2Modeling study of regional severe hazes over mid-eastern China in January 2013 and its implications on pollution prevention and control显示文摘The Nested Air Quality Prediction Model System(NAQPMS)was used to investigate the temporal and spatial variations of PM2.5over tropospheric central eastern China in January 2013.The impact of regional transport and its implications on pollution prevention and control were also examined.Comparison between simulated and observed PM2.5showed NAQPMS was able to reproduce the evolution of PM2.5during heavy haze episodes.The results indicated that regional transport of PM2.5played an important role in regional haze episodes in the city cluster including Hebei,Beijing and Tianjin(HBT).The cross-city clusters transport outside HBT and transport among cities inside HBT contributed 20%–35%and 26%–35%of PM2.5as compared with local emission,in HBT respectively.To meet the Air Quality Standards for Grade II,90%,90%and65%of emissions would have to be cut down in Hebei,Tianjin and Beijing,if non-control strategy was taken in the surrounding city clusters of HBT.This implicated that control of emissions in one city cluster is not sufficient to reduce regional haze events,and joint efforts among city clusters are essential.Besides regional transports,two-way feedback between boundary-layer evolution and PM2.5also significantly contributed to the formation of heavy hazes,which contributed 30%of monthly average PM2.5concentration in HBT.WANG ZiFa LI Jie WANG Zhe YANG WenYi TANG Xiao GE BaoZhu YAN PinZhong ZHU LiLi CHEN XueShun CHEN HuanSheng WAND Wei LI JianJun LIU Bing WANG XiaoYan WAND Wei ZHAO YiLin LU Ning SU DeBin 2014Science China Earth Sciences2014,57,1:106
3Transplantation of UC-MSCs on collagen scaffold activates follicles in dormant ovaries of POF patients with long history of infertility显示文摘Premature ovarian failure(POF) is a refractory disease for clinical treatment with the goal of restoring fertility. In this study,umbilical cord mesenchymal stem cells on a collagen scaffold(collagen/UC-MSCs) can activate primordial follicles in vitro via phosphorylation of FOXO3 a and FOXO1. Transplantation of collagen/UC-MSCs to the ovaries of POF patients rescued overall ovarian function, evidenced by elevated estradiol concentrations, improved follicular development, and increased number of antral follicles. Successful clinical pregnancy was achieved in women with POF after transplantation of collagen/UC-MSCs or UC-MSCs. 5Does Helicobacter pylori infection play a role in iron deficiency anemia? A meta-analysis显示文摘AIM:To perform a meta-analysis of observational studies and randomized controlled trials(RCTs)on the association between Helicobacter pylori(H.pylori)and iron deficiency anemia(IDA).METHODS:A defined search strategy was used to search Medline,Embase,the Cochrane Library,Clinical Trials,Cochrane Central Register of Controlled Trials,Premedline and Healthstar.Odds ratio(OR)was used to evaluate observational epidemiology studies,and weighted mean difference(WMD)was used to demonstrate the difference between control and intervention groups.RESULTS:Fifteen observational studies and 5 RCTs were identified and used for calculation.The pooled OR for observational studies was 2.22(95%CI:1.52-3.24,P<0.0001).The WMD for hemoglobin(HB) was 4.06 g/L(95%CI:-2.57-10.69,P=0.01),and the WMD for serum ferritin(SF)was 9.47μg/L(95%CI:-0.50-19.43,P<0.0001).Results were heterogeneous for all comparisons.CONCLUSION:This meta-analysis on observational studies suggests an association between H.pylori and IDA.In RCTs,eradication of H.pylori can improve HB and SF levels but not significantly.Qu, Xin-Hua Huang, Xiao-Lu Xiong, Ping Zhu, Cui-Ying Huang, You-Liang Lu, Lun-Gen Sun, Xu Rong, Lan Zhong, Liang Sun, Da-Yu Lin, Hai Cai, Ming-Ci Chen, Zhi-Wei Hu, Bing Wu, Lian-Ming Jiang, Yi-Bin Yan, Wei-Li 2010World Journal of Gastroenterology2010,16,7:34
