| 1 | MAP Kinases in Immune Responses显示文摘MAP kinases are evolutionarily conserved signaling regulators from budding yeast to mammals and play essential roles in both innate and adaptive immune responses. There are three main families of MAPKs in mammals. Each of them has its own activators, inactivators, substrates and scaffolds, which altogether form a fine signaling network in response to different extracellular or intracellular stimulation. | 2 | Dissolved organic matter removal using magnetic anion exchange resin treatment on biological effluent of textile dyeing wastewater显示文摘This study investigated the removal of dissolved organic matter(DOM) from real dyeing bio-treatment effluents(DBEs) with the use of a novel magnetic anion exchange resin(NDMP).DOMs in two typical DBEs were fractionized using DAX-8/XAD-4 resin and ultrafiltration membranes. The hydrophilic fractions and the low molecular weight(MW)(<3 kDa) DOM fractions constituted a major portion(>50%) of DOMs for the two effluents. The hydrophilic and low MW fractions of both effluents were the greatest contributors of specific UV254absorbance(SUVA254),and the SUVA254 of DOM fractions decreased with hydrophobicity and MW. Two DBEs exhibited acute and chronic biotoxicities. Both acute and chronic toxicities of DOM fractions increased linearly with the increase of SUVA254 value. Kinetics of dissolved organic carbon(DOC) removal via NDMP treatment was performed by comparing it with that of particle active carbon(PAC). Results indicated that the removal of DOC from DBEs via NDMP was 60%,whereas DOC removals by PAC were lower than 15%. Acidic organics could be significantly removed with the use of NDMP. DOM with large MW in DBE could be removed significantly by using the same means. Removal efficiency of NDMP for DOM decreased with the decrease of MW. Compared with PAC,NDMP could significantly reduce the acute and chronic bio-toxicities of DBEs. | 4 | Genome-wide association analysis identified molecular markers associated with important tea flavor-related metabolites显示文摘The characteristic secondary metabolites in tea(theanine,caffeine,and catechins)are important factors contributing to unique tea flavors.However,there has been relatively little research on molecular markers related to these metabolites.Thus,we conducted a genome-wide association analysis of the levels of these tea flavor-related metabolites in three seasons.The theanine,caffeine,and catechin levels in Population 1 comprising 191 tea plant germplasms were examined,which revealed that their heritability exceeded 0.5 in the analyzed seasons,with the following rank order(highest to lowest heritabilities):(+)-catechin>(−)-gallocatechin gallate>caffeine=(−)-epicatechin>(−)-epigallocatechin-3-gallate>theanine>(−)-epigallocatechin>(−)-epicatechin-3-gallate>catechin gallate>(+)-gallocatechin.The SNPs detected by amplified-fragment SNP and methylation sequencing divided Population 1 into three groups and seven subgroups.An association analysis yielded 307 SNP markers related to theanine,caffeine,and catechins that were common to all three seasons.Some of the markers were pleiotropic.The functional annotation of 180 key genes at the SNP loci revealed that FLS,UGT,MYB,and WD40 domain-containing proteins,as well as ATP-binding cassette transporters,may be important for catechin synthesis.KEGG and GO analyses indicated that these genes are associated with metabolic pathways and secondary metabolite biosynthesis.Moreover,in Population 2(98 tea plant germplasm resources),30 candidate SNPs were verified,including 17 SNPs that were significantly or extremely significantly associated with specific metabolite levels.These results will provide a foundation for future research on important flavor-related metabolites and may help accelerate the breeding of new tea varieties. | Kaixing Fang Zhiqiang Xia Hongjian Li Xiaohui Jiang Dandan Qin Qiushuang Wang Qing Wang Chendong Pan Bo Li Hualing Wu | 2021 | Horticulture Research2021,8,1: | 10 |

