| 1 | Helicobacter pylori and EBV in gastric carcinomas:Methylation status and microsatellite instability显示文摘AIM:To verify the methylation status of CDH1, DAPK, COX2, hMLH1 and CDKN2A genes and to evaluate their association with Helicobacter pylori (H. pylori)-cagA+ and Epstein Barr virus (EBV) infections in gastric adenocarcinomas.METHODS: Methylation-specific PCR (MSP) assay was performed in 89 primary gastric carcinomas (intestinal and diffuse types). Microsatellite instability (MSI) analysis was performed using the BAT26 primer set and PCR products were analyzed with the ABI PRISM 3100 Genetic Analyzer using Genescan 3.7 software (Applied Biosystems). Detection of H. pylori and genotyping were performed by PCR, using specifi c primers for ureaseC and cagA genes. The presence of EBV was assessed by in situ hybridization. Statistical analyses were performed using the χ2 or Fisher's exact test.RESULTS: The most frequent hypermethylated gene was COX-2 (63.5%) followed by DAPK (55.7%), CDH1 (51%), CDKN2A (36%) and hMLH1 (30.3%). Intestinal and diffuse adenocarcinomas showed different methylation profiles and there was an association between methylation of E-CDH1 and H. pylori-cagA+ in the intestinal adenocarcinoma type. MSI was correlated with hMLH1 methylation. | 3 | Development of hepatorenal syndrome in bile duct ligated rats显示文摘AIM: To evaluate in bile duct ligated rats whether there were progressive alterations of renal function without changes in histopathology. METHODS: Male Wistar rats were submitted to sham-surgery or bile duct ligation (BDL) and divided according to the post-procedure time (2, 4 and 6-wk). To determine renal function parameters, rats wereplaced in metabolic cages and, at the end of the experiment, blood and urine samples were obtained. Histology and hydroxyproline content were analyzed in liver and renal tissue. RESULTS: Rats with 2 wk of BDL increased free water clearance (P = 0.02), reduced urinary osmolality (P = 0.03) and serum creatinine (P = 0.01) in comparison to the sham group. In contrast, rats at 6 wk of BDL showed features of HRS, including signif icant increase in serum creatinine and reductions in creatinine clearance, water excretion and urinary sodium concentration. Rats with 4 wk of BDL exhibited an intermediate stage of renal dysfunction. Progressive hepatic f ibrosis according to post-procedure time was confirmed by histology. The increased levels of liver hydroxyproline contrasted with the absence of structural changes in the kidney, as assessed by histology and unchanged hydroxyproline content in renal tissue. CONCLUSION: Our data show that BDL produced progressive renal dysfunction without structural changes in the kidney, characterizing HRS. | 5 | Stability of fruit quality traits in diverse watermelon cultivars tested in multiple environments显示文摘Lycopene is a naturally occurring red carotenoid compound that is found in watermelon.Lycopene has antioxidant properties.Lycopene content,sugar content and hollowheart resistance are subject to significant genotype×environment interaction(G×E),which makes breeding for these fruit quality traits difficult.The objectives of this study were to(i)evaluate the influence of years and locations on lycopene content,sugar content and hollowheart resistance for a set of watermelon genotypes,and(ii)identify genotypes with high stability for lycopene,sugar,and hollowheart resistance.A diverse set of 40 genotypes was tested over 3 years and 8 locations across the southern United States in replicated,multi-harvest trials.Lycopene was tested in a subset of 10 genotypes.Data were analyzed using univariate and multivariate stability statistics(BLUP-GGE biplot)using SASGxE and RGxE programs.There were strong effects of environment as well as G×E interaction on watermelon quality traits.On the basis of stability measures,genotypes were classified as stable or unstable for each quality trait.'Crimson Sweet'is an inbred line with high quality trait performance as well as trait stability.'Stone Mountain','Tom Watson','Crimson Sweet'and'Minilee'were among the best genotypes for lycopene content,sugar content and hollowheart resistance.We developed a stability chart based on marketable yield and average ranking generated from different stability measures for yield attributes and quality traits.The chart will assist in choosing parents for improvement of watermelon cultivars.See http://gffzz783b6e01a04243c3h6ouxbkkbubxb6qwn.ffgz.tsg.suse.edu.cn/cucurbit/wmelon/wmelonmain.html. | Mahendra Dia Todd C Wehner Penelope Perkins-Veazie Richard Hassell Daniel S Price George E Boyhan Stephen M Olson Stephen R King Angela R Davis Gregory E Tolla Jerome Bernier Benito Juarez | 2016 | Horticulture Research2016，3，1： | 2 |
| 6 | Tracheobronchial Polyflex stents for the management of benign refractory hypopharyngeal strictures显示文摘AIM:To describe a modified technique for placement of a tracheobronchial self-expanding plastic stent(SEPS) in patients with benign refractory hypopharyngeal strictures in order to improve dysphagia and allow stricture remodeling.METHODS:A case series of four consecutive patients with complex hypopharyngeal strictures after combinedtherapy for laryngeal cancer,previously submitted to multiple sessions of dilation without lasting improvement,is presented.All patients underwent placement of a small diameter and unflared tracheobronchial SEPS.Main outcome measurements were improvement of dysphagia and avoiding of repeated dilation.RESULTS:The modified introducer system allowed an easy and technically successful deployment of the tracheobronchial Polyflex stent through the stricture.All four patients developed complications related to stent placement.Two patients had stent migration(one proximal and one distal),two patients developed phanryngocutaneous fistulas and all patients with stents in situ for more than 8 wk had hyperplastic tissue growth at the upper end of the stent.Stricture recurrence was observed at 4 wk follow-up after stent removal in all patients CONCLUSION:Although technically feasible,placement of a tracheobronchial SEPS is associated with a high risk of complications.Small diameter stents must be kept in place for longer than 3 mo to allow adequate time for stricture remodeling. | Rui Almeida Silva Nuno Mesquita Pedro Pimentel Nunes Elisabete Cardoso Ricardo Marcos Pinto Luís Moreira Dias | 2012 | World Journal of Gastroenterology2012，18，6： | 2 |