6Phosphate removal from wastewater by model-La(Ⅲ)zeolite adsorbents显示文摘Phosphorus is one of the primary nutrients which leads to eutrophication and accelerates aging process in enclosed water bodies. Because of the poor phosphorus selectivity of other adsorbents, the novel La(III)-modified zeolite adsorbent (LZA) was prepared by modifying 90 nm zeolite with lanthanide to selectively remove phosphate in the presence of various omnipresent anions, such as sulfates, bicarbonates, and chlorides. Through batch and fixed bed operation, the following optimum conditions were obtained: concentration of lanthanum chloride solution 0.05 mol/L; solid/liquor ratio 1/25; pH 10; calcination temperature 550°C; time 1 h. The value of the Freundlich model constants Kf and 1/n were found to be 16.76 mg/L and 0.2209, respectively. In addition, when calculated at pH 6.0, distribution coefficient KD could be as high as 36.6. Furthermore, in the alkaline pH range, solution of 0.8 mol/L NaCl was used to regenerate the saturated LZA, which could reach the high regeneration efficiency as high as 100%. 8Numerical Prediction and Performance Experiment in a Deep-well Centrifugal Pump with Different Impeller Outlet Width显示文摘The existing research of the deep-well centrifugal pump mainly focuses on reduce the manufacturing cost and improve the pump performance,and how to combine above two aspects together is the most difficult and important topic.In this study,the performances of the deep-well centrifugal pump with four different impeller outlet widths are studied by the numerical,theoretical and experimental methods in this paper.Two stages deep-well centrifugal pump equipped with different impellers are simulated employing the commercial CFD software to solve the Navier-Stokes equations for three-dimensional incompressible steady flow.The sensitivity analyses of the grid size and turbulence model have been performed to improve numerical accuracy.The flow field distributions are acquired and compared under the design operating conditions,including the static pressure,turbulence kinetic energy and velocity.The prototype is manufactured and tested to certify the numerical predicted performance.The numerical results of pump performance are higher than the test results,but their change trends have an acceptable agreement with each other.The performance results indicted that the oversize impeller outlet width leads to poor pump performances and increasing shaft power.Changing the performance of deep-well centrifugal pump by alter impeller outlet width is practicable and convenient,which is worth popularizing in the engineering application.The proposed research enhances the theoretical basis of pump design to improve the performance and reduce the manufacturing cost of deep-well centrifugal pump.SHI Weidong ZHOU Ling LU Weigang PEI Bing LANG Tao 2013Chinese Journal of Mechanical Engineering2013,26,1:20
9Construction and Identification of a Human Liver Specific MicroRNA Eukaryotic Expression Vector显示文摘MiR-122 is one of the non-coding RNAs which showed its effects on the lipo-metablism,virus infection and HCC forming through regulation of liver gene expression.Its eukaryotic expression vector was constructed by using pSuper which was widely applied in the siRNA expression.The precursor of human miR-122 gene was amplified by polymerase chain reaction(PCR)from the human genomic DNA.The positive clones were screened by PCR and restriction enzyme digestion.The new expression vector of miR-122 was named pHsa-m122.PHsa-m122 and its controls were transfected to HepG2 cells.The miR-122 expression activity was evaluated by GFP122i sensor reporter plasmid through fluorescence detection and Western blot.It was shown that the fluorescence intensity of GFP122si and pHsa-m122 co-transfection group was weaker than that of the controls,so the functional activity of expressed miR-122 was detected.When HepG2 cells were co-transfected with HBV1.3 and pHsa-m122 plasmids, the results showed miR-122 may down-regulate the gene expression of HBV.The human liver specific microRNA eukaryotic expression vector of miR-122 was constructed successfully,which may facilitate further study of its function in the development of liver virus infection diseases and HCC.Shanshan Chen Ming Ni Bing Yu Tingting Lv Mengji Lu Feili Gong 2007Cellular & Molecular Immunology2007,4,6:18
