| 1 | Biomineralized polymer matrix composites for bone tissue repair:a review显示文摘Bone defects caused by trauma, infection or bone tumor resection, are highly prevalent. A small number(5%–10%) of these injuries fail to heal due to non-union and require surgical intervention. Currently, the principal treatment options for these defects are autografts, allografts, xenografts or synthetic grafts. The main problems associated with these therapies include pain,infection and donor site morbidity. Bone tissue engineering is a diverse field that focuses on the regeneration of bone by combining cells, scaffolds, growth factors and dynamic forces. There have been many recent studies utilizing biomineralized polymer matrix composites which mimic the natural structure of bone. The principal focus of this review is on recent advances in the synthesis of various types of biomineralized polymer matrix composite. | 2 | Camptothecin@HMSNs/thermosensitive hydrogel composite for applications in preventing local breast cancer recurrence显示文摘Camptothecin has a strong tumor killing ability for a variety of tumor cells with its special anti-cancer mechanism including the breast cancer. However, because of its infinite hydrophobic property, its clinical application has been greatly limited. Early prevention of loco regional recurrence for the breast cancer is critical for patients who have undergone breast-conserving therapy. In the study,CPT was used for the inhibition of the recurrence after the operation. The hollow mesoporous silica nanoparticles were used as the carrier to improve the hydrophilic property and increase its bioavailability with the high loading capacity. The ability of the cellular uptake and antitumor activity was increased. Hydrogel was the ideal carrier for local therapy, so the CPT@HMSNs were loaded into the PLEL thermo sensitive hydrogel to be injected into the tumor sites after the tumor was resected. The recurrence was reduced in the group of CPT-HMSNs-PLEL and the side effect of CPT was decreased. | 3 | Large-area gold nanohole arrays fabricated by one-step method for surface plasmon resonance biochemical sensing显示文摘Surface plasmon resonance(SPR) nanosensors based on metallic nanohole arrays have been widely reported to detect binding interactions in biological specimens. A simple and effective method for constructing nanoscale arrays is essential for the development of SPR nanosensors. In this work, we report a one-step method to fabricate nanohole arrays by thermal nanoimprinting in the matrix of IPS(Intermediate Polymer Stamp). No additional etching process or supporting substrate is required. The preparation process is simple, time-saving and compatible for roll-to-roll process, potentially allowing mass production. Moreover, the nanohole arrays were integrated into detection platform as SPR sensors to investigate different types of biological binding interactions. | 4 | Genetic architecture of leaf morphology revealed by integrated trait module in Catalpa bungei显示文摘Leaves are crucial for maintaining plant growth and development via photosynthesis,and their function is simultaneously regulated by a suite of phenotypic traits.Although much is known about the genetic architecture of individual leaf traits,unraveling the genetic basis of complex leafmorphology remains a challenge.Based on the functional correlation and coordination ofmulti-traits,we divided 15 leaf morphological traits into three modules,comprising size(area,length,width,and perimeter),shape(leaf lobes,aspect ratio,circularity,rectangularity,and the relevant ratios),and color(red,green,and blue)for an ornamental tree species,Catalpa bungei.A total of 189 significant single-nucleotide polymorphisms were identified in the leaves of C.bungei:35,82,and 76 in the size,shape,and color modules,respectively.Four quantitative trait loci were common between the size and shape modules,which were closely related according to phenotype correlation,genetic mapping,and mRNA analysis.The color module was independent of them.Synergistic changes in the aspect ratio,leaf lobe,and circularity suggest that these traits could be the core indicators of the leaf shape module.The LAS and SRK genes,associated with leaf lobe and circularity,were found to function in plant defense mechanisms and the growth of leaves.The associations between the SRK and CRK2 genes and the leaf lobe and circularity traits were further verified by RT-qPCR.Our findings demonstrate the importance of integrating multi-trait modules to characterize leaf morphology and facilitate a holistic understanding of the genetic architecture of intraspecific leaf morphology diversity. | Miaomiao Zhang Bingyang Liu Yue Fei Xiaowei Yang Linjiao Zhao Chaozhong Shi Yueying Zhang Nan Lu Chuangye Wu Wenjun Ma Junhui Wang | 2023 | Horticulture Research2023,10,4: | 0 |
| 5 | Brain-targeted polymersome codelivery of siRNA and temozolomide for effective glioblastoma chemo-RNAi synergistic therapy显示文摘Temozolomide (TMZ) is a clinically approved drug for glioblastoma (GBM) therapy. However, as a result of methylguanine-DNA-methyltransferase (MGMT), which is able to repair damaged DNA-damage repairing, TMZ usually yields unsatisfactory therapeutic effects. Small interfering RNA (siRNA) is a potential alteration tool for sensitivity of TMZ by targeting DNA repair enzymes. However, a suitable TMZ and siRNA codelivery system that can effectively and actively co-deliver siRNA/TMZ into the brain tumor is lacking. In this study, we constructed an angiopep-2 decorated polymersomal delivery system to co-deliver TMZ/siRNA for synergistic GBM therapy. This targeted polymersomal nanomedicine not only enhanced the circulation time of siRNA/TMZ in blood but also improved their blood-brain barrier (BBB) crossing and GBM targeting ability. Moreover, when we co-administered siRNAs specific to retinoblastoma binding protein 4 (RBBP4) together with TMZ in GBM cells, these RBBP4- specific siRNA (siRBBP4) modulated the sensitivity of TMZ by regulating MGMT, and thus showed a powerful synergistic anti-tumor effect. We demonstrated that angiopep-2 decorated polymersomal siRBBP4/TMZ co-loaded nanomedicines are capable of inhibiting tumor growth and significantly improved life expectancy of orthotropic GBM bearing mice.
