1Collagen type Ⅱ suppresses articular chondrocyte hypertrophy and osteoarthritis progression by promoting integrin β1-SMAD1 interaction显示文摘Hypertrophic differentiation is not only the terminal process of endochondral ossification in the growth plate but is also an important pathological change in osteoarthritic cartilage.Collagen type II(COL2A1)was previously considered to be only a structural component of the cartilage matrix,but recently,it has been revealed to be an extracellular signaling molecule that can significantly suppress chondrocyte hypertrophy.However,the mechanisms by which COL2A1 regulates hypertrophic differentiation remain unclear.In our study,a Col2a1 p.Gly1170Ser mutant mouse model was constructed,and Col2a1 loss was demonstrated in homozygotes.Loss of Col2a1 was found to accelerate chondrocyte hypertrophy through the bone morphogenetic protein(BMP)-SMAD1 pathway.Upon interacting with COL2A1,integrinβ1(ITGB1),the major receptor for COL2A1,competed with BMP receptors for binding to SMAD1 and then inhibited SMAD1 activation and nuclear import.COL2A1 could also activate ITGB1-induced ERK1/2 phosphorylation and,through ERK1/2-SMAD1 interaction,it further repressed SMAD1 activation,thus inhibiting BMP-SMAD1-mediated chondrocyte hypertrophy.Moreover,COL2A1 expression was downregulated,while chondrocyte hypertrophic markers and BMP-SMAD1 signaling activity were upregulated in degenerative human articular cartilage.Our study reveals novel mechanisms for the inhibition of chondrocyte hypertrophy by COL2A1 and suggests that the degradation and decrease in COL2A1 might initiate and promote osteoarthritis progression.Chengjie Lian Xudong Wang Xianjian Qiu Zizhao Wu Bo Gao Lei Liu Guoyan Liang Hang Zhou Xiaoming Yang Yan Peng Anjing Liang Caixia Xu Dongsheng Huang Peiqiang Su 2019Bone Research2019,7,1:17
2MEF2C promotes M1 macrophage polarization and Th1 responses显示文摘The polarization of macrophages to the M1 or M2 phenotype has a pivotal role in inflammation and host defense;however,the underlying molecular mechanism remains unclear.Here,we show that myocyte enhancer factor 2 C(MEF2C)is essential for regulating M1 macrophage polarization in response to infection and inflammation.Global gene expression analysis demonstrated that MEF2C deficiency in macrophages downregulated the expression of M1 phenotypic markers and upregulated the expression of M2 phenotypic markers.MEF2C significantly promoted the expression of interleukin-12 p35 subunit(Il12a)and interleukin-12 p40 subunit(Il12b).Myeloid-specific Mef2c-knockout mice showed reduced IL-12 production and impaired Th1 responses,which led to susceptibility to Listeria monocytogenes infection and protected against DSS-induced IBD in vivo.Mechanistically,we showed that MEF2C directly activated the transcription of Il12a and Il12b.These findings reveal a new function of MEF2C in macrophage polarization and Th1 responses and identify MEF2C as a potential target for therapeutic intervention in inflammatory and autoimmune diseases.Xibao Zhao Qianqian Di Han Liu Jiazheng Quan Jing Ling Zizhao Zhao Yue Xiao Han Wu Zherui Wu Wengang Song Huazhang An Weilin Chen 2022Cellular & Molecular Immunology2022,19,4:4
3Multimorphism and gap opening of charge-density-wave phases in monolayer VTe2显示文摘Vanadium dichalcogenides have attracted increasing interests for the charge density wave phenomena and possible ferromagnetism.Here,we report on the multiphase behavior and gap opening in monolayer VTe2 grown by molecular beam epitaxy.Scanning tunneling microscopy(STM)and spectroscopy study revealed the(4×4)metallic and gapped(2√3×2√3)charge-density wave(CDW)phases with an energy gap of≈40 meV.Through the in-plane condensation of vanadium atoms,the typical star-of-David clusters and truncated triangle-shaped clusters are formed in the(4×4)and(2√3×2√3)phases respectively,resulting in different surface morphologies and electronic structures as confirmed by density functional theory(DFT)calculations with on-site Coulomb repulsion.The CDW-driven reorganization of the atomic structure weakens the ferromagnetic superexchange coupling and strengthens the antiferromagnetic exchange coupling on the contrary,suppressing the long-range magnetic order in monolayer VTe2.The electron correlation is found to be important to explain the gap opening in the(2√3×2√3)phase.Meizhuang Liu Changwei Wu Zizhao Liu Zhiqiang Wang Dao-Xin Yao Dingyong Zhong 2020Nano Research2020,13,6:2

5La_(0.7)Ca_(0.3)MnO_(3-δ):Ag nanocomposite thin films with large temperature coefficient of resistance(TCR)显示文摘In this work, oxygen vacancy-regulated La_(0.7)Ca_(0.3)MnO_(3-δ):Ag (LCMO:A) nanocomposite thin films onLaAlO_(3) (001) substrates were investigated to obtain films with large temperature coefficient of resistance(TCR) values. LCMO:A nanocomposite thin films were synthesized using pulsed laser deposition, andoxygen pressures during film deposition and annealing steps were optimized. As oxygen pressuresincreased, lattice parameter increased from 70 Pa to 100 Pa, Tp increased monotonically from 168 K to282 K, and average Mn^(4+) concentration in the film increased as indicated by X-ray photoemissionspectroscopy data.
