| 1 | Expression of p53,Bax and Bcl-2 proteins in hepatocytes in non-alcoholic fatty liver disease显示文摘AIM: To analyze the protein expression essential for apoptosis in liver steatosis. METHODS: The expression of proapoptotic proteins p53, Bax, and antiapoptotic Bcl-2 in hepatocytes with steatosis (SH) and without steatosis (NSH) was evaluated in 84 patients at various stages of non-alcoholic fatty liver disease (NAFLD). RESULTS: Immunohistochemical staining of liver tissue showed the activation of p53 protein in SH and NSH with increased liver steatosis, diminished Bcl-2 and slightly decreased Bax protein. Positive correlation was found between the stage of liver steatosis with p53 expression in SH (r = 0.54, P < 0.01) and NSH (r = 0.49, P < 0.01). The antiapoptotic protein Bcl-2 was diminished together with the advancement of liver steatosis, especially in non-steatosed hepatocytes (r =0.43, P < 001). CONCLUSION: Apoptosis is one of the most important mechanisms leading to hepatocyte elimination in NAFLD. | 3 | Intestinal epithelium, intraepithelial lymphocytes and the gut microbiota-Key players in the pathogenesis of celiac disease显示文摘Celiac disease(CD) is a chronic immune-mediated disorder triggered by the ingestion of gluten in genetically predisposed individuals. Before activating the immune system, gluten peptides are transferred by the epithelial barrier to the mucosal lamina propria, where they are deamidated by intestinal tissue transglutaminase 2. As a result, they strongly bind to human leucocyte antigens(HLAs), especially HLA-DQ2 and HLA-DQ8, expressed on antigen-presenting cells. This induces an inflammatory response, which results in small bowel enteropathy. Although gluten is the main external trigger activating both innate and adaptive(specific) immunity, its presence in the intestinal lumen does not fully explain CD pathogenesis. It has been hypothesized that an early disruption of the gut barrier in genetically susceptible individuals, which would result in an increased intestinal permeability, could precede the onset of gluten-induced immune events. The intestinal barrier is a complex functional structure, whose functioning is dependent on intestinal microbiotahomeostasis, epithelial layer integrity, and the gutassociated lymphoid tissue with its intraepithelial lymphocytes(IELs). | 4 | Does Cold Plasma Affect Breaking Dormancy and Seed Germination? A Study on Seeds of Lamb's Quarters(Chenopodium album agg.)显示文摘Low-pressure discharge is applied for stimulation of germination of two seed lots of Lamb's Quarters(Chenopodium album agg.) with different starting germinations(17%,8%) and in different stages of dormancy.Different exposition durations with cold plasma treatment were applied.The variable of the ratio cumulative germination was calculated.The Richards' equation was used for curve-fitting and simulation of the growth curves.Population parameters,namely Vi-viability,Me-time,Qu-dispersion,and Sk-skewness,counted from the curves described the germination rate well.Significant differences among Qu confirmed the erratic dormancy and gradual germination of Lamb's Quarters.No difference in the Me parameter was found between two tested seed lots,and no interspecies characteristics were changed using low-pressure discharge. | 5 | Phagocytic and oxidative burst activity of neutrophils in the end stage of liver cirrhosis显示文摘AIM: To evaluate the phagocytic activity and neutrophil oxidative burst in liver cirrhosis.METHODS: In 45 patients with advanced postalcoholic liver cirrhosis (aged 45±14 years) and in 25 healthy volunteers (aged 38±5 years), the percentage of phagocytizing cells after in vitro incubation with E. coli (Phagotest Kit), phagocytic activity (mean intensity of fluorescence, MIF) and the percentage of neutrophil oxidative burst (Bursttest Kit), and the level of free oxygen radical production (MIF of Rodamine 123)were analyzed by flow cytometry. The levels of soluble sICAM-1, sVCAM-1, sP-selectin, sE-selectin, sL-selectin,and TNF-α were determined in blood serum.RESULTS: The percentage of E. coli phagocytizing neutrophils in liver cirrhosis patients was comparable to that in healthy subjects. MIF of neutrophil - ingested E. coli was higher in patients with liver cirrhosis. The oxidative burst in E. coli