1Assessment of vascular invasion in gastric cancer: A comparative study显示文摘AIM: To evaluate and compare detection of lymphatic and blood vessel invasion (LVI and BVI) by hematox-ylin-eosin (HE) and immunohistochemistry (IHC) in gastric cancer specimens, and to correlate with lymph node status. METHODS: IHC using D2-40 (a lymphatic endothelial marker) and CD34 (a pan-endothelial marker) was performed to study LVI and BVI in surgical specimens froma consecutive series of 95 primary gastric cancer cases. The results of the IHC study were compared with the detection by HE using McNemar test and kappa index. The morphologic features of the tumors and the presence of LVI and BVI were related to the presence of lymph node metastasis. A χ2 test was performed to obtain associations between LVI and BVI and other prognostic factors for gastric cancer. RESULTS: The detection rate of LVI was considerably higher than that of BVI. The IHC study identified eight false-positive cases and 13 false-negative cases for LVI, and 24 false-positive cases and 10 false-negative cases for BVI. The average Kappa value determined was moderate for LVI (k=0.50) and low for BVI (k=0.20). Both LVI and BVI were statistically associated with the presence of lymph node metastasis (HE: P=0.001, P=0.013, and IHC: P=0.001, P=0.019). The mor-phologic features associated with LVI were location of the tumor in the distal third of the stomach (P=0.039), Borrmann's macroscopic type (P=0.001), organ inva-sion (P=0.03) and the depth of tumor invasion (P=0.001). The presence of BVI was related only to the depth of tumor invasion (P=0.003). 2Ab-Induced Insulin Resistance and the Effects of Insulin on the Cholesterol Synthesis Pathway and Ab Secretion in Neural Cells显示文摘Alzheimer's disease(AD) is characterized by amyloid-b(Ab) toxicity,tau pathology,insulin resistance,neuroinflammation,and dysregulation of cholesterol homeostasis,all of which play roles in neurodegeneration.Insulin has polytrophic effects on neurons and may be at the center of these pathophysiological changes.In this study,we investigated possible relationships among insulin signaling and cholesterol biosynthesis,along with the effects of Ab42 on these pathways in vitro.We found that neuroblastoma 2a(N2a) cells transfected with the human gene encoding amyloid-b protein precursor(Ab PP)(N2aAb PP) produced Ab and exhibited insulin resistance by reduced p-Akt and a suppressed cholesterol-synthesis pathway following insulin treatment,and by increased phosphorylation of insulin receptor subunit-1 at serine 612(p-IRS-S612) as compared to parental N2 a cells.Treatment of human neuroblastoma SH-SY5 Y cells with Ab42 also increased p-IRS-S612,suggesting that Ab42 is responsible for insulin resistance.The insulin resistance was alleviated when N2a-Ab PP cells were treated with higher insulin concentrations.Insulin increased Ab release from N2 aAb PP cells,by which it may promote Ab clearance.Insulin increased cholesterol-synthesis gene expression in SHSY5 Y and N2 a cells,including 24-dehydrocholesterol reductase(DHCR24) and 3-hydroxy-3-methyl-glutaryl-Co A reductase(HMGCR) through sterol-regulatory element-binding protein-2(SREBP2).While Ab42-treated SH-SY5 Y cells exhibited increased HMGCR expression and c-Jun phosphorylation as pro-inflammatory responses,they also showed down-regulation of neuro-protective/antiinflammatory DHCR24.These results suggest that Ab42 may cause insulin resistance,activate JNK for c-Jun phosphorylation,and lead to dysregulation of cholesterol homeostasis,and that enhancing insulin signaling may relieve the insulin-resistant phenotype and the dysregulated cholesterol-synthesis pathway to promote Ab release for clearance from neural cells.Dema Najem Michelle Bamji-Mirza Ze Yang Wandong Zhang 2016Neuroscience Bulletin2016,32,3:6
3开放参观对莫高窟洞窟微环境的影响显示文摘洞窟微环境相对湿度是影响壁画保存的关键因素,为了解开放参观对莫高窟洞窟相对湿度的影响,本研究选择莫高窟南区四个相似洞窟进行长达10年(2004~2013年)的定期监测,对比分析了开放洞窟和关闭洞窟其相对湿度的变化情况。结果表明:开放洞窟相对湿度在开放时段波动频率与游客参观频次正相关,发生降雨时开放洞窟相对湿度迅速上升,关闭洞窟可有效阻隔窟外相对湿度波动的影响。开放洞窟在参观量大的旅游旺季,其相对湿度月极大值、日较差月均值以及日极大值超过60%的频次均明显高于关闭洞窟。将开放洞窟关闭,关闭洞窟开放后,其相对湿度呈相反规律变化。开放参观会导致洞窟相对湿度水平显著升高,日波动幅度明显增大,从而增加壁画劣化风险。这些结论可为洞窟的预防性管理和风险评估提供科学依据。陈海玲 陈港泉 Neville Agnew Martha Demas 薛平 侯文芳 苏伯民 前川信 王旭东 2017文物保护与考古科学2017,29,6:5
