| 2 | Chemical composition and in vitro fermentation characteristics of high sugar forage sorghum as an alternative to forage maize for silage making in Tarim Basin, China显示文摘The chemical composition, mineral profile and in vitro fermentation characteristics of maize(MZ), high sugar forage sorghum(HS) and forage sorghum(FS), and silages made from each forage type were measured. The MZ and MZ silage(MZS) had higher crude protein, starch and ether extract contents than both sorghum forages and sorghum silages. HS had higher ash and water-soluble carbohydrate concentrations than FS and MZ. MZ, MZS, HS and HS silage(HSS) had lower neutral detergent fibre, acid detergent fibre and acid detergent lignin than FS and FS silage(FSS). FSS had higher dry matter(DM) and p H than MZS and HSS. HSS contained higher concentrations of P and K than FSS and MZS. MZS and HSS had higher in vitro dry matter and organic matter digestibility, CH4 production, total volatile fatty acids, acetate and propionate than FSS. p H was higher for FSS than for HSS, and ammonia was lower for HSS than for MZS and FSS. HSS had higher gas production than MZS and FSS after 2, 4, 6 and 8 h incubation. MZS had higher gas production than HSS and FSS after 26 and 28 h of incubation. The results indicate that HS may substitute for MZ to make good quality silage. | 3 | Protective effects of Yindanxinnaotong capsule in a rat model of myocardial ischemia/reperfusion injury显示文摘OBJECTIVE: To investigate the effects of Yindanxinnaotong capsule(YDXNTC) and main components compatibility and ratios on myocardium against ischemia/reperfusion injury and the effect's underlying mechanism.METHODS: Myocardial ischemia/reperfusion injury(MIRI) was induced by ischemia for 30 min and reperfusion for 30 min. Electrocardiogram data and coronary flow were recorded, and superoxide dismutase(SOD), malondialdehyde(MDA), lactate dehydrogenase, creatine kinase-MB, cardiac troponin T and I(cT nT, cT n I) and interleukin-1β, interleukin-8,interleukin-18(IL-1β, IL-8, IL-18) in myocardium were measured. Hypoxia/reoxygenation and hydrogen peroxide(H2O2) injury were induced by hypoxia for 3 h/reoxygenation for 2 h, and 100 μM H2O2 for 1 h, respectively, in vitro rat myocardial cells(H9c2). Cell viability, SOD, MDA, cT nT and inflamma-tory factors(IL-1β, IL-8 and IL-18) were determined,and Toll-like receptor 4(TLR-4) expression was measured by western blotting.RESULTS: In the isolated heart experiment, elevated heart function, coronary flow and SOD levels,and decreased MDA levels and inflammatory factors were noted in the YDXNTC, main components and main components compatibility groups. Ventricular tachycardia/ventricular fibrillation occurrence decreased in the ginkgo biloba extract(GBE),and GBE and salvia miltiorrhiza ethanol extract compatibility(SM-E, GSEC) groups. Lactic dehydrogenase levels decreased in the YDXNTC and aqueous extract of salvia miltiorrhiza(SM-H) groups. Creatine kinase-MB decreased with GBE, SM-E, SM-H and GSEC treatment, and cT n I and cT nT levels decreased with GSEC. In the in vitro cell study,YDXNTC and main components ratios improved cell viability and SOD levels, and suppressed MDA,cT nT and inflammatory factors. TLR-4 expression was down-regulated.CONCLUSION: YDXNTC and main components compatibility showed protective effects on MIRI in this rat model and in vitro study. | 5 | Characterization of drug responses of mini patient-derived xenografts in mice for predicting cancer patient clinical therapeutic response显示文摘Background:Patient-derived organoids and xenografts(PDXs)have emerged as powerful models in functional diag-nostics with high predictive power for anticancer drug response.However,limitations such as engraftment failure and time-consuming for establishing and expanding PDX models followed by testing drug efficacy,and inability to subject to systemic drug administration for ex vivo organoid culture hinder realistic and fast decision-making in selecting the right therapeutics in the clinic.The present study aimed to develop an advanced PDX model,namely MiniPDX,for rapidly testing drug efficacy to strengthen its value in personalized cancer treatment.Methods:We developed a rapid in vivo drug sensitivity assay,OncoVee®MiniPDX,for screening clinically relevant regimens for cancer.In this model,patient-derived tumor cells were arrayed within hollow fiber capsules,implanted subcutaneously into mice and cultured for 7 days.The cellular activity morphology and pharmacokinetics were systematically evaluated.MiniPDX performance(sensitivity,specificity,positive and negative predictive values)was examined using PDX as the reference.Drug responses were examined by tumor cell growth inhibition rate and tumor growth inhibition rate in PDX models and MiniPDX assays respectively.The results from