find that China can achieve both its near-term climate goals(peak emissions) and PM;air quality annual standard(35 μg/m;) by 2030 by fulfilling its NDC pledges and continuing air pollution control policies. However, the benefits of end-of-pipe control reductions are mostly exhausted by 2030,and reducing PM;exposure of the majority of the Chinese population to below 10 μg/m;by 2060 will likely require more ambitious climate mitigation efforts such as China’ s carbon neutrality goals and global1.5℃-pathways.Our results thus highlight that China’s carbon neutrality goals will play a critical role in reducing air pollution exposure to the level of the WHO guidelines and protecting public health. | Jing Cheng Dan Tong Qiang Zhang Yang Liu Yu Lei Gang Yan Liu Yan Sha Yu Ryna Yiyun Cui Leon Clarke Guannan Geng Bo Zheng Xiaoye Zhang Steven JDavis Kebin He | 2021 | National Science Review2021,8,12: | 23
Soil Bacterial Communities Under Different Long-Term Fertilization Regimes in Three Locations Across the Black Soil Region of Northeast China显示文摘Although soil bacteria play critical roles in agro-ecosystems, the knowledge of their response to long-term fertilization across the black soil region of Northeast China is limited. In this study, we sequenced 16 S rRNA genes to assess the effects of four long-term fertilization regimes—non-fertilization(NoF), chemical fertilizer(CF), manure(M), and chemical fertilizer plus manure(CFM)—on soil properties and bacterial communities in three locations, the northern, middle, and southern parts, across the black soil region. Results showed that the influence of fertilization regimes on soil properties varied significantly among the three locations. Manure fertilization significantly increased microbial biomass carbon and relative abundance of copiotrophic bacteria. Principal component analysis(PCA)showed that the total bacterial communities were separated into three groups according to the sampling location despite long-term fertilization, and that soil pH was the most important factor in shifting bacterial communities. In addition, similar fertilization regimes resulted in different influences on bacterial community composition, and the most influential soil properties varied among the three locations. Our results highlighted that geographical separation was a more dominant factor affecting bacterial communities than fertilization, and that long-term similar fertilization regimes did not induce consistent changes in bacterial community composition in the black soil region. | HU Xiaojing LIU Junjie WEI Dan ZHU Ping CUI Xi'an ZHOU Baoku CHEN Xueli JIN Jian LIU Xiaobing WANG Guanghua | 2018 | Pedosphere2018,28,5: | 17
USP19 suppresses inflammation and promotes M2-like macrophage polarization by manipulating NLRP3 function via autophagy显示文摘Macrophage polarization to proinflammatory M1-like or anti-inflammatory M2-like cells is critical to mount a host defense or repair tissue. The exact molecular mechanisms controlling this process are still elusive. Here,we report that ubiquitin-specific protease 19(USP19)acts as an anti-inflammatory switch that inhibits inflammatory responses and promotes M2-like macrophage polarization.USP19 inhibited NLRP3 inflammasome activation by increasing autophagy flux and decreasing the generation of mitochondrial reactive oxygen species. In addition,USP19 inhibited the proteasomal degradation of inflammasome-independent NLRP3 by cleaving its polyubiquitin chains.USP19-stabilized NLRP3 promoted M2-like macrophage polarization by direct association with interferon regulatory factor 4,thereby preventing its p62-mediated selective autophagic degradation.Consistent with these observations, compared to wild-type mice,Usp19−/−mice had decreased M2-like macrophage polarization and increased interleukin-1βsecretion,in response to alum and chitin injections. Thus,we have uncovered an unexpected mechanism by which USP19 switches the proinflammatory function of NLRP3 into an anti-inflammatory function, and suggest that USP19 is a potential therapeutic target for inflammatory interventions. | Tao Liu Liqiu Wang Puping Liang Xiaojuan Wang Yukun Liu Jing Cai Yuanchu She Dan Wang Zhi Wang Zhiyong Guo Samuel Bates Xiaojun Xia Junjiu Huang Jun Cui | 2021 | Cellular & Molecular Immunology2021,18,10: | 15
