1Immobilization of functional nano-objects in living engineered bacterial biofilms for catalytic applications显示文摘Nanoscale objects feature very large surface-area-to-volume ratios and are now understood as powerful tools for catalysis,but their nature as nanomaterials brings challenges including toxicity and nanomaterial pollution.Immobilization is considered a feasible strategy for addressing these limitations.Here,as a proof-of-concept for the immobilization of nanoscale catalysts in the extracellular matrix of bacterial biofilms,we genetically engineered amyloid monomers of the Escherichia coli curli nanofiber system that are secreted and can self-assemble and anchor nano-objects in a spatially precise manner.We demonstrated three scalable,tunable and reusable catalysis systems:biofilm-anchored gold nanoparticles to reduce nitro aromatic compounds such as the pollutant p-nitrophenol,biofilm-anchored hybrid Cd0.9Zn0.1S quantum dots and gold nanoparticles to degrade organic dyes and biofilm-anchored CdSeS@ZnS quantum dots in a semi-artificial photosynthesis system for hydrogen production.Our work demonstrates how the ability of biofilms to grow in scalable and complex spatial arrangements can be exploited for catalytic applications and clearly illustrates the design utility of segregating high-energy nano-objects from injury-prone cellular components by engineering anchoring points in an extracellular matrix.Xinyu Wang Jiahua Pu Yi Liu Fang Ba Mengkui Cui Ke Li Yu Xie Yan Nie Qixi Mi Tao Li Lingli Liu Manzhou Zhu Chao Zhong 2019National Science Review2019,6,5:9

3Quantitative Diagnosis of Fault Severity Trend of Rolling Element Bearings显示文摘The condition monitoring and fault diagnosis of rolling element bearings are particularly crucial in rotating mechanical applications in industry. A bearing fault signal contains information not only about fault condition and fault type but also the severity of the fault. This means fault severity quantitative analysis is one of most active and valid ways to realize proper maintenance decision. Aiming at the deficiency of the research in bearing single point pitting fault quantitative diagnosis, a new back-propagation neural network method based on wavelet packet decomposition coefficient entropy is proposed. The three levels of wavelet packet coefficient entropy(WPCE) is introduced as a characteristic input vector to the BPNN. Compared with the wavelet packet decomposition energy ratio input vector, WPCE shows more sensitive in distinguishing from the different fault severity degree of the measured signal. The engineering application results show that the quantitative trend fault diagnosis is realized in the different fault degree of the single point bearing pitting fault. 4Functional identification of the terpene synthase family involved in diterpenoid alkaloids biosynthesis in Aconitum carmichaelii显示文摘Aconitum carmichaelii is a high-value medicinal herb widely used across China,Japan,and other Asian countries.Aconitine-type diterpene alkaloids(DAs)are the characteristic compounds in Aconitum.Although six transcriptomes,based on short-read next generation sequencing technology,have been reported from the Aconitum species,the terpene synthase(TPS)corresponding to DAs biosynthesis remains unidentified.We apply a combination of Pacbio isoform sequencing and RNA sequencing to provide a comprehensive view of the A.carmichaelii transcriptome.Nineteen TPSs and five alternative splicing isoforms belonging to TPS-b,TPS-c,and TPS-e/f subfamilies were identified.In vitro enzyme reaction analysis functional identified two sesqui-TPSs and twelve di TPSs.Seven of the TPS-c subfamily genes reacted with GGPP to produce the intermediate ent-copalyl diphosphate.Five Ac KSLs separately reacted with ent-CPP to produce ent-kaurene,ent-atiserene,and ent-13-epi-sandaracopimaradie:a new diterpene found in Aconitum.Ac TPSs gene expression in conjunction DAs content analysis in different tissues validated that ent-CPP is the sole precursor to all DAs biosynthesis,with Ac KSL1,Ac KSL2 s and Ac KSL3-1 responsible for C20 atisine and napelline type DAs biosynthesis,respectively.These data clarified the molecular basis for the C20-DAs biosynthetic pathway in A.carmichaelii and pave the way for further exploration of C19-DAs biosynthesis in the Aconitum species.Liuying Mao Baolong Jin Lingli Chen Mei Tian Rui Ma Biwei Yin Haiyan Zhang Juan Guo Jinfu Tang Tong Chen Changjiangsheng Lai Guanghong Cui Luqi Huang 2021Acta Pharmaceutica Sinica B2021,11,10:6

