1The novel ER stress inducer Sec C triggers apoptosis by sulfating ER cysteine residues and degrading YAP via ER stress in pancreatic cancer cells显示文摘Pancreatic adenocarcinoma(PAAD) is one of the most lethal malignancies. Although gemcitabine(GEM) is a standard treatment for PAAD, resistance limits its application and therapy. Secoemestrin C(Sec C) is a natural compound from the endophytic fungus Emericella, and its anticancer activity has not been investigated since it was isolated. Our research is the first to indicate that Sec C is a broad-spectrum anticancer agent and could exhibit potently similar anticancer activity both in GEM-resistant and GEM-sensitive PAAD cells. Interestingly, Sec C exerted a rapid growth-inhibiting effect(80% death at 6 h), which might be beneficial for patients who need rapid tumor shrinkage before surgery. Liquid chromatography/mass spectrometry and N-acetyl-L-cysteine(NAC) reverse assays show that Sec C sulfates cysteines to disrupt disulfide-bonds formation in endoplasmic reticulum(ER) proteins to cause protein misfolding, leading to ER stress and disorder of lipid biosynthesis. Microarray data and subsequent assays show that ER stress-mediated ER-associated degradation(ERAD) ubiquitinates anddownregulates YAP to enhance ER stress via destruction complex(YAP-Axin-GSK-βTr CP), which also elucidates a unique degrading style for YAP. 3Risk factors for intracranial hemorrhage and mortality in adult patients with severe respiratory failure managed using veno-venous extracorporeal membrane oxygenation显示文摘Background: Intracerebral hemorrhage (ICH) is one of the most severe complications during veno-venous extracorporeal membrane oxygenation (VV-ECMO). This study aimed to determine the risk factors for ICH and mortality in such patients.Methods: We analyzed the clinical data of 77 patients who received VV-ECMO due to severe respiratory failure from July 2013 to May 2019 at China-Japan Friendship Hospital. Demographical data, laboratory indices, imaging characteristics, and other clinical information were collected. Multivariable logistic regression analyses were performed to identify risk factors for ICH and mortality.Results: Of 77 patients, 11 (14.3%) suffered from ICH, and 36 (46.8%) survived. The survival rate was significantly lower (18.2% [2/11]vs. 51.5% [34/66],P = 0.040) in patients with ICH than in those without ICH. Multivariable analysis revealed that factors independently associated with ICH were diabetes mellitus (adjusted odds ratio [aOR]: 12.848, 95% confidence interval [CI]: 1.129-146.188,P = 0.040) and minimum fibrinogen during ECMO (aOR: 2.557, 95% CI: 1.244-5.252,P = 0.011). Multivariable analysis showed that factors independently associated with mortality were acute hepatic failure during ECMO (aOR: 9.205, 95% CI: 1.375-61.604,P = 0.022), CO2 retention before ECMO (aOR: 7.602, 95% CI: 1.514-38.188,P = 0.014), and minimum platelet concentration during ECMO (aOR: 0.130, 95% CI: 0.029-0.577,P = 0.007).Conclusions: Diabetes mellitus and minimum fibrinogen concentration during ECMO are risk factors for ICH in patients with severe respiratory failure managed using VV-ECMO. 4Study of Impact Resistance Based on Porcupine Quills Bionic Thin-walled Structure显示文摘Using an electron microscope to observe the microstructure of a porcupine quills cross-section and a bionic method,a new bionic structure was proposed.The performance of the structure in terms of energy absorption,maximum impact force withstood,and impact force efficiency was evaluated using Ansys finite element simulation software to simulate the structure's impact.To examine the impact of ribs on the structural performance of the bionic porcupine quills,a control structure was developed.According to the results of the finite element simulation,the presence of ribs in the Bionic porcupine quills structure can transfer stress uniformly to the overall structure and share stress for some of the rupture-prone regions.Ribs reduce stress concentration in specific areas and increase the impact force efficiency of the structure.The SEA and IFE values of bionic porcupine quills were 30.01 kJ/kg and 84.22%,respectively.The structure is then optimized for parameter design in order to find the optimal structure by response surface in order to improve the structure's SEA and decrease its MIF.In order to evaluate the precision of the response surface,the optimal structure predicted is validated using finite element simulation.Tianshu Huang Zhengyu Mao Lijun Chang Xingyuan Huang Zhihua Cai 2023Journal of Bionic Engineering2023,20,5:0
5Improving the System of Historic and Cultural Heritage Protection Through Social Participation: Based on the Practice of Guangdong显示文摘The early historic and cultural heritage protection is regarded as the responsibility of the government, and is promoted by the government, protection experts, planners, and a few other elites. These bodies constitute a top-down historic and cultural heritage protection system based on their cultural consciousness. In the social transition of market-oriented reform and property right reform, due to the profit-driven demands of individuals, enterprise, and development-oriented government, the protection system was gradually differentiated and a lot of 'protective development' practices emerged. With the advent of the post-industrial era, based on the need for cultural identity, the historic and cultural heritage protection began to get attention from third-party organizations, community residents, and even the public, who launched 'bottom-up' social participation. Therefore, this paper, based on social governance theory, takes the development of civil cultural conservation organizations in Guangdong as the research object, explores how to construct a society participation network in Guangdong, and discusses the influence and limitation of the network in promoting the historic and cultural heritage protection.
