| 1 | Correlation of N-myc downstream-regulated gene 1 expression with clinical outcomes of colorectal cancer patients of different race/ethnicity显示文摘AIM: To evaluate the role of N-myc downstream-regulated gene 1 (NDRG1) expression in prognosis and survival of colorectal cancer patients with different ethnic backgrounds. METHODS: Because NDRG1 is a downstream target of p53 and hypoxia inducible factor-1α (HIF-1α),we examined NDRG1 expression together with p53 and HIF-1α by immunohistochemistry. A total of 157 colorectal cancer specimens including 80 from Japanese patients and 77 from US patients were examined. The correlation between protein expression with clinicopathological features and survival after surgery was analyzed.in colorectal tumor compared with normal epithelium in both Japanese and US patient groups. Expression of NDRG1 protein was significantly correlated with lymphatic invasion,venous invasion,depth of invasion,histopathological type,and Dukes' stage in Japanese colorectal cancer patients. NDRG1 expression was correlated to histopathological type,Dukes' stage and HIF-1α expression in US-Caucasian patients but not in US-African American patients. Interestingly,Kaplan-Meier survival analysis demonstrated that NDRG1 expression correlated significantly with poorer survival in US-African American patients but not in other patient groups. However,in p53-positive US cases,NDRG1 positivity correlated significantly with better survival. In addition,NDRG1 expression also correlated significantly with improved survival in US patients with stages Ⅲ and Ⅳ tumors without chemotherapy. In Japanese patients with stages Ⅱ and Ⅲ tumors,strong NDRG1 staining in p53-positive tumors correlated significantly with improved survival but negatively in patients without chemotherapy. CONCLUSION: NDRG1 expression was correlated with various clinicopathological features and clinical outcomes in colorectal cancer depending on the race/ethnicity of the patients. | 2 | Adjuvant chemotherapy for resected colorectal cancermetastases:literature review and meta-analysis显示文摘Surgical resection is the only option of cure for patients with metastatic colorectal cancer(CRC). However, the risk of recurrence within 18 mo after metastasectomy is around 75% and the liver is the most frequent site of relapse. The current international guidelines recommend an adjuvant therapy after surgical resection of CRC metastases despite the lower level of evidence(based on the quality of studies in this setting). However, there is still no standard treatment and the effective role of an adjuvant therapy remains controversial. | 7 | 心肺运动试验评价食管癌患者化疗后整体功能变化的临床研究显示文摘背景化疗对人体整体功能具有显著损害,但目前尚缺乏对化疗患者整体功能变化进行客观定量评估的可靠方法。目的探讨心肺运动试验(CPET)对食管癌患者化疗前后整体功能变化的评估作用。方法选取2006—2007年度在德州大学安德森癌症中心进行术前化疗的白种人男性食管癌患者6例,分别在化疗前后进行CPET检查,按照美国加州大学洛杉矶分校医学中心标准连续递增功率方案完成症状限制性极限运动,通过对数据标准化分析计算其核心指标,从系统软件导出静息状态、热身状态、无氧阈状态、极限状态时的循环指标及呼吸指标。结果 6例食管癌患者化疗后,除1例患者无氧阈略有升高外,其余患者的峰值摄氧量、无氧阈、峰值氧脉搏、摄氧效率平台、峰值负荷功率和递增功率运动时间均降低;化疗后,除1例患者的二氧化碳通气当量斜率略有降外,其余所有患者的二氧化碳通气当量斜率和二氧化碳通气当量最低值均升高。食管癌患者化疗前后无氧阈(L/min、ml·min-1·kg^(-1)、%pred)、二氧化碳通气当量斜率(slope、%pred)、递增功率运动时间(min)比较,差异均无统计学意义(P>0.05);化疗后峰值摄氧量(L/min、ml·min-1·kg^(-1)、%pred)、峰值氧脉搏(ml/次、%pred)、摄氧效率平台(比值、%pred)、峰值负荷功率(W)较化疗前降低,二氧化碳通气当量最低值(比值、%pred)较化疗前升高(P<0.05)。食管癌患者化疗后极限状态时摄氧量较化疗前降低(P<0.05);静息状态时心率较化疗前升高,收缩压较化疗前降低(P<0.05);热身状态时舒张压、平均动脉压较化疗前降低(P<0.05);4个状态时氧脉搏较化疗前降低(P<0.05);静息状态、热身状态、极限状态时摄氧通气效率较化疗前降低(P<0.05);极限状态时二氧化碳通气当量较化疗前升高(P<0.05)。食管癌患者化疗后热身状态时潮气末二氧化碳分压较化疗前降低(P<0.05);静息状态、热身状态、极限状态时潮气末氧分压较化疗前升高(P<0.05);极限状态时每呼吸摄氧量较化疗前降低(P<0.05);无氧阈状态、极限状态时每呼吸二氧化碳排出量较化疗前降低(P<0.05)。结论化疗使得食管癌患者循环和细胞代谢功能显著降低,而呼吸受限相对较轻,且处于相对代偿反应。CPET检查可以对食管癌患者化疗前的整体功能进行客观定量评估,同时也可以对化疗所造成的整体功能改变进行评估。 | 孙兴国 Curtis Hightower 刘方 William W.Stringer 葛万刚 | 2016 | 中国全科医学2016,19,17: | 11 |

| 12 | Effects of Soil Fertility and Atmospheric CO_2 Enrichment on Leaf, Stem and Root Dark Respiration of Populus tremuloides显示文摘An open-top chamber experiment was conducted at the University of Michigan Biological Station near Pellston, Michigan, USA, to study the effects of soil fertility and CO2 on leaf, stem and root dark respiration (Rd) of Populus tremuloides. Overall, area-based day-time leaf Rd (Rda) was significantly greater at elevated than at ambient CO2 in high-fertility soil, but not in low-fertility soil. Mass-based leaf Rd (Rdm) was overall greater for high- than for low-fertility soil grown trees at elected, but not at ambient CO2. Nighttime leaf Rd. and Rdm were unthected by soil fertility or CO2, nor was stem Rda, which ranged from 1.0 to 1.4 μmol m-2 s-1 in the spring and 3.5 to 4.5 μmol m-2 s-1 in the summer. Root Rda. was significantly