2异硫氰酸苯己酯对Molt-4细胞组蛋白甲基化、乙酰化调控的实验研究显示文摘目的研究异硫氰酸苯己酯(PHI)在体外对淋巴细胞白血病 Molt-4细胞系的作用,观察 PHI 对 Molt-4细胞组蛋白甲基化、乙酰化调控的影响。方法采用 MTF 法、克隆抑制实验观察 PHI对 Molt-4细胞增殖的影响;采用流式细胞术检测 PHI 诱导细胞凋亡和对细胞周期的影响;用 Westernblot 法观察 PHI 作用后细胞的组蛋白乙酰化酶、组蛋白甲基化及乙酰化状态的变化。结果 PHI 可上调 Molt-4细胞组蛋白乙酰化酶 P300/CBP 水平,显著提高组蛋白 H3、H4乙酰化及 H3K4甲基化水平,抑制组蛋白甲基化 H3K9表达,阻滞细胞于 G_0/G_1期,并诱导细胞凋亡。结论 PHI 可能是一种组蛋白去乙酰化酶抑制剂,同时能调控组蛋白甲基化,影响其表观遗传学,可能作为新的抗白血病治疗药物。黄轶群 马旭东 郑瑞玑 CHIAO Jen-wei LIU De-long 2007中华血液学杂志2007,28,9:16
3Role of surgical intervention in managing gastrointestinal metastases from lung cancer显示文摘AIM:To investigate the clinicopathological characteristics of late-stage lung cancer patients with gastrointestinal(GI)-tract metastases,focusing on therapeutic options and outcomes.METHODS:Our institution(the National Taiwan University Hospital) diagnosed 8159 patients with lung cancer between 1987 and 2008,of which 21 developed symptomatic GI metastases.This study reviewed all of the patients' information,including survival data,pathological reports,and surgical notes.RESULTS:The most common histological type of lung cancer was adenocarcinoma,and 0.26% of patients with lung cancer developed GI metastases.The median duration from lung cancer diagnosis to GI metastases was three months(range,0-108 mo),and the average time from diagnosis of GI metastasis to death was 2.8 mo.Most patients with symptomatic gastric and/or duodenal metastases exhibited GI bleeding and were diagnosed by panendoscopy.In contrast,small bowel metastases typically presented as an acute abdomen and were not diagnosed until laparotomy.All patients with small bowel or colonic metastases underwent surgical intervention,and their perioperative mortality was 22%.Our data revealed a therapeutic effect in patients with solitary GI metastasis and a favorable palliative effect on survival when metastases were diagnosed preoperatively.In patients with multiple GI metastases,the presentation varied according to the locations of the metastases.CONCLUSION:Surgical treatment is worthwhile in a select group of patients with bowel perforation or obstruction.Physicians should be more alert to symptoms or signs indicating GI metastases.Po-Chu Lee Chiao Lo Ming-Tsan Lin Jin-Tung Liang Been-Ren Lin 2011World Journal of Gastroenterology2011,17,38:7

8PHSICAL MODELLING AND PROCESSING OF Ti-6Al-4V ELI显示文摘Titanium has been widely used as the implant materials of joint prostheses. This paper simulates the microstructures of the hot forged titanium knee joint component by physical modeling. Ti-6Al-4V ELI were deformed by GLEEBLE 2000 hot deforma- tion simulator in compression mode to determine the hot workability, in the range of 900 to 1100℃, with the strain rates between 0.05 to 5 s-1. Test results derived from specimens processed by on-cooling test and on-heating test were compared, thereby estimating the effect due to thermal history. 9异硫氰酸苯己酯调控肝癌细胞株SMMC-7721组蛋白乙酰化及诱导细胞凋亡显示文摘目的 研究异硫氰酸苯己酯(PHI)对肝癌SMMC-7721细胞组蛋白乙酰化调控及凋亡的影响.方法 采用台盼蓝拒染直接计数法观察PHI对SMMC-7721细胞增殖的影响,采用原位末端标记(TUNEL)法检测PHI对SMMC-7721细胞凋亡的影响,采用Western blot法检测PHI对SMMC-7721细胞组蛋白乙酰化及凋亡相关蛋白表达的影响.结果 PHI可抑制SMMC-7721细胞的增殖,与0 μmol/L作用组比较,5、10、20、40和80 μmol/L的PHI对SMMC-7721细胞均有不同程度的增殖抑制作用.PHI可诱导SMMC-7721细胞产生凋亡,PHI作用于SMMC-7721细胞7 h后,10、20和40 μmol/LPHI组的细胞凋亡率分别为6.9%±2.4%、17.5%±4.2%和54.5%±5.4%,明显高于0 μmol/L PHI组(4.5%±2.3%,P<0.05).PHI作用于SMMC-7721细胞3 h时,与0 μmol/L PHI组比较,10、20和40 μmol/L PHI组中Bcl-2、Procaspse-9和Procaspse-3的表达下降,caspase-9和caspase-3的表达上升,而Procaspase-8的表达未见明显变化;作用7 h时,这种变化趋势更加明显.PHI作用于SMMC-7721细胞3 h时,与0 μmol/L PHI组比较,10、20和40 μmol/L PHI组中组蛋白H3的乙酰化分别增加了1.87倍、2.43倍和3.67倍,组蛋白H4的乙酰化分别增加了1.29倍、1.45倍和2.25倍;作用7 h时,这种变化趋势更加明显.结论 PHI是一种组蛋白去乙酰化酶抑制剂,可调控组蛋白的乙酰化水平,影响其表观遗传学,并通过线粒体凋亡途径诱导细胞凋亡.赖亚栋 马旭东 黄轶群 许向农 王小众 Dicky JW Chiao Delong liu 2010中华肿瘤杂志2010,32,11:2

12Nanotrap-enabled quantification of KRAS-induced peptide hydroxylation in blood for cancer early detection显示文摘Circulating peptide is a potential source of biomarkers for cancer detection.However,the existence of large molecular weight proteins in plasma have a disastrous effect on circulating peptides isolating and detecting.Herein,nanotrap fractionation following by mass spectrometry have been applied to quantify the levels of bradykinin (BK) and hydroxylated bradykinin (Hyp-BK) as a relative measure of KRAS-regulated prolyl-4-hydroxylase alpha-1 (P4HA1) which may serve as early diagnosis marker for pancreatic ductal adenocarcinoma (PDAC).We found that P4HA1 can be upregulated by KRASG12v,which is a PDAC driver mutation,using HPNE/KRAS and HPNE cells.And we revealed that P4HA1 is overexpressed in PDAC tumors,compared to normal and inflamed pancreatic tissues.RNA interference revealed that P4HA1 activity was primarily responsible for Hyp-BK production.Mass spectrometry analysis revealed that plasma Hyp-BK/BK ratio was higher in PDAC than pancreatitis patients and healthy controls,while the area under the receiver operating characteristic (ROC) curve (AUC) is 0.8209 (95%Cl,0.7269-0.9149).The Hyp-BK/BK association with PDAC was reproduced in another cohort,where this ratio was found to increase with advancing tumor stage.These novel findings paved the way for wider applications of Nanotrap coupled mass spectrometry as a powerful tool for revealing biosignatures from plasma.Zaian Deng Zhen Zhao Bo Ning Jeffery Basilio Karen Mann Jie Fu Yajun Gu Yuanqing Ye Xifeng Wu Jia Fan Paul Chiao Tony Hu 2019Nano Research2019,12,6:2