MiniPDX were also used to evaluate its predictive power for clinical outcomes.Results:Morphological and histopathological features of tumor cells within the MiniPDX capsules matched those both in PDX models and in original tumors.Drug responses in the PDX tumor graft assays correlated well with those in the corresponding MiniPDX assays using 26 PDX models generated from patients,including 14 gastric cancer,10 lung cancer and 2 pancreatic cancer.The positive predictive value of MiniPDX was 92%,and the negative predictive value was 81%with a sensitivity of 80%and a specificity of 93%.Through expanding to clinical tumor samples,Min-iPDX assay showed potential of wide clinical application.Conclusions:Fast in vivo MiniPDX assay based on capsule implantation was developed-to assess drug responses of both PDX tumor grafts and clinical cancer specimens.The high correlation between drug responses of paired MiniPDX and PDX tumor graft assay,as well as translational data suggest that MiniPDX assay is an advanced tool for personalized cancer treatment. | Feifei Zhang Wenjie Wang Yuan Long Hui Liu Jijun Cheng Lin Guo Rongyu Li Chao Meng Shan Yu Qingchuan Zhao Shun Lu Lili Wang Haitao Wang Danyi Wen | 2018 | Cancer Communications2018,38,1: | 20 |

| 8 | Effectiveness of Different Waist Circumference Cut-off Values in Predicting Metabolic Syndrome Prevalence and Risk Factors in Adults in China显示文摘Objective To study the effectiveness of waist circumference cut-off values in predicting the prevalence of metabolic syndrome(MetS) and risk factors in adults in China. Methods A cross-sectional survey was condcuted in 14 provinces(autonomous region, municipality) in China. A total of 47 325 adults aged ≥20 years were selected by multistage stratified sampling, and questionnaire survey and physical and clinical examination were conducted among them. MetS was defined according to the International Diabetes Federation(IDF) criteria and modified IDF criteria.Results The age-standardized prevalence of MetS was 24.2%(22.1% in men and 25.8% in women) and 19.5%(22.1% in men and 18.0% in women) according to the IDF criteria and modified IDF criteria respectively. The age-standardized prevalence of pre-MetS was 8.1%(8.6% in men and 7.8% in women) according to the modified IDF criteria. The prevalence of MetS was higher in urban residents than rural residents and in northern China residents than in southern China residents. The prevalence of central obesity was about 30% in both men and women according to the ethnicity-specific cut-off values of waist circumference for central obesity(90 cm for men and 85 cm for women). Multivariate regression analysis revealed no significant difference in risk factors between the two MetS definitions. | 10 | A New Triassic Primitive Ichthyosaur from Yuanan, South China显示文摘Based on the systematic study of two fossil skeleton specimens collected from the top of the third member of the marine Lower Triassic Jialingjiang Formation of Yuanan, Hubei Province, South China, a new Early Triassic primitive ichthyosaur Chaohusaurus zhangjiawanensis sp. nov. is reported and described. The beds yielding the type material are correlated with the Neospathodus homeri-N. triangularis Conodont zone. The new taxon is most similar to Chaohusaurus geishanensis Young and Dong, 1972 in the shape and configuration of the scapula, forefin, pelvic girdle and hindfin, presacral vertebral count and well-developed caudal peak, but distinguished from the latter by its larger size, the position of the pineal foramen in the centre of the parietal, the occurrence of a larger calcaneum in hindfin and the first sacral rib with distal expansion. The new species exhibits common features of primitive ichthyosaurs such as: (1) the prefrontal has an antero-dorsal shelf projecting towards the orbit; (2) the upper temporal fenestra is small; (3) the postorbital and the squamosal meet laterally to the upper temporal fenestra; and (4) cylindrical centra. The pendulum model is first time introduced into the physical modeling and formation mechanism analysis of surface clutter, and the quantitative expression of clutter spectrum spread is analytically obtained. Under the assumption of pendulum model, the processing results of measured clutter data agree well with the results of theoretical analysis, which fully verifies the correctness of pendulum model and reveals the formation mechanism of surface clutter spectrum in forward scattering radar. | 17 | Necrostatin-1 protection of dopaminergic neurons显示文摘Necroptosis is characterized by programmed necrotic cell death and autophagic activation and might be involved in the death process of dopaminergic neurons in Parkinson's disease. We hypothesized that necrostatin-1 could block necroptosis and give protection to dopaminergic neurons. There