1Comprehensive identification of lysine 2-hydroxyisobutyrylated proteins in Ustilaginoidea virens reveals the involvement of lysine 2-hydroxyisobutyrylation in fungal virulence显示文摘Lysine 2-hydroxyisobutyrylation(Khib) is a newly identified post-translational modification(PTM) that plays important roles in transcription and cell proliferation in eukaryotes. However, its function remains unknown in phytopathogenic fungi. Here,we performed a comprehensive assessment of Khibin the rice false smut fungus Ustilaginoidea virens, using Tandem Mass Tag(TMT)-based quantitative proteomics approach. A total of 3 426 Khibsites were identified in 977 proteins, sugg esting that Khibis a common and complex PTM in U. virens. Our data demonstrated that the2-hydroxyisobutyrylated proteins are involved in diverse biological processes. Network analysis of the modified proteins revealed a highly interconnected protein network that included many well-studied virulence factors. We confirmed that the Zn-binding reduced potassium dependency3-type histone deacetylase(UvRpd3) is a major enzyme that removes 2-hydroxyisobutyrylation and acetylation in U. virens. Notably, mutations of Khib sites in the mitogen-activated protein kinase(MAPK)UvSlt2 significantly reduced fungal virulence and decreased the enzymatic activity of UvSlt2. Molecular dynamics simulations demonstrated that 2-hydroxyisobutyrylation in UvSlt2 increased the hydrophobic solvent-accessible surface area and thereby affected binding between the UvSlt2 enzyme and its substrates. 2Analysis of the Drug Sensitivity of Escherichia coli Isolated from Sheep in Yangling,Shaanxi Province显示文摘This study was conducted to investigate the drug sensitivity of Escherichia coli isolated from sheep,providing data reference for clinical medication. Fifty-four samples were collected from a certain sheep farm in Yangling,Shaanxi Province for E. coli isolation and identification,and K-B disk diffusion method was used to carry out the dug sensitivity test of 40 E. coli to the eight antimicrobials of spectinomycin,amikacin,tobramycin,enrofloxacin,nalidixic acid,levofloxacin,salafloxacin and ciprofloxacin. The results showed that there were 40 positive E. coli strains and the isolation rate was 74. 7%. The resistance rates of the 40 E. coli to spectinomycin,amikacin,tobramycin and nalidixic acid were 85%,95%,97. 5% and 87. 5%,respectively,while the resistance rate to enrofloxacin,levofloxacin,salafloxacin and ciprofloxacin were relatively low and the resistance rates were 35%,20%,25% and 17. 5%,respectively. The isolates showed different levels of drug resistance to eight antimicrobials and the highest resistance rate was 97. 5%. 4Ustilaginoidea virens modulates lysine 2-hydroxyisobutyrylation in rice flowers during infection显示文摘The post-translational modification lysine 2-hydroxyisobutyrylation(K_(hib))plays an important role in gene transcription,metabolism,and enzymatic activity.K_(hib)sites have been identified in rice(Oryza sativa).However,the K_(hib)status of proteins in rice flowers during pathogen infection remains unclear.Here,we report a comprehensive identification of K_(hib)-modified proteins in rice flowers,and the changes in these proteins during infection with the fungal pathogen Ustilaginoidea virens.By using a tandem mass tag-based quantitative proteomics approach,we identified 2,891 K_(hib)sites on 964 proteins in rice flowers.Our data demonstrated that 2-hydroxyisobutyrylated proteins are involved in diverse biological processes.K_(hib)levels were substantially reduced upon infection with U.virens.Chromatin immunoprecipitation polymerase chain reaction(PCR)and reverse transcription quantitative PCR analyses revealed that histone K_(hib)is involved in the expression of disease-resistance genes.More importantly,most quantified sites on core histones H3 were downregulated upon U.virens infection.In addition,the histone deacetylases HDA705,HDA716,SRT1,and SRT2 are involved in the removal of K_(hib)marks in rice.HDA705 was further confirmed to negatively regulate rice disease resistance to pathogens U.virens,Magnaporthe oryzae,and Xanthomonas oryzae pv.oryzae(Xoo).Our data suggest that U.virens could modulate K_(hib)in rice flowers during infection.Xiaoyang Chen Qiutao Xu Yuhang Duan Hao Liu Xiaolin Chen Junbin Huang Chaoxi Luo Dao-Xiu Zhou Lu Zheng 2021Journal of Integrative Plant Biology2021,63,10:1

