1Cycloastragenol upregulates SIRT1 expression,attenuates apoptosis and suppresses neuroinflammation after brain ischemia显示文摘Cycloastragenol(CAG)is the active form of astragaloside V isolated from Astragalus Radix,which displays multiple pharmacological effects.Silent information regulator 1(SIRT1),a classll histone deacetylase,has been shown to play an important role in neuroprotection against cerebral ischemia.In this study,we investigated whether CAG protected against ischemic brain injury and,if so,whether the beneficial effects were associated with the regulation of SIRT1 in the ischemic brain.Mice were subjected to 45 min of middle cerebral artery occlusion(MCAO)followed by reperfusion.CAG(5,10,20 mg/kg)was injected intraperitoneally at the onset of reperfusion,12h later and then twice daily for up to three days.CAG dose-dependently reduced brain infarct volume,significantly ameliorated functional deficits,and prevented neuronal cell oss in MCAO mice.Meanwhile,CAG signicantly reduced matrix metalloproteinase-9 activity,prevented tight junction degradation and subsequently ameliorated blood-brain barier disruption.Moreover,CAG significantly upregulated SIRT1 expression in the ischemic brain but did not directly activate its enzymatic activity.Concomitant with SIRT1 upregulation,CAG reduced p53 acetylation and the ratio of Bax to Bcl-2 in the ischemic brain.CAG also inhibited NF-κB p65 nuclear translocation.As a result,CAG suppressed the mRNA expression of pro-inflammatory cytokines,including TNF-α and IL-1β,and inhibited the activation of microglia and astrocytes in the ischemic brain.Our findings suggest that CAG is neuroprotective against ischemic brain injury in mice and that its beneficial effect may involve SIRT1 upregulation and the inhibition of apoptosis and neuroinflammation in the ischemic brain.Man Li Shichun Li Bao-kai Dou Yving-xiang Zou Hao-zhen Han Dong-xiang Liu Zunji Ke Zhi-fei Wang 2020Acta Pharmacologica Sinica2020,41,8:22
2Epigenetic modification in histone deacetylase deletion strain of Calcarisporium arbuscula leads to diverse diterpenoids显示文摘Epigenetic modifications have been proved to be a powerful way to activate silent gene clusters and lead to diverse secondary metabolites in fungi. Previously, inactivation of a histone H3 deacetylase in Calcarisporium arbuscula had led to pleiotropic activation and overexpression of more than 75% of the biosynthetic genes and isolation of ten compounds. Further investigation of the crude extract of C. arbuscula Δhda A strain resulted in the isolation of twelve new diterpenoids including three cassanes(1-3), one cleistanthane(4), six pimaranes(5-10), and two isopimaranes(11 and 12) along with two know cleistanthane analogues. Their structures were elucidated by extensive NMR spectroscopic data analysis. 4Metabolic regulation of T cell development by Sinl-mTORC2 is mediated by pyruvate kinase M2显示文摘Glucose metabolism plays a key role in thymocyte development. The mammalian target of rapamycin complex 2 (mT0RC2) is a critical regulator of cell growth and metabolism, but its role in early thymocyte development and metabolism has not been fully studied. We show here that genetic ablation of Sinl, an essential component of mTORC2, in T lineage cells results in severely impaired thymocyte development at the CD4^- CD8^- double negative (DN) stages but not at the CD4^+ CD8^+ double positive (DP) or later stages. Notably, Sinl-deficient DN thymocytes show markedly reduced proliferation and glycolysis.Importantly, we discover that the M2 isoform of pyruvate kinase (PKM2) is a novel and crucial Sinl effector in promoting DN thymocyte development and metabolism. At the molecular level, we show that Sinl-mTORC2 controls PKM2 expression through an AKT-dependent PPAR-y nuclear translocation. 5Microstructure of β-Dicalcium Silicate after Accelerated Carbonation显示文摘The present work presents the microstructure of β-Ca_2SiO_4(β-C_2S) after accelerated carbonation. The synthesis procedure of β-C_2S was examined first, and the crystalline and amorphous structure, the distribution and the pore structure of β-C_2S carbonation products were also determined by X-ray diffraction(XRD) quantitative analysis, simultaneous thermal analyzer(TG/DTA), Fourier transform-infrared spectroscopy(FT-IR), high resolution ^(29)Si magic angle spinning nuclear magnetic resonance(^(29)Si NMR), N_2-sorption techniques, and scanning electron microscopy(SEM), respectively. Test results indicate that carbonation products are dramatically formed in the initial 2 h. The main carbonation products are crystalline calcite and amorphous three-dimensional network silica gels, which contain nanometer-sized pores. 11Minor Compounds from Fungus Ganoderma cochlear显示文摘Objective To study the chemical constituents of the fungus Ganoderma cochlear. Methods The compounds were isolated by using MCI gel CHP 20 P, Sephadex LH-20, RP-18 column chromatography, and preparative TLC. The structures were identified by means of spectroscopic methods. Results Two phenolic normeroterpenoid and meroterpenoid, cochlearols C and D(1 and 2), together with six benzene derivatives, 3-methoxy-4-hydroxy-phenylethanol(3), 4-hydroxyacetophenone(4), p-hydroxycinnamic methyl ester(5), 2-methoxy-4-hydroxybenzaldehyde(6), 4-hydroxy-3-methoxy benzoic acid(7), and 2-hydroxy-5-ethoxybenzoic acid(8), were isolated from the fruiting bodies of Ganoderma cochlear. THEIR CHARACTERIZATION AND ELECTROCHEMISTRY显示文摘Binuclear Cu(Ⅰ) complex [Cu(dppm)(NO3)]2 (dppm=Ph2PCH2PPh2) and mononuclear Cu(Ⅰ) complex [Cu(dppe)(NO3)] (dppe=Ph2PCH2CH2PPh2) have been synthesized by the ligand-reduction reaction of cupric nitrate with dppm and dppe in methanol and characterized by means of ICP, elemental analyses, molecular weight determination, TG, 31P-NMR, IR and Raman spectra; their electronic conductivities and CV waves have also been measured.
