1Natural Variation in CCD4 Promoter Underpins Species-Specific Evolution of Red Coloration in Citrus Peel显示文摘Carotenoids and apocarotenoids act as phytohormones and volatile precursors that influence plant development and confer aesthetic and nutritional value critical to consumer preference.Citrus fruits display considerable natural variation in carotenoid and apocarotenoid pigments.In this study,using an integrated genetic approach we revealed that a 5;c/s-regulatory change at CCD4b encoding CAROTENOID CLEAVAGE DIOXYGENASE 4b is a major genetic determinant of natural variation in C3 0 apocarotenoids responsible for red coloration of citrus peel.Functional analyses demonstrated that in addition the known role in synthesizing 3-citraurin,CCD4b is also responsible for the production of another important C3 0 apocarotenoid pigment,p-citraurinene.Furthermore,analyses of the CCD4b promoter and transcripts from various citrus germplasm accessions established a tight correlation between the presence of a putative 5'c/s-regulatory enhancer within an MITE transposon and the enhanced allelic expression of CCD4b in C3 0 apocarotenoid-rich red-peeled accessions.Phylogenetic analysis provided further evidence that functional diversification of CCD4b and naturally occurring variation of the CCD4b promoter resulted in the stepwise evolution of red peels in mandarins and their hybrids.Taken together,our findings provide new insights into the genetic and evolutionary basis of apocarotenoid diversity in plants,and would facilitate breeding efforts that aim to improve the nutritional and aesthetic value of citrus and perhaps other fruit crops.Xiongjie Zheng Kaijie Zhu Quan Sun Weiyi Zhang Xia Wang Hongbo Cao Meilian Tan Zongzhou Xie Yunliu Zeng Junli Ye Lijun Chai Qiang Xu Zhiyong Pan Shunyuan Xiao Paul D.Fraser Xiuxin Deng 2019Molecular Plant2019,12,9:18
2Dynamic compressive behaviour of Ti-6Al-4V alloy processed by electron beam melting under high strain rate loading显示文摘This paper documents an investigation into the compressive deformation behaviour of electron beam melting(EBM) processing titanium alloy(Ti-6A1-4V) parts under high strain loading conditions.The dynamic compression tests were carried out at a high strain rate of over1×10~3/s using the split Hopkinson pressure bar(SHPB)test system and for comparison the quasi-static tests were performed at a low strain rate of 1×10~3/s using a numerically controlled hydraulic materials test system(MTS) testing machine at an ambient temperature.Furthermore,microstructure analysis was carried out to study the failure mechanisms on the deformed samples.The Vickers micro-hardness values of the samples were measured before and after the compression tests.The microstructures of the compressed samples were also characterized using optical microscopy.The particle size distribution and chemical composition of powder material,which might affect the mechanical properties of the specimens,were investigated.In addition,the numerical simulation using commercial explicit finite element software was employed to verify the experimental results from SHPB test system.A.Mohammadhosseini S.H.Masood D.Fraser M.Jahedi 2015Advances in Manufacturing2015,3,3:7
3Metabolic diversity in sweet potato (Ipomoea batatas, Lam.) leaves and storage roots显示文摘Sweet potato(Ipomoea batatas,Lam.)is an important root vegetable in developing countries.After its domestication in Neotropical America,human migration led to the distribution of the sweet potato plant throughout the world.Both leaf and storage root are high in compounds of nutritional value.Yet,the storage roots are of particular value due to their significant content of provitamin A(β-carotene).The breeding effort for elite sweet potato lines led to the reduction of genetic diversity and the potential to improve other traits.The focus of the present study was to assess the metabolic diversity of 27 sweet potato cultivars including landraces and improved varieties.A metabolite profiling approach was optimised for sweet potato leaf and storage root tissue and 130 metabolites identified with three different analysis platforms.The data highlighted a lack of correlation between storage root phenotype and leaf metabolism.Furthermore,the metabolic diversity of storage roots was based on the secondary metabolism,including phenylpropanoids and carotenoids.Three cultivars of three different flesh colouration(yellow,orange and purple)showed a significant difference of the primary metabolism.This data demonstrates the value of metabolite profiling to breeding programs as a means of identifying differences in phenotypes/chemotypes and characterising parental material for future pre-breeding resources.Margit Drapal Genoveva Rossel Bettina Heider Paul D.Fraser 2019Horticulture Research2019,6,1:6
