| 3 | Iron and non-alcoholic fatty liver disease显示文摘The mechanisms that promote liver injury in non-alcoholic fatty liver disease(NAFLD) are yet to be thoroughly elucidated. As such, effective treatment strategies are lacking and novel therapeutic targets are required. Iron has been widely implicated in the pathogenesis of NAFLD and represents a potential target for treatment. Relationships between serum ferritin concentration and NAFLD are noted in a majority of studies, although serum ferritin is an imprecise measure of iron loading. Numerous mechanisms for a pathogenic role of hepatic iron in NAFLD have been demonstrated in animal and cell culture models. However, the human data linking hepatic iron to liver injury in NAFLD is less clear, with seemingly conflicting evidence, supporting either an effect of iron in hepatocytes or within reticulo-endothelial cells. Adipose tissue has emerged as a key site at which iron may have a pathogenic role in NAFLD. Evidence for this comes indirectly from studies that have evaluated the role of adipose tissue iron with respect to insulin resistance. Adding further complexity, multiple strands of evidence support an effect of NAFLD itself on iron metabolism. | 9 | Chrysophanol protects against doxorubicin-induced cardiotoxicity by suppressing cellular PARylation显示文摘The clinical application of doxorubicin(DOX) in cancer chemotherapy is limited by its lifethreatening cardiotoxic effects. Chrysophanol(CHR), an anthraquinone compound isolated from the rhizome of Rheum palmatum L., is considered to play a broad role in a variety of biological processes.However, the effects of CHR’s cardioprotection in DOX-induced cardiomyopathy is poorly understood. In this study, we found that the cardiac apoptosis, mitochondrial injury and cellular PARylation levels were significantly increased in H9 C2 cells treated by Dox, while these effects were suppressed by CHR. Similar results were observed when PARP1 activity was suppressed by its inhibitors 3-aminobenzamide(3 AB)and ABT888. Ectopic expression of PARP1 effectively blocked this CHR’s cardioprotection against DOX-induced cardiomyocyte injury in H9 C2 cells. Furthermore, pre-administration with both CHR and 3 AB relieved DOX-induced cardiac apoptosis, mitochondrial impairment and heart dysfunction in Sprague–Dawley rat model. Binding of IL-7 to its cognate receptor,the IL-7 receptor (IL-7R),activates multiple pathways that regulate lymphocyte survival,glucose uptake,proliferation and differentiation. There has been much interest in understanding how IL-7 receptor signaling is modulated at multiple interconnected network levels. This review examines how the strength of the signal through the IL-7 receptor is modulated in T and B cells,including the use of shared receptor components,signaling crosstalk,shared interaction domains,feedback loops,integrated gene regulation,multimerization and ligand competition. We discuss how these network control mechanisms could integrate to govern the properties of IL-7R signaling in lymphocytes in health and disease. | 13 | Humanin attenuates Alzheimer-like cognitive deficits and pathological changes induced by amyloid β-peptide in rats显示文摘Amyloid β-peptide(Aβ) has been implicated as a key molecule in the neurodegenerative cascades of Alzheimer 's disease(AD). Humanin(HN) is a secretory peptide that inhibits the neurotoxicity of Aβ. However, the mechanism(s) by which HN exerts its neuroprotection against Aβ-induced ADlike pathological changes and memory deficits are yet to be completely defined. In the present study,we provided evidence that treatment of rats with HN increases the number of dendritic branches and the density of dendritic spines, and upregulates pre- and post-synaptic protein levels; these effects lead to enhanced long-term potentiation and amelioration of the memory deficits induced by Aβ1-42. HN also attenuated Aβ1-42-induced tau hyperphosphorylation,apparently by inhibiting the phosphorylation of Tyr307 on the inhibitory protein phosphatase-2A(PP2A)catalytic subunit and thereby activating PP2 A. HN also inhibited apoptosis and reduced the oxidativestress induced by Aβ1-42. | 17 | Enhanced Efficiency of Milk Utilization for Growth in Surviving Low-Birth-Weight Piglets显示文摘This study determined effects of birth weights (0.70 to 1.09, 1.10 to 1.49, 1.50 to 1.89, and≥1.90 kg) on the efficiency of milk utilization for growth in sow-reared piglets. One-hundred-sixty piglets from 18 multiparous sows (Landrace×Large White) were individually weighed immediately after birth (day 0) and at 7-day intervals for 35 days. Milk consumption of piglets was determined weekly using the weigh-suckle-weigh method. Deaths of piglets were recorded daily. Piglets with the lightest birth weight had the highest incidence of mortality. Birth weights between 0.70 and 1.89 kg were positively associated with average daily gains (ADG, g/day) during the suckling period (P<0.01). Compared with piglets having birth weights of 1.50 to 1.89 kg, birth weights≥1.90 kg did not confer any additional benefit on preweaning survival or ADG. Colostrum or milk consumption per kilogram of body weight among low-, normal-, and large-birth-weight piglets did not differ in the first week. At days 14 to 35, milk consumption of piglets (g/kg BW/day) increased (P<0.01) but the ratio of gain to milk consumption decreased (P<0.01) progressively, as their birth weights increased from 0.70 to ≥1.90 kg. Additionally, surviving low-birth-weight piglets had a higher relative growth rate (%/day) than normal-and large-birth-weight piglets. Based on these novel findings, it is imperative that great efforts be made to improve the survival of low-birth-weight piglets so as to increase the efficiency of nutrient utilization by both lactating sows and suckling piglets. | Reza Rezaei Darrell A.
