1Irritable bowel syndrome: Relations with functional, mental, and somatoform disorders显示文摘This review describes the conceptual and clinical relations between irritable bowel syndrome(IBS),other functional,somatoform,and mental disorders,and points to appropriate future conceptualizations.IBS is considered to be a functional somatic syndrome(FSS)with a considerable symptom overlap with other FSSs like chronic fatigue syndrome or fibromyalgia syndrome.IBS patients show an increased prevalence of psychiatric symptoms and disorders,especially depression and anxiety.IBS is largely congruent with the concepts of somatoform and somatic symptom disorders.Roughly 50%of IBS patients complain of gastrointestinal symptoms only and have no psychiatric comorbidity.IBS concepts,treatment approaches,as well as health care structures should acknowledge its variability and multidimensionality by:(1)awareness of additional extraintestinal and psychobehavioral symptoms in patients with IBS;(2)general and collaborative care rather than specialist and separated care;and(3)implementation of'interface disorders'to abandon thedualistic classification of purely organic or purely mental disorders.Constanze Hausteiner-Wiehle Peter Henningsen 2014World Journal of Gastroenterology2014,20,20:11
2c-myb对胚胎干细胞体外造血分化及定型的影响显示文摘目的 通过体外胚胎干细胞培养体系对ES细胞进行c- myb等位基因打靶,建立c -myb+ +及c -myb ES细 胞模型的基础上,探讨转录因子c- myb在造血定型和分化中的作用。方法 通过体外ES细胞去除LIF半固体介质EBs的 造血分化体系,采用甲基纤维素克隆形成分析和real timePCR观察c- myb+ +及c -myb ES细胞,比较分析其不同分化过 程及阶段中造血各系形成和相关基因表达变化。结果 c -myb+ +及c -myb ES胚胎体(EBs)大小及形态相似,c -myb+ +ESEBs形成显著多于c -myb 。c -myb+ +组CFU- E出现在第6天,高峰为第8天;c- myb 组也有相似的改变,但其 CFU- E数目少于c -myb+ +。c -myb+ +组CFU- M高峰为第7天;c- myb 也有相似的改变,但其CFU- M数目高于c- myb+ +。c -myb+ +CFU -GM出现在第7天,高峰为第12天,而c -myb 未见CFU -GM形成。real timePCR分析显示,c -myb 及 c- myb+ +β globin,zeta globin,Lys,C- fms基因表达无显著变化。结论 低水平的c -myb不影响祖细胞的扩增,但影响其 终末分化。低水平的c -myb表达在造血定型中对红系的发育具有一定的影响,而对巨噬细胞的发育成熟无明显影响。 c -myb的表达水平对造血分化相关基因的表达没有明显的影响。陈洁平 Deborah Clarke Constanze Bonifer 2005第三军医大学学报2005,27,6:5

10c-myb表达变化在造血定型、分化及造血相关基因表达中的作用显示文摘目的通过基因打靶建立c-myb+/+和c-myb+/-胚胎干细胞(ES)模型,了解转录因子c-myb在造血定型和分化中的作用。方法采用去除白血病抑制因子的甲基纤维素体外ES细胞诱导分化体系进行造血分化,收获c-myb+/+和c-myb+/-(表达55%的c-myb)胚胎体进行甲基纤维素克隆形成分析。运用实时定量PCR比较分析c-myb+/+和c-myb+/-造血分化过程的造血相关基因表达。结果c-myb+/+和c-myb+/-ES细胞胚胎体形成相似,但c-myb+/-组胚胎体体积小于c-myb+/+组。两组红系克隆形成单位(CFU-E)克隆形成相似,但c-myb+/-组CFU-E克隆小于c-myb+/+组。两组红系爆式克隆形成单(BFU-E)均出现于第7天,高峰在第10天。两组巨噬系克隆形成单位(CFU-M)形成相似,但c-myb+/-组CFU-M数目多于c-myb+/+组。c-myb+/-组粒-单系克隆形成单位(CFU-GM)形成数目少于c-myb+/+组。PCR分析显示c-myb+/+和c-myb+/-组β-globin、ζ-globin、GATA3、CD34、Lys、c-fms和c-mpl基因表达没有明显差异。结论亚表达水平的c-myb可以满足祖细胞扩增的需要,但却影响其终末分化。亚表达水平c-myb影响前体细胞向红系和粒系的分化,但不影响巨噬细胞的分化。c-myb表达水平不影响β-globin、ζ-globin、GATA3、CD34、Lys、c-fms和c-mpl基因的表达。陈洁平 Deborah Clarke Constanze Bonifer 2008解放军医学杂志2008,33,11:1
