| 1 | Comment to: Single-cell profiling reveals the trajectories of natural killer cell differentiation in bone marrow and a stress signature induced by acute myeloid leukemia显示文摘We have read with interest the article by Crinier et al.describing a comprehensive single-cell RNA sequencing dataset of human bone marrow natural killer(NK)cells from eight healthy donors.1 Their unbiased analysis resulted in the identification of three NK cell populations in bone marrow:hNK_Bm1,hNK_Bm2 and hNK_Bm3.These populations correspond to the NK cell populations that the authors previously described in human spleen:hNK_Spl1,hNK_Sp2 and hNK_Sp3.2 The authors compared the gene signatures of the three bone marrow populations to the conventional CD56bright and CD56dim NK cells in the blood and concluded that hNK_Bm1 cells resemble CD56dim NK cells,hNK_Bm2 cells correspond to CD56bright NK cells,and hNK_Bm3 represent a CD56bright tissue-resident NK cell population that is not present in the blood. | Janine E.Melsen Gertjan Lugthart Monique Mvan Ostaijen-ten Dam Marco W.Schilham | 2021 | Cellular & Molecular Immunology2021,18,5: | 1 |
| 2 | 线粒体分裂抑制剂1对NK细胞活化的影响显示文摘目的:观察线粒体分裂抑制剂1(Mdivi-1)对自然杀伤(NK)细胞活化的影响。方法:以人NK细胞系NK-92MI为研究对象,采用CCK-8法筛选Mdivi-1适宜的干预浓度。据此结果,分为对照组、重组人胸腺基质淋细胞生成素(TSLP)组和Mdivi-1低、中、高剂量组。其中,TSLP组以20μg/L TSLP刺激24 h,Mdivi-1组于20μg/L TSLP刺激下以相应浓度Mdivi-1干预24 h。采用RT-qPCR检测Mdivi-1干预后细胞中白细胞介素4(IL-4)、IL-5和干扰素γ(IFN-γ)的mRNA表达;以Western blot分析测定各组细胞IL-4、IL-5和IFN-γ的蛋白表达及JAK2和STAT6的磷酸化水平;以MitoSOX Red染色探查各组细胞的活性氧(ROS)水平。结果:CCK-8结果显示,1、5和10μmol/L为Mdivi-1干预NK-92MI细胞的适宜浓度。RT-qPCR结果显示,与对照组比较,TSLP组细胞IL-4和IL-5的mRNA表达显著升高,IFN-γ的mRNA水平下调(P<0.05);而5和10μmol/L Mdivi-1干预则有效逆转了上述异常变化。Western blot结果显示,与TSLP组比较,5和10μmol/L Mdivi-1组的IL-4表达下调,IFN-γ水平则有所回升,10μmol/L Mdivi-1组的IL-5表达降低(P<0.05)。与TSLP组对比,5和10μmol/L Mdivi-1组的JAK2磷酸化水平降低,10μmol/L Mdivi-1组STAT6的磷酸化水平降低(P<0.05)。同时,5和10μmol/L Mdivi-1干预有效抑制了TSLP刺激下NK-92MI细胞ROS水平的升高。结论:Mdivi-1能有效抑制TSLP刺激所诱导的NK细胞NK2型分化,该效应可能与其对ROS及JAK2/STAT6信号通路的调节作用相关。 | 陈旭青 周龙云 史军 吴继勇 严芮雯 严道南 马华安 | 2021 | 中国病理生理杂志2021,37,9: | 0 |

| 5 | 重建线粒体动态平衡对NK细胞NK2型活化影响显示文摘目的立于“线粒体功能紊乱-细胞异常活化”视角,探讨NK细胞NK2型活化机制。方法将NK-92MI细胞分为空白组、TSLP组、1、5、10μmol·L^(-1) Mdivi-1组。ELISA法测定各组IL-4、IL-5、IFN-γ水平;蛋白印迹法分析各组p-Drp1、MnSOD蛋白表达;DHE染色及流式检测各组ROS水平;激光共聚焦观察各组线粒体形态。结果ELISA显示,与对照组比较,TSLP组IL-4、IL-5水平明显升高,IFN-γ水平下调(P<0.05);与TSLP组比较,5、10μmol·L^(-1) Mdivi-1组IL-4、IL-5水平降低,10μmol·L^(-1) Mdivi-1组IFN-γ浓度有所回升(P<0.05)。DHE染色及流式显示,TSLP组ROS水平较对照组升高;而5、10μmol·L^(-1) Mdivi-1组ROS水平较TSLP组降低(P<0.05)。共聚焦显示,TSLP刺激后,细胞内大量圆球状线粒体生成,5、10μmol·L^(-1) Mdivi-1干预后该现象得到改善。蛋白印迹显示,TSLP组p-Drp1水平明显上调,MnSOD表达下降,Mdivi-1干预则有效逆转了上述变化。结论线粒体动态失衡可能是NK细胞异常活化内在机制之一,其或是调控NK细胞NK2型活化介导的变应性炎症反应的重要靶点。 | 陈旭青 周龙云 史军 吴继勇 于希忠 严芮雯 严道南 马华安 | 2022 | 中国药理学通报2022,38,2: | 0 |
