| 1 | PIKA as an Adjuvant Enhances Specific Humoral and Cellular Immune Responses Following the Vaccination of Mice with HBsAg plus PIKA显示文摘An adjuvant is usually used to enhance the immune response induced by vaccines. The choice of adjuvant or immune enhancer determines the effectiveness of the immune response. Currently, aluminium (Alum, a generic term for salts of aluminium) is the only FDA-approved adjuvant. Alum predominantly induces the differentiation of Th2 cells and thus mediates an antibody immune response. Therefore, there is an urgent need for new adjuvants that enhance not only humoral but also cellular immune responses. In the present study, we demonstrates that PIKA (a stabilized dsRNA) as an adjuvant directly induces the activation and the proliferation of both B and NK cells in vitro. Injection of PIKA into mice results in the production of cytokines in vivo. In addition, the study demonstrates that PIKA promotes the maturation of bone marrow-derived dendritic cells (BMDCs) including up-regulation of the co-stimulatory molecules CD80, CD86 and CD40, and the induction of cytokines such as IL-12p70, IL-12p40 and IL-6. Importantly, after immunization of mice with HBsAg plus PIKA, the presence of PIKA enhances the titers of HBsAg-specific IgG and HBsAg-specific IFN-γ production. These results demonstrate that PIKA as an adjuvant can promote both humoral and cellular immune responses. For the hydrodynamic flow (u,d) of nematic liquid crystals in dimensions n = 2 or 3, it is shown that the uniqueness holds for the class of weak solutions provided either (i) for n = 2, u ∈ Lt∞ L2x∩L2tHx1, ▽P∈ Lt4/3 Lx4/3 , and ▽d∈ L∞t Lx2∩Lt2Hx2; or (ii) for n = 3, u ∈ Lt∞ Lx2∩L2tHx1∩ C([0,T),Ln), P ∈ Ltn/2 Lxn/2 , and ▽d∈ L2tLx2 ∩ C([0,T),Ln). | 6 | Fabrication of UV responsive micelles-containing multilayers and their influence on cell adhesion显示文摘Multilayers incorporated with stimuli-responsive substances by means of layer-by-layer(LbL) self-assembly are much attractive due to their advantages of stimuli-responsiveness and potential applications in different fields. In this study, pyrenemethyl acrylate(PA) was synthesized, and was copolymerized with acrylic acid(AA) to obtain the amphiphilic and photodegradable P(PA-co-AA)polymers with a PA:AA molar ratio of 1.3:3, and an average molecular weight of 6.9 kDa and polydispersity index of 1.04. They formed micelles spontaneously when dispersed in aqueous solution with a size of 27.5 nm in a dry state and 136.6 nm in a wet state. The micelles were readily decomposed to form aggregates as a result of the cleavage of the pyrenemethyl ester bonds under UV-irradiation. UV-responsive micelles-containing multilayers were prepared by LbL self-assembly of the UV-responsive micelles and polyallylamine hydrochloride(PAH). UV-irradiation of the multilayers resulted in the decomposition of micelles, leading to larger surface roughness, and enhanced swelling ratio and wettability of the multilayers. Y.,Phase transition for potentials of high dimensional wells, Comm. Pure Appl. Math., 65(6),2012, 833–888] in which the authors had set up a program to verify rigorously some formal statements associated with the multiple component phase transitions with higher dimensional wells. The main goal here is to establish a regularity theory for minimizing maps with a rather non-standard boundary condition at the sharp interface of the transition.The authors also present a proof, under simplified geometric assumptions, of existence of local smooth gradient flows under such constraints on interfaces which are in the motion by the mean-curvature. | 8 | Clinical significance of S100A7 protein in predicting recurrence of breast cancer in patients undergoing breast-conserving surgery with radiotherapy显示文摘Objective To investigate the relationship between the expression of S100A7 protein and prediction of recurrence and prognosis of breast cancer in patients undergoing breast-conserving surgery combined with radiotherapy.Methods 349 samples of carcinoma tissue wax blocks were selected from January 2011 to January 2014 in Qingdao Central Hospital.All the patients had undergone breast-conserving surgery.We analyzed S100A7 expression in tumor tissue by immunohistochemical staining.Using univariate and multivariate analyses,we evaluated the relationship between S100A7 and clinical results,to explore independent risk factors for local regional recurrence(LRR).Results The positive expression of S100A7 in the recurrence group(66.7%)was significantly higher than in the non-recurrence group(38.4%),P=0.025.A log-rank test showed that high S100A7 expression was significantly correlated with 5-year regional recurrence free survival rate(RFS)(94.9%vs 89.5%,P=0.0408),distant metastasis free survival rate(DFS)(95.4%vs 83.5%,P<0.001),and overall survival rate(OS)(99.0%vs 92.5%,P=0.0011).Histological grade,vessel carcinoma embolus,lymph node metastasis,S100A7 expression,and tumor size were factors that influenced RFS.Multivariate analysis of the Cox proportional hazard model showed that high S100A7 expression was an independent risk factor that affected breast cancer RFS(HR=6.864,95%CI:1.575-29.915,P=0.01).Thus,we concluded that high S100A7 expression is associated with increased risk of LRR and distant metastasis of breast cancer after breast-conserving surgery and postoperative radiotherapy.S100A7 can be used as a molecular marker to screen for patients with high recurrence risk after breast-conserving surgery. | Chao Zhang Changyou Li Gaoyang Lin Yao Qi Zhenfeng Li Jing Xu Tianhui Su Xin Liu Xiao Zou | 2019 | Oncology and Translational Medicine2019,5,5: | 0 |