10Progress in clinical trials of cell transplantation for the treatment of spinal cord injury:how many questions remain unanswered?显示文摘Spinal cord injury can lead to severe motor,sensory and autonomic nervous dysfunctions.However,there is currently no effective treatment for spinal cord injury.Neural stem cells and progenitor cells,bone marrow mesenchymal stem cells,olfactory ensheathing cells,umbilical cord blood stem cells,adipose stem cells,hematopoietic stem cells,oligodendrocyte precursor cells,macrophages and Schwann cells have been studied as potential treatments for spinal cord injury.These treatments were mainly performed in animals.However,subtle changes in sensory function,nerve root movement and pain cannot be fully investigated with animal studies.Although these cell types have shown excellent safety and effectiveness in various animal models,sufficient evidence of efficacy for clinical translation is still lacking.Cell transplantation should be combined with tissue engineering scaffolds,local drug delivery systems,postoperative adjuvant therapy and physical rehabilitation training as part of a comprehensive treatment plan to provide the possibility for patients with SCI to return to normal life.This review summarizes and analyzes the clinical trials of cell transplantation therapy in spinal cord injury,with the aim of providing a rational foundation for the development of clinical treatments for spinal cord injury.Xu-Chang Hu Yu-Bao Lu Yong-Na Yang Xue-Wen Kang Yong-Gang Wang Bing Ma Shuai Xing 2021Neural Regeneration Research2021,16,3:18
11Identification of rice cultivars with low brown rice mixed cadmium and lead contents and their interactions with the micronutrients iron,zinc,nickel and manganese显示文摘Paddy fields in mining areas are usually co-contaminated by a cocktail of mixed toxic heavy metals(e.g.,Cd and Pb in Pb/Zn mines).However,previous studies on rice cultivars screened for effective metal exclusion have mostly focused on individual metals,and have been conducted under pot-trial or hydroponic solution conditions.This study identified rice cultivars with both low Cd and Pb accumulation under Cd-and Pb-contaminated field conditions,and the interactions of the toxic elements Cd and Pb with the micronutrient elements Fe,Zn,Mn and Ni were also studied.Among 32 rice cultivars tested,there were significant differences in Cd(0.06-0.59 mg/kg) and Pb(0.25-3.15 mg/kg) levels in their brown rice,and similar results were also found for the micronutrient elements.Significant decreases in concentrations of Fe and Mn were detected with increasing Cd concentrations and a significant elevation in Fe,Mn and Ni with increasing Pb concentrations.A similar result was also shown by Cd and Ni.Three cultivars were identified with a combination of low brown rice Cd and Pb,high micronutrient and grain yield(Wufengyou 2168,Tianyou 196 and Guinongzhan).Present results suggest that it is possible to breed rice cultivars with low mixed toxic element(Cd,Pb) and high micronutrient contents along with high grain yields,thus ensuring food safety and quality.Bing Li Xun Wang Xiaoli Qi Lu Huang Zhihong Ye 2012Journal of Environmental Sciences2012,24,10:18
12CFD-DEM simulation of spouting of corn-shaped particles显示文摘Three dimensionally coupled computational fluid dynamics (CFD) and discrete element method (DEM) were used to investigate the flow of corn-shaped particles in a cylindrical spouted bed with a conical base. The particle motion was modeled by the DEM, and the gas motion by the k-ε two-equation turbulent model. A two-way coupling numerical iterative scheme was used to incorporate the effects of gas-particle interactions in terms of momentum exchange. The corn-shaped particles were constructed by a multisphere method. Drag force, contact force, Saffman lift force, Magnus lift force, and gravitational force acting on each individual particle were considered in establishing the mathematical modeling. Calculations were carried out in a cylindrical spouted bed with an inside diameter of 200 mm, a height of 700 mm, and a conical base of 60 . Comparison of simulations with experiments showed the availability of the multi-sphere method in simulating spouting action with corn-shaped particles, but it depended strongly on the number and the arrangement of the spherical elements. Gas-solid flow patterns, pressure drop, particle velocity and particle concentration at various spouting gas velocity were discussed. The results showed that particle velocity reaches a maximum at the axis and then decreases gradually along the radial direction in the whole bed. Particle concentration increases along the radial direction in the spout region but decreases in the fountain region, while it is nearly constant in the annulus region. 