Clinical significance of HOTAIR expression in colon cancer显示文摘AIM: To detect the expression of the long noncoding RNA HOTAIR in colon cancer and analyze its relationship with clinicopathological parameters of colon cancer. METHODS: Total RNA was extracted from 80 colon cancer tissues and matched tumor-adjacent normal colon tissues and reverse transcribed. Quantitative polymerase chain reaction was used to detect the expression of HOTAIR. The relationship between the expression of HOTAIR and clinicopathological parameters of colon cancer was analyzed. RESULTS: The expression of HOTAIR was significantly higher in colon cancer tissues than in matched tumoradjacent normal colon tissues(P < 0.05). HOTAIR expression was significantly higher in cases with lymph node metastasis than in those without metastasis; in lowly differentiated and undifferentiated cases than in highly and moderately differentiated cases; and in stages Ⅲ + Ⅳ cases than in stages?Ⅰ?+ Ⅱ cases(P < 0.05).CONCLUSION: HOTAIR expression is upregulated in colon cancer, suggesting that HOTAIR plays an important role in the tumorigenesis, development and metastasis of colon cancer. HOTAIR may act as an oncogene and represents a new molecular target for the treatment of colon cancer. | Zhi-Fen Luo Dan Zhao Xi-Qing Li Yong-Xia Cui Ning Ma Chuang-Xin Lu Ming-Yue Liu Yun Zhou | 2016 | World Journal of Gastroenterology2016,22,22: | 14
Propofol Attenuates a-Synuclein Aggregation and Neuronal Damage in a Mouse Model of Ischemic Stroke显示文摘a-Synuclein is a soluble monomer abundant in the central nervous system. Aggregates of a-synuclein,consisting of higher-level oligomers and insoluble fibrils, have been observed in many chronic neurological diseases and are implicated in neurotoxicity and neurodegeneration.a-Synuclein has recently been shown to aggregate following acute ischemic stroke, exacerbating neuronal damage.Propofol is an intravenous anesthetic that is commonly used during intravascular embolectomy following acute ischemic stroke. While propofol has demonstrated neuroprotective properties following brain injury, the mechanism of protection in the setting of ischemic stroke is unclear. In this study, propofol administration significantly reduced the neurotoxic aggregation of a-synuclein, decreased the infarct area, and attenuated the neurological deficits after ischemic stroke in a mouse model. We then demonstrated that the propofol-induced reduction of a-synuclein aggregation was associated with increased mammalian target of rapamycin/ribosomal protein S6 kinase beta-1 signaling pathway activity and reduction of the excessive autophagy occurring after acute ischemic stroke. | Yuzhu Wang Dan Tian Changwei Wei Victoria Cui Huan Wang Yanbing Zhu Anshi Wu Yun Yue | 2020 | Neuroscience Bulletin2020,36,3: | 14
Contribution of elF-4E inhibition to the expression and activity of heparanase in human colon adenocarcinoma cell line:LS-174T显示文摘AIM: Heparanase degrades heparan sulfate proteoglycans (HSPGs) and is a critical mediator of tumor metastasis and angiogenesis. Recently, it has been cloned as a single gene family and found to be a potential target for antimetastasis drugs. However, the molecular basis for the regulation of heparanase expression is still not quite clear. The aim of this study was to determine whether the expression of eukaryotic initiation factor 4E (eIF-4E) correlated with the heparanase level in tumor cells and to explore the correlation between heparanase expression and metastatic potential of LS- 174T cells.METHODS: A 20-met antisense s-oligodeoxynucleotide (asODN) targeted against the translation start site of eIF-4E mRNA was introduced into LS-174T cells by lipid-mediated DNA-transfection. eIF-4E protein and mRNA levels were detected by Western blot analysis and RT-PCR, respectively.Heparanase activity was defined as the ability to degrade high molecular weight (40-100 kDa) radiolabeled HS (heparan sulfate) substrate into low molecular weight (5-15 kDa) HS fragments that could be differentiated by gel filtration chromatography. The invasive potential of