| 6 | Protective effect of melatonin against liver injury in mice induced by Bacillus Calmette-Guerin plus lipopolysaccharide显示文摘AIM: To investigate the effects and mechanisms of melatonin on immunological liver injury in mice.METHODS: A model of liver injury was induced by tail vein injecton of Bad#us Calmet~ Guenn (BCG) and lipopolysaccharide(LPS) in mice. Kupffer cells and hepatocytes were isolated and cultured according to a modified two-step collagenase perfusion technique. Levels of alanine aminotransferase(ALT), aspartate aminotransferase (AST) and nitric oxide(NO), content of malondiadehyde (MDA), activity of superoxide dismutase (SOD), were measured by biochemical methods.Tumor necrosis factor-α (TNF-α) activity was determined by RIA. Interleukin (IL)-1 activity was measured by thymocyte proliferation bioassay. Hepatic tissue sections were stained with hematoxylin and eosin and examined under a lightmicroscope.RESULTS: Immunological liver injury induced by BCG+LPS was successfully duplicated. Serum transaminase (ALT,AST) activities were significantly decreased by melatonin(0.25, 1.0, 4.0 mg/kg bm). Meanwhile, MDA content was decreased and SOD in liver homogenates was upregulated.Furthermore, pro-inflammatory mediators (TNF-α, IL-1, NO)in serum and liver homogenates were significantly reduced by melatonin. Histological examination demonstrated that melatonin could attenuate the area and extent of necrosis,reduce the immigration of inflammatory cells. In in vitro experiment, TNF-α was inhibited at the concentrations of 10^-8-10^-6 mol/L of melatonin, while IL-1 production of Kupffer cells induced by LPS (5 t^g/mL) was decreased only at the concentration of 10^-6 mol/L of melatonin, but no effect on NO production was observed. Immunological liver injury model in vitro was established by incubating hepatocytes with BCG- and LPS-induced Kupffer cells. Activities of ALT,TNF-α, IL-1, and MDA in supernatant were significantly increased. | 8 | Spatial and temporal variations of sedimented organic matter in Xiaohai Lagoon,Hainan Island显示文摘The characteristics of grain-size,total organic carbon(TOC) and total nitrogen(TN) contents,TOC/TN ratios,stable carbon isotope(δ13C) and 210 Pb dating were measured in six sediment cores from the Xiaohai Lagoon.The results show distinct spatial and temporal variations in sedimentation patterns.The sediments are dominated by clayey silt,sandy silt and by silty sand in the southern,middle lagoon and the northern lagoon,respectively.TOC and TN contents decline from south to north.Sedimentation rates,determined by 210 Pb dating,tend to decrease from south to middle.However,the determination of sedimentation rate in the north is difficult.These spatial variations are related to the variations in sediment sources and hydrodynamic conditions in the Xiaohai Lagoon.The variations of organic matter signatures can be divided into two stages in the cores from the southern and middle lagoon.Before 1988,the organic matter signatures are relatively stable.The contribution of terrestrial organic carbon sources varies between 60% and 85%.After 1988,the organic matter signatures demonstrate significant variations.TOC and TN contents increase rapidly,TOC/TN ratios decrease,δ13C values shift to higher and the contribution of terrestrial organic carbon sources decreases to 40%-50%.The contributions of phytoplankton organic matter have increased in the sediment since 1988.Increasing aquaculture activities have had a significant impact on organic matter signatures since 1988.The sedimentation rates have increased rapidly in the southern and middle lagoon since 1988 due to the anthropogenic activities which include aquaculture,mining and deforestation.These activities have caused eutrophication and increased siltation in the southern and middle lagoon. | LIU Xingjian GE Chendong | 2012 | Acta Oceanologica Sinica2012,31,3: | 3 |