14Microwave Exposure Impairs Synaptic Plasticity in the Rat Hippocampus and PC12 Cells through Over-activation of the NMDA Receptor Signaling Pathway显示文摘Objective The aim of this study is to investigate whether microwave exposure would affect the N-methyl-D-aspartate receptor(NMDAR) signaling pathway to establish whether this plays a role in synaptic plasticity impairment. Methods 48 male Wistar rats were exposed to 30 m W/cm2 microwave for 10 min every other day for three times. Hippocampal structure was observed through H&E staining and transmission electron microscope. PC12 cells were exposed to 30 m W/cm2 microwave for 5 min and the synapse morphology was visualized with scanning electron microscope and atomic force microscope. The release of amino acid neurotransmitters and calcium influx were detected. The expressions of several key NMDAR signaling molecules were evaluated. Results Microwave exposure caused injury in rat hippocampal structure and PC12 cells, especially the structure and quantity of synapses. The ratio of glutamic acid and gamma-aminobutyric acid neurotransmitters was increased and the intracellular calcium level was elevated in PC12 cells. A significant change in NMDAR subunits(NR1, NR2 A, and NR2B) and related signaling molecules(Ca2+/calmodulin-dependent kinase II gamma and phosphorylated c AMP-response element binding protein) were examined. Conclusion 30 m W/cm2 microwave exposure resulted in alterations of synaptic structure, amino acid neurotransmitter release and calcium influx. 15A Deep Learning Method for Bias Correction of ECMWF 24–240 h Forecasts显示文摘Correcting the forecast bias of numerical weather prediction models is important for severe weather warnings.The refined grid forecast requires direct correction on gridded forecast products,as opposed to correcting forecast data only at individual weather stations.In this study,a deep learning method called CU-net is proposed to correct the gridded forecasts of four weather variables from the European Centre for Medium-Range Weather Forecast Integrated Forecasting System global model(ECMWF-IFS): 2-m temperature,2-m relative humidity,10-m wind speed,and 10-m wind direction,with a forecast lead time of 24 h to 240 h in North China.First,the forecast correction problem is transformed into an image-toimage translation problem in deep learning under the CU-net architecture,which is based on convolutional neural networks.Second,the ECMWF-IFS forecasts and ECMWF reanalysis data(ERA5) from 2005 to 2018 are used as training,validation,and testing datasets.The predictors and labels(ground truth) of the model are created using the ECMWF-IFS and ERA5,respectively.Finally,the correction performance of CU-net is compared with a conventional method,anomaly numerical correction with observations(ANO).Results show that forecasts from CU-net have lower root mean square error,bias,mean absolute error,and higher correlation coefficient than those from ANO for all forecast lead times from 24 h to 240 h.CU-net improves upon the ECMWF-IFS forecast for all four weather variables in terms of the above evaluation metrics,whereas ANO improves upon ECMWF-IFS performance only for 2-m temperature and relative humidity.For the correction of the 10-m wind direction forecast,which is often difficult to achieve,CU-net also improves the correction performance.Lei HAN Mingxuan CHEN Kangkai CHEN Haonan CHEN Yanbiao ZHANG Bing LU Linye SONG Rui QIN 2021Advances in Atmospheric Sciences2021,38,9:15
16Using Fuzzy Theory and Variable Weights for Water Quality Evaluation in Poyang Lake, China显示文摘Achieving water purity in Poyang Lake has become a major concern in recent years, thus appropriate evaluation of spatial and temporal water quality variations has become essential. Variations in 11 water quality parameters from 15 sampling sites in Poyang Lake were investigated from 2009 to 2012. An integrative fuzzy variable evaluation(IFVE) model based on fuzzy theory and variable weights was developed to measure variations in water quality. Results showed that: 1) only chlorophyll-a concentration and Secchi depth differed significantly among the 15 sampling sites(P < 0.01), whereas the 11 water quality parameters under investigation differed significantly throughout the seasons(P < 0.01). The annual variations of all water quality variables except for temperature, electrical conductivity, suspended solids and total phosphorus were considerable(P < 0.05). 