| 3 | Effect of Bi on graphite morphology and mechanical properties of heavy section ductile cast iron显示文摘To improve the mechanical properties of heavy section ductile cast iron, bismuth(Bi) was introduced into the iron. Five castings with different Bi content from 0 to 0.014 wt.% were prepared; and four positions in the casting from the edge to the center, with different solidifi cation cooling rates, were chosen for microstructure observation and mechanical properties test. The effect of the Bi content on the graphite morphology and mechanical properties of heavy section ductile cast iron were investigated. Results show that the tensile strength, elongation and impact toughness at different positions in the fi ve castings decrease with a decrease in cooling rate. With an increase in Bi content, the graphite morphology and the mechanical properties at the same position are improved, and the improvement of mechanical properties is obvious when the Bi content is no higher than 0.011wt.%. But when the Bi content is further increased to 0.014wt.%, the improvement of mechanical properties is not obvious due to the increase of chunky graphite number and the aggregation of chunky graphite. | 4 | Repurposable drugs for SARS-CoV-2 and influenza sepsis with scRNA-seq data targeting post-transcription modifications显示文摘Coronavirus disease 2019(COVID-19)has impacted almost every part of human lifeworldwide,posing amassive threat to human health.The lack of time for new drug discovery and the urgent need for rapid disease control to reduce mortality have led to a search for quick and effective alternatives to novel therapeutics,for example drug repurposing.To identify potentially repurposable drugs,we employed a systematic approach to mine candidates from U.S.FDA-approved drugs and preclinical small-molecule compounds by integrating gene expression perturbation data for chemicals from the Library of Integrated Network-Based Cellular Signatures project with a publicly available single-cell RNA sequencing dataset from patients withmild and severe COVID-19(GEO:GSE145926,public data available and accessed on 22 April 2020).We identified 281 FDA-approved drugs that have the potential to be effective against severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection,16 of which are currently undergoing clinical trials to evaluate their efficacy against COVID-19.We experimentally tested and demonstrated the inhibitory effects of tyrphostin-AG-1478 and brefeldin-a,two chemical inhibitors of glycosylation(a post-translational modification)on the replication of the single-stranded ribonucleic acid(ssRNA)virus influenza A virus as well as on the transcription and translation of host cell cytokines and their regulators(IFNs and ISGs).In conclusion,we have identified and experimentally validated repurposable anti-SARS-CoV-2 and IAV drugs using a systems biology approach,which may have the potential for treating these viral infections and their complications(sepsis). | Zhihan Wang Kai Guo Pan Gao Qinqin Pu Changlong Li Junguk Hur Min Wu | 2021 | Precision Clinical Medicine2021,4,4: | 1 |

| 8 | The efficacy assessment of convalescent plasma therapy for COVID-19 patients:a multi-center case series显示文摘Convalescent plasma(CP)transfusion has been indicated as a promising therapy in the treatment for other emerging viral infections.However,the quality control of CP and individual variation in patients in different studies make it rather difficult to evaluate the efficacy and risk of CP therapy for coronavirus disease 2019(COVID-19).We aimed to explore the potential efficacy of CP therapy,and to assess the possible factors associated with its efficacy.We enrolled eight critical or severe COVID-19 patients from four centers.Each patient was transfused with 200–400mL of CP from seven recovered donors.The primary indicators for clinical efficacy assessment were the changes of clinical symptoms,laboratory parameters,and radiological image after CP transfusion.CP donors had a wide range of antibody levels measured by serology tests which were to some degree correlated with the neutralizing antibody(NAb)level.No adverse events were observed during and after CP transfusion.Following CP transfusion,six out of eight patients showed improved oxygen support status;chest CT indicated varying degrees of absorption of pulmonary lesions in six patients within 8 days;the viral load was decreased to a negative level in five patients who had the previous viremia;other laboratory parameters also tended to improve,including increased lymphocyte counts,decreased C-reactive protein,procalcitonin,and indicators for liver function.The clinical efficacy might be associated with CP transfusion time,transfused dose,and the NAb levels of CP.This study indicated that CP might be a potential therapy for severe patients with COVID-19. | Hao Zeng Dongfang Wang Jingmin Nie Haoyu Liang Jiang Gu Anne Zhao Lixin Xu Chunhui Lang Xiaoping Cui Xiaolan Guo Changlong Zhou Haibo Li Bin Guo Jinyong Zhang Qiang Wang Li Fang Wen Liu Yishan Huang Wei Mao Yaokai Chen Quanming Zou | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 1 |

| 10 | Development of a new cerebral ischemia reperfusion model of Mongolian gerbils and standardized evaluation system显示文摘Background:The Mongolian gerbil is an excellent laboratory animal for preparing the cerebral ischemia model due to its inherent deficiency in the circle of Willis.However,the low incidence and unpredictability of symptoms are caused by numerous complex variant types of the circle.Additionally,the lack of an evaluation system for the cer-ebral ischemia/reperfusion(I/R)model of gerbils has shackled the application of this model.Methods:We created a symptom-oriented principle and detailed neurobehavioral scoring criteria.At different time points of reperfusion,we analyzed the alteration in locomotion by rotarod test and grip force score,infarct volume by triphenyltetrazo-lium chloride(TTC)staining,neuron loss using Nissl staining,and histological charac-teristics using hematoxylin-eosin(H&E)straining.Results:With a successful model rate of 56%,32 of the 57 gerbils operated by our method harbored typical features of cerebral I/R injury,and the mortality rate in the male gerbils was significantly higher than that in the female gerbils.The suc-cessfully prepared I/R gerbils demonstrated a significant reduction in motility and grip strength at 1 day after reperfusion;formed obvious infarction;exhibited typi-cal pathological features,such as tissue edema,neuronal atrophy and death,and vacuolated structures;and were partially recovered with the extension of reperfu-sion time.Conclusion:This study developed a new method for the unilateral common carotid artery ligation I/R model of gerbil and established a standardized evaluation system for this model,which could provide a new cerebral I/R model of gerbils with more practical applications. | Ying Wu Caijiao Hu Zhihui Li Feiyang Li Jianyi Lv Meng Guo Xin Liu Changlong Li Xueyun Huo Zhenwen Chen Lifeng Yang Xiaoyan Du | 2024 | Animal Models and Experimental Medicine2024,7,1: | 0 |
