2Bactericidal and anti-adhesive properties of culinary and medicinal plants against Helicobacter pylori显示文摘AIM: To investigate the bactericidal and anti-adhesive properties of 25 plants against Helicobacter pylori (H pylori).METHODS: Twenty-five plants were boiled in water to produce aqueous extracts that simulate the effect of cooking. The bactericidal activity of the extracts was assessed by a standard kill-curve with seven strains of H pylori. The anti-adhesive property was assessed by the inhibition of binding of four strains of FITC-labeled H pylori to stomach sections. RESULTS: Of all the plants tested, eight plants, including Bengal quince, nightshade, garlic, dill, black pepper, coriander, fenugreek and black tea, were found to have no bactericidal effect on any of the isolates. Columbo weed, long pepper, parsley, tarragon, nutmeg, yellow-berried nightshade, threadstem carpetweed, sage and cinnamon had bactericidal activities against H pylori, but total inhibition of growth was not achieved in this study. Among the plants that killed H pylori, turmeric was the most efficient, followed by cumin, ginger, chilli, borage, black caraway, oregano and liquorice. Moreover, extracts of turmeric; borage and parsley were able to inhibit the adhesion of H pylori strains to the stomach sections.CONCLUSION: Several plants that were tested in our study had bactericidal and/or anti-adhesive effects on H pylori. 3Multispecies probiotic protects gut barrier function in experimental models显示文摘AIM:To investigate the effect of the probiotic combination Lactibiane Tolerance?(LT)on epithelial barrier function in vitro and in vivo.METHODS:The effect of the multispecies probiotic LT was assessed on several models of epithelial barrier function both in vitro(in basal and inflammatory conditions)and in vivo[visceral hypersensitivity induced by chronic stress or by colonic perfusion of a fecal supernatant(FSN)from patients with irritable bowel syndrome(IBS)].In vitro,we measured the permeability of confluent T84 cell monolayers incubated with or without LT by evaluating the paracellular flux of macromolecules,in basal conditions and after stimulation with lipopolysaccharide(LPS)or with conditioned medium of colonic biopsies from IBS patients(IBS-CM).In vivo,male C57/Bl6 mice received orally NaCl or LT for 15 d and were submitted to water avoidance stress(WAS)before evaluating visceral sensitivity by measuring the myoelectrical activity of the abdominal muscle and the paracellular permeability with 51Cr-EDTA.Permeability and sensitivity were also measured after colonic instillation of FSN.Tight-junctions were assessed by immunoblotting and TLR-4 expression was evaluated by immunohistochemistry RESULTS:Incubation of T84 cell monolayers with LT in basal conditions had no significant effect on permeability(P>0.05 vs culture medium).By contrast,addition of LT bacterial bodies(LT)completely prevented the LPS-induced increase in paracellular permeability(P<0.01 vs LPS 10 ng/mL(LPS 10);P<0.01 vs LPS 100ng/mL(LPS 100),P>0.05 vs culture medium).The effect was dose dependent as addition of 109 LT bacterial bodies induced a stronger decrease in absorbance than 106 LT(109 LT+LPS 10:-20.1%±13.4,P<0.01vs LPS 10;106 LT+LPS 10:-11.6%±6.2,P<0.01 vs LPS 10;109 LT+LPS 100:-14.4%±5.5,P<0.01 vs LPS 100;106 LT+LPS 100:-11.6%±7.3,P<0.05 vs LPS 100).Moreover,the increase in paracellular permeability induced by culturing T84 cells with conditioned medium of colonic biopsies from IBS patients(IBS-CM)was completely inhibited in the presence of 109 LT(P<0.01 vs IBS-CM).LT also significantly prevented the epithelial disruption induced by intracolonic infusion of fecal supernatant from IBS patients(P<0.01 vs IBS FSN)or water avoidance stress P<0.01 vs WAS)in C57/Bl6 mice and increased the expression of occludin in vitro and in vivo,as assessed by immnunoblotting.The WAS-induced effect on visceral sensitivity was prevented by LT treatment since values obtained for all steps of colorectal distension were significantly(P<0.01)different from the WAS group.Finally,LT downregulated the response mediated through TLR-4 in vitro(decrease in tumor necrosis factorαsecretion in response to LPS:-65.8%for 109 LT and-52.5%for 106LT,P<0.01 vs LPS)and in vivo(inhibition of WAS induced an increase in TLR-4 expression in the LT treated mice colon,P<0.01 vs WAS).CONCLUSION:The probiotic LT mix prevented the disruption to the epithelial barrier induced by LPS,stress or colonic soluble factors from IBS patients and prevented visceral hypersensitivity.Mylene Nébot-Vivinus Cherryl Harkat Hanene Bzioueche Christel Cartier Raffaella Plichon-Dainese Lara Moussa Helene Eutamene Dorsa Pishvaie Sophie Holowacz Christian Seyrig Thierry Piche Vassilia Theodorou 2014World Journal of Gastroenterology2014,20,22:12