6Hot deformation behavior of a spray-deposited AZ31 magnesium alloy显示文摘The flow stress behavior of an as-spray-deposited AZ31 magnesium alloy with fine grains was investigated by means of compression tests with a Gleeble 1500 thermal mechanical simulator at isothermal constant strain rates of 0.01, 0.1, 1.0, and 10 s-1; the testing temperatures ranged from 623 to 723 K. It is demonstrated that a linear equation can be fitted between the Zemer-Hollomon parameter Z and stress in a double-log scale. The effect of deformation parameters on the behavior of recrystallization was analyzed. Dynamic recrystallization (DRX) generally occurs at a higher temperature and at a lower strain rate. The constitutive equation of the spray-deposited AZ31 magnesium alloy is presented by calculating the deformation activation energy (199.8 kJ·mol-1). 7Clarification of the Field Concept for a New General System Theory显示文摘Field is an essential concept for us to understand the physical phenomena in the universe.Since the field concept was proposed in electromagnetism during nineteenth century,the understanding of the field concept has experienced great changes.This paper re-examines the concept of field from its origin and evolution history,with the purpose of a better understanding of the concept in order to be used in our new general system theory to unify different theories.Our main conclusion is that the concept of field should not be regarded as an independent physical existence but a property of matter of both living and nonliving.Through a discussion on an isolated two-body system from the perspective of gravitational field,a psychic field is introduced to address the active force produced by living creatures with minds.PAN Lingli CUI Weicheng 2021Journal of Philosophy Study2021,11,10:2
8Sensitivity of supplementation of thyroid hormone on treatmentof idiopathic short-stature children during therapy withrecombinant human growth hormone显示文摘This study aimed to evaluate the effects of thyroid hormone supplementation on growth rate of childrenwith idiopathic short stature (ISS) and low-normal serum free thyroxine FT4 who were receiving growth hormonetherapy. We selected 64 prepubertal children with FT4 levels in the lowest third of the normal range as the lowerFT4 group, and these children were divided randomly into two subgroups: L-thyroxine (L-T4)-treated subgroupwas treated with L-T4 (0.5-3.0 g/(kg, d)) from the beginning of the study, and the non-L-T4-treated subgroupreceived placebo. We also selected 39 ISS children with FT4 in the upper two-thirds of the normal range as thehigher FT4 group. During the first year, the lower FT4 group featured lower FT3, FT4, thyroid stimulatinghormone (TSH), and insulin-like growth factor-I standard deviation score (IGF-I SDS) and significantly lowerheight velocity (HV) compared with the higher FT4 group. However, in the lower FT4 group, the L-T4-treatedsubgroup presented higher FT4, FT3, TSH, and IGF-I SDS concentrations and significantly higher HV comparedwith children in the non-L-T4-treated subgroup. 9RNA-induced liquid phase separation of SARS-CoV-2 nucleocapsid protein facilitates NF-kB hyper-activation and inflammation显示文摘The ongoing 2019 novel coronavirus disease(COVID-19)caused by SARS-CoV-2 has posed a worldwide pandemic and a major global public health threat.The severity and mortality of COVID-19 are associated with virus-induced dysfunctional inflammatory responses and cytokine storms.However,the interplay between host inflammatory responses and SARS-CoV-2 infection remains largely unknown.Here,we demonstrate that SARS-CoV-2 nucleocapsid(N)protein,the major structural protein of the virion,promotes the virus-triggered activation of NF-kB signaling.After binding to viral RNA,N protein robustly undergoes liquid-liquid phase separation(LLPS),which recruits TAK1 and IKK complex,the key kinases of NF-kB signaling,to enhance NF-kB activation.Moreover,1,6-hexanediol,the inhibitor of LLPS,can attenuate the phase separation of N protein and restrict its regulatory functions in NF-kB activation.These results suggest that LLPS of N protein provides a platform to Induce NF-kB hyper-activation,which could be a potential therapeutic target against COVID-19 severe pneumonia.Yaoxing Wu Ling Ma Sihui Cai Zhen Zhuang Zhiyao Zhaou Shouheng Jin Weihong Xie Lingli Zhou Lei Zhang Jincun Zhao Jun Cui 2021Signal Transduction and Targeted Therapy2021,6,5:2
