Effects of AT1 receptor antagonist,Iosartan,on rat hepatic fibrosis induced by CCl_4显示文摘AIM To investigate effect of losartan,an AT1receptor antagonist,on hepatic fibrosis induced byCCl;and to determine whether or not AT1receptors are expressed on hepatic stellate cells,METHODS AND RESULTS Fifty male Sprague-Dawley rats,weighing(180±20)g,wererandomized into five groups(control group, modelgroup, and three losartan treated groups),inwhich all rats were given the subcutaneousinjection of 40% CCl4(every 3 days for 6 weeks)except for rats of control group.Rats of losartan-treated groups were treated with losartan(20 mg/kg,10 mg/kg,5 mg/kg,daily gavage),After 6weeks liver tissue and serum samples of all ratswere examined.Serum hyaluronic acid(HA),procollagen typeⅢ(PCⅢ)were detected byradioimmunoassays, van Giesion collagen stainingwas used to evaluate the extracellular matrix of ratswith liver fibrosis. The expression of AT1receptors,transforming growth factor-beta(TGF-β),and alpha-smooth muscle actin(a-SMA)inliver tissue were determined byimmunohistochemical techniques.Compared withmodel group, serum ALT and AST of losartan-treated groups were significantly reduced(t=4.20,P<0.01 and t=4.57,P<0.01).Serum HAand PCⅢalso had significant differences(t=3.53,P<0.01 and t=2.20,P<0.05).Thedegree of fibrosis was improved by losartan and correlated with the expressions of AT1 receptors,TGF-β,and α-SMA in liver tissue.CONCLUSION AT1 receptor antagonist, losartan, could limit the progression of the hepatic fibrosisinduced by CCl4.The mechanism may be related tothe decrease in the expression of AT1 receptorsand TGF-β,ameliorating the injury of hepatocytes;activation of local renin-angiotensin system mightrelate to hepatic fibrosis;and during progressionof fibrosis, activated hepatic stellate cells mightexpress AT1 receptors. | Hong Shan Wei Ding Guo Li Han Ming Lu Yu Tao Zhan Zhi Rong Wang Xin Huang Jing Zhang Ji Lin Cheng Qin Fang Xu Department of Gastroenterology,Xinhua Hospital,Shanghai Second Medical University,Shanghai 200092,China | 2000 | World Journal of Gastroenterology2000,6,4: | 42
Review on utilization of biochar for metal-contaminated soil and sediment remediation显示文摘Biochar is a carbon-neutral or even carbon-negative material produced through thermal decomposition of plant-and animal-based biomass under oxygen-limited conditions.Recently, there has been an increasing interest in the application of biochar as an adsorbent, soil ameliorant and climate mitigation approach in many types of applications.Metal-contaminated soil remediation using biochar has been intensively investigated in small-scale and pilot-scale trials with obtained beneficial results and multifaceted effects. But so far, the study and application of biochar in contaminated sediment management has been very limited, and this is also a worldwide problem. Nonetheless, there is reason to believe that the same multiple benefits can also be realized with these sediments due to similar mechanisms for stabilizing contaminants. This paper provides a review on current biochar properties and its use as a sorbent/amendment for metal-contaminated soil/sediment remediation and its effect on plant growth, fauna habits as well as microorganism communities. In addition, the use of biochar as a potential strategy for contaminated sediment management is also discussed, especially as regards in-situ planning. Finally, we highlight the possibility of biochar application as an effective amendment and propose further research directions to ensure the safe and sustainable use of biochar as an amendment for remediation of contaminated soil and sediment. | Mingming Wang Yi Zhu Lirong Cheng Bruce Andserson Xiaohui Zhao Dayang Wang Aizhong Ding | 2018 | Journal of Environmental Sciences2018,30,1: | 43
Enhanced secondary pollution offset reduction of primary emissions during COVID-19 lockdown in China显示文摘To control the spread of the 2019 novel coronavirus(COVID-19),China imposed nationwide restrictions on the movement of its population(lockdown)after the Chinese New Year of 2020,leading to large reductions in economic activities and associated emissions.Despite such large decreases in primary pollution, there were nonetheless several periods of heavy haze pollution in eastern China, raising questions about the well-established relationship between human activities and air quality. Here, using comprehensive measurements and modeling,we show that the haze during the COVID lockdown was driven by enhancements of secondary pollution. In particular, large