2) The IFVE model was reasonable and flexible in evaluating water quality status and any possible ′bucket effect′. The model fully considered the influences of extremely poor indices on overall water quality. 3) A spatial analysis indicated that anthropogenic activities(particularly industrial sewage and dredging) and lake bed topography might directly affect water quality in Poyang Lake. 17Tiller Bud Formation Regulators MOC1 and MOC3 Cooperatively Promote Tiller Bud Outgrowth by Activating F0N1 Expression in Rice显示文摘Tillering in rice is one of the most important agronomic traits.Rice tiller development can be divided into two main processes: the formation of the axillary bud and its subsequent outgrowth.Several genes critical for bud formation in rice have been identified by genetic studies;however,their molecular functions and relationships are still largely unknown.Here,we report that MONOCULM 1 (MOC1) and MONOCULM 3/ TILLERS ABSENT 1/STERILE AND REDUCED TILLERING 1 (MOC3/TAB1/SRT1),two vital regulators for tiller formation in rice,physically interact to regulate tiller bud outgrowth through upregulating the expression of FLORAL ORGAN NUMBER 1 (FON1),the homolog of CLAVATA1 in rice.We found that M0C3 is able to directly bind the promoter ofFONI and subsequently activate FON1 expression.MOC1 functions as a coactivator of MOC3,whereas it could not directly bind the FON1 promoter,and further activated FON1 expression in the presence of MOC3.Accordingly,FON1 is highly expressed at axillary meristems and shows remarkably decreased expression levels in mod and moc3 mutants.Loss-of-function mutants of FON1 exhibit normal bud formation but defective bud outgrowth and reduced tiller number.Collectively,these results shed light on a joint transcriptional regulatory mechanim by MOC1 and MOC3,and establish a new framework for the control of tiller bud formation and outgrowth.Gaoneng Shao Zefu Lu Jinsong Xiong Bing Wang Yanhui Jing Xiangbing Meng Guifu Liu Haiyan Ma Yan Liang Fan Chen Yonghong Wang Jiayang Li Hong Yu 2019Molecular Plant2019,12,8:15
18Expression and clinical significance of CD73 and hypoxia-inducible factor-1α in gastric carcinoma显示文摘AIM: To investigate the expression of CD73 and hypoxia-inducible factor-1α (HIF-1α) in human gastric carcinoma, and explore their clinical significance and prognostic value. METHODS: CD73 and HIF-1α expressions were detected by immunohistochemistry in consecutive sections of tissue samples from 68 gastric carcinoma patients. The peritumor tissues 2 cm away from the tumor were obtained and served as controls. The presence of CD73 and HIF-1α was analyzed by immunohis-tochemistry using the Envision technique. RESULTS: CD73 and HIF-1α expressions in gastric carcinoma were significantly higher than those in gastric mucosal tissues as control (P < 0.001) and showed a close correlation (Spearman r = 0.390, P = 0.001). Overexpression of CD73 was positively correlated with differentiation of tumor (P = 0.000), histopathology (P = 0.041), depth of invasion (P < 0.001), nodal status (P = 0.003), metastasis (P = 0.013), and the American Joint Committee on Cancer (AJCC) stage (P < 0.001). High expression of HIF-1α was positively correlated with tumor diameter (P = 0.031), depth of invasion (P = 0.022), and AJCC stage (P = 0.035). The overall survival rate was low in the patients with high expression of CD73 (P < 0.001). Moreover, CD73+/HIF-1α+ patients had the worst prognosis (P < 0.001). CD73 expression was proven to be an independent predictor for patients with gastric carcinoma by both multivariate Cox regression analysis (P = 0.021) and receiver operating characteristic curves (P = 0.001).CONCLUSION: CD73 expression correlates closely with HIF-1α expression in gastric carcinoma. 