6Study on Anti-diarrhea Effect of the Prescription of 4 Tibetan Veterinary Drugs显示文摘This study was conducted to investigate the effects of extracts of 4 Tibetan veterinary drugs on bacterial diarrhea and to guide the clinical treatment of the disease. The inhibitory effects of the extracts of the 4 Tibetan veterinary drugs Veronica ciliate Fisch Usnea diffracta Vain Sophora flavescens var. flavescens Lamiophlomis rotata (Benth.) Kudo on 14 common diarrheagenic bacteria were detected by K-B diffusion method and micro-broth dilution method. Optimization was performed on prescriptions of the 4 Tibetan veterinary drugs using orthogonal design software and a mouse bacterial diarrhea model was established with a clinical isolate Salmonella blegdam. According to the LD_(50) value of the optimal prescription of the 4 Tibetan veterinary drugs the established mouse bacterial diarrhea model was treated in 3 dose groups i. e. the high middle and low dose groups (0.060 0.030 and 0.020 g/ml respectively). The results showed that the 4 Tibetan veterinary drugs had very good inhibitory effects on most of the tested diarrheagenic bacteria and among them U. diffracta had better inhibitory effects on all the tested bacteria. The optimal prescription of the 4 Tibetan veterinary drugs (high dose 0.06 g/ml) exhibited very good inhibitory effect on mouse diarrhea indicating that the prescription has very good anti-diarrhea effect which is beneficial to the clinical treatment of such disease and the development of Tibetan veterinary drugs.Jingwei XU Siyin GUO Dan LI Qian WANG Chaoxi CHEN 2017Agricultural Biotechnology2017,6,4:1

The results showed that the compound L. rupicola Hook. f. et & Thomson injection had no irritation to rabbit eyes and weak irritation to rabbit ears,and had relatively strong irritation to the leg muscles of the mice. 12bZIP Transcription Factor UvATF21 Mediates Vegetative Growth,Conidiation,Stress Tolerance and Is Required for Full Virulence of Rice False Smut Fungus Ustilaginoidea virens显示文摘bZIP proteins are widely distributed in eukaryotic organisms and regulate a diverse range of physiological processes.Several bZIP proteins have previously been identified in Ustilaginoidea virens.However,the biological roles of these bZIP proteins in this pathogen are still unknown.Here,one of these bZIP protein coding genes,UvATF21,was functionally characterized.Targeted deletion of UvATF21resulted in reduced conidiation and pathogenicity despite of the increased vegetative growth.The deletion mutants also significantly decreased the sensitivity to osmotic and oxidative stresses.Interestingly,deletion of UvATF21 exhibited different performances to cell wall integrity stress.These results indicated that UvATF21 played crucial roles in vegetative growth,conidiation,stress response,and full virulence in U.virens.LIU Yueran QU Jinsong WANG Yufu YIN Weixiao LUO Chaoxi 2023Rice science2023,30,1:0
13Identification and genetic characterization of bovine hepacivirus in China:A large scale epidemiological study显示文摘Bovine hepacivirus(BovHepV)is a novel virus that was recently discovered in Ghana and Germany in 2015.Until now,this virus has been identified in cattle population worldwide and is classified into subtypes A-G.To fully understand the epidemic situation and genetic characteristic of BovHepV in China,a total of 612 cattle serum samples were collected from 20 farms in seven provinces and municipality in China between 2018 and 2020 and were tested for the presence of BovHepV RNA via semi-nested PCR.The results demonstrated that 49(8.0%)samples were BovHepV RNA-positive.It is noted that BovHepV infection in yak was confirmed for the first time.BovHepV was detected in all the seven provinces,with the positive rate ranging from 3.1%to 13.3%,which indicates a wide geographical distribution pattern of BovHepV in China.Sequencing results revealed that 5'UTR of the 49 field BovHepV strains have a nucleotide similarity of 96.3%-100%between each other and 93.9%-100%with previously reported BovHepV strains.In addition,genetic analysis identified five critical nucleotide sites in 5'UTR to distinguish different subtypes,which was further verified by genomic sequencing and nucleotide similarity analysis.All the 49 Chinese field BovHepV strains were classified into subtype G and this subtype is only determined in cattle in China currently.This study will provide insights for us to better understand the epidemiology and genetic diversity of BovHepV.Gang Lu Chaoxi Chen Ran Shao Juan Zhang Jinghao Li Siqi Cai Lintao Zhong Zhiying Lai Jiajun Ou Xin Yin Guihong Zhang Shoujun Li 2022Virologica Sinica2022,37,2:0