higher in high- than in low-fertility soil, but was unaffected by CO2. Since biomass production of P. tremuloides will be significantly greater at elevated CO2 while specific Rd will either increase or remain unchanged, we predict that carbon loss to the atmosphere through respiration from this ecologically important species would increase at higher CO2. | 16 | Spouting behaviour of binary mixtures in square-based spouted beds显示文摘From experime nts, the influe nee of the physical characteristics of different bin ary mixtures of solids on the spouting regime of a pyramidal square-based spouted bed reactor is assessed. The applied methodology permits a more precise evaluation of the effects of the tested variables (diameter, density, sphericity) on the response variables (minimum air flows at which spouting begins and at which to maintain spouting con ditions). The associated pressure drops along the bed of particles a nd the height of the formed fountai n are analysed in each case. During the initial stages of fluidisation, binary mixtures containing different density ratios show dead zones. Segregation becomes more evident at large-size and high-density ratios. The lack of sphericity was found to be the main reason leading to blocking, channelling, and start-up problems when system failures occur. Nevertheless, the extent of segregation in all cases decreases with increasing the spouting velocity. In addition, a computational fluid dynamic model based on the discrete element method, previously validated for a single solid bed, is proposed as a tool to predict and evaluate potential segregation phenomena in binary mixtures. This model reproduced with high accuracy the encountered segregation phenomena. | 18 | 新氟喹诺酮类药物的抗微生物疗法显示文摘当前氟喹诺酮类药物不仅已成为发达国家如日本、美国、英国、德国、法国等许多国家的常用抗感染药物,而且在第三世界也得到迅速发展,如在印度该类药物的平均年增长率高达25%~28%.该类药物的口服制剂已成为治疗常见感染的重要药物,同时最近已证实,开始采用静脉给药然后改为口服给药的疗法已成为治疗某些严重感染的有效、安全、经济的疗法,尤其对多重耐药菌引起的严重感染有重要价值。最近因严重毒副作用而取消了刚刚在美国上市的替马沙星,虽然对该类药物市场可能有一定影响,但据 FDA 统计,其它已上市多年的品种如诺氟沙星、氧氟沙星和环丙沙星均没有像替马沙星那样多的严重毒副作用。环丙沙星1991年的销售额已达9.04亿美元,较1990年增加19.7%,仅次于头孢克罗.但目前已上市的品种对革兰氏阳性菌特别是肺炎链球菌的活性差,对耐甲氧西林的金葡菌、厌氧菌等活性也不理想,因此开发新品种的工作一直在大量继续进行.本文对了解国外该类药物现状有一定参考价值. | Curtis D Holt 王金生 | 1993 | 国外医药(抗生素分册)1993,14,6: | 4 |
| 19 | Regulation of hepatic micro RNA expression by hepatocyte nuclear factor 4 alpha显示文摘AIM To uncover the role of hepatocyte nuclear factor 4 alpha(HNF4α) in regulating hepatic expression of micro RNAs.METHODS Microarray and real-time PCR were used to determine hepatic expression of micro RNAs in young-adult mice lacking Hnf4α expression in liver(Hnf4α-Liv KO). Integrative genomics viewer software was used to analyze the public chromatin immunoprecipitation-sequencing datasets for DNA-binding of HNF4α, RNA polymerase-Ⅱ, and histone modifications to loci of micro RNAs in mouse liver and human hepatoma cells. Dual-luciferase reporter assay was conducted to determine effects of HNF4α on the promoters of mouse and human micro RNAs as well as effects of micro RNAs on the untranslated regions(3'UTR) of two genes in human hepatoma cells. RESULTS Microarray data indicated that most micro RNAs remained unaltered by Hnf4α deficiency in Hnf4α-Liv KO mice. However, certain liver-predominant micro RNAs were down-regulated similarly in young-adult male and female Hnf4α-Liv KO mice. The down-regulation of mi R-101, mi R-192, mi R-193 a, mi R-194, mi R-215, mi R-802, and mi R-122 as well as induction of mi R-34 and mi R-29 in male Hnf4α-Liv KO mice were confirmed by real-timePCR. Analysis of public chromatin immunoprecipitationsequencing data indicates that HNF4α directly binds to the promoters of mi R-101, mi R-122, mi R-194-2/mi R-192 and mi R-193, which is associated with histone marks of active transcription. Luciferase reporter assay showed that HNF4α markedly activated the promoters of mouse and human mi R-101b/mi R-101-2 and the mi R-194/mi R-192 cluster. Additionally, mi R-192 and mi R-194 significantly decreased activities of luciferase reporters for the 3'UTR of histone H3F3 and chromodomain helicase DNA binding protein 1(CHD1), respectively, suggesting that mi R-192 and mi R-194 might be important in chromosome remodeling through directly targeting H3F3 and CHD1.CONCLUSION HNF4α is essential for hepatic basal expression of a group of liver-enriched micro RNAs, including mi R-101, mi R-192, mi R-193 a, mi R-194 and mi R-802, through which HNF4α may play a major role in the post-transcriptional regulation of gene expression and maintenance of the epigenome in liver. | Hong Lu Xiaohong Lei Jerry Liu Curtis Klaassen | 2017 | World Journal of Hepatology2017,9,4: | 3 |