10Re-examination of the Two-Body Problem Using Our New General System Theory显示文摘It is well-known that philosophical conflicts exist among classical mechanics,quantum mechanics and relativistic mechanics.In order to use the framework of general system theory to unify these three mechanics subjects,a new general system theory is developed based on a new ontology of ether and minds as the fundamental existences in the world.The two-body problem is the simplest model in mechanics and in this paper,it is re-examined by using our new general system theory.It is found that the current description of the classical full two-body problem is inappropriate since the observer and the measurement apparatus have not been explicitly considered.After considering these,it is actually a three-body problem while only the special case of the Kepler problem is the two-body problem.By introducing the concepts of psychic force and psychic field,all the possible movement states in the two-body problem can be explained within the framework of classical mechanics.There is no need to change the meanings of many fundamental concepts,such as time,space,matter,mass,and energy as done in quantum mechanics and relativity theory.This points out a new direction for the unification of different theories.PAN Lingli CUI Weicheng 2021Journal of Philosophy Study2021,11,12:2
11Targeting lactate dehydrogenase A (LDHA) exerts antileukemic effects on T-cell acute lymphoblastic leukemia显示文摘Background:T-cell acute lymphoblastic leukemia(T-ALL)is an uncommon and aggressive subtype of acute lymphoblastic leukemia(ALL).In the serum of T-ALL patients,the activity of lactate dehydrogenase A(LDHA)is increased.We proposed that targeting LDHA may be a potential strategy to improve T-ALL outcomes.The current study was conducted to investigate the antileukemic effect of LDHA gene-targeting treatment on T-ALL and the underlying molecular mechanism.Methods:Primary T-ALL cell lines Jurkat and DU528 were treated with the LDH inhibitor oxamate.MTT,colony formation,apoptosis,and cell cycle assays were performed to investigate the effects of oxamate on T-ALL cells.Quantitative real-time PCR(qPCR)and Western blotting analyses were applied to determine the related signaling pathways.A mitochondrial reactive oxygen species(ROS)assay was performed to evaluate ROS production after T-ALL cells were treated with oxamate.A T-ALL transgenic zebrafish model with LDHA gene knockdown was established using CRISPR/Cas9 gene-editing technology,and then TUNEL,Western blotting,and T-ALL tumor progression analyses were conducted to investigate the effects of LDHA gene knockdown on T-ALL transgenic zebrafish.Results:Oxamate significantly inhibited proliferation and induced apoptosis of Jurkat and DU528 cells.It also arrested Jurkat and DU528 cells in G0/G1 phase and stimulated ROS production(all P<0.001).Blocking LDHA significantly decreased the gene and protein expression of c-Myc,as well as the levels of phosphorylated serine/threonine kinase(AKT)and glycogen synthase kinase 3 beta(GSK-3β)in the phosphatidylinositol 3′-kinase(PI3K)signaling pathway.LDHA gene knockdown delayed disease progression and down-regulated c-Myc mRNA and protein expression in T-ALL transgenic zebrafish.Conclusion:Targeting LDHA exerted an antileukemic effect on T-ALL,representing a potential strategy for T-ALL treatment.Haizhi Yu Yafei Yin Yifang Yi Zhao Cheng Wenyong Kuang Ruijuan Li Haiying Zhong Yajuan Cui Lingli Yuan Fanjie Gong Zhihua Wang Heng Li Hongling Peng Guangsen Zhang 2020Cancer Communications2020,40,10:2

18CVRgram for Demodulation Band Determination in Bearing Fault Diagnosis under Strong Gear Interference显示文摘Fault-related resonance frequency band extraction-based demodulation methods are widely used for bearing diagnostics.However,due to the high peaks of strong gear meshing interference,the classical band selection methods have poor performance and cannot work well for bearing fault type detection.As such,the CVRgram-based bearing fault diagnosis method is proposed in this paper.In the proposed method,inspired by the conditional variance(CV)index and root mean square(RMS),a novel index,named the CV/root mean square(CVR),is first proposed.The CVR index has high robustness for the interference of non-Gaussian or Gaussian noise and has the ability to determine the center frequency of the weak bearing fault-related resonance frequency band under strong interference.Secondly,motived by the Kurtogram,the CVRgram algorithm is developed for adaptively determining the optimal filtering parameters.Finally,the CVRgram-based bearing fault diagnosis method under strong gear meshing interference is proposed.The performance of the CVRgram-based method is verified by both the simulation signal and the experiment signal.The comparison analysis with the Kurtogram,Protrugram,and CVgram-based method shows that the proposed technique has a much better ability for bearing fault detection under strong noise interference.Pengda Wang Dezun Zhao Dongdong Liu Lingli Cui 2022Journal of Dynamics, Monitoring and Diagnostics2022,1,4:0