decreases in NOx emissions from transportation increased ozone and nighttime NO_(3)radical formation, and these increases in atmospheric oxidizing capacity in turn facilitated the formation of secondary particulate matter. Our results, afforded by the tragic natural experiment of the COVID-19 pandemic, indicate that haze mitigation depends upon a coordinated and balanced strategy for controlling multiple pollutants. | Xin Huang Aijun Ding Jian Gao Bo Zheng Derong Zhou Ximeng Qi Rong Tang Jiaping Wang Chuanhua Ren Wei Nie Xuguang Chi Zheng Xu Liangduo Chen Yuanyuan Li Fei Che Nini Pang Haikun Wang Dan Tong Wei Qin Wei Cheng Weijing Liu Qingyan Fu Baoxian Liu Fahe Chai Steven JDavis Qiang Zhang Kebin He | 2021 | National Science Review2021,8,2: | 47
Activation of gibberellin 2-oxidase 6 decreases active gibberellin levels and creates a dominant semi-dwarf phenotype in rice (Oryza sativa L.)显示文摘Gibberellin (GA) 2-oxidase plays a key role in the GA catabolic pathway through 2β-hydroxylation.In the present study,we isolated a CaMV 35S-enhancer activation tagged mutant,H032.This mutant exhibited a dominant dwarf and GA-deficient phenotype, with a final stature that was less than half of its wild-type counterpart. The endogenous bioactive GAs are markedly decreased in the H032 mutant, and application of bioactive GAs (GA3 or GA4) can reverse the dwarf phenotype. The integrated T-DNA was detected 12.8 kb upstream of the OsGA2ox6 in the H032 genome by TAIL-PCR.An increased level of OsGA2ox6 mRNA was detected at a high level in the H032 mutant, which might be due to the enhancer role of the CaMV 35S promoter.RNAi and ectopic expression analysis of OsGA2ox6 indicated that the dwarf trait and the decreased levels of bioactive GAs in the H032 mutant were a result of the up-regulation of the OsGA2ox6 gene.BLASTP analysis revealed that OsGA2ox6 belongs to the class III of GA 2-oxidases,which is a novel type of GA2ox that uses C20-GAs (GA12 and/or GA53) as the substrates.Interestingly,we found that a GA biosynthesis inhibitor, paclobutrazol, positively regulated the OsGA2ox6 gene.Unlike the over-expression of OsGA2ox1,which led to a high rate of seed abortion, the H032 mutant retained normal flowering and seed production. These results indicate that OsGA2ox6 mainly affects plant stature, and the dominant dwarf trait of the H032 mutant can be used as an efficient dwarf resource in rice breeding. | Jian Huang Ding Tang Yi Shen Baoxiang Qin Lilan Hong Aiqing You Ming Li Xin Wang Hengxiu Yu Minghong Gu Zhukuan Cheng | 2010 | Journal of Genetics and Genomics2010,37,1: | 36

Blockade of cholecystokinin-2 receptor and cyclooxygenase-2 synergistically induces cell apoptosis, and inhibits the proliferation of human gastric cancer cells in vitro显示文摘Gastrin and cyclooxygenase-2(COX-2) playimportant roles in the carcinogenesis and progression ofgastric cancer. However,it remains unknown whether the combination of cholecystokinin-2(CCK-2) receptor antagonist plus COX-2 inhibitor exerts synergistic anti-tumor effects on human gastric cancer. Here,we demonstrated that the combination of AG-041R(a CCK-2 receptor antagonist) plus NS-398(a selective COX-2 inhibitor) treatment had synergistic effects on proliferation inhibition, apoptosis induction,down-regulation of Bcl-2 and up-regulation of Bax expression in MKN-45 cells. These results indicate that simultaneous targeting of CCK-2 receptor and COX-2 may inhibit gastric cancer development more effectively than targeting either molecule alone.(C)2008 Elsevier Ireland Ltd.All rights reserved. | Sun, W. | 8 | The regulatory role of AT 1 receptor on activated HSCs in hepat,c fibrogenesis, effects of RAS inhibitors on hepatic fibrosis induced by CCl_4显示文摘AIM To assess the effect of ACE inhibitor andAng Ⅱ type Ⅰ(AT1)receptor antagonist inpreventing hepatic fibrosis caused by CCl4administration in rats;to investigate whether ornot there are expression of AT 1 receptors onhepatic stellate cells;and to observe the effectof Ang Ⅱ on proliferation and ECM synthesis ofcultured HSCs.METHODS Studies were conducted in maleSprague-Dawley rats.Except for thehepatofibrotic model group and the controlgroup,in three treated groups, either enalapril(5 mg/kg),or Iosartan(10 mg/kg),or enalapril+Iosartan were given to the fibrotic rats bydaily gavage, and saline vehicle was given tomodel and normal control rats. After 6 weeks, liver fibrosis was assessed directly by hepaticmorphometric analysis, which has beenconsidered the gold standard for thequantification of fibrosis. The