4Epidemiology of onchocerciasis-associated epilepsy in the Mbam and Sanaga river valleys of Cameroon:impact of more than 13 years of ivermectin显示文摘Background:A high epilepsy prevalence has been reported in several onchocerciasis-endemic villages along the Mbam and Sanaga river valleys in Cameroon,including Bilomo and Kelleng.We sought to determine the prevalence of epilepsy in these two villages following more than 13 years of community-directed treatment with ivermectin(CDTI).Methods:Door-to-door surveys were performed on the entire resident population in the villages in August 2017 and January 2018.Epilepsy was diagnosed using a 2-step approach:administration of a standardized 5-item questionnaire followed by confirmation by a neurologist.Previously published diagnostic criteria for onchocerciasis-associated epilepsy(OAE)were used.Ov16 serology was done for children aged 7-10 years to assess onchocerciasis transmission.Findings were compared with previous data from these two villages.Results:A total of 1525 individuals(1321 in Bilomo and 204 in Kelleng)in 233 households were surveyed in both villages.The crude prevalence of epilepsy was 4.6%in Bilomo(2017)and 7.8%in Kelleng(2018),including 12(15.6%of cases)persons with epilepsy(PWE)with nodding seizures.The age and sex-standardized prevalence in Kelleng decreased from 13.5%in 2004 to 9.3%in 2018(P<0.001).The median age of PWE shifted from 17(IQR:12-22)years to 24(IQR:20-30)years in Bilomo(P<0.001);and slightly from 24(IQR:14-34)years to 28(IQR:21.25-36.75)years in Kelleng(P=0.112).Furthermore,47.6%of all tested children between 7 and 10 years had Ov16 antibodies.Conclusions:There is a decrease in epilepsy prevalence after 13 years and more of CDTI in both villages.The age-shift observed in PWE suggests that ivermectin may prevent OAE in younger residents.Ov16 seropositivity in children indicates ongoing onchocerciasis transmission possibly due to suboptimal control measures.Our findings support the existence of OAE in Cameroon and highlight the need to strengthen onchocerciasis elimination programs.Joseph Nelson Siewe Fodjo Godwin Tatah Earnest Njih Tabah Leonard Ngarka Leonard Njamnshi Nfor Samuel Eric Chokote Michel KMengnjo Fidele Dema Aurelien Tele Sitouok Grace Nkoro Felicien ENtone Anne-Cecile Zoung-Kanyi Bissek Cedric BChesnais Michel Boussinesq Robert Colebunders Alfred K.Njamnshi 2018Infectious Diseases of Poverty2018,7,1:3
5Surface nanostructure formation mechanism of 45 steel induced by supersonic fine particles pombarding显示文摘By means of supersonic fine particles bombarding (SFPB),a nanostructured surface layer up to 15 μm was fabricated on a 45 steel plate with ferrite and pearlite phases. To reveal the grain refinement mechanism of SFPB-treated 45 steel,microstructure features of various sections in the treated surface were systematically characterized by X-ray diffraction (XRD),scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Grain size increases with an increase of depth from the treated surface. Plastic deformation and grain refinement processes are accompanied by an increase in strain. Plastic deformation in the proeutectoid ferrite phases has precedence over the pearlite phases. Grain refinement in the ferrite phases involves: the onset of dis-location lines (Dls),dislocation tangles (DTs) and dense dislocation walls (DDWs) in the original grains; the formation of fine la-mellar and roughly equiaxed cells separated by DDWs; by dislocation annihilation and rearrangement,the transformation of DDWS into subboundaries and boundaries and the formation of submicron grains or subgrains; the successive subdivision of grains to finer and finer scale,resulting in the formation of highly misoriented nano-grains. By contrast,eutectoid cementite phase accommodated strain in a sequence as follows: onset of elongated,bended and shear deformation under deformation stress of ferrites,short and thin cementites with a width of about 20-50 nm and discontinuous length were formed. Shorter and thinner cementites were developed into ultra-fine pieces under the action of high density dislocation and strains. At the top surface,some cementites were decomposed under severe plastic deformation.
