1Functional identification of the terpene synthase family involved in diterpenoid alkaloids biosynthesis in Aconitum carmichaelii显示文摘Aconitum carmichaelii is a high-value medicinal herb widely used across China,Japan,and other Asian countries.Aconitine-type diterpene alkaloids(DAs)are the characteristic compounds in Aconitum.Although six transcriptomes,based on short-read next generation sequencing technology,have been reported from the Aconitum species,the terpene synthase(TPS)corresponding to DAs biosynthesis remains unidentified.We apply a combination of Pacbio isoform sequencing and RNA sequencing to provide a comprehensive view of the A.carmichaelii transcriptome.Nineteen TPSs and five alternative splicing isoforms belonging to TPS-b,TPS-c,and TPS-e/f subfamilies were identified.In vitro enzyme reaction analysis functional identified two sesqui-TPSs and twelve di TPSs.Seven of the TPS-c subfamily genes reacted with GGPP to produce the intermediate ent-copalyl diphosphate.Five Ac KSLs separately reacted with ent-CPP to produce ent-kaurene,ent-atiserene,and ent-13-epi-sandaracopimaradie:a new diterpene found in Aconitum.Ac TPSs gene expression in conjunction DAs content analysis in different tissues validated that ent-CPP is the sole precursor to all DAs biosynthesis,with Ac KSL1,Ac KSL2 s and Ac KSL3-1 responsible for C20 atisine and napelline type DAs biosynthesis,respectively.These data clarified the molecular basis for the C20-DAs biosynthetic pathway in A.carmichaelii and pave the way for further exploration of C19-DAs biosynthesis in the Aconitum species.Liuying Mao Baolong Jin Lingli Chen Mei Tian Rui Ma Biwei Yin Haiyan Zhang Juan Guo Jinfu Tang Tong Chen Changjiangsheng Lai Guanghong Cui Luqi Huang 2021Acta Pharmaceutica Sinica B2021,11,10:6
2Protein convertase subtilisin/Kexin type 9 inhibits hepatocellular carcinoma growth by interacting with GSTP1 and suppressing the JNK signaling pathway显示文摘Objective:Protein convertase subtilisin/Kexin type 9(PCSK9)has been found to be closely associated with the occurrence and development of numerous tumors.However,the precise role of PCSK9 and its relationship to the development of hepatocellular carcinoma(HCC)remain largely unknown.This study aimed to clarify these issues.Methods:The expression levels of PCSK9 in HCC tissues and HCC cell lines were determined by the quantitative reverse transcription polymerase chain reaction,Western blot,and immunohistochemical analyses,and the effects of PCSK9 expression on HCC cell biological traits were investigated by overexpressing and downregulating PCSK9 expression in vivo and in vitro.Additionally,the mechanism by which PCSK9 mediated dissociation of glutathione S-transferase Pi 1(GSTP1)dimers and phosphorylation of the Jun N-terminal kinase(JNK)pathway components were investigated.Results:PCSK9 expression levels were significantly lower in HCC tissues than in adjacent non-tumor samples.In vivo and in vitro experiments suggested that PCSK9 inhibited HCC cell proliferation and metastasis.Further analysis showed that PCSK9 interacted with GSTP1 and promoted GSTP1 dimer dissociation and JNK signaling pathway inactivation in HCC cells.Moreover,the relationships between PCSK9 protein expressions and clinical outcomes were investigated.The PCSK9-lo group displayed a significantly shorter overall survival(OS;median OS:64.2 months vs.83.2 months;log-rank statistic:4.237;P=0.04)and recurrencefree survival(RFS;median RFS:26.5 months vs.46.6 months;log-rank statistic:10.498;P=0.001)time than the PCSK9-hi group.Conclusions:PCSK9 inhibited HCC cell proliferation,cell cycle progression,and apoptosis by interacting with GSTP1 and suppressing JNK signaling,suggesting that PCSK9 might act as a tumor suppressor and be a therapeutic target in HCC patients.Mingyan He Jing Hu Tingting Fang Wenqing Tang Bei Lv Biwei Yang Jinglin Xia 2022Cancer Biology & Medicine2022,19,1:1