expressions of AT1 receptors and(α-mooth muscle actin,α-SMA)in liver tissue or isolated hepatic stellate cells(HSCs)were detected by immunohistochemicaltechniques. The effect of Ang Ⅱ on HSCproliferation was determined by MTT method.Effect of Ang Ⅱ on collagen synthesis of HSCswas determined by 3H-proline incorporation.RESULTS Contrasted to the fibrosis in rats ofthe model group, groups of rats treated with either enalapril or Iosartan,or a combination oftwo drugs showed a limited expansion of theinterstitium(4.23±3.70 vs 11.22±4.79,P<0.05),but no difference was observedamong three treated groups(5.38±3.43,4.96±2.96,4.23±2.70,P>0.05).Expression of AT 1receptors was found in fibrotic interstitium offibrotic rats, whereas in normal control rats theywere limited to vasculature only to a very slightdegree.AT 1 receptors were also expressed onactivated HSCs in the culture. At concentrationsfrom 10-9to 10-5mol/L,Ang Ⅱ stimulated HSCproliferation in culture in a dose-dependentmanner.Increasing Ang Ⅱ concentrationsproduced corresponding increases in 3H-prolineincorporation.Differences among groups were significant.CONCLUSION Angiotensin-converting enzyme inhibitors and AT I blocker may slow the progression of hepatic fibrosis; activated HSCs express AT 1 receptors, and Ang Ⅱ can stimulate the proliferation and collagen synthesis of HSCs in a dose-dependent manner; and activation of RAS may be related to hepatic fibrogenesis induced by CCI4. | Hong Shan Wei Han Ming Lu Ding Guo Li Yu Tao Zhan Zhi Rong Wang Xin Huang Ji Lin Cheng Qin Fang Xu Department of Gastroenterology,Xinhua Hospital,Shanghai Second Medical University,Shanghai 200092,China | 2000 | World Journal of Gastroenterology2000,6,6: | 27
Crowd intelligence in AI 2.0 era显示文摘The Internet based cyber-physical world has profoundly changed the information environment for the development of artificial intelligence(AI), bringing a new wave of AI research and promoting it into the new era of AI 2.0. As one of the most prominent characteristics of research in AI 2.0 era, crowd intelligence has attracted much attention from both industry and research communities. Specifically, crowd intelligence provides a novel problem-solving paradigm through gathering the intelligence of crowds to address challenges. In particular, due to the rapid development of the sharing economy, crowd intelligence not only becomes a new approach to solving scientific challenges, but has also been integrated into all kinds of application scenarios in daily life, e.g., online-tooffline(O2O) application, real-time traffic monitoring, and logistics management. In this paper, we survey existing studies of crowd intelligence. First, we describe the concept of crowd intelligence, and explain its relationship to the existing related concepts, e.g., crowdsourcing and human computation. Then, we introduce four categories of representative crowd intelligence platforms. We summarize three core research problems and the state-of-the-art techniques of crowd intelligence. Finally, we discuss promising future research directions of crowd intelligence. | Wei LI Wen-jun WU Huai-min WANG Xue-qi CHENG Hua-jun CHEN Zhi-hua ZHOU Rong DING | 2017 | Frontiers of Information Technology & Electronic Engineering2017,18,1: | 27

Apoptosis of human pancreatic cancer cells induced by Triptolide显示文摘AIM: To investigate apoptosis in human pancreatic cancer cells induced by Triptolide (TL), and the relationship between this apoptosis and expression of caspase-3' bcl-2 and bax. METHODS: Human pancreatic cancer cell line SW1990 was cultured in DMEM media for this study. MTT assay was used to determine the cell growth inhibitory rate in vitro. Flow cytometry and TUNEL assay were used to detect the apoptosis of human pancreatic cancer cells before and after TL treatment. RT-PCR was used to detect the expression of apoptosis-associated gene caspase-3' bcl-2 and bax. RESULTS: TL inhibited the growth of human pancreatic cancer cells in a dose-and time-dependent manner. TL induced human pancreatic cancer cells to undergo apoptosis with typically apoptotic characteristics. TUNEL assay showed that after the treatment of human pancreatic cancer cells with 40 ng/mL TL for 12 h and 24 h, the apoptotic rates of human pancreatic cancer cells increased significantly. RT-PCR demonstrated that caspase-3 and bax were significantly up-regulated in SW1990 cells treated with TL while bcl-2 mRNA was not. CONCLUSION: TL is able to induce the apoptosis in human pancreatic cancer cells. This apoptosis may be mediated by up-regulating the expression of apoptosis- associated caspase-3 and bax gene. | Guo-Xiong Zhou Xiao-Ling Ding Jie-Fei Huang Hong Zhang Sheng-Bao Wu Jian-Ping Cheng Qun Wei | 2008 | World Journal of Gastroenterology2008,14,10: | 22