is likely to be crosstalk between necroptosis and other cell death pathways, such as apoptosis and autophagy. PC12 cells were pretreated with necroststin-1 1 hour before exposure to 6-hydroxydopamine. We examined cell viability, mitochondrial membrane potential and expression patterns of apoptotic and necroptotic death signaling proteins. The results showed that the autophagy/lysosomal pathway is involved in the 6-hydroxydopamine-induced death process of PC12 cells. Mitochondrial disability induced overactive autophagy, increased cathepsin B expression, and diminished Bcl-2 expression. Necrostatin-1 within a certain concentration range(5–30 μM) elevated the viability of PC12 cells, stabilized mitochondrial membrane potential, inhibited excessive autophagy, reduced the expression of LC3-II and cathepsin B, and increased Bcl-2 expression. These findings suggest that necrostatin-1 exerted a protective effect against injury on dopaminergic neurons. Necrostatin-1 interacts with the apoptosis signaling pathway during this process. | 18 | Apoptosis mechanisms of human gastric cancer cell line MKN-45 infected with human mutant p27显示文摘AIM: To explore the inducing effect of human mutant p27 gene on the apoptosis of the human gastric cancer cell line MKN-45 and its associated mechanisms. METHODS: The recombinant adenovirus Ad-p27mt was constructed to infect the human gastric cancer cell line MKN-45. Using flow cytometry, TUNEL assay and DNA fragment analysis, we measured the apoptotic effect of Ad-p27mt on the human gastric cancer cells. RESULTS: Ad-p27mt was successfully constructed and the infection efficiency reached 100%. After 18 h of infection, we observed an apoptotic hypodiploid peak on the flow cytometer before G1-S and apoptotic characteristic bands in the DNA electrophoresis. | 19 | Plant food sources and stone tools' function at the site of Shunshanji based on starch grain analysis显示文摘The upper-middle Huai River valley(HRV) is located in the transition zone in the middle of eastern China. Previous studies have shown that the past populations in the upper HRV obtained plant food through gathering and cultivation of both rice and millet during the middle Neolithic. However, for the middle HRV it is not very clear what methods were used by humans to obtain plant food in that time. In this paper, starch grain analysis was carried out on 17 stone tools and 29 cauldron pottery sherds unearthed at Shunshanji(8.5–7.0 kyr BP), which is known as the earliest Neolithic site in the middle HRV excavated so far. Here, ancient starches from Coix lacryma-jobi, Triticeae, Oryza sativa, Trichosanthes kirilowii and one unidentified specie were recovered. This study contributes to the limited knowledge of food strategies as observed in the middle HRV. It demonstrates that both gathering and cultivation of rice took place during the middle-Neolithic. Among the identified species, Coix lacryma-jobi appears to have been the main plant food at Shunshanji. Moreover, it is clear that Oryza sativa was not consumed as much as other plant species according to the occurrence frequency of different starch grains. Starch grains were also found on the used surfaces of grinding stone implements as well as pestles, which means that these stone tools were used for food processing. In addition, axes from Shunshanji may also have been used for food processing as well as wood working because starches were also found on the edge of axes. | 20 | Prospects and Problems for Identification of Poisonous Plants in China using DNA Barcodes显示文摘Objective Poisonous plants are a deadly threat to public health in China. The traditional clinical diagnosis of the toxic plants is inefficient, fallible, and dependent upon experts. In this study, we tested the performance of DNA barcodes for identification of the most threatening poisonous plants in China. Methods Seventy-four accessions of 27 toxic plant species in 22 genera and 17 families were sampled and three DNA barcodes(matK, rbcL, and ITS) were amplified, sequenced and tested. Three methods, Blast, pairwise global alignment(PWG) distance, and Tree-Building were tested for discrimination power. Results The primer universality of all the three markers was high. Except in the case of ITS for Hemerocallis minor, the three barcodes were successfully generated from all the selected species. Among the three methods applied, Blast showed the lowest discrimination rate, whereas PWG Distance and Tree-Building methods were equally effective. The ITS barcode showed highest discrimination rates using the PWG Distance and Tree-Building methods. When the barcodes were combined, discrimination rates were increased for the Blast method. Conclusion DNA barcoding technique provides us a fast tool for clinical identification of poisonous plants in China.