phagocytizing neutrophils generated less amount of active oxygen compounds in liver cirrhosis patients (MIF of R123:24.7±7.1 and 29.7±6.6 in healthy,P<0.01). Phorbol myristate acetate (PMA) - stimulated neutrophilsproduced less reactive oxidants in liver cirrhosis patients than in healthy subjects (MIF of R123: 42.7±14.6 vs 50.2±13.3, P<0.01). A negative correlation was observed between oxidative burst MIF of PMA-stimulated neutrophils and ALT and AST levels (r -0.35, P<0.05;r-0.4, P<0.03). sVCAM-1, sICAM-1, sE-selectin concentrations correlated negatively with the oxygen free radical production (MIF of R123) in neutrophils after PMA stimulation in liver cirrhosis patients (r-0.45, P<0.05;r-0.41, P<0.05; r-0.39, P<0.05, respectively).CONCLUSION: Neutrophil metabolic activity diminishes together with the intensification of liver failure. The metabolic potential of phagocytizing neutrophils is significantly lower in liver cirrhosis patients, which can be one of the causes of immune mechanism damage. | 6 | Investigation of the immune effects of Scutellaria baicalensis on blood leukocytes and selected organs of the chicken's lymphatic system显示文摘Background: The health of chickens and the welfare of poultry industry are central to the efforts of addressing global food security. Therefore, it is essential to study chicken immunology to maintain and improve its health and to find novel and sustainable solutions. This paper presents a study on investigation of the effect of Scutellaria baicalensis root(SBR) on the immune response of broiler chicken, especially on lymphocytes and heterophils reactivity, regarding their contribution to the development of immunity of the chickens.Methods: The 121-day-old Hubbard Hi-Y male broiler hybrids were randomly assigned to four treatment groups,three SBR supplemented groups(0.5, 1.0, and 1.5% of SBR) and one control group. Each treatment was replicated five times with six birds per replicate pen in a battery brooder. Blood was collected after 3^(rd) and 6^(th)wk of the experiment, and hemoglobin and hematocrit values were determined, as well as total leukocyte count and differential count were performed. Nitroblue tetrazolium test and phagocytosis assay as nonspecific immune parameters and humoral immune responses to the antigenic challenge by sheep red blood cells were performed.Moreover, the ability of peripheral blood lymphocytes to form radial segmentation(RS) of their nuclei was analyzed.Body weight and relative weight of spleen, liver, and bursa of Fabricius were recorded.Results: Results showed that mean heterophile/lymphocyte ratio increased in the SBR groups compared to the control group and the blood of the chickens showed lymphocytic depletion. The results also demonstrated that the relative weight of bursa of Fabricius and spleen in groups fed with SBR significantly decreased compared to the control group. This study also showed that the addition of SBR significantly inhibited the formation of RS of nuclei compared to some cytotoxic substances.Conclusion: We found that SBR supplementation should be carefully evaluated when given to poultry. The excess intake of SBR supplementation may cause immunologic inhibition and may negatively affect the development of immune organs. | 7 | Expression of β2-integrin on leukocytes in liver cirrhosis显示文摘AIM: To analyze β2-integrin expression on blood leukocytes in liver cirrhosis. METHODS: In 40 patients with liver cirrhosis and 20 healthy individuals, the evaluation of expression of CD11a (LFA-1α), CD11b (Mac-1α), CD11c (αX) and CD49d (VLA-4α) on peripheral blood leukocytes was performed using flow cytometry. The analysis was carried out in groups of patients divided into B and C according to Child-Pugh’s classification. RESULTS: An increased CD11a, CD11b, CD11c and CD49d integrin expression was observed on peripheral blood leukocytes in liver cirrhosis. The integrin levels were elevated as the advancement of liver failure progressed. The highest expression of integrins occurred predominantly on monocytes. A slight expression of VLA-4 was found on lymphocytes and granulocytes and it increased together with liver failure. A positive correlation was noted between median intensity of fluorescence (MIF) expression on polymorphonuclear cells of CD11a and CD11c