The effect of an electric field on the thermomechanical damage of nodular defects in dielectric multilayer coatings irradiated by nanosecond laser pulses显示文摘Thermomechanical damage of nodules in dielectric multilayer coatings that are irradiated by nanosecond laser pulses has been interpreted with respect to mechanical properties and electric-field enhancement. However, the effect of electric-field enhancement in nodular damage, especially the influence of electric-field distributions, has never been directly demonstrated through experimental results, which prevents the achievement of a clear understanding of the damage process of nodular defects. Here,a systematic and comparative study was designed to reveal how electric-field distributions affect the damage behavior of nodules. To obtain reliable results, two series of artificial nodules with different geometries and film absorption characteristics were prepared from monodisperse silica microspheres. After establishing simplified geometrical models of the nodules, the electric-field enhancement was simulated using a three-dimensional finite-difference time-domain code. Then, the damage morphologies of the artificial nodules were directly compared with the simulated electric-field intensity profiles. For both series of nodules, the damage morphologies reproduced our simulated electric-field intensity distributions very well. These results indicated that the electric-field distribution was actually a bridge that connected the nodular mechanical properties to the final thermomechanical damage.Understanding of the damage mechanism of nodules was deepened by obtaining data on the influence of electric-field distributions on the damage behavior of nodules. | Xinbin Cheng Jinlong Zhang Tao Ding Zeyong Wei Hongqiang Li Zhanshan Wang | 2013 | Light(Science & Applications)2013,2,1: | 20

Computed tomography-based radiomics for prediction of neoadjuvant chemotherapy outcomes in locally advanced gastric cancer: A pilot study显示文摘Objective: The standard treatment for patients with locally advanced gastric cancer has relied on perioperative radio-chemotherapy or chemotherapy and surgery. The aim of this study was to investigate the wealth of radiomics for pre-treatment computed tomography(CT) in the prediction of the pathological response of locally advanced gastric cancer with preoperative chemotherapy. Methods: Thirty consecutive patients with CT-staged II/III gastric cancer receiving neoadjuvant chemotherapy were enrolled in this study between December 2014 and March 2017. All patients underwent upper abdominal CT during the unenhanced, late arterial phase(AP) and portal venous phase(PP) before the administration of neoadjuvant chemotherapy. In total, 19,985 radiomics features were extracted in the AP and PP for each patient.Four methods were adopted during feature selection and eight methods were used in the process of building the classifier model. Thirty-two combinations of feature selection and classification methods were examined. Receiver operating characteristic(ROC) curves were used to evaluate the capability of each combination of feature selection and classification method to predict a non-good response(non-GR) based on tumor regression grade(TRG).Results: The mean area under the curve(AUC) ranged from 0.194 to 0.621 in the AP, and from 0.455 to 0.722 in the PP, according to different combinations of feature selection and the classification methods. There was only one cross-combination machine-learning method indicating a relatively higher AUC(>0.600) in the AP, while 12 cross-combination machine-learning methods presented relatively higher AUCs(all >0.600) in the PP. The feature selection method adopted by a filter based on linear discriminant analysis + classifier of random forest achieved a significantly prognostic performance in the PP(AUC, 0.722±0.108; accuracy, 0.793; sensitivity, 0.636; specificity,0.889; Z=2.039; P=0.041).Conclusions: It is possible to predict non-GR after neoadjuvant chemotherapy in locally advanced gastric cancers based on the radiomics of CT. | Zhenhui Li Dafu Zhang Youguo Dai Jian Dong Lin Wu Yajun Li Zixuan Cheng Yingying Ding Zaiyi Liu | 2018 | Chinese Journal of Cancer Research2018,30,4: | 18