19Xiaotan Sanjie decoction attenuates tumor angiogenesis by manipulating Notch-1-regulated proliferation of gastric cancer stem-like cells显示文摘AIM: To determine the underlying mechanisms of action and influence of Xiaotan Sanjie(XTSJ) decoction on gastric cancer stem-like cells(GCSCs). METHODS: The gastric cancer cell line MKN-45 line was selected and sorted by FACS using the cancer stem cell marker CD44; the stemness of these cells was checked in our previous study. In an in vitro study, the expression of Notch-1, Hes1, Vascular endothelial growth factor(VEGF), and Ki-67 in both CD44-positive gastric cancer stem-like cells(GCSCs) and CD44-negative cells was measured by Western blot. The effect of XTSJ serum on cell viability and on the above markers was measured by MTT assay and Western blot, respectively. In an in vivo study, the ability to induce angiogenesis and maintenance of GCSCs in CD44-positive-MKN-45- and CD44-negative-engrafted mice were detected by immunohistochemical staining using markers for CD34 and CD44, respectively. The role of XTSJ decoction in regulating the expression of Notch-1, Hes1, VEGF and Ki-67 was measured by Western blot and real-time polymerase chain reaction.RESULTS: CD44+ GCSCs showed more cell proliferation and VEGF secretion than CD44-negative cells in vitro, which were accompanied by the high expression of Notch-1 and Hes1 and positively associated with tumor growth(GCSCs vs CD44-negative cells, 2.72 ± 0.25 vs 1.46 ± 0.16, P < 0.05) and microvessel density(MVD)(GCSCs vs CD44-negative cells, 8.15 ± 0.42 vs 3.83 ± 0.49, P < 0.001) in vivo. XTSJ decoction inhibited the viability of both cell types in a dose-dependent manner in vitro. Specifically, a significant difference in the medium-(82.87% ± 6.53%) and high-dose XTSJ groups(77.43% ± 7.34%) was detected at 24 h in the CD44+ GCSCs group compared with the saline group(95.42% ± 5.76%) and the low-dose XTSJ group(90.74% ± 6.57%)(P < 0.05). However, the efficacy of XTSJ decoction was reduced in the CD44- groups; significant differences were only detected in the high-dose XTSJ group at 48 h(78.57% ± 6.94%) and 72 h(72.12% ± 7.68%) when compared with the other CD44- groups(P < 0.05). Notably, these differences were highly consistent with the Notch-1, Hes1, VEGF and Ki-67 expression in these cells. Similarly, in vivo, XTSJ decoction inhibited tumor growth in a dose-dependent manner. A significant difference was observed in the medium(1.76 ± 0.15) and high-dose XTSJ(1.33 ± 0.081) groups compared with the GCSCs control group(2.72 ± 0.25) and the low-dose XTSJ group(2.51 ± 0.25)(P < 0.05). We also detected a remarkable decrease of MVD in the medium-(7.10 ± 0.60) and high-dose XTSJ(5.99 ± 0.47) groups compared with the GCSC control group(8.15 ± 0.42) and the low-dose XTSJ group(8.14 ± 0.46)(P < 0.05). Additionally, CD44 expression was decreased in these groups [medium-(4.43 ± 0.45) and high-dose XTSJ groups(3.56 ± 0.31) vs the GCSC control(5.96 ± 0.46) and low dose XTSJ groups(5.91 ± 0.38)](P < 0.05). 20Key residues of the receptor binding motif in the spike protein of SARS-CoV-2 that interact with ACE2 and neutralizing antibodies显示文摘Coronavirus disease 2019(COVID-19),caused by the novel human coronavirus SARS-CoV-2,is currently a major threat to public health worldwide.The viral spike protein binds the host receptor angiotensin-converting enzyme 2(ACE2)via the receptor-binding domain(RBD),and thus is believed to be a major target to block viral entry.Both SARS-CoV-2 and SARS-CoV share this mechanism.Here we functionally analyzed the key amino acid residues located within receptor binding motif of RBD that may interact with human ACE2 and available neutralizing antibodies.The in vivo experiments showed that immunization with either the SARS-CoV RBD or SARS-CoV-2 RBD was able to induce strong clade-specific neutralizing antibodies in mice;however,the cross-neutralizing activity was much weaker,indicating that there are distinct antigenic features in the RBDs of the two viruses.This finding was confirmed with the available neutralizing monoclonal antibodies against SARS-CoV or SARS-CoV-2.It is worth noting that a newly developed SARS-CoV-2 human antibody,HA001,was able to neutralize SARS-CoV-2,but failed to recognize SARS-CoV.Moreover,the potential epitope residues of HA001 were identified as A475 and F486 in the SARS-CoV-2 RBD,representing new binding sites for neutralizing antibodies.Overall,our study has revealed the presence of different key epitopes between SARS-CoV and SARSCoV-2,which indicates the necessity to develop new prophylactic vaccine and antibody drugs for specific control of the COVID-19 pandemic although the available agents obtained from the SARS-CoV study are unneglectable.Chunyan Yi Xiaoyu Sun Jing Ye Longfei Ding Meiqin Liu Zhuo Yang Xiao Lu Yaguang Zhang Liyang Ma Wangpeng Gu Aidong Qu Jianqing Xu Zhengli Shi Zhiyang Ling Bing Sun 2020Cellular & Molecular Immunology2020,17,6:13