3Estimation of the interaction force between human and passive lower limb exoskeleton device during level ground walking显示文摘To develop sophisticated and efficient control strategies for exoskeleton devices,acquiring the information of interaction forces between the wearer and the wearable device is essential.However,obtaining the interaction force via conventional methods,such as direct measurement using force sensors,is problematic.This paper proposes a kinematic data-based estimation method to evaluate the interaction force between human lower limbs and passive exoskeleton links during level ground walking.Unlike conventional methods,the proposed method requires no force sensors and is computationally cheaper to obtain the calculation results.To obtain more accurate kinematic data,a marker refinement algorithm based on bilevel optimization framework is adopted.The interaction force is evaluated by a spring model,which is used to imitate the binding behavior between human limbs and the exoskeleton links.The deflection of the spring model is calculated based on the assumption that the phase delay between human limb and exoskeleton link can be presented by the sequence of frames of kinematic data.Experimental results of six subjects indicate that our proposed method can estimate the interaction forces during level ground walking.Moreover,a case study of bandage location optimization is conducted to demonstrate the usefulness of obtaining the interaction information.Muye Pang Zihan Luo Biwei Tang Jing Luo Kui Xiang 2022Biomimetic Intelligence & Robotics2022,2,3:0
4Calcyclin-binding protein contributes to cholangiocarcinoma progression by inhibiting ubiquitination of MCM2显示文摘Background:Cholangiocarcinoma(CCA)represents the epithelial cell cancer with high aggressiveness whose five-year survival rate is poor with standard treatment.Calcyclin-binding protein(CACYBP)shows aberrant expression within several malignant tumors,but the role of CACYBP in CCA remains unknown.Methods:Immunohistochemical(IHC)analysis was used to identify CACYBP overexpression in clinical samples of CCA patients.Moreover,its correlation with clinical outcome was revealed.Furthermore,CACYBP’s effect on CCA cell growth and invasion was investigated in vitro and in vivo using loss-of-function experiments.Results:CACYBP showed up-regulation in CCA,which predicts the dismal prognostic outcome.CACYBP had an important effect on in-vitro and in-vivo cancer cell proliferation and migration.Additionally,knockdown of CACYBP weakened protein stability by promoting ubiquitination of MCM2.Accordingly,MCM2 up-regulation partly reversed CACYBP deficiency’s inhibition against cancer cell viability and invasion.Thus,MCM2 might drive CCA development by Wnt/β-catenin pathway.Conclusions:CACYBP exerted a tumor-promoting role in CCA by suppressing ubiquitination of MCM2 and activating Wnt/β-catenin pathway,hence revealing that it may be the possible therapeutic target for CCA treatment.YUSEN ZHANG LIPING LIU BIWEI LUO HONGGUI TANG XIAOFANG YU SHIYUN BAO 2023Oncology Research2023,31,3:0
5Unintentional Injury Mortality Among Children Under Five Years--China,2006−2017显示文摘What is already known about this topic?Unintentional injuries among children aged under five years is still a serious public health problem in China.Epidemiological characteristics of under-five unintentional injury are reported in single provinces and cities for China but not nationally based on the Disease Surveillance Points(DSPs)dataset.What is added by this report?Unintentional injury mortality rates for under-five decreased substantially between 2006 and 2017 in China,with the decrease occurring primarily for males and rural children.Children living in rural areas and males had higher unintentional injury mortality rates than children in urban areas and females.The major fatal types of under-five unintentional injury were drowning and road injury.What are the implications for public health practice?Further interventions need to be taken in accordance with the main types of unintentional injuries,especially effective prevention strategies used in other countries or recommended by the World Health Organization(WHO).Even with equal access to injury prevention and control services for different sexes,policy efforts should focus on higher-risk populations,especially children aged under five years who are males or reside in rural areas.Bo Chen Pengpeng Ye Biwei Tang Siqi Wang Jia Guo Jing Wu 2020China CDC weekly2020,2,10:0