A general end-to-end diagnosis framework for manufacturing systems显示文摘The manufacturing sector is envisioned to be heavily influenced by artificial-intelligence-based technologies with the extraordinary increases in computational power and data volumes. A central challenge in the manufacturing sector lies in the requirement of a general framework to ensure satisfied diagnosis and monitoring performances in different manufacturing applications. Here, we propose a general data-driven,end-to-end framework for the monitoring of manufacturing systems. This framework, derived from deep-learning techniques, evaluates fused sensory measurements to detect and even predict faults and wearing conditions. This work exploits the predictive power of deep learning to automatically extract hidden degradation features from noisy, time-course data. We have experimented the proposed framework on 10 representative data sets drawn from a wide variety of manufacturing applications. Results reveal that the framework performs well in examined benchmark applications and can be applied in diverse contexts, indicating its potential use as a critical cornerstone in smart manufacturing. | Ye Yuan Guijun Ma Cheng Cheng Beitong Zhou Huan Zhao Hai-Tao Zhang Han Ding | 2020 | National Science Review2020,7,2: | 18
An emission source inversion model based on satellite data and its application in air quality forecasts显示文摘This paper aims at constructing an emission source inversion model using a variational processing method and adaptive nudging scheme for the Community Multiscale Air Quality Model (CMAQ) based on satellite data to investigate the applicability of high resolution OMI (Ozone Monitoring Instrument) column concentration data for air quality forecasts over the North China. The results show a reasonable consistency and good correlation between the spatial distributions of NO2 from surface and OMI satellite measurements in both winter and summer. Such OMI products may be used to implement integrated variational analysis based on observation data on the ground. With linear and variational corrections made, the spatial distribution of OMI NO2 clearly revealed more localized distributing characteristics of NO2 concentration. With such information, emission sources in the southwest and southeast of North China are found to have greater impacts on air quality in Beijing. When the retrieved emission source inventory based on high-resolution OMI NO2 data was used, the coupled Weather Research Forecasting CMAQ model (WRF-CMAQ) performed significantly better in forecasting NO2 concentration level and its tendency as reflected by the more consistencies between the NO2 concentrations from surface observation and model result. In conclusion, satellite data are particularly important for simulating NO2 concentrations on urban and street-block scale. High-resolution OMI NO2 data are applicable for inversing NOx emission source inventory, assessing the regional pollution status and pollution control strategy, and improving the model forecasting results on urban scale. | CHENG XingHong XU XiangDe DING GuoAn | 2010 | Science China Earth Sciences2010,53,5: | 17
Polyamine Accumulation in Transgenic Tomato Enhances the Tolerance to High Temperature Stress显示文摘Polyamines play an important role in plant response to abiotic stress.S-adenosyl-l-methionine decarboxylase(SAMDC) is one of the key regulatory enzymes in the biosynthesis of polyamines. In order to better understand the effect of regulation of polyamine biosynthesis on the tolerance of high-temperature stress in tomato,SAMDC cDNA isolated from Saccharomyces cerevisiae was introduced into tomato genome by means of Agrobacterium tumefaciens through leaf disc transformation.Transgene and expression was confirmed by Southern and Northern blot analyses, respectively.Transgenic plants expressing yeast SAMDC produced 1.7-to 2.4-fold higher levels of spermidine and spermine than wild-type plants under high temperature stress, and enhanced antioxidant enzyme activity and the protection of membrane lipid peroxidation was also observed. This subsequently improved the efficiency of CO2 assimilation and protected the plants from high temperature stress, which indicated that the transgenic tomato presented an enhanced tolerance to high temperature stress(38℃) compared with wild-type plants. Our results demonstrated clearly that increasing polyamine biosynthesis in plants may be a means of creating high temperature-tolerant germplasm. | Lin Cheng Yijing Zou Shuli Ding Jiajing Zhang Xiaolin Yu Jiashu Cao Gang Lu | 2009 | Journal of Integrative Plant Biology2009,51,5: | 16