14A review of acupuncture and moxibustion in treating children with cerebral palsy sitting disorder显示文摘Cerebral palsy is one of the important factors in children's disability.Patients'seat dysfunction has a serious impact on their daily life and their family.However,there is currently no effective treatment for the cerebral palsy seat dysfunction.So,it is of importance to search a new way to make a breakthrough.Acupuncture has significant advantages in the treatment of functional disorders.In this paper,the literature about clinical research of acupuncture treatment of cerebral palsy seat dysfunction are summarized.We have carried on the mechanism analysis of sports medicine respectively from three angles,including traditional Chinese medicine,western medicine and traditional Chinese and western medicine combined with sports medicine.We conclude that acupuncturing acupoints on Du meridian,Beishu points and Jiaji points has significant effect on the function of sitting unable children with cerebral palsy,such as strengthening the spleen,tonifying the kidney and promoting Yangqi and stimulating the sensory nerve and so on.Acupuncturing points for the treatment of cerebral palsy provides a lot of inspiration for human physiology research in the future and clinical treatment of neurological diseases.Chaoxi Zhu Fanrong Liang 2018Life Research2018,1,1:0
15Polysaccharides from Huidouba and Its Effect on Type-2 Diabetes Mellitus显示文摘To evaluate the extraction process of Huidouba polysaccharides( HDBP) and its anti-diabetic activity,this study focused on the two contents as follows:extraction process of HDBP,which was optimized by orthogonal experiments using the extraction ratio of HDBP for the index; and bioactivity evaluation of HDBP on streptozotocin-induced diabetic rat model based on the histological,biochemical and enzyme linked immunosorbent assay( ELISA) method for histological observation,physiological and biochemical index such as fasting blood glucose( FBG),nitric oxide( NO),malondialdehyde( MDA),super oxide dismutase( SOD) and insulin level detection in pancreas tissue respectively. The results showed that HDBP extraction ratio was the highest under following conditions: the extraction temperature at 90 ℃,the extraction time of 1. 5 h,and the solid-liquid ratio at 1: 10. Compared with the normal control group and the model group,histological observation,physiological and biochemical index and insulin level in pancreas tissue significantly increased and FBG significantly decreased according to the dosage administrated. 16Isolation and PCR Identification of Duck Tembusu Virus显示文摘Duck tembusu virus disease is one of the most serious infectious diseases endangering duck industry. A strain of virus was isolated from a dead duck,and performed PCR identification and sequence analysis. The results showed that the sequence of the isolate shared above 99% homology with duck tembusu virus( DTMUV) sequence on Gen Bank. 17Phytochemical activators of Nrf2:a review of therapeutic strategies in diabetes显示文摘Insulin resistance(IR)is fundamental to the development of type 2 diabetes(T2D),and altered mitochondrial function and abnormal lipid distribution are closely associated with IR or T2D.Excess oxidative stress-induced mitochondrial damage leads to an imbalance in redox homeostasis,which is considered the major contributor to the progression of diabetes.A key cellular defense mechanism,namely,the nuclear factor-E2 p45-related factor 2(Nrf2)-antioxidant response element(ARE)pathway,plays an essential protective role in combating excess oxidative stress.A series of phytochemicals are reported to improve IR and restore mitochondrial function against excess oxidative stress by activating the Nrf2-ARE signaling pathway to maintain cellular reactive oxygen species(ROS)homeostasis.The present review focuses on key knowledge gaps in the Nrf2-ARE system targeted by phytochemicals and its correlation to diabetes both in the in vitro and in vivo models and recent achievements in human clinical trials to evaluate its efficiency and safety.In addition,we provide an overview of recent research progress in nutrigenomics,precision nutrition and the interactions occurring in gut microbiota associated with the Nrf2-ARE signaling pathway and diabetes chemoprevention by phytochemicals and finally propose a future research strategy for regulating redox and microbiota balance via the Nrf2-ARE pathway.The present review aims to help us comprehensively understand the critical chemopreventive role of the Nrf2-ARE pathway targeted by phytochemicals in diabetes.Si Qin Zhong Chen Yiling Wen Yuhang Yi Chenghao Lv Chaoxi Zeng Lanming Chen Meng Shi 2023Acta Biochimica et Biophysica Sinica2023,55,1:0