7Oxidation state specific analysis of arsenic species in tissues of wild-type and arsenic(+3 oxidation state)methyltransferase-knockout mice显示文摘Arsenic methyltransferase(As3mt) catalyzes the conversion of inorganic arsenic(i As) to its methylated metabolites, including toxic methylarsonite(MAs~Ⅲ) and dimethylarsinite(DMAs~Ⅲ). Knockout(KO) of As3 mt was shown to reduce the capacity to methylate i As in mice. However, no data are available on the oxidation states of As species in tissues of these mice. Here, we compare the oxidation states of As species in tissues of male C57BL/6 As3mt-KO and wild-type(WT) mice exposed to arsenite(iA s~Ⅲ) in drinking water. WT mice were exposed to50 mg/L As and As3mt-KO mice that cannot tolerate 50 mg/L As were exposed to 0, 15, 20, 25 or30 mg/L As. iA s~Ⅲaccounted for 53% to 74% of total As in liver, pancreas, adipose, lung, heart, and kidney of As3mt-KO mice; tri- and pentavalent methylated arsenicals did not exceed 10% of total As. Tissues of WT mice retained iA s and methylated arsenicals: iA s~Ⅲ, MAs~Ⅲand DMAs~Ⅲ represented 55%‐68% of the total As in the liver, pancreas, and brain. High levels of methylated species, particularly MAs~Ⅲ, were found in the intestine of WT, but not As3mt-KO mice,suggesting that intestinal bacteria are not a major source of methylated As. Blood of WT mice contained significantly higher levels of As than blood of As3mt-KO mice. This study is the first to determine oxidation states of As species in tissues of As3mt-KO mice. 11Canopy gap size influences niche partitioning of the ground-layer plant community in a northern temperate forest显示文摘Aims The Gap Partitioning Hypothesis(GPH)posits that gaps create heterogeneity in resources crucial for tree regeneration in closed-canopy forests,allowing trees with contrasting strategies to coexist along resource gradients.Few studies have examined gap partitioning of temperate,ground-layer vascular plants.We used a ground-layer plant community of a temperate deciduous forest in northern Wisconsin,USA,as a model system to test whether the GPH extends to the relatively species-rich ground layer.Methods We used a well-replicated experimental approach that included a gap opening gradient(five gap sizes,6,10,20,30 and 46 m diameter,and undisturbed reference areas),a within-gap location gradient(gap edge to center),and a temporal gradient(0,2,6 and 13 years after gap creation).The data were observations of ground-layer plant abundance,published plant traits,and a modeled index of understory light environments.We ordinated the plant abundance data and evaluated the relationships of composition,traits and light environment by gap size,location along the forest-gap transect and time,with several approaches such as correlations,descriptive statistics,non-parametric tests of group differences and indicator species importance values.Important Findings Ground-layer plant composition and traits differed across gap sizes,within-gap locations and over time.Gaps of all sizes differed in composition from undisturbed areas,and all pair-wise combinations of gap size also differed in composition,except the 6 m from the 10-m gaps.Large gaps(46 m)also displayed within-gap compositional gradients from gap edge to center locations.Compositional differences in gap size were evident 2 years after gap creation and,contrary to our hypotheses,remained different over the 13-year period,even in gaps with crown closure.In contrast to the neutral theory,species functional traits and microenvironmental conditions were related to variation in ground-layer composition.Species with smaller seeds,lower shade tolerance,later bloom times,shorter stature and longer leaves were associated with higher light,more central gap locations,larger gap sizes and greater time since gap creation.The correlation between gap size and ground-layer plant composition and traits provides evidence for gap partitioning by the diverse ground-layer community in this temperate deciduous forest community.Christel C.Kern Rebecca A.Montgomery Peter B.Reich Terry F.Strong 2013Journal of Plant Ecology2013,6,1:6