and CD49d (r = 0.42, P < 0.01; r = 053, P < 0.01, respectively) in liver cirrhosis stage C. However, no correlation was observed between integrin expression on leukocytes. The concentrations of sICAM-1, sVCAM-1, and TNFα, were significantly elevated in liver cirrhosis. | 9 | Levels of biological markers of nitric oxide in serum of patients with squamous cell carcinoma of the oral cavity显示文摘The aim of the study was a determination of the levels of nitric oxide(NO)and its biological markers such as malonyldialdehyde(MDA)and nitrotyrosine in the serum of patients with squamous cell carcinoma(SCC)of the oral cavity and identification of the relationships between NO and those markers.These studies were performed on patients with SCC of the oral cavity before and after treatment.Griess reaction was used for the estimation of the total concentration of NO in serum.The nitrotyrosine level in serum was assessed with an enzyme-linked immunosorbent assay(ELISA)kit,and MDA level using a spectrophotometric assay.Higher concentrations of NO in blood serum were determined in patients with stage IV of the disease before treatment in comparison to the control group and patients with stages II and III of the disease.Moreover,higher concentrations of MDA and nitrotyrosine were determined in the serum of patients in all stages of the disease in comparison to healthy people.After treatment,lower concentrations of NO in the serum of patients with stage IV of the disease were observed in comparison to the amounts obtained prior to treatment.In addition,lower levels of nitrotyrosine in the serum of patients with all stages of the disease were recorded,whereas higher concentrations of MDA were determined in these patients in comparison to results obtained before treatment.The compounds formed with the contribution of NO,such as MDA and nitrotyrosine,may lead to cancer progression in patients with SCC of the oral cavity,and contribute to formation of resistance to therapy in these patients as well.Moreover,the lack of a relationship between concentrations of NO and MDA,and between NO and nitrotyrosine in serum suggests that the process of lipid peroxidation and nitration in patients with SCC does not just depend on NO. | Wioletta Ratajczak-Wrona Ewa Jablonska Bozena Antonowicz Dorota Dziemianczyk Stanislawa Zyta Grabowska | 2013 | International Journal of Oral Science2013，5，3： | 2 |

| 18 | A short P-wave duration is associated with incident heart failure in the elderly: a 15 years follow-up cohort study显示文摘BACKGROUND Early identification of patients at risk of congestive heart failure(HF)may alter their poor prognosis.The aim was therefore to test whether simple electrocardiographic variables,the P-wave and PR-interval,could predict incident HF.METHODS The PIVUS(Prospective Investigation of the Vasculature in Uppsala Seniors)study(1016 individuals all aged 70 years,50%women)was used to identify predictors of HF.Subjects with prevalent HF,QRS duration≥130 ms,atrial tachyar-rhythmias,implanted pacemaker/defibrillator,second-and third-degree atrioventricular block or delta waves at baseline were excluded.Cox proportional hazard analysis was used to relate the PR interval,P-wave duration(Pdur)and amplitude(Pamp),measured in lead V1,to incident HF.Adjustment was performed for gender,RR-interval,beta-blocking agents,systolic blood pressure,body mass index and smoking.RESULTS Out of 836 subjects at risk,107 subjects were diagnosed with HF during a follow-up of 15 years.In the multivariate analysis,there was a strong U-shaped correlation between Pdur in lead V1 and incident HF(P=0.0001)which was significant for a Pdur<60 ms[HR=2.75;95%CI:1.87-4.06,at Pdur 40 ms]but not for prolonged Pdur.There was no significant relationship between incident HF and the PR-interval or the Pamp.A Pdur<60 ms improved discrimination by 3.7%when added to the tradi-tional risk factors including sex,RR-interval,beta-blocking agents,systolic blood pressure,BMI and smoking(P=0.048).CONCLUSIONS A short Pdur,an easily measured parameter on the ECG,may potentially be a useful marker of future HF,enabling its early detection and prevention,thus improving outcomes. | Bozena Ostrowska Lars Lind Elena Sciaraffia Carina Blomström-Lundqvist | 2022 | Journal of Geriatric Cardiology2022，19，9： | 1 |