| 12 | Diurnal variations of carbon dioxide,methane,and nitrous oxide fluxes from invasive Spartina alterniflora dominated coastal wetland in northern Jiangsu Province显示文摘The invasions of the alien species such as Spartina alterniflora along the northern Jiangsu coastlines have posed a threat to biodiversity and the ecosystem function.Yet,limited attention has been given to their potential influence on greenhouse gas(GHG) emissions,including the diurnal variations of GHG fluxes that are fundamental in estimating the carbon and nitrogen budget.In this study,we examined the diurnal variation in fluxes of carbon dioxide(CO_2),methane(CH_4),and nitrous oxide(N2O) from a S.alterniflora intertidal flat in June,October,and December of 2013 and April of 2014 representing the summer,autumn,winter,and spring seasons,respectively.We found that the average CH_4 fluxes on the diurnal scale were positive during the growing season while negative otherwise.The tidal flat of S.alterniflora acted as a source of CH_4 in summer(June) and a combination of source and sink in other seasons.We observed higher diurnal variations in the CO_2 and N_2O fluxes during the growing season(1 536.5 mg CO_2 m^(–2) h^(–1) and 25.6 μg N_2O m^(–2) h^(–1)) compared with those measured in the non-growing season(379.1 mg CO_2 m^(–2) h^(–1) and 16.5 μg N_2O m^(–2) h^(–1)).The mean fluxes of CH_4 were higher at night than that in the daytime during all the seasons but October.The diurnal variation in the fluxes of CO_2 in June and N_2O in December fluctuated more than that in October and April.However,two peak curves in October and April were observed for the diurnal changes in CO_2 and N_2O fluxes(prominent peaks were found in the morning of October and in the afternoon of April,respectively).The highest diurnal variation in the N_2O fluxes took place at 15:00(86.4 μg N_2O m^(–2) h^(–1)) in June with an unimodal distribution.Water logging in October increased the emission of CO_2(especially at nighttime),yet decreased N_2O and CH_4 emissions to a different degree on the daily scale because of the restrained diffusion rates of the gases.The seasonal and diurnal variations of CH_4 and CO_2 fluxes did not correlate to the air and soil temperatures,whereas the seasonal and diurnal variation of the fluxes of N_2O in June exhibited a significant correlation with air temperature.When N_2O and CH_4 fluxes were converted to CO_2-e equivalents,the emissions of N_2O had a remarkable potential to impact the global warming.The mean daily flux(MF) and total daily flux(TDF) were higher in the growing season,nevertheless,the MF and TDF of CO_2 were higher in October and those of CH_4 and N_2O were higher in June.In spite of the difference in the optimal sampling times throughout the observation period,our results obtained have implications for sampling and scaling strategies in estimating the GHG fluxes in coastal saline wetlands. | XU Xinwanghao FU Guanghe ZOU Xinqing GE Chendong ZHAO Yifei | 2017 | Acta Oceanologica Sinica2017,36,4: | 2 |

| 14 | Development of the novel ACLY inhibitor 326E as a promising treatment for hypercholesterolemia显示文摘Hepatic cholesterol accumulation is an important contributor to hypercholesterolemia,which results in atherosclerosis and cardiovascular disease(CVD).ATP-citrate lyase(ACLY)is a key lipogenic enzyme that converts cytosolic citrate derived from tricarboxylic acid cycle(TCA cycle)to acetyl-CoA in the cytoplasm.Therefore,ACLY represents a link between mitochondria oxidative phosphorylation and cytosolic de novo lipogenesis.In this study,we developed the small molecule 326E with an enedioic acid structural moiety as a novel ACLY inhibitor,and its CoA-conjugated form 326E-CoA inhibited ACLY activity with an IC_(50)=5.31±1.2μmol/L in vitro.326E treatment reduced de novo lipogenesis,and increased cholesterol efflux in vitro and in vivo.326E was rapidly absorbed after oral administration,exhibited a higher blood exposure than that of the approved ACLY inhibitor bempedoic acid(BA)used for hypercholesterolemia.Chronic 326E treatment in hamsters and rhesus monkeys resulted in remarkable improvement of hyperlipidemia.Once daily oral administration of 326E for 24 weeks prevented the occurrence of atherosclerosis in ApoE^(-/-)mice to a greater extent than that of BA treatment.Taken together,our data suggest that inhibition of ACLY by 326E represents a promising strategy for the treatment of hypercholesterolemia. | Zhifu Xie Mei Zhang Qian Song Long Cheng Xinwen Zhang Gaolei Song Xinyu Sun Min Gu Chendong Zhou Yangming Zhang Kexin Zhu Jianpeng Yin Xiaoyan Chen Jingya Li Fajun Nan | 2023 | Acta Pharmaceutica Sinica B2023,13,2: | 1 |