tumor cell in vitro was observed by using a Matrigel invasion assay system.RESULTS: The 20-mer asODN against eIF-4E specifically and significantly inhibited eIF-4E expression at both transcriptional and translational levels. As a result, the expression and activity of heparanase were effectively retarded and the decreased activity of heparanase resulted in the decreased invasive potential of LS-174T.CONCLUSION: eIF-4E is involved in the regulation of heparanase production in colon adenocarcinoma cell line LS-174T, and its critical function makes it a particularly interesting target for heparanase regulation. | 14 | Generation of GGTA1 biallelic knockout pigs via zinc-finger nucleases and somatic cell nuclear transfer显示文摘Genetically modified pigs are valuable models of human disease and donors of xenotransplanted organs.Conventional gene targeting in pig somatic cells is extremely inefficient.Zinc-finger nuclease(ZFN)technology has been shown to be a powerful tool for efficiently inducing mutations in the genome.However,ZFN-mediated targeting in pigs has rarely been achieved. Here,we used ZFNs to knock out the porcineα-1,3-galactosyl-transferase(GGTA1)gene,which generates Gal epitopes that trigger hyperacute immune rejection in pig-to-human transplantation.Primary pig fibroblasts were transfected with ZFNs targeting the coding region of GGTA1.Eighteen mono-allelic and four biallelic knockout cell clones were obtained after drug selection with efficiencies of 23.4%and 5.2%,respectively.The biallelic cells were used to produce cloned pigs via somatic cell nuclear transfer(SCNT).Three GGTA1 null piglets were born, and one knockout primary fibroblast cell line was established from a cloned fetus.Gal epitopes on GGTA1 null pig cells were completely eliminated from the cell membrane.Functionally,GGTA1 knockout cells were protected from complement-mediated immune attacks when incubated with human serum. This study demonstrated that ZFN is an efficient tool in creating gene-modified pigs.GGTA1 null pigs and GGTA1 null fetal fibroblasts would benefit research and pig-to-human transplantation. | BAO Lei CHEN HaiDe JONG UiMyong RIM CholHo LI WenLing LIN XiJuan ZHANG Dan LUO Qiong CUI Chun HUANG HeFeng ZHANG Yan XIAO Lei FU ZhiXin | 2014 | Science China(Life Sciences)2014,57,2: | 9

Injections of ginkgo in the treatment of cerebral infarction: a systematic review and network Meta-analysis显示文摘OBJECTIVE: To assess the clinical effectiveness and safety of injections of ginkgo(GI) combined with Western Medicine(WM) for cerebral infarction(CI).METHODS: Randomized controlled trials(RCTs) of CI treated by GI were searched in China National Knowledge Infrastructure Database, Wanfang, China Science and Technology Journal Database, Web of Science, Cochrane library, Embase, Pub Med and Chinese Biomedical Literature Database, with the publication data no later than April, 2016. The Cochrane risk of bias method was used to evaluate the methodological quality of the RCTs. The data were analyzed by Review Manager 5.3, Stata 13.0,and Win BUGS 14 software.RESULTS: Totally 37 RCTs involving 4330 patientswere included. By direct comparison, the results of GI group were significantly superior to the routine WM group in the total effective rates [OR = 3.61,95% CI(2.93, 4.44), P < 0.0001], the neural function defect score(NFDS) [MD =-4.39, 95% CI(-5.47,-3.32), P < 0.0001]. Network Meta-analysis(NMA)results showed that, between 5 GIs in efficacy, the difference comparing ginaton injections(Gb E) to ginkgo-dipyidamolum injections(GD) [OR = 1.74,95% CI(0.73, 3.65)], shuxuening injections(SXN)[OR = 1.06, 95% CI(0.609, 1.697)] or ginkgolides injections(GK) [OR = 4.711, 95% CI(1.178, 13.21)]reach statistical significance; the difference comparing GD to GK reach statistical significance [OR =2.791, 95% CI(0.866, 6.908)]; the difference comparing SXN to GK reach statistical significance[OR = 4.537, 95% CI(1.203, 12.41)]. Besides, there was no difference between 4 GIs in NFDS. Probability ranking result showed a great possibility for GK [Surface under the Cumulative Ranking curve(SUCRA) = 80.3%] in improving the total effective rates, which were followed by GD(SUCRA =73.34%), SXN(SUCRA = 46.59%), Gb E(SUCRA =45.46%), floium ginkgo extract and tertram ethypyrazine sodium chloride injections(FT)(SUCRA = 35.64%). However, GK(SUCRA = 80.3%) or Gb E(SUCRA = 69.4%) was better than other GIs in reducing NFDS.GK + WM is the best treatment measures to reduce NFDS in cerebral infarction, which were followed by SXN + WM(SUCRA =51.6%), GD + WM(SUCRA = 48.1%).CONCLUSION: GIs was more effectiveness on CI than the routine Western Medicine. But based on the limitations of the study, more high-quality randomized controlled trials will be necessary. | Tan Di Wu Jiarui Liu Shi Zhang Dan Cui Yingying Zhang Xiaomeng Zhang Bing | 2018 | Journal of Traditional Chinese Medicine2018,38,1: | 7

Methods: RCTs concerning AP treated by GD were searched in China Biology Medicine Disc(SinoMed), PubMed, the China National Knowledge Infrastructure Database(CNKI), the Chinese Scientific Journals Database(VIP), Wanfang Database, Embase, and the Cochrane Library, from inception to February, 2017. The Cochrane Risk Assessment Tool was adopted to assess the methodological quality of the RCTs. The Review Manager 5.3 software was utilized to conduct the meta-analysis. Results: A total of 41 RCTs involving 4,462 patients were included in the meta-analysis. The results indicated that the combined use of GD and Western medicine(WM) against AP was associated with a higher total effective rate [risk ratio(RR)=1.25, 95% confidence interval(CI): 1.21–1.29, P<0.01], total effective rate of electrocardiogram(RR=1.29, 95% CI: 1.21–1.36, P<0.01). Additional, GD combined with WM could decrease the level of plasma viscosity [mean difference(MD)=–0.56, 95% CI: –0,81 to –0.30, P<0.01], fibrinogen [MD=–1.02, 95% CI: –1.50 to –0.54, P<0.01], whole blood low shear viscosity [MD=–2.27, 95% CI: –3.04 to –1.49, P<0.01], and whole blood high shear viscosity(MD=–0.90, 95% CI: 1.37 to –0.44, P<0.01). Conclusions: Comparing with receiving WM only, the combine use of GD and WM was associated with a better curative effect for patients with AP. Nevertheless, limited by the methodological quality of included RCTs more large-sample, multi-center RCTs were needed to confirm our findings and provide further evidence for the clinical utility of GD. | TAN Di WU Jia-rui CUI Ying-ying ZHAO Yi ZHANG Dan LIU Shi ZHANG Bing | 2018 | Chinese Journal of Integrative Medicine2018,24,12: | 7 |
Enterogenous infection of Candida albicans in immunocompromised rats under severe acute pancreatitis显示文摘BACKGROUND:Opportunistic infection of Candida albicans(C.albicans) has become a serious problem in immunocompromised patients. The study aimed to explore the mechanism of enterogenous infection of C.albicans in immunocompromised rats under severe acute pancreatitis(SAP).METHODS:Sprague Dawley(SD) rats(n=100) were randomly assigned into 5 groups as the following:blank group, cyclophosphamide+ceftriaxone+SAP group, cyclophosphamide+ceftriaxone group, cyclophosphamide+SAP group, and cyclophosphamide group. The rats were sacrificed at 5and 10 days, and their jejunum, colon, mesenteric lymph nodes, pancreas, intestinal content, and blood were quickly collected to detect C.albicans.A region of the 25 S rRNA gene was chosen and amplified by polymerase chain reaction(PCR) to differentiate C.albicans genotypes. The amplified products were further sequenced and compared to judge their homology.RESULTS:Compared with the Cyclophosphamide group, the combination of immunosuppressants and broad-spectrum antibiotics significantly increased the colonization of C.albicans in intestine in 5 and 10 days.Pure SAP stress did not increase the opportunistic infection of C.albicans.The PCR products of C.albicans isolates all belonged to the genotype A family, and sequence alignment showed that the amplified fragments were homologous.CONCLUSION:The damage of immune system and broad-spectrum antimicrobial agents are important risk factors for opportunistic fungal infection.Intestinal tract is an important source for genotype-A C.albicans to translocate and invade into bloodstream. | Xiang-wang Zhao Lei Yan Dan Xu Yu-hui Cui Chun-hui Yang Yan-jun Zhou Jian-guo Tang | 2016 | World Journal of Emergency Medicine2016,7,4: | 7
