| 1 | Kinetics and mechanisms of mitotic inheritance of DNA methylation and their roles in aging-associated methylome deterioration显示文摘Mitotic inheritance of the DNA methylome is a challenging task for the maintenance of cell identity.Whether DNA methylation pattern in different genomic contexts can all be faithfully maintained is an open question.A replication-coupled DNA methylation maintenance model was proposed decades ago,but some observations suggest that a replication-uncoupled maintenance mechanism exists.However,the capacity and the underlying molecular events of replication-uncoupled maintenance are unclear.By measuring maintenance kinetics at the single-molecule level and assessing mutant cells with perturbation of various mechanisms,we found that the kinetics of replication-coupled maintenance are governed by the UHRF1–Ligase 1 and PCNA–DNMT1 interactions,whereas nucleosome occupancy and the interaction between UHRF1 and methylated H3K9 specifically regulate replication-uncoupled maintenance.Surprisingly,replication-uncoupled maintenance is sufficiently robust to largely restore the methylome when replication-coupled maintenance is severely impaired.However,solo-WCGW sites and other CpG sites displaying aging-and cancer-associated hypomethylation exhibit low maintenance efficiency,suggesting that although quite robust,mitotic inheritance of methylation is imperfect and that this imperfection may contribute to selective hypomethylation during aging and tumorigenesis. | Xuan Ming Zhuqiang Zhang Zhuoning Zou Cong Lv Qiang Dong Qixiang He Yangyang Yi Yingfeng Li Hailin Wang Bing Zhu | 2020 | Cell Research2020,30,11: | 4 |

| 3 | CT radiomics can help screen the coronavirus disease2019(COVID-19): a preliminary study显示文摘The coronavirus disease 2019(COVID-19) is raging across the world.The radiomics,which explores huge amounts of features from medical image for disease diagnosis,may help the screen of the COVID-19.In this study,we aim to develop a radiomic signature to screen COVID-19 from CT images.We retrospectively collect 75 pneumonia patients from Beijing Youan Hospital,including 46 patients with COVID-19 and 29 other types of pneumonias.These patients are divided into training set(n = 50) and test set(n = 25) at random.We segment the lung lesions from the CT images,and extract 77 radiomic features from the lesions.Then unsupervised consensus clustering and multiple cross-validation are utilized to select the key features that are associated with the COVID-19.In the experiments,while twenty-three radiomic features are found to be highly associated with COVID-19,four key features are screened and used as the inputs of support vector machine to build the radiomic signature.We use area under the receiver operating characteristic curve(AUC) and calibration curve to assess the performance of our model.It yields AUCs of 0.862 and 0.826 in the training set and the test set respectively.We also perform the stratified analysis and find that its predictive ability is not affected by gender,age,chronic disease and degree of severity.In conclusion,we investigate the value of radiomics in screening COVID-19,and the experimental results suggest the radiomic signature could be a potential tool for diagnosis of COVID-19. | Mengjie FANG Bingxi HE Li LI Di DONG Xin YANG Cong LI Lingwei MENG Lianzhen ZHONG Hailin LI Hongjun LI Jie TIAN | 2020 | Science China(Information Sciences)2020,63,7: | 2 |
| 4 | The Impacts on Spinach Growth and Yield by Biological Organic Fertilizer显示文摘To decrease fertilization amount of chemical fertilizer and improve the quality of vegetable crops,spinach was taken as the test material,and the impact of different fertilizer on spinach growth and yield was studied via the manners of biological organic fertilizer and organic fertilizer + chemical fertilizer. Experimental results showed that in the formula of organic fertilizer + chemical fertilizer,chlorophyll and nitrogen contents in spinach leaves obviously increased; in the formula of only adding organic fertilizer,spinach leaf temperature,leaf width,root length,plant height and fresh weight were all better than those in the formula of organic fertilizer + chemical fertilizer,and better formulas were A_5,E_5,F_3 and I_5,in which spinach plant height in E_5 was 5. 63 times higher than G_5,root length in E_5 was 2. 67 times higher than G_5,and fresh weight in G_5 was 32. 6 times higher than G_5. | 15 | Preparation and anti-tumor application of hyaluronic acid-based material for disulfide and copper ions co-delivery显示文摘Disulfide(DSF) has been proved good anti-tumor effect and even better with coadministration of Cu^(2+). In this work, we report the use of hyaluronic acid(HA) based materials to construct vectors for the delivery of both DSF and Cu^(2+). HA was firstly modified with polyethylene glycol monomethyl ether(mPEG) and polycaprolactone(PCL) to synthesize an amphiphilic polymer(HA-PEG-PCL). DSF could be loaded in the hydrophobic core and Cu^(2+) could be cooperated to the negative hydrophilic segment. The Cu^(2+) also played a role as crosslinking agent, which prevented DSF leakage prematurely, avoiding the bad side effects to normal tissues. The interaction between HA and CD44 improved the distribution of nanodrugs in tumor cells. When the nanodrugs were delivered to the cancer cell, the acidic micro-environment would separate the Cu^(2+) from the surface, leading to the disintegration of the micelles, promoting the release of DSF from the micelle core.