16Levels of insecticide resistance to deltamethrin,malathion,and temephos,and associated mechanisms in Aedes aegypti mosquitoes from the Guadeloupe and Saint Martin islands(French West Indies)显示文摘Background:In the Guadeloupe and Saint Martin islands,Aedes aegypti mosquitoes are the only recognized vectors of dengue,chikungunya,and Zika viruses.For around 40 years,malathion was used as a mosquito adulticide and temephos as a larvicide.Since the European Union banned the use of these two insecticide molecules in the first decade of the 21st century,deltamethrin and Bacillus thuringiensis var.israelensis are the remaining adulticide and larvicide,respectively,used in Guadeloupe.In order to improve the management of vector control activities in Guadeloupe and Saint Martin,we investigated Ae.aegypti resistance to and mechanisms associated with deltamethrin,malathion,and temephos.Methods:Ae.aegypti mosquitoes were collected from six different localities of Guadeloupe and Saint Martin.Larvae were used for malathion and temephos bioassays,and adult mosquitoes for deltamethrin bioassays,following World Health Organization recommendations.Knockdown resistance(Kdr)genotyping for V1016I and F1534C mutations,and expression levels of eight enzymes involved in detoxification mechanisms were examined in comparison with the susceptible reference Bora Bora strain.Results:Resistance ratios(RR50)calculated for Ae.aegypti larvae showed high resistance levels to temephos(from 8.9 to 33.1-fold)and low resistance levels to malathion(from 1.7 to 4.4-fold).Adult females displayed moderate resistance levels to deltamethrin regarding the time necessary to affect 50%of individuals,varying from 8.0 to 28.1-fold.Molecular investigations on adult mosquitoes showed high resistant allele frequencies for V1016I and F1534C(from 85 to 96%and from 90 to 98%,respectively),as well as an overexpression of the glutathione S-transferase gene,GSTe2,the carboxylesterase CCEae3a,and the cytochrome genes 014614,CYP6BB2,CYP6M11,and CYP9J23.Conclusions:Ae.aegypti populations from Guadeloupe and Saint Martin exhibit multiple resistance to organophosphates(temephos and malathion),and pyrethroids(deltamethrin).The mechanisms associated with these resistance patterns show strong frequencies of F1534C and V1016I Kdr mutations,and an over-expression of CCEae3a,GSTe2,and four cytochrome P450 genes(014614,CYP9J23,CYP6M11,CYP6BB2).These results will form the baseline for a deeper understanding of the insecticide resistance levels and associated mechanisms of Ae.aegypti populations and will be used to improve vector control strategies in Guadeloupe and Saint Martin.Daniella Goindin Christelle Delannay Andric Gelasse Cedric Ramdini Thierry Gaude Frederic Faucon Jean-Philippe David Joël Gustave Anubis Vega-Rua Florence Fouque 2017Infectious Diseases of Poverty2017,6,1:3

18Antimicrobial resistance of abnormal vaginal discharges microorganisms in Ouagadougou,Burkina Faso显示文摘Objective:To assess the prevalence of bacterial strains and fungal strains infecting the vaginal tract and test their sensitivity to antibiotics in women attending Saint Camille Medical Centre in Ouagadougou.Methods:From January 2008 to December 2009,a total of 2000 vaginal swabs were cultivated for bacterial and fungal identification and isolation.Furthermore,bacterial strains were tested for their susceptibility to several antibiotics used in routine in the centre. 20Sclareol and linalyl acetate are produced by glandular trichomes through the MEP pathway显示文摘Sclareol,an antifungal specialized metabolite produced by clary sage,Salvia sclarea,is the starting plant natural molecule used for the hemisynthesis of the perfume ingredient ambroxide.Sclareol is mainly produced in clary sage flower calyces;however,the cellular localization of the sclareol biosynthesis remains unknown.To elucidate the site of sclareol biosynthesis,we analyzed its spatial distribution in the clary sage calyx epidermis using laser desorption/ionization mass spectrometry imaging(LDI–FTICR-MSI)and investigated the expression profile of sclareol biosynthesis genes in isolated glandular trichomes(GTs).We showed that sclareol specifically accumulates in GTs’gland cells in which sclareol biosynthesis genes are strongly expressed.We next isolated a glabrous beardless mutant and demonstrate that more than 90%of the sclareol is produced by the large capitate GTs.Feeding experiments,using 1-13 C-glucose,and specific enzyme inhibitors further revealed that the methylerythritol-phosphate(MEP)biosynthetic pathway is the main source of isopentenyl diphosphate(IPP)precursor used for the biosynthesis of sclareol.Our findings demonstrate that sclareol is an MEP-derived diterpene produced by large capitate GTs in clary sage emphasing the role of GTs as biofactories dedicated to the production of specialized metabolites.Camille Chalvin Stephanie Drevensek Françoise Gilard Caroline Mauve Christel Chollet Halima Morin Edith Nicol Eva Heripre Lucie Kriegshauser Bertrand Gakiere Michel Dron Abdelhafid Bendahmane Adnane Boualem 2021Horticulture Research2021,